Effect of dietary sesame (Sesame indicum L) seed meal level supplemented with lysine and phytase on performance traits and antioxidant status of late-phase laying hens

  • Objective: This study was performed to investigate the effects of supplementing sesame seed meal (SSM) with phytase and lysine on performance, egg quality, blood biochemical and antioxidant status of laying hens. Methods: A total of 960, 56-wk-old laying hens were divided into 12 dietary groups with eight replicates per group (10 birds per replicate). A completely randomized design with factorial arrangement 2×3×2 consisted of two levels of lysine supplement (0% and 10% over requirement), three SSM levels (0%, 10%, and 20%) with or without phytase (0 and 300 g/ton). The feeding trial lasted 10 weeks. Results: Birds fed diets with 10% SSM had higher feed intake than groups fed 0% and 20% SSM. The addition of phytase to experimental feeds, improved feed conversion ratio, increased egg weight and mass (p<0.01). Egg quality criteria was not affected by supplementing phytase; however, supplementing 300 g/ton phytase to hens diet, led to a significant (p<0.05) increase in egg shell strength. Egg yolk cholesterol and serum low-density lipoprotein cholesterol, atherogenic index and total cholesterol were decreased (p<0.01) by diet containing 20% SSM. The high-density lipoprotein cholesterol was increased (p<0.05) in serum of hens fed 20% SSM than the other groups. It was also observed that total antioxidant capacity and total superoxide dismutase content of hens fed 20% SSM was significantly higher than control group (p<0.05). Conclusion: As from results, dietary supplementation of SSM and phytase had no negative effects on laying hens performance or egg quality while improving the egg oxidative stability.

Effects of Dietary Combinations of Vitamin A, E and Methionine on Growth Performance, Meat Quality and Immunity in Commercial Broilers

  • The experiment was conducted to study the effect of dietary combinations of vitamin A (VA), vitamin E (VE) and methionine (Met) on growth performance, meat quality and immunity in commercial broilers. Ross chicks (n=3,630) were allocated to five experimental treatments with three replicates per diet. The dietary treatments were: VA 8,000 IU, VE 10 IU (diet 1); VA 12,000 IU, VE 10 IU (diet 2); VA 8,000 IU, VE 100 IU (diet 3); VA 12,000 IU, VE 100 IU (diet 4) and; VA 12,000 IU, VE 100 IU/kg diet and 20% Met higher than other groups (diet 5). The Met content in diet 1 to diet 4 were as per the requirement suggested by NRC. Separate vitamin premixes were prepared for each treatment diet as per the requirement of study. The 35 d study revealed significantly (p<0.0001) higher weight gains in broilers fed diet 3 and diet 5, than in the rest of the groups during starter phase (0-3 weeks) only. The feed intake did not vary significantly at all phases of study, but feed efficiency was significantly (p<0.05) lower in diet 1 during starter and overall phase (4-5 weeks). The bone strength and bone composition, except bone calcium, remained unaffected due to experimental diets studied after 35 d of experimental feeding. The thio-barbituric acid reactive substances (TBARS) were significantly (p=0.0013) lower in the breast meat in group 5, followed by group 3, than in the rest of the groups. The immune studies conducted, antibody titers to sheep red blood cells, thickness index to phytohaemagglutinin-P, and heterophil: lymphocyte ratio, did not show any significant difference among treatments. It could be concluded that supplementation of VA, VE and Met at higher levels could be beneficial to broilers only during the starter phase.

New composite traits for joint improvement of milk and fertility trait in Holstein dairy cow

  • Objective: The objective of this study was to define a new composite trait for Holstein dairy cows and evaluate the possibility of joint improvement in milk and fertility traits. Methods: A data set consisting 35,882 fertility related records (days open [DO], calving interval [CI], and number of services per conception [NSC], and total milk yield in each lactation [TMY]) was collected from 1998 to 2016 in Polish Holstein-Friesian breed herds. In this study TMY, DO, CI, and lactation length of each cow was used to obtain composite milk and fertility traits (CMF). Results: Moderate heritability (0.15) was estimated for composite trait that was higher than heritability of female fertility related traits: DO 0.047, CI 0.042, and NSC 0.014, and slightly lower than heritability of TMY 0.19. Favourable genetic correlations (-0.87) were estimated between CMF with TMY. Spearman rank correlation coefficients between breeding value of CMF with DO, CI, and TMY were high (>0.94) but with NSC were moderate (0.64). Selection on CMF caused favourable correlated genetic gains for DO, CI, and TMY. Different selection indices with different emphasis on fertility and milk production were constructed. The amount of correlated genetic gains obtained for DO and total milk production according to selection in CMF were higher than of genetic gains obtained for DO and TMY in selection indices with different emphasis on milk and fertility. Conclusion: The animal selection only based on a composite trait - CMF proposed in current study would simultaneously lead to favourable genetic gains for both milk and fertility related traits. In this situation CMF introduced in current study can be used to overcome to limitations of selection index and CMF could be useful for countries that have problems in recording traits, especially functional traits.

