인체 혈장에서 분리한 LDL과 LDL의 지방산 조성과 기능성의 변화 (Fatty Acid Composition and Functional Properties of Low Density Lipoprotein and Oxidized LDL from Human Plasma)

  • 인체의 혈장 저밀도 지단백(LDL)은 관상동맥경화 발병의 주 요인이다. 그러나 최근의 연구들은, 정상적인 LDL은 산소 자류라디칼에 의해 쉽게 산화되며, 결과 LDL 수용채와 결합하지 못한다고 밝히고 있다. 따라서 이 변형된 형태의 산화된 LDL은 macrophage scavernger receptor에 의해 인식되어 foam cell을 형성하여, 동맥혈관이 좁아지는 역할을 수행한다고 알려지고 있다. 지리과 산화에는 지방산이 중요한 작용을 하므로, 한국인의 LDL의 지방산 조성을 분석하여 서양인과 비교하였다. 결과, 한국인의 불포화 지방산의 비율이 총 지방산 함량의 약 30%인 반면 서양인은 약 70%의 분포를 갖고 있는 것으로 발표되었다. 따라서 한국인이 서양인에 비해 LDL의 산화에 대한 영향을 적게 받을 수 있으며, 따라서 동맥경화나 심장병의 발생률이 훨씬 적을 것으로 결론을 내릴 있다. 정상적인 LDL을 황산구리와 함께 배양하여, 지방의 산화를 유도하였으며 이의 정도를 지방산 산화의 생성물인 TBARS를 측정하여, LDL이 산화될 때 생성되는 자유라디칼의 양을 측정하므로서 비교하였다. 이 때, 항상화제인 비타민 C; 비타민 E와 히알우로닉산을 첨가하면 LDL의 산화가 억제되는 효과를 확인하였다. 자유 라디탈이 증가함에 따라 산화의 정도도 증가하였으며, 자유라디칼 형성의 경시적 변화는 TBARS와 유사하였다. 따라서 luminometer에 의한 자유라디칼의 정량은 TBARS에 의한 것보다 훨씬 간편한 것으로 나타났다.

에스트로겐 수용체를 통한 카드뮴 독성 및 항산화제에 의한 독성경감에 관한 연구 (Study on the Estrogen Receptor Mediated Toxicity of Cadmium and Protective Effects of Antioxidant)

  • Cadmium, a human carcinogen, can induce toxicity in various cell lines and organs. Despite extensive research, the mechanisms of cadmium-induced cell toxicity and estrogenic potential in human are not clear. This study was performed to investigate cadmium-induced toxicity on human breast cancer cells: MCF-7 cells, an estrogen receptor (ER) positive breast cancer cells, and MDA-MB-231 cells, an ER negative breast cancer cells. MCF-7 cells was proved to be more sensitive than the other cell lines (IC50 = $50\;{\mu}M$ at MCF-7 cells and $120{\mu}M$ at MDA-MB-231). The expression of JNK and AP-1 transcription factors such as c-Jun and c-Fos dependent transcription were increased by cadmium treatment. Inhibition of ER activation by ER antagonist (tamoxifen or ICI 182,780) significantly recovered the viablity and inhibited apoptotic cell death. This suggested that cadmium-induced cell death in ER (+) cells was mediated by JNK/AP-1 pathway and this pathway was more stimulated by ER activated by cadmium. Co-treatment of antioxidants such as selenium (Se), butylated hydroxyanisole (BHA), glutathione (GSH), or N-acetyl-L-cysteine (NAC) recovered the cadmium-induced cell death in MCF-7 cells. Cadmium-induced lipid peroxidation was decreased by GSH, NAC, or BHA in MCF-7 cells. The expression of SOD protein was decreased by cadmium ($100{\mu}M$) but recovered by GSH, NAC, BHA, or Se. Our data showed that the cadmium-induced cell toxicity in human breast cancer cells could be protected by the antioxidants (Se, BHA, NAC, GSH, or NAC) and ER antagonist (tamoxifen or ICI 182,780). Therefore, toxicity of cadmium in breast cancer were mediated by oxidative stress and $ER{\alpha}$.

