  • Previously, three kinds of lipases, lipAD1, lipAD2, and lipAD3, and lipase chaperone, lipBD, of Acinetobacter schindleri DYL129 isolated from soil sample were reported. In this report, three lipase and lipase chaperone were cloned into the pET32a(+) or pGEX-6P-1 vectors for protein expression in Escherichia coli, and named as pETLAD1, pETLAD2, pETLAD3 and pETLBD or pGEXLAD1, pGEXLAD 2, pGEXLAD3 and pGEXLBD, respectively. Protein expression rate was higher in pET system than in pGEX system. Although LipAD1 and LipAD2 were produced as inclusion bodies, their expression levels were high. So LipAD1 and LipAD2 could be solubilized in 8 M urea buffer and purified. LipAD3 and LipBD were overexpressed in soluble form and purified. Those proteins were purified by His-tag affinity chromatography connected in AKTA prime system. The activities of the purified lipases were demonstrated with 1% tributyrin agar plate. After purification, molecular mass was determined with sodium dodecyl sulfate-polyacrylamide gel electrophoresis. LipAD1 showed high activity toward ${\rho}$-nitrophenyl acetate and ${\rho}$-nitrophenyl butyrate, LipAD2 showed high activity toward ${\rho}$-nitrophenyl acetate and ${\rho}$-nitrophenyl myristate, and LipAD3 showed high activity toward ${\rho}$-nitrophenyl acetate, ${\rho}$-nitrophenyl butyrate, and ${\rho}$-nitrophenyl miristate, respectively. Three lipases, LipAD1, LipAD2, and LipAD3, showed optimal reaction at $50^{\circ}C$ using ${\rho}$-nitrophenyl butyrate, as substrate.

  • Though it has been widely known the nitrogen effects are influenced by soils, varieties, and mineral nutrients in the rice culture, few analyses in relation to the factors increasing nitrogen effect have been studied in Korea. The effects of potassium and silica on the factors increasing nitrogen effects in paddy soils were investigated in accordance with soil improvement practices and nitrogen application methods for the cultivated varieties. The results obtained are as follows. 1. For 413 paddy fields, the yield from soils without nitrogen application ranged from 200 to 850kg/10a and that from nitrogen application did 350 to 1,051kg/10a. The yield increament by nitrogen application varied 50 to 650kg/10a depending on soils. 2. Soil chemical characteristics for high yield were different between with nitrogen and without nitrogen application. In the without nitrogen application, however, contents of organic matter, phosphorous, potassium and calcium of high yield soils were lower than those of low yield, while the available silica content was higher in the former. 3. The yield increased with nitrogen application up to 22.4kg/10a and thereafter it decreased. These phenomena were supposed to be not be decrease of nitrogen uptake but by lowered silica uptake. 4. Clay soil incorporation, deep plough, and inorganic constituents control such as Ca, Mg, and $Sio_2$ were effective as soil improvement praitices. It was appeared that increases of silica content and Ca/Mg ratio were important to increase nitrogen effects. 5. For the correlation between yield and yield components, it was high between yield and panicle in low nitrogen level and so was it between grain yield and ripening rate in high nitrogen. 6. In the urea and super granule urea application plot, recovery rate of nitrogen by plant and soil was high and yield was remarkable high. 7. Regardless of fertilizer types such as ammonium sulfate and urea, the residual nitrogen was about 4kg/10a in both plots of 5.8 and 11.6kg/10a. N applied. 8. The potassium application to soil enhanced the nitrogen efficiency. It was more effective in low potassium soil. 9. Optimum pH value for gel formation in the 4% sodium silicate solution was approximately 6.6. 10. It was suggested that silica could affect to rice plant growth as the inorganic and organic chemical components.