Genome-wide association study for the free amino acid and nucleotide components of breast meat in an F2 crossbred chicken population

  • Flavor is an important sensory trait of chicken meat. The free amino acid (FAA) and nucleotide (NT) components of meat are major factors affecting meat flavor during the cooking process. As a genetic approach to improve meat flavor, we performed a genome-wide association study (GWAS) to identify the potential candidate genes related to the FAA and NT components of chicken breast meat. Measurements of FAA and NT components were recorded at the age of 10 weeks from 764 and 767 birds, respectively, using a White leghorn and Yeonsan ogye crossbred F2 chicken population. For genotyping, we used 60K Illumina single-nucleotide polymorphism (SNP) chips. We found a total of nine significant SNPs for five FAA traits (arginine, glycine, lysine, threonine content, and the essential FAAs and one NT trait (inosine content), and six significant genomic regions were identified, including three regions shared among the essential FAAs, arginine, and inosine content traits. A list of potential candidate genes in significant genomic regions was detected, including the KCNRG, KCNIP4, HOXA3, THSD7B, and MMUT genes. The essential FAAs had significant gene regions the same as arginine. The genes related to arginine content were involved in nitric oxide metabolism, while the inosine content was possibly affected by insulin activity. Moreover, the threonine content could be related to methylmalonyl-CoA mutase. The genes and SNPs identified in this study might be useful markers in chicken selection and breeding for chicken meat flavor.

Genetic Differentiation among Sheep Populations from Near-sea Mainland in East Asia

  • Using the method of 'random sampling in typical colonies of the central area of the habitat', 60 Small-tailed Han sheep were obtained in Jining city, Shangdong province. The variations of Small-tailed Han sheep at 12 structural loci encoding blood proteins were detected by several electrophoresis techniques and their gene frequencies were then estimated. The same data of four other sheep populations from Near-sea Mainland in East Asia were cited for the analysis of genetic differentiation. The average heterozygosities of five populations, namely Kharkhorin sheep, Ulaanbaatar sheep, Small-tailed Han sheep, Hu sheep and Cham Tribe sheep were 0.3447, 0.3285, 0.3157, 0.3884 and 0.2300, respectively. The coefficient of gene differentiation among four populations, Kharkhorin sheep, Ulaanbaatar sheep, Small-tailed Han sheep and Hu sheep, was 0.045557, and that between these four breeds and Cham Tribe sheep was 0.088005, indicating that the level of gene differentiation among the former four sheep populations of Mongolian group was comparatively lower than that between Cham Tribe sheep and other four sheep populations. The origin of Cham Tribe sheep deserve further research. The documentary research on the evolution of Small-tailed Han sheep and Hu sheep from Mongolian sheep was further verified by the biochemical experiments in the study. It was reasonably deduced that Hu sheep, Small Tailed Han sheep and Cham Tribe sheep were decreasingly influenced by the bloodline of Mongolian sheep.

  • It is very difficult to get the information about semen quality analysis in transgenic pigs because of limited numbers and research facilities. Therefore, in the present study, we analyzed the semen quality of transgenic boars generated for xenotransplantation research. Briefly, the semen samples were collected from 5 homozygous ${\alpha}1,3$-Galactosyltransferase knock-out ($GalT^{-/-}$) transgenic boars and immediately transported to the laboratory. These semen samples were decupled with DPBS and conducted to analyze semen parameters by a computer-assisted semen analysis (CASA) system. The boar semen were examined all 12 parameters such as total motility (TM), curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP), and hyperactivated (HYP), etc. In results, among the 5 $GalT^{-/-}$ boars, three boars (#134, 144, and 170) showed normal range of semen parameters, but #199 and 171 boars showed abnormal ranges of semen parameters according to standard ranges of semen parameters. Unfortunately, #171 boar showed azoospermia symptom with rare sperm counts in the original semen. Conclusively, assessment of semen parameters by CASA system is useful to pre-screening of reproductively healthy boar prior to natural mating and artificial insemination for multiplication and breeding.