라론 증후군의 구강 악안면 증상 (ORAL AND MAXILLOFACIAL MANIFESTATIONS OF LARON SYNDROME)

  • Growth hormone insensitivity(GHI) 증후군은 Laron에 의해 처음 보고된 질환으로 성장 호르몬 결핍증과 유사하게 악악면 양상과 심각한 성장 지연을 보이지만, 이와는 달리 혈중 성장호르몬은 정상이거나 증가되어 있고 혈중 Insulin like growth factor-I(IGF-I)과 성장 호르몬 결합 단백질은 감소되어 있는 특징을 보인다. 이 중에서 원발성으로 GHI 증후군을 보이는 경우를 라론 증후군으로 분류하고 있으며, 세계적으로 약 200여 증례가 보고되고 있으나, 치의학적인 보고는 극히드물다. 본 증례는 라론 증후군을 보이는 두 증례를 관찰한 바 작은 두개저와 상하악골, 왜소치, 구근상 치관, 침상 치근 등의 치아형태 이상 등의 다양한 구강 악안면 이상을 보였기에 이를 보고하는 바이다.

착상전 생쥐 자궁에서 콜라겐의 변화 (Altering of Collagens in Early Pregnant Mouse Uterus)

  • 착상기 이전 자궁에서 특이적 자궁내막 준비가 진행되어야 하는데, 이는 자궁 내막의 점진적 분화로 배아의 착상과 성공적 임신에 절대적으로 필요하다. 배아 발생 동안에 관찰되는 조직의 재구성은 세포외 기질을 포함한 다양한 요인에 의해 조절된다. 임신 동안에 관찰되는 극적인 변화로는 배아의 이동, 탈락막 반응, 태반의 분화를 그 예로 들 수 있다. 배아와 자궁간의 성공적 착상을 위한 변화들은 배아와 자궁의 착상을 위한 능력 갖출 수 있도록 한다. 이러한 변화과정 중에, 콜라겐이 주성분인 세포외 기질의 극적인 변화가 진행된다. 이러한 변화는 매우 복잡하여 그 기작을 밝히는 것은 쉽지 않으나, 최근 들어 PCR-select cDNA subtraction 방법, microarry 방법 등 대단위 유전자 동정 방법들을 이용하여 많은 후보 유전자가 동정되었다. 스테로이드 호르몬은 임신과 임신 유지에 중요한 역할을 수행하며, 세포외 기질의 재구성을 엄격하게 성스테로이드 호르몬에 의한 유전자 네트워크를 통하여 조절한다. 자궁의 세포외 기질의 병리적 조절이 당뇨병 등에서 보고되고 있다. 세포외 기질의 재구성은 착상과 태아와 자궁의 발달을 이해하는 데 중요하고, 또한 생식과 관련된 질병을 극복하는 데 중요하다. 비록 세포외 기질의 구성성분이 매우 다양하고 복잡하여 논의할 것이 무척 많으나, 본 종설에서는 착상기를 전후한 시기에 콜라겐의 변화를 중심으로 논하였다.

Inhibition of eNOS/sGC/PKG Pathway Decreases Akt Phosphorylation Induced by Kainic Acid in Mouse Hippocampus