  • The Sr and Pb isotopic ratios and chemical composition were measured for atmospheric bulk deposition samples collected in the Jeonju, Gunsan and Namweon areas over a period of one year. Acidity of deposition ranged pH $4\~7$ with little higher in dry season, and around pH 5.0 in rainy season. The EC and TDS of rainy season was low showing dilution effect, and increased during dry season. Sulfate $(SO_4)\;and\;NO_3$ are atmospheric aerosols largely of anthropogenic origin in winter. Sodium was concentrated in winter deposition, Ca was concentrated in spring to summer deposition. Namweon has lower EC and TDS than those of other, and Jeonju has higher. Namweon was concentrated in $HCO_3$ and Cunsan was concentrated in Cl. Aluminium, Cu, and Zn show good correlation index with TDS, indicating of their origin atmospheric. $^{87}Sr/^{86}Sr$ ratios of bulk deposition ranged from 0.7109 to 0.7128. The isotopic variations are correlated with mixing of isotopic compositions of local soils, road deposit and biogenic aerosol. In order to constrain further the origin of aerosols in rainwater, it will be necessary to collect additional Sr isotopic data for aerosols. Lead isotope ratios for all areas were similar and belonged to Pb isotope ratios of Seoul's aerosols, but little different with Beijing's aerosols. It showing that Pb in the Korea mainly derived from the gasoline combustion, not exclusively from the Beijing.

  • This study was conducted to identify the effects of caffeine on the lipid and protein components or blood chemistry levels of the serum as well as the total homogenate, mitochondrial and microsomal fraction of the rat(Sprague-Dawley, female) liver. Acute test were conducted to determine those effects. The acute test was conducted by dividing rats into 7 groups according to the time lapsed after a single oral administration of 100mg/kg caffeine(that is control, 2hrs, 4hrs, 8hrs, 24hrs, 48hrs and 72hrs lapsed group). The concentrations of glucose, urea nitrogen, uric acid, creatinine, total protein, albumin, A/G ratio, triglyceride, total cholesterol, HDL-cholesterol, free fatty acid, phospholipid as well as the activities of alanine aminotransferase(ALT), aspartate aminotransferase(AST) and alkaline phosphatase(ALP) were measured in the serum of each experimental groups. The concentrations of the carbonyl group, malondialdehyde(MDA) and the patterns of the SDS-PAGE(Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis) were analyzed to determine the oxidative damages and metabolic changes on the lipid and protein components in the serum, and total homogenate, mitochondrial and microsomal fractions of the rat liver. The results obtained from this study were summarized as follows; 1. The concentrations of serum glucose were significantly higher(p<0.01) between 4(143.0mg/dl) and 8hrs(138.0mg/dl) in comparison to that of the control(101.1mg/dl) after a single oral administration of caffeine(100mg/kg). While on the other, there were no significant differences in the concentrations of urea nitrogen, uric acid, creatinine, total protein, albumin and albumin/globulin(A/G) ratio in comparison to those of the control. 2. The concentrations of total cholesterol and HDL-cholesterol in serum were significantly higher(p<0.01) between 4(77.4mg/dl, total cholesterol) and 8hrs(64.7mg/dl, HDL-cholesterol) in comparison to those of the control(62.8, 46.7mg/dl) after a single oral administration of caffeine(100mg/kg). On the other hand, the concentrations of triglyceride in serum were significantly lower(p<0.01) after 8hrs(38.8mg/dl) in comparison to that of the control(66.5mg/dl). 3. The activities of AST in serum was significantly higher(p<0.05) from 2hrs(149U/L) to 8hrs(178U/L) in comparison to the control(112U/L) after a single oral administration of caffeine(100mg/kg). The activities of ALT in serum were significantly higher(p<0.01) at 4(45.5U/L), 24(49.3U/L), 48(46.8U/L) and 72 hrs(42.3U/L) in comparison to that of the control(39.7U/L) after a single oral administration of caffeine(100mg/kg). • The purpose of this study was to evaluate the possibile clinical application of electroplating to increase diameter of an orthodontic wire, through examining the change of physical properties. The diameter of stainless steel orthodontic wire was increased from 0.016 inch to 0.018 inch by electroplating in a bath of nickel sulfate 100g/L, nickel chloride 60g/L, boric acid 30g/L, and sodium chloride 50g/L, under the conditions of 1.7V, $25\~29^{\circ}C\;and\;3.1\~3.3pH$. During the electroplating, the rate of diameter increase was measured every minute. To investigate uniformity, the diameter was measured at three different locations of each wire specimen aster electroplating. An X-ray diffraction test was performed to analyze the nature of the electroplated metal. Following heat treatment to improve adhesion between the wire and electroplated metal, a three-point bending test was conducted to compare stiffness, field strength, and ultimate strength among four wire groups; 0.016 inch, electroplated 016, electroplated and heat-treated 016, and 0.018 inch wires. Through the comparison of each wire group, following results were obtained. 1. In the load-deflection graph, the curve of the electroplated group was Placed between that of the 0.016 inch group and the 0.018 inch group, and the owe was closer to the 0.018 inch group by heat treatment. 2. In the electroplated and heat-treated 016 wire group, the values of stiffness, yield strength and ultimate strength showed higher tendency than in the original 0.016 Inch group. Stiffness and ultimate strength showed statistically significant differences between two groups. 3. Stiffness, yield strength, and ultimate strength of electroplated wire presented lower values than those of 0.018 inch wire group. 4. Stiffness, yield strength, and ultimate strength of electroplated and heat-treated wire showed higher tendency than those of electroplated wire group, and ultimate strength showed statistically significant difference between two groups. 5. After electroplating, the difference in diameter between the three locations was within $0.1\~0.3\%$ variation, and showed no statistical significance.

