The relationship between odd- and branched-chain fatty acids and microbial nucleic acid bases in rumen

  • Objective: This study aims to identify the relationship between odd- and branched-chain fatty acids (OBCFAs) and microbial nucleic acid bases in the rumen, and to establish a model to accurately predict microbial protein flow by using OBCFA. Methods: To develop the regression equations, data on the rumen contents of individual cows were obtained from 2 feeding experiments. In the first experiment, 3 rumen-fistulated dry dairy cows arranged in a $3{\times}3$ Latin square were fed diets of differing forage to concentration ratios (F:C). The second experiment consisted of 9 lactating Holstein dairy cows of similar body weights at the same stage of pregnancy. For each lactation stage, 3 cows with similar milk production were selected. The rumen contents were sampled at 4 time points of every two hours after morning feeding 6 h, and then to analyse the concentrations of OBCFA and microbial nucleic acid bases in the rumen samples. Results: The ruminal bacteria nucleic acid bases were significantly influenced by feeding diets of differing forge to concentration ratios and lactation stages of dairy cows (p<0.05). The concentrations of OBCFAs, especially odd-chain fatty acids and C15:0 isomers, strongly correlated with the microbial nucleic acid bases in the rumen (p<0.05). The equations of ruminal microbial nucleic acid bases established by ruminal OBCFAs contents showed a good predictive capacity, as indicated by reasonably low standard errors and high R-squared values. Conclusion: This finding suggests that the rumen OBCFA composition could be used as an internal marker of rumen microbial matter.

Effects of different dietary ratio of metabolizable glucose and metabolizable protein on growth performance, rumen fermentation, blood biochemical indices and ruminal microbiota of 8 to 10-month-old dairy heifers

  • Objective: The aim of this experiment was to evaluate the effects of different dietary ratio of metabolizable glucose (MG) to metabolizable protein (MP) on growth performance, blood metabolites, rumen fermentation parameters and the ruminal microbial community of 8 to 10-month-old heifers. Methods: A total of 24 Holstein heifers weighing an average of 282.90 kg (8 month of age) were randomly assigned to four groups of six. The heifers were fed one of four diets of different dietary MG/MP (0.97, 1.07, 1.13, and 1.26). Results: The results showed that the ratio of MG/MP affected the growth performance, blood metabolites, rumen fermentation parameters and the ruminal microbial community of heifers. The average daily gain of heifers was enhanced by increasing the ratio of MG/MP (p<0.05). The concentration of blood urea nitrogen, cholesterol, and low density lipoprotein cholesterol as well as the concentration of total volatile fatty acid in the rumen fluid of heifers decreased with the improvement in the ratio of dietary MG/MP (p<0.05). However, the relative amount of Ruminococcus albus and Butyrivibrio fibrisolvens in the rumen of heifers was increased significantly (p<0.05) when the dietary MG/MP increased. At the same time, with the improvement in dietary MG/MP, the amount of Fibrobacter succinogenes increased (p = 0.08). Conclusion: A diet with an optimal ratio (1.13) of MG/MP was beneficial for the improvement of growth, rumen fermentation, dietary protein and energy utilization of 8 to 10-month-old dairy heifers in this experiment.

Effects of Selenizing Codonopsis pilosula Polysaccharide on Macrophage Modulatory Activities

  • The purpose of the present study was to investigate the immune-enhancing activity of selenizing Codonopsis pilosula polysaccharide (sCPPS5) in nonspecific immune response. In in vitro experiment, the results showed that sCPPS5 could promote the phagocytic uptake, NO production, and TNF-α and IL-6 secretion of RAW264.7 cells. sCPPS5 could also strongly increase the IκB-α degradation in the cytosol and the translocation of NF-κB p65 subunit into the nucleus of RAW264.7 cells. In the vivo experiment, sCPPS5 at medium doses could significantly improve the phagocytic index of peritoneal macrophages and induce the secretion of TNF-α and IL-6. Moreover, the effect of sCPPS5 was significantly better than Codonopsis pilosula polysaccharide (CPPS). These results indicated that selenylation modification could significantly enhance the immune-enhancing activity of CPPS in the nonspecific immune response.

  • The present experiment was conducted to evaluate the effects of single-sex or mixed rearing on the growth and laying performance, blood parameters, egg quality, and feather scores of laying hens in an aviary system. A total of 2,928 (females, 2,856; males, 72), Hy-Line Brown chickens at 20 weeks were evenly assigned to two treatments: termed here the single-sex (only female) and mixed-sex (female to male 20 : 1) conditions. They were allowed an ad libitum diet for 20 weeks in an aviary system. There were eight replicates of each treatment, each consisting of 12.42 m2·birds-1 (n=183), respectively. The body weight and uniformity in the mixed-sex condition were higher than those in the single-sex condition, but egg production did not show any significant differences between the two types of treatment for 20 weeks. The mixed-sex group showed an effect through stress parameters (corticosterone), whereas the feather score on the back was low at the end of the experiment. These results suggest that the presence of males affect corticosterone level for mating but can reduce the vigilance and aggressive behavior of laying hens.