Phenotypic characterization of Hanwoo (native Korean cattle) cloned from somatic cells of a single adult

  • We investigated phenotypic differences in Hanwoo cattle cloned from somatic cells of a single adult. Ten genetically identical Hanwoo were generated by somatic cell nuclear transfer from a single adult. Weights at birth, growing pattern, horn and noseprint patterns were characterized to investigate phenotypic differences. The weights of clones at 6 and 12 months were slightly heavier than that of the donor. A horn pattern analysis revealed that seven clones had exactly the same horn pattern as the donor cow, whereas three were different. Although similarities such as general appearance can often be used to identify individual cloned animals, no study has characterized noseprint patterns for this end. A noseprint pattern analysis of all surviving clones showed that all eight animals had distinct noseprints. Four were similar to the donor, and the remaining four had more secondary-like characteristics.

Metabolomics Analysis of the Beef Samples with Different Meat Qualities and Tastes

  • The purpose of this study was to investigate the meat metabolite profiles related to differences in beef quality attributes (i.e., high-marbled and low-marbled groups) using nuclear magnetic resonance (NMR) spectroscopy. The beef of different marbling scores showed significant differences in water content and fat content. High-marbled meat had mainly higher taste compounds than low-marbled meat. Metabolite analysis showed differences between two marbling groups based on partial least square discriminant analysis (PLS-DA). Metabolites identified by PLS-DA, such as N,N-dimethylglycine, creatine, lactate, carnosine, carnitine, sn-glycero-3-phosphocholine, betaine, glycine, glucose, alanine, tryptophan, methionine, taurine, tyrosine, could be directly linked to marbling groups. Metabolites from variable importance in projection plots were identified and estimated high sensitivity as candidate markers for beef quality attributes. These potential markers were involved in beef taste-related pathways including carbohydrate and amino acid metabolism. Among these metabolites, carnosine, creatine, glucose, and lactate had significantly higher in high-marbled meat compared to low-marbled meat (p<0.05). Therefore, these results will provide an important understanding of the roles of taste-related metabolites in beef quality attributes. Our findings suggest that metabolomics analysis of taste compounds and meat quality may be a powerful method for the discovery of novel biomarkers underlying the quality of beef products.

Survey on Feeding Facilities and Animal Welfare Level of Laying Hen Welfare Certified Farms (국내 동물복지 인증 산란계 농가의 사육시설 및 동물복지 수준 현황 조사)

  • The purpose of this study was to investigate farmers' field survey to improve animal welfare certification standards and to obtain basic data on the animal welfare level for certified farmers to develop evaluation methods of animal welfare level suitable for domestic farm environment. The subjects of the study were selected 10 animal welfare laying certified farmhouses. The farming certified farming facility survey collected breeding form, head, feed and drink space, breeding density, length and shape of perch. Animal welfare was assessed by the presence of sand bath, spawning, enrichment measures, free range, feathers pecking. The results of the study showed that most the certified animal welfare laying hens complied with the certification standards. All the farms were providing the nest box, but there were farms with more than 20% of the spawning rate. The perches were provided in sufficient length, but only three of ten farms were using rounded perches. Feather damage has been identified in most survey farms, which is likely to be due to feather damage caused by roosters producing both fertilized eggs. In this study, 10 farm households were surveyed. It was not possible to represent the whole certified farmhouse. Therefore, it should be analyzed thoroughly to evaluate the level of animal welfare.

Porcine growth hormone induces the nuclear localization of porcine growth hormone receptor in vivo

  • Objective: Recent studies have challenged the traditional paradigm that growth hormone receptor (GHR) displays physiological functions only in the cell membrane. It has been demonstrated that GHR localizes to the cell nucleus and still exhibits important physiological roles. The phenomenon of nuclear localization of growth hormone (GH)-induced GHR has previously been described in vitro. However, until recently, whether GH could induce nuclear localization of GHR in vivo was unclear. Methods: In the present study, we used pig as an animal model, and porcine growth hormone (pGH) or saline was injected into the inferior vena cava. We subsequently observed the localization of porcine growth hormone receptor (pGHR) using multiple techniques, including, immunoprecipitation and Western-blotting, indirect immunofluorescence assay and electronmicroscopy. Results: The results showed that pGH could induce nuclear localization of pGHR. Taken together, the results of the present study provided the first demonstration that pGHR was translocated to cell nuclei under pGH stimulation in vivo. Conclusion: Nuclear localization of pGHR induced by the in vivo pGH treatment suggests new functions and/or novel roles of nuclear pGHR, which deserve further study.
