Anaerobic Bacterial Degradation for the Effective Utilization of Biomass

  • Biomass is originally photosynthesized from inorgainic compounds such as $CO_2$, minerals, water and solar energy. Recent studies have shown that anaerobic bacteria have the ability to convert recalcitrant biomass such as cellullosic or chitinoic materials to useful compounds. The biomass containing agricultural waste, unutilized wood and other garbage is expected to utilize as feed, food and fuel by microbial degradation and other metabolic functions. In this study we isolated several anaerobic, cellulolytic and chitinolytic bacteria from rumen fluid, compost and soil to study their related enzymes and genes. The anaerobic and cellulolytic bacteria, Clostridium thermocellum, Clostridium stercorarium, and Clostridium josui, were isolated from compost and the chitinolytic Clostridium paraputrificum from beach soil and Ruminococcus albus was isolated from cow rumen. After isolation, novel cellulase and xylanase genes from these anaerobes were cloned and expressed in Escherichia coli. The properties of the cloned enzymes showed that some of them were the components of the enzyme (cellulase) complex, i.e., cellulosome, which is known to form complexes by binding cohesin domains on the cellulase integrating protein (Cip: or core protein) and dockerin domains on the enzymes. Several dockerin and cohesin polypeptides were independently produced by E. coli and their binding properties were specified with BIAcore by measuring surface plasmon resonance. Three pairs of cohesin-dockerin with differing binding specificities were selected. Two of their genes encoding their respective cohesin polypeptides were combined to one gene and expressed in E. coli as a chimeric core protein, on which two dockerin-dehydrogenase chimeras, the dockerin-formaldehyde dehydrogenase and the dockerin-NADH dehydrogenase are planning to bind for catalyzing $CO_2$ reduction to formic acid by feeding NADH. This reaction may represent a novel strategy for the reduction of the green house gases. Enzymes from the anaerobes were also expressed in tobacco and rice plants. The activity of a xylanase from C. stercorarium was detected in leaves, stems, and rice grain under the control of CaMV35S promoter. The digestibility of transgenic rice leaves in goat rumen was slightly accelerated. C. paraputrificum was found to solubilize shrimp shells and chitin to generate hydrogen gas. Hydrogen productivity (1.7 mol $H_2/mol$ glucos) of the organism was improved up to 1.8 times by additional expression of the own hydrogenase gene in C. paraputrficum using a modified vector of Clostridiu, perfringens. The hydrygen producing microflora from soil, garbage and dried pelletted garbage, known as refuse derived fuel(RDF), were also found to be effective in converting biomass waste to hydrogen gas.

Dose-Response of Chitooligosaccharide on Gut Ecology and Lipid Status in Rats (흰쥐에서 키토올리고당의 장 생태와 지질 상태에 미치는 용량에 따른 효과)

  • This study was carried out to investigate the dose-response of chitooligosaccharide (with a molecular weight of 1~3 kDa) on antimicrobial activity and lipid lowering functions in rats. Sprague-Dawley male rats were given experimental diets containing 0 (control), 0.5, 2, or 5% chitooligosaccharide (COS) for 5 weeks. Weight gain and food intake were significantly lower in rats fed 5% COS than control rats and rats fed 0.5 and 2% COS. The numbers of fecal bacteria, including bifidobacteria, lactobacilli, bacteroides, total anaerobes, and total aerobes, which reflect gut microbiota, were significantly decreased in rats fed 5% COS. Plasma triglyceride concentrations significantly decreased in a dose-dependent manner in rats fed 2% or 5% COS, while plasma total cholesterol was not significantly different among groups. The hepatic concentration of triglycerides was lower in rats fed 5% COS, and fecal triglycerides significantly increased in rats fed 5% COS. These results indicate that 5% COS supplementation in a diet may exert antimicrobial activity in vivo, and inhibit the proliferation of typical gut microbes, while lowering lipids.

Biogas Production and Utilization Technologies from Organic Waste (유기성폐기물을 이용한 바이오가스 생산 및 활용기술)

  • Anaerobic digestion (AD) is the most promising method of treating and recycling of different organic wastes, such as OFMSW, household wastes, animal manure, agro-industrial wastes, industrial organic wastes and sewage sludge. During AD, i.e. degradation in the absence of oxygen, organic material is decomposed by anaerobes forming degestates such as an excellent fertilizer and biogas, a mixture of carbon dioxide and methane. AD has been one of the leading technologies that can make a large contribution to producing renewable energy and to reducing $CO_2$ and other GHG emission, it is becoming a key method for both waste treatment and recovery of a renewable fuel and other valuable co-products. A classification of the basic AD technologies for the production of biogas can be made according to the dry matter of biowaste and digestion temperature, which divide the AD process in wet and dry, mesophilic and thermophilic. The biogas produced from AD plant can be utilized as an alternative energy source, for lighting and cooking in case of small-scale, for CHP and vehicle fuel or fuel in industrials in case of large-scale. This paper provides an overview of the status of biogas production and utilization technologies.