Effects of Replacing Corn with Chinese Brown Rice on Growth Performance and Apparent Fecal Digestibility of Nutrients in Weanling Pigs

  • This experiment was conducted to study the effects of replacing corn with Chinese brown rice on performance and apparent fecal digestibility of nutrients in weanling pigs. Seventy-two cross-bred (Duroc${\times}$Landrace${\times}$Large White) weanling pigs, weighing an average of $7.38{\pm}0.06kg$, were randomly assigned to either a corn-soybean meal diet (treatment 1), a corn-brown ricesoybean meal diet (treatment 2) and a brown rice-soybean meal diet (treatment 3). The brown rice used in this experiment was husked from one kind of early, long grain, non-glutinous rice (ELGNR, indica rice). The growth performance of weanling pigs in the experiment was similar during weeks 1 and 2, but feed conversion ratio (FCR) was significantly better in treatment 2 during weeks 3 and 4. During week 2, only the digestibility of organic matter in treatments 2 and 3 was significantly higher than that in treatment 1. During week 4, the digestibility of gross energy, ether extract, organic matter and dry matter in brown rice treatment, especially in treatment 3, were significantly higher than in treatment 1. At the end of the experiment, the concentration of serum urea nitrogen of pigs in treatment 2 was lower than that in treatment 1 and 3 and no difference was found in serum glucose and phosphorus. It could be concluded from this experiment that replacing 50% of corn with brown rice in the corn-soybean meal diet caused no negative effects to weanling pig performance, and apparent fecal digestibility was unaffected. It is feasible to replace corn with brown rice in weanling pig diets in southern China.

  • This study was carried out to investigate the effects of semen on reproductive ability in crossbred Korean native chicken (KNC, 58-wk old). The body weight, volume of semen and concentration of spermatozoa, were 2.96 g, 0.40 ml, $36.58{\times}10^8/ml$, respectively, in KNC. The fertility and hatchability were 94.8% and 78.8% respectively in crossbred KNC. KNC(Y) was high compared to other strains in fertility. The other strains were not significantly different among 6 strains. The results of this experiment indicated that hatchability of (G) was high compared to other strains. The result of this study could be available to genetic improvement of reproductive traits as a basic reference in KNC strains. To achieve the more effective improvement of reproductive traits, addition research such as genetic parameter evaluation should be performed.

Influence of Phytase and Xylanase Supplementation on Growth Performance and Nutrient Utilisation of Broilers Offered Wheat-based Diets

  • Individual and combined supplementation of phosphorus-adequate, wheat-based broiler diets with exogenous phytase and xylanase was evaluated in three experiments. The effects of the enzyme combination in lysine-eficient diets containing wheat and sorghum were more pronounced than those of the individual feed enzymes. The inclusion of phytase plus xylanase improved (p<0.05) weight gains (7.3%) and feed efficiency (7.0%) of broilers (7-28 days post-hatch) and apparent metabolisable energy (AME) by 0.76 MJ/kg DM. Phytase plus xylanase increased (p<0.05) the overall, apparent ileal digestibility of amino acids by 4.5% (0.781 to 0.816); this was greater than the responses to either phytase (3.6%; 0.781 to 0.809) or xylanase (0.7%; 0.781 to 0.784). Absolute increases in amino acid digestibility with the combination exceeded the sum of the individual increases generated by phytase and xylanase for alanine, aspartic acid, glutamic acid, glycine, histidine, isoleucine, phenylalanine, threonine, tyrosine and valine. These synergistic responses may have resulted from phytase and xylanase having complementary modes of action for enhancing amino acid digestibilities and/or facilitating substrate access. The two remaining experiments were almost identical except wheat used in Experiment 2 had a higher phytate concentration and a lower estimated AME content than wheat used in Experiment 3. Individually, phytase and xylanase were generally more effective in Experiment 2, which probably reflects the higher dietary substrate levels present. Phytase plus xylanase increased (p<0.05) gains (15.4%) and feed efficiency (7.0%) of broiler chicks from 4-24 days post-hatch in Experiment 2; whereas, in Experiment 3, the combination increased (p<0.05) growth to a lesser extent (5.6%) and had no effect on feed efficiency. This difference in performance responses appeared to be 'rotein driven'as the combination increased (p<0.05) nitrogen retention in Experiment 2 but not in Experiment 3; whereas phytase plus xylanase significantly increased AME in both experiments. In Experiments 2 and 3 the combined inclusion levels of phytase and xylanase were lower that the individual additions, which demonstrates the benefits of simultaneously including phytase and xylanase in wheat-based poultry diets.