  • The serine/threonine kinase Akt has been shown to play a role of multiple cellular signaling pathways and act as a transducer of many functions initiated by growth factor receptors that activate phosphatidylinositol 3-kinase (PI3K). It has been reported that phosphorylated Akt activates eNDS resulting in the production of NO and that NO stimulates soluble guanylate cyclase (sGC), which results in accumulation of cGMP and subsequent activation of the protein kinase G (PKG). It has been also reported that PKG activates PI3K/Akt signaling. Therefore, it is possible that PI3K, Akt, eNOS, sGC, and PKG form a loop to exert enhanced and sustained activation of Akt. However, the existence of this loop in eNOS-expressing cells, such as endothelial cells or astrocytes, has not been reported. Thus, we examined a possibility that Akt phosphorylation might be enhanced via eNOS/sGC/PKG/PI3K pathway in astrocytes in vivo and in vitro. Phosphorylation of Akt was detected in astrocytes after KA treatment and was maintained up to 72 h in mouse hippocampus. 2 weeks after KA treatment, astrocytic Akt phosphorylation was normalized to control. The inhibition of eNOS, sGC, and PKG significantly decreased Akt and eNDS phosphorylation induced by KA in astrocytes. In contrast, the decreased phosphorylation of Akt and eNDS by eNDS inhibition was significantly reversed with PKG activation. The above findings in mouse hippocampus were also observed in primary astrocytes. These data suggest that Akt/eNOS/sGC/PKG/PI3K pathway may constitute a loop, resulting in enhanced and sustained Akt activation in astrocytes.

$\beta$-Subunit 94~96 Residues of Tethered Recombinant Equine Chorionic Gonadotropin are Important Sites for Luteinizing Hormone and Follicle Stimulating Hormone like Activities

  • Equine chorionic gonadotropin (eCG) is a heavily glycosylated glycoprotein composed of non-covalently linked $\alpha$- and $\beta$-subunits. To study the function and signal transduction of tethered recombinant-eCG (rec-eCG), a single chain eCG molecule was constructed, and the rec-eCG protein was prepared. In this study, we constructed 5 mutants (${\Delta}1$, ${\Delta}2$, ${\Delta}3$, ${\Delta}4$, and ${\Delta}5$) of rec-eCG using data about known glycoprotein hormones to analyze the role of specific follicle stimulating homone (FSH)-like activity. Three amino acids of certain specific sites were replaced with alanine. The expression vectors were transfected into CHO cells and subjected to G418 selection for 2~3 weeks. The media were collected and the quantity of secreted tethered rec-eCGs was quantified by ELISA. The LH- and FSH-like activities were assayed in terms of cAMP production by rat LH/CG and rat FSH receptors. Then, the metabolic clearance rate analyzed by the injection of rec-eCG (5 IU) into the tail vein was analyzed. The mutant eCGs (${\Delta}l$, ${\Delta}4$, and ${\Delta}5$) were transcripted, but not translated into proteins. Rec-eCG A2 was secreted in much lower amounts than the wild type. Only the rec-eCG ${\Delta}3$ ($\beta$-subunit: $Gln^{94}-Ile^{95}-Lys^{96}{\rightarrow}Ala^{94}-Ala^{95}-Ala^{96}$) was efficiently secreted. Although activity is low, its LH-like activity was similar to that of tethered $eCG{\beta\alpha}$. However, the FSH-like activity of rec-$eCG{\beta\alpha\Delta}3$ was completely flat. The result of the analysis of the metabolic clearance rate shoed the persistence of the mutant in the blood until 4 hours after the injection. After then, it almost disappeared at 8 hours. Taken together, these data suggest that 94~96 amino acid sequences in eCG $\beta$-subunit appear to be of utmost importance for signal transduction of the FSH receptor.

Lidocaine이 아미노산 신경전도물질의 유리, 수용체 결합, 및 섭취에 미치는 효과에 관한 시험관내 실험에 관한 연구 (Effect of Lidocaine on the Release, Receptor Binding and Uptake of Amino Acid Neurotransmitters In vitro)