  • Mouse calvarial osteoblast cells were isolated and cultured. To examine whether the cells produce active gelatinases in culture medium or not,the cells were analyzed using by zymograsphic analysis on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). We show that mouse calvarial osteoblasts in culture constitutively synthesize progelatinase- A. Then, mouse osteoblasts, which were stimulated by PTH, $1,25(OH)_2D_3$, mononuclear cell conditioned medium (MCM) and IL-1 as bone resorption agent's, showed increased collagenolysis by producing the active gelatinase. However, treatment of indomethacin and dexamethasone significantly decreased those effects of collagenolysis in mouse osteoblastic cells. On the other hand, IL-1 in stimulating bone resorption was examined using fetal mouse long bone organ culture. IL-1 stimulated bone resorption and produced marked resorption when present simultaneously. Furthermore, when it was examined the effects of indomethacin and dexamethasone on the dose dependent responses of $IL-1{\alpha}$, indomethacin and dexametasone produced a rightward shift in the IL-1 dose response curve. The results of in vitro cytotoxicities showed that Taeyoungjon-Jahage water extracts(T.Y.J-J.H.G extracts) have no any cytotoxicities in concentrations of $1-200\;{\mu}g/ml$ and furthermore there is no any cytotoxicity even in concentration of $300\;{\mu}g/ml$ on mouse calvarial bone cells. T.Y.J.-J.H.G. extracts had protective activity against PTH (2 units/mI), or MCM (5%, v/v), or $rhIL-1{\alpha}$ (1 ng/mI) or $1,25(OH)_2D3$ (10 ng/ml) , $IL-1{\alpha}$ and $IL-1{\beta}-induced$ collagenolysis in the mouse calvarial cells. Pretreatment of the T.Y.J.-J.H.G.extracts for 1 h, which by itself had little effect on cell survival, did not enhance the collagenolysis, nor significantly reduced the collagenolysis by pretreatment. Furthermore. the medicinal extracts were shown to have the protective effects against collagenolysis induced by $IL-1{\alpha}$ and $IL-1{\beta}$. Pretreatment of the extracts for 1 h significantly reduced the collagenolysis. Interestingly, the T.Y.J.-J.H.G. extracts were shown to have the inhibiting effects against gelatinase enzyme and processing activity induced by the bone resortion agents of PTH, $1,25(OH)_2D_3$, $IL-1{\beta}$ and $IL-1{\alpha}$, with strong protective effect in pretreatment with the extracts. T.Y.J.-J.H.G. extracts were shown to have the inhibiting effects against $IL-1{\alpha}-$ and $IL-1{\beta}-stimulated$ bone resorption and the effect of the pretreatment with a various concentrations of the medicinal extracts were significant. The inhibition extent and phenomena of IL-1 stimulated bone resorption by nonsteroidal anti-inflammatory agents of indomethacin and dexamethasone were similar to those obtained by T.Y.J.-J.H.G. extracts treatment in the mouse calvarial tissue culture system. These results indicated that the T.Y.J.-J.H.G.-water extracts are highly stable and applicable to clinical uses in osteoporosis.

