The OECD acute toxicity guideline has been revised recently to protect animal welfare. The GLP authority of the Ministry of Environment, the National Institute for Environmental Research, recommended GLP laboratories in Korea to ufo the revised acute toxicity guideline. This study was carried out to optimize newly adopted OECD test guideline 420 (TG 420). Bisphenol A was selected for test chemical. Following TG420, Bisphenol A was classified as class 5/unclassified group. The revised TG 420 was very effective test in minimizing animal number and classifying chemicals. The method, however had short-coming in evaluation of test results statistically because the test had no control group, and the test should be stopped when animals were dead at the lowest dose or alive at the highest dose. TG 420 required at Least 20 animals to complete the test, but it could result in producing unused animals that need to sacrifice.
A rapid and simple analytical method, using liquid chromatography tandem mass spectrometry (LC-MS/MS), was developed to detect myo-inositol (MI) in infant formulas. For protein removal: acid hydrolysis and lipid removal through organic solvent extraction. The operating conditions for instrumental analysis were determined based on previously reported analogous methods that used LC-MS/MS. Quantitative analysis was used for the detection limit test, infant formula recovery test, and standard reference material (SRM) 1849a to verify the validity of our LC-MS/MS analytical method, which was developed to quantify MI. For validation, the results of our method were compared with the results of quantitative analyses of certified values. The test results showed that the limit of detection was 0.05 mg/L, the limit of quantitation was 0.17 mg/L, and the method detection limit was 17 mg/kg. The recovery test exhibited a recovery between 98.07-98.43% and a relative standard deviation between 1.93-2.74%. Therefore, the result values were good. Additionally, SRM 1849a was measured to have an MI content of 401.84 mg/kg and recovery of 98.25%, which is comparable to the median certified value of 409 mg/kg. From the aforementioned results, we judged that the instrumental analysis conditions and preparation method used in this study were valid. The rapid analytical method developed herein could be implemented in many laboratories that seek to save time and labor.
The objectives of the study were to examine non-genetic factors that influence somatic cell counts in dairy cattle and to estimate the genetic parameters of somatic cell counts. A total of 34, 097-test day somatic cell count records were obtained from the Zimbabwe Dairy Services Association (ZDSA). The data were from 5, 615 Holstein daughters of 390 sires and 2, 541 dams tested between May 1994 and December 1998. First lactation cows contributed 22, 147 records to the data set, while 11, 950 records were from second and later parity cows. The model for analysis included fixed effects of month of calving, year of calving, stage of lactation, calving interval and test date. Milk yield and age on test day were fitted in the model as covariates. The additive genetic effects pertaining to cows, sires and dams and the residual error were the random effects. The Average Information Restricted Maximum Likelihood algorithm was used for analysis. The heritability of somatic cell scores was low at $0.027{\pm}0.013$ for parity one cows and $0.087{\pm}0.031$ for parity two and above. Repeatability estimates were $0.22{\pm}0.01$ and $0.30{\pm}0.01$ for the two lactation groups, respectively. Genetic and phenotypic correlations between the somatic cell scores and test day milk production were small and negative. It seems that there is no genetic link between somatic cell counts and milk yield in Holstein cattle in Zimbabwe. The results also seem to indicate that somatic cell count is a trait that is mainly governed by environmental factors.

Purpose: In EU, the animal welfare regulations are strengthening. Also, in Korea, for strengthening of animal protection laws, animal welfare farm certification scheme is being prepared to enforce. To get the certification of animal welfare farm, especially for sows, individual specific feeding in sow group is necessary in accordance with the animal welfare regulation. Also, liquid feeding is reported to be more effective for farrowing as well as dry sow. Therefore, in this study, a electronic liquid sow feeder using RFID which could be fed to farrowing sow simultaneously is developed. Methods : We manufactured a prototype of appropriate elements which were evaluated in the preliminary test for selecting RFID tag, supply pump, mixing type, etc. With the prototype, the performance test for liquid feeding gilt was done in group raising training barn. Results : The performance test result shows the C.V.(coefficient of variation) of liquid mixing and feeding is 0.77~1.97% and 2.4~5.3%, respectively, which means the system could feed sows uniformly. The feeding time of pneumatically activated ball valve is 2 seconds per 0.9 kg of liquid feed. The eating time for gilt were 65 seconds in average. Conclusions : The prototype could feed sow uniformly, and deliver the liquid feed 9 times for a sow continuously. Also, total eating time for a sow was 18 minutes in one visit to the prototype of feeding station.
Objective: The objective was to determine standardized ileal digestibility (SID) of amino acids (AA) in 11 plant protein sources fed to growing pigs. Methods: Eleven feed ingredients used were sesame meal, two sources of soybean meal (SBM) produced in the Republic of Korea, a source of SBM produced in India, high-protein distillers dried grains (HPDDG), perilla meal, canola meal, copra meal, corn germ meal, palm kernel expeller, and tapioca distillers dried grains (TDDG). Experimental diets were prepared to contain each test ingredient as a sole source of AA, and a nitrogen-free diet was also prepared to estimate the basal ileal endogenous losses of AA. Twelve barrows surgically fitted with T-cannulas at the distal ileum with an initial body weight of 29.0 kg (standard deviation = 3.0) were individually housed in metabolism crates equipped with a feeder and a nipple drinker. A $12{\times}9$ incomplete Latin square design was employed with 12 experimental diets, 12 animals, and 9 periods. After a 5-d adaptation period, ileal digesta were collected on d 6 and 7 in each experimental period. Results: Values for apparent ileal digestibility of most indispensable AA in three sources of SBM were greater compared with other test ingredients except HPDDG and canola meal (p<0.05). Pigs fed diets containing SBM sources had also greater SID of most indispensable AA compared with those fed diets containing other test ingredients (p<0.05) except for HPDDG and canola meal. There was no difference in the apparent ileal digestibility and SID of AA among sources of SBM. The TDDG had the least value for the SID of methionine among test ingredients (p<0.05). Conclusion: The SID of most AA in SBM, HPDDG, and canola meal were greater than those in sesame meal, perilla meal, copra meal, and TDDG.