  • Lidocaine 투여에 의한 전신경련의 작용기전을 추구하고자 흰쥐의 전체뇌를 또는 선조체, 해마, 및 중뇌를 부위별로 적출하여 synaptosomes를 마련하고 $20{\mu}M$ veratrine또는 $5{\mu}M\;K^+$ 첨가에 의한 신경 전달물질 (Aspartic acid, Glutamic acid, GABA, Norepinephrine)의 유리촉진작용에 미치는 lidocaine, propranolol, norepinephrine 또는 serotonin의 억제효과를 관찰하였고 $[^3H]M$$[^3H]-glutamic$ acid의 synaptosomes로의 섭취에 미치는 lidocaine의 영향도 관찰하였다. 아울러 crude synaptic membrane을 이용하여 $[^3H]-GABA$$[^3H]-glutamic$ acid의 수용체 결합에 미치는 lidocaine의 작용도 실험하여 다음과 같은 결과를 얻었다. 1. Lidocaine과 propranolol은 veratrine에 의한 aspartate, glutamate, GABA 및 norepinephrine의 유리를 억제하였고, 그중 GABA 유리에 대한 억제작용이 가장 현저하였다. 2. Norepinephrine과 serotonin은 $100{\mu}M$의 농도에서 veratrine에 의한 aspartate, glutamate 및 GABA의 유리촉진 작용을 억제하였다. 3. Lidocaine은 veratrine에 의한 아미노산 유리촉진 효과에 대해서 보다 과 $K^+$ 에 의한 유리촉진 효과를 더욱 약하게 억제하였고 특히 GABA 유리에 대한 억제작용이 가장 약했다. 4. GABA와 glutamic acid의 수용체 결합과 synaptosomes로의 섭취는 1 mM 이하의 lidocaine농도에서 크게 면화가 없었다. 이상의 결과로 보아 신경전도물질의 veratrine에 의한 유리가 과 $K^+$에 의한 유리보다 더욱 생리적이라는 점을 고려한다면, lidocaine 경련은 lidocaine이 흥분성 전도물질인 aspartate나glutamate보다 억제성 전도물질인 GABA의 유리를 더욱 현저하게 억제함으로서 나타남을 시사한다.

Effect of Doxorubicin on Catecholamine Release in the Isolated Perfused Rat Adrenal Gland

  • The present study was undertaken to investigate the effect of doxorubicin (DX) on secretion of catecholamines (CA) evoked by ACh, high $K^+,$ DMPP and McN-A-343 from the isolated perfused rat adrenal gland and to establish the mechanism of its action. DX $(10^{-7}{\sim}10^{-6}\;M)$ perfused into an adrenal vein for 60 min produced relatively dose- and time-dependent inhibition of CA secretory responses evoked by ACh $(5.32{\times}10^{-3}\;M),$ DMPP $(10^{-4}\;M)$ and McN-A-343 $(10^{-4}\;M).$ However, lower dose of DX did not affect CA secretion by high $K^+\;(5.6{\times}10^{-2}\;M),$ but its higher doses depressed time-dependently CA secretion evoked by high $K^+.$ DX itself did also fail to affect basal CA output. In adrenal glands loaded with DX $(3{\times}10^{-7}\;M),$ CA secretory responses evoked by Bay-K-8644, an activator of L-type $Ca^{2+}$ channels and cyclopiazonic acid, an inhibitor of cytoplasmic $Ca^{2+}-ATPase$ were time-dependently inhibited. Furthermore, daunorubicin $(3{\times}10^{-7}\;M),$ given into the adrenal gland for 60 min, attenuated CA secretory responses evoked by ACh, high $K^+,$ DMPP and McN-A-343. Taken together, these results suggest that DX causes relatively dose- and time-dependent inhibition of CA secretory responses evoked by stimulation of cholinergic (both nicotinic and muscarinic) receptors from the isolated perfused rat adrenal gland. However, lower dose of DX did not affect CA secretion by high $K^+,$ and higher doses of DX reduced time-dependently CA secretion of high $K^+.$ It is thought that these effects of DX may be mediated by inhibiting both influx of extracellular calcium into the rat adrenomedullary chromaffin cells and intracelluar calcium release from the cytoplasmic store. Also, there was no difference in the mode of action between DX and daunorubicin in rat adrenomedullary CA secretion.