  • Inflammatory bowel disease is an immune disorder associated with chronic mucosal inflammation and severe ulceration in the gastrointestinal tract. Antibodies against proinflammatory cytokines, including TNF${\alpha}$, are currently used as promising therapeutic agents against the disease. Stabilization of the transcript is a crucial post-transcriptional process in the expression of proinflammatory cytokines. In the present study, we assessed the expression and histological distribution of the HuR protein, an important transcript stabilizer, in tissues from experimental animals and patients with Crohn's disease. The total and cytosolic levels of the HuR protein were enhanced in the intestinal epithelia from dextran sodium sulfate (DSS)-treated mice compared to those in control tissues from normal mice. Moreover, the expression of HuR was very high only in the mucosal and glandular epithelium, and the relative localization of the protein was sequestered in the lower parts of the villus during the DSS insult. The expression of HuR was significantly higher in mucosal lesions than in normal-looking areas. Consistent with the data from the animal model, the expression of HuR was confined to the mucosal and glandular epithelium. These results suggest that HuR may contribute to the post-transcriptional regulation of proinflammatory genes during early mucosal insults. More mechanistic investigations are warranted to determine the potential use of HuR as a predictive biomarker or a promising target against IBD.

  • This study was conducted to investigate meat quality and sensory characteristics between castrated and non-castrated dairy goats. Dairy goat of Saanen breeds was slaughtered at an age of 6 mon. Then, characteristics of dairy goat meat were analyzed to chemical compositions, collagen content, pH, meat color, cooking loss, water-holding capacity, shear force, protein solubility, and myofibrillar protein fractions by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Also, odor from dairy goat meats was analyzed by sensory evaluation and volatile substances by gas chromatography-mass spectrometry (GC-MS). As a result, the chemical compositions and physicochemical characteristics were not significantly different between castrated and non-castrated dairy goats meat. Also, there is no difference protein solubility (sarcoplasmic, myofibrillar and total protein) and protein fraction by SDS-PAGE. Sensory evaluation results in odour scores are highly (p<0.05) non-castration dairy goat meat better than castration. As a result, overall palatability was higher (p<0.05) in castrated goat meat when compared with non-castrated one. The indole and octadecanoic acid by GC-MS based on sensory evaluation results were only detected in non-castrated dairy goat meat. Therefore, distribution for goat meats castrated compared to non-castrated dairy goat meat is expected to be able to get a good response to the Korean consumer.

  • Improving flour quality is one of the major targets of wheat breeding programs. This study determined the optimum high-molecular-weight glutenin subunits (HMW-GS) to improve flour quality, and analyzed the correlation between agronomic and quality traits in Korea. A total of 180 wheat varieties, including 55 Korean and 125 foreign cultivars, carrying various Glu-1 alleles, were evaluated for their quality and agronomic traits. Results indicated that Glu-A1b, Glu-B1b, and Glu-D1f were the most prevailing alleles for each Glu-1 locus for Korean wheat cultivars. Korean wheat cultivars recorded shorter days to heading (DTH) and longer days to maturity (DTM) compared to foreign cultivars. In addition, an interaction effect was found between Glu-A1 and Glu-B1 alleles on several quality parameters. The combination of Glu-A1c and Glu-B1i showed a higher protein content, dry gluten content, and higher sodium dodecyl sulfate (SDS) sedimentation value than other Glu-A1×Glu-B1 combinations. Cultivars carrying Glu-A1a or Glu-A1b, Glu-B1i or Glu-B1al, and Glu-D1d for each Glu-1 locus exhibited a longer mixing time and stronger mixing tolerance. The DTM positively correlated with the protein content, gluten index and SDS sedimentation value. However, a negative correlation was observed between DTH and quality traits. Owing to the above results, this study suggests that an increase in the frequency of Glu-B1i or Glu-B1al, Glu-D1d coupled with a short DTH and long DTM could significantly improve wheat quality properties.