Objective: The objectives of the present study were to determine digestible energy (DE), metabolizable energy (ME), and standardized total tract digestibility (STTD) of phosphorus (P) in meat meal (MM) and to compare these values with those in fish meal (FM), and soybean meal (SBM) fed to pigs. Methods: Two experiments were conducted to determine energy concentrations and STTD of P in MM, FM, and SBM fed to growing pigs. In Exp. 1, DE and ME in the 3 test ingredients were measured using 24 barrows with initial body weight (BW) of 77.7±8.3 kg. A corn-based diet and 3 diets containing corn and 22% to 30% of each test ingredient were prepared. In Exp. 2, the STTD of P in the 3 test ingredients was measured using 24 barrows (90.9±6.6 kg BW). Three diets were formulated to contain each test ingredient as the sole source of P. Results: In Exp. 1, the DE and ME values in MM (3,310 and 2,856 kcal/kg dry matter [DM]) were less (p<0.05) than those in FM (4,121 and 3,572 kcal/kg DM) and SBM (4,390 and 4,032 kcal/kg DM). In Exp. 2, FM (64.3%) had greater (p<0.05) STTD of P than SBM (44.8%) with MM (55.8%) having intermediate STTD of P. Conclusion: The MM contains less energy concentrations compared with FM and SBM, and digestibility of phosphorus in MM does not differ from that in FM and SBM.

The aim of this study was to compare two random regression models (RRM) fitted by fourth ($RRM_4$) and fifth-order Legendre polynomials ($RRM_5$) with a lactation model (LM) for evaluating Holstein cattle in Brazil. Two datasets with the same animals were prepared for this study. To apply test-day RRM and LMs, 262,426 test day records and 30,228 lactation records covering 305 days were prepared, respectively. The lowest values of Akaike's information criterion, Bayesian information criterion, and estimates of the maximum of the likelihood function (-2LogL) were for $RRM_4$. Heritability for 305-day milk yield (305MY) was 0.23 ($RRM_4$), 0.24 ($RRM_5$), and 0.21 (LM). Heritability, additive genetic and permanent environmental variances of test days on days in milk was from 0.16 to 0.27, from 3.76 to 6.88 and from 11.12 to 20.21, respectively. Additive genetic correlations between test days ranged from 0.20 to 0.99. Permanent environmental correlations between test days were between 0.07 and 0.99. Standard deviations of average estimated breeding values (EBVs) for 305MY from $RRM_4$ and $RRM_5$ were from 11% to 30% higher for bulls and around 28% higher for cows than that in LM. Rank correlations between RRM EBVs and LM EBVs were between 0.86 to 0.96 for bulls and 0.80 to 0.87 for cows. Average percentage of gain in reliability of EBVs for 305-day yield increased from 4% to 17% for bulls and from 23% to 24% for cows when reliability of EBVs from RRM models was compared to those from LM model. Random regression model fitted by fourth order Legendre polynomials is recommended for genetic evaluations of Brazilian Holstein cattle because of the higher reliability in the estimation of breeding values.

The development of effective genetic evaluations and selection of sires requires accurate estimates of genetic parameters for all economically important traits in the breeding goal. The main objective of this study was to assess the relative performance of the traditional lactation average model (LAM) against the random regression test-day model (RRM) in the estimation of genetic parameters and prediction of breeding values for Holstein Friesian herds in Ethiopia. The data used consisted of 6,500 test-day (TD) records from 800 first-lactation Holstein Friesian cows that calved between 1997 and 2013. Co-variance components were estimated using the average information restricted maximum likelihood method under single trait animal model. The estimate of heritability for first-lactation milk yield was 0.30 from LAM whilst estimates from the RRM model ranged from 0.17 to 0.29 for the different stages of lactation. Genetic correlations between different TDs in first-lactation Holstein Friesian ranged from 0.37 to 0.99. The observed genetic correlation was less than unity between milk yields at different TDs, which indicated that the assumption of LAM may not be optimal for accurate evaluation of the genetic merit of animals. A close look at estimated breeding values from both models showed that RRM had higher standard deviation compared to LAM indicating that the TD model makes efficient utilization of TD information. Correlations of breeding values between models ranged from 0.90 to 0.96 for different group of sires and cows and marked re-rankings were observed in top sires and cows in moving from the traditional LAM to RRM evaluations.