한우, 칡소 및 제주 흑우 Calpain-Calpastatin 유전자 다양성 (Diversity of Calpain-Calpastatin gene frequencies in Brown, Brindle and Jeju Black Hanwoo)

  • The aim of study was to investigate genetic diversity for the calpain/calpastatin gene in three Hanwoo breeds [(Brown (n=62), Brindle (n=81) and Jeju Black (n=30)]. Random samples from three breeds of Hanwoo were selected and genotyped for the 7 SNPs of calpain/calpastatin using TaqMan method. Allele frequencies were investigated for CAPN1/CAST gene. Allele frequency of CAST2 SNP was 0.75, 0.59 and 0.22 for Brown, Brindle and Jeju black, respectively. The CAST3 revealed allele frequency of 0.59 and 0.57 in Brown and Jeju Black, while it showed very low allele frequency (0.07) in Brindle. In particular, favorable allele (G allele) for the CAPN1-2 SNP which was shown a strong association with tenderness in Taurine and Indicine cattle revealed 16% and 17% higher allele frequency in Brown Hanwoo (0.82) comparing Brindle (0.66) and Jeju Black Hanwoo (0.65). AMOVA demonstrated that among population variance occupied only 10% of total variance and among individual variance was 0%, while within individual variance was 90% of total variance. This result showed that population effect contributed very small portion of genetic to these three Hanwoo breeds, while within individual variance contributed large portion of genetic diversity within these Hanwoo breeds. In conclusion, three Hanwoo breeds (Brown, Brindle and Jeju black) showed a genetically homogeneous based on the 7 SNPs of CAPN1/CAST gene and it came from same ancestor to form modern Hanwoo breed.

ALKALINE HYDROGEN PEROXIDE TREATED ASPEN AS A RUMINANT FEED

  • Two experiments were conducted to evaluate the nutritive value of a diet containing 20% alkaline hydrogen peroxide(AHP) treated aspen sawdust for ruminants. In experiment 1, sheep fed treated aspen(treated) had higher (p < 0.05) average daily gain and improved feed/gain ratio compared to animals fed untreated aspen (untreated), Apparent NDF and ADF digestibilities of treated aspen were higher (p < 0.05) than for untreated material. A similar trend was observed for DM, organic matter and cellulose digestibilities coefficients. In experiment 2, the soluble and degradable DM and crude protein (CP) fractions tended to increase with AHP treatment. Treatment also increased (p<0.05) the degradation rate of the degradable fraction. Results of these experiment indicate that AHP treatment results in a substantial improvement in the extent of utilization of aspen sawdust by ruminants.

Performance of Weaner Lambs on Conventional Feeds or Supplemented with Mango Seed Kernel (Mangifera indica) and Babul Pods Chuni (Acacia nilotica) under Intensive Production System

  • Twenty four weaner lambs, eight each of Marwari, Patanwadi and Merino${\times}$Patanwadi breeds (9.9 to 10.8 kg) were randomly divided into two dietary treatments on body weight basis Viz. $T_1$-conventional (Maize-38%, GN Cake-25%, Rice Polish-24%, Jaggery solution-10%, Mineral mixture-3%) and $T_2$-supplemented non conventional group (GN Cake-25%, Rice Polish-14%, Mango seed kernel-25%, Babul Pods chuni-23%, Jaggery solution-10%, Mineral mixture-3%). The Jaggery solution was prepared by mixing 6.5 kg Jaggery and 3.5 kg water. The average final body weight at the end of the experiment was recorded to be $19.33{\pm}0.76$ and $19.72{\pm}0.8kg$ in conventional and non-conventional groups, respectively. The total dry matter intake (DMI) during the entire experiment period was recorded to be $89.56{\pm}5.19$ and $95.08{\pm}1.06$ (kg/head) and $532.83{\pm}9.94$ and $566.75{\pm}10.49g/d$ in conventional and nonconventional groups, respectively. The body weight gain and body measurements did not influenced by diet. The ration effect was found to be significant when the DMI was expressed in terms of g/d. The crude protein (CP) and digestible crude protein (DCP) intake/head and per kg gain observed during experiment under conventional and non-conventional treatment group did not differ from each other. However, the total digestible nutrients (TDN) intake per kg gain was significantly (p<0.05) higher in supplemented non-conventional group. The intake values of DCP and TDN were more or less in agreement with Indian Council Agricultural Research (1985) recommendations. The estimated total feed cost (Rs./animal) for experimental lambs was $274.16{\pm}8.57$ and $242.67{\pm}5.10$ in conventional and non- group, respectively. The non-conventional group had significantly (p<0.05) lower feed cost (11.6%). The return as percent of feed cost and feed cost/kg dressed weight were $92.89{\pm}5.58$ (%) and Rs. $35.40{\pm}1.11$ and $122.61{\pm}5.06$ (%) and Rs.$30.47{\pm}1.71$ in conventional and non-conventional group (p<0.05), respectively which is the reflection of significantly lower total feed cost incurred during feeding in non-conventional group. Lambs fed non-conventional based diet had similar live weights as those fed conventional diets but costed less money to achieve those weights.