수소이온농도 변화의 수축물질에 따른 가토신동맥 수축에 미치는 영향과 기전 (Effects of $H^{+}$ on the Contraction Induced by Various Agonists in the Renal Artery of a Rabbit)

  • The effects of $H^{+}$ on the arterial contraction and their mechanisms were investigated in the renal artery of a rabbit. The helical strips of isolated renal artery were immersed in the HEPES-buffered or $CO_{2}/HCO_{3}^{-}$-buffered Tyrode's solution. The contractions induced by agonists (norepinephrine, histamine, serotonin and angiotensin II) or high $K^{+}$ were observed with change of extracellular or intracellular $H^{+}$ concentration. The contractions induced by norepinephrine, histamine, serotonin, angiotensin II or high $K^{+}$ in HEPES-buffered Tyrode's solution were inhibited by increase in extracellular $H^{+}$ concentration and potentiated by decrease in extracellular $H^{+}$ concentration. The degrees of these effects were most evident in the contraction induced by serotonin and angiotensin II, moderate in those by histamine and high $K^{+}$, and least in those by norepinephrine. Maximal contraction by norepinephrine, histamine and high $K^{+}$ were not influenced by change in extracellular $H^{+}$ concentration, but influenced in those contration by serotonin and angiotensin II. The attenuated contractions by an acidic pH were not returned to the level of contraction at normal pH (7.4) by elevation of extracellular $Ca{2+}$ concentration. The agonists (norepinephrine, histamine and serotonin)-induced contractions in $Ca{2+}$-free Tyrode's solution were also attenuated by increase in extracellular $H^{+}$ concentration and potentiated by decrease in extracellular $H^{+}$ concentration. Elevation of $Pco_{2}$ in the $CO_{2}/HCO_{3}^{-}$-buffered Tyrode's solution, which increase the intracellular $H^{+}$ concentration, at constant extracellular pH (7.4), increased the contraction by 30 mM $K^{+}$. From the above results, it is suggested that the decrease in contractions by increase in extracellular $H^{+}$ concentration may be resulted from that $H^{+}$ make the receptors less sensitive to agonists and cell membrane hyperpolarize and then inhibit the $Ca{2+}$ influx as well as $Ca{2+}$ release from intracellular $Ca{2+}$ storage site.

한국인에서 건선과 KIR (Killer Cell Immunoglobulin-like Receptor) 유전자형 사이의 연관성 (Association of KIR (Killer Cell Immunoglobulin-like Receptor) Genotype with Psoriasis in Korean Population)

  • Background: Psoriasis is a multifactorial autoimmune skin disease with a pathogenesis that has remained obscure. Recently, T cells bearing natural killer receptors (NKRs) were precisely and strongly targeted as new putative pathogenic immunocytes in psoriasis. Among NKRs, killer cell immunoglobulin-like receptor (KIR) is the major molecule recognizing HLA class I allotypes and might be closely related to psoriasis. Methods: To investigate the association of KIR genotype and patients with psoriasis in Korean, we defined the 14 KIR genotypes in 96 patients with psoriasis and 86 healthy controls using PCR-SSP methods. Results: The frequencies of KIR2DS4 and KIR3DL1 were significantly decreased in psoriasis compared with controls (RR=0.21, p<0.02). When patients were divided into two subgroups at the age of onset, type I (<30 years) and type II ($({\geq}30$ years) respectively, these phenomena were similarly observed independent of groups divided (type I: RR=0.26, p<0.005; type II: RR=0.14, p<0.0006). When the patients were divided into subgroups according to the age of onset and family history, the frequencies of KIR2DS4, KIR3DL1, and KIR2DS3 were significantly decreased in type I compared with type II psoriasis (3DL1, 2DS4: p<0.004; 2DS3: p<0.04) and were significantly decreased in psoriasis without family history compared to with family history (3DL1, 2DS4: p<0.007; 2DS3: p<0.05). The frequency of haplotype combination BB was significantly increased in psoriasis compared with controls (RR=2.74, p<0.009). Conclusion: These results suggest that KIR genotype is a factor for the occurrence and development of psoriasis and in future how combinations of HLA and KIR genes influence psoriasis needs to be defined.