  • There are many of anaerobic culture methods and equipments for isolation and cultivation of anaerabic bacteria, but most of these methods are used without pre-reduced media. Gas-jet method is a recommend. able method for the culture of anaerobes, resently developed. Bacteriological studies were experimented of liver abscess of cattle by the use of gas. jet method. The results were summarised as follows; 1. Gas-jet method for anaerobic culture are expedient for the making of pre-reduced media, maintaining of oxygen free condition in the culture tube, picking of bacteria from colony and colony counting etc. 2. A 121 strains of facultative anaerobic and anaerobic bacteria were isolated from liver abscess of 27 head of cattle, and the isolated anaerobic bacteria were as follows. Peptostreptococcus spp. 7 strains Acid aminococcus fermentans 1 Veillonella spp. 1 Bacterioides spp. 6 Bifidobacterium spp. 4 Arachinia propionica 2 Lactobacillus spp. 4 Propionibacterium acnes 1 3. Liver abscess were infected with many of bacteria, about $10^3-10^9$ numbers per gram of abcessed tissue. Almost of abscess were mixed infection of various bacterial species rather than simple species.

Pathogenicity and Bacteriological Characterization of Erwinia carotovora subsp. carotovora isolated from Mulberry Trees (뽕나무에서 Erwinia carotovora subsp. carotovora의 분리)

  • The bacteria isolated from mulberry trees which had shoot soft rot symptoms occurred after intermediate cutting during autumn rearing season were indentified. External morphology of the bacteria showed rod shape with peritrichous flagella, and were facultative anaerobes in oxygen requirement test. The baacteria easily putrefied the slices of potato and carrot when they were placed on to the center of sliced surfaces, and the bacteria, also, iniciated rapid development of soft rot symptoms on mulberry leaves in pathogenesity test. The results of morphological, physiological and pathological tests on the bacteria suggested that the isolate could be a strain of Erwinia carotovora subsp. carotovora.

  • Anaerobic bacteria are suggested to be potential source for new antibiotics. In order to search for antibiotics from domestic origin, we collected 800 soil samples across Korean locations and could isolate as many as 989 anaerobic strains. Among them 10, strains were found to have good producing capacity of antibiotics. An anaerobe was finally selected due to secreting antibiotics having high antimicrobial activity towards multiple resistant microorganism(E coli JM 83) transformed by genetic engineering technique. Its morphological, physiological and biochemical charateristics were investigated, together with antimicrobial spectrum therefrom. On antimicrobial spectrum study, substance secreted from this strain, had no activities to fungus and yeast. The selected strain showed G(+) and coccal shape, on Gram, staining and electron scanning microscopy, respectively. Biochemically this strain utilized glucose, fructose lactose, sucrose, but did not arabinose, cellulose, rhamnose, sorbitol, trehalose, mannitol. Catalase test showed negative property. Optimal growth temperature was $37^{\circ}C$. The results obtained above suggest this strain Streptococcus faecium subspp. and we named it Streptococcus sp. An-21-1.

  • Metronidazole is an antimicrobial drug widely used against various types of infectious agents, including protozoa, amoeba, Helicobacter pylori, and anaerobes. Metronidazole may produce some adverse effects on hematologic, immunologic, neurologic and other systems. We report a case of reversible metronidazole-induced encephalopathy. The toxic dose of metronidazole and the onset of encephalopathy were variable. Two patients showed abnormally high signal intensity in the bilateral dentate nucleus of cerebellum, and characteristic abnormalities were detected by brain magnetic resonance imaging (MRI) and T2-weighted images, fluid-attenuated inversion recovery images and/or diffusion weighted imaging (DWI). Discontinuation of metronidazole resulted in the improvement of the neurologic symptoms over a period of two to three weeks. We followed up the brain MRI with DWI in one case following obvious clinical improvement, and the previously detected lesion had disappeared.

Isolation of Anaerobic Bacteria from Oral Pyogenic Infections (구강 화농성 감염에서 혐기성 세균의 배양분리)

  • Strict anaerobic procedures and anaerobic chamber were employed in order to improve the isolation of obligate anaerobes from oral pyogenic infections. Also different culture media were utilized to maximize bacterial recovery. Two blood media: nalidixic acid Tween blood agar (NATB) and plain blood agar (BA), two non-blood media: nalidixic acid Tween agar (NAT) and Gifu anaerobic medium (GAM) were used and compared for their isolation efficacy. Specimens from seven patients were collected with syringe. After collection, the needle was sealed with sterilixed silicone rubber and brought to labortory. The time spent from specimen collection to its processing in anaerobic chamber usually was 15 min. Identification of isolated bacterial strains was done with the API-20A system. Increase in isolation of anaerobic vacteria was achieved. Obligate anaerobic bacteria isolated were 3.3 strains per specimen. This figure falls within the range of 1.9-4.4 strains per specimen reported in other countries. With respect to the media utilized, blood media were superior to non-blood media. NATB medium was effective especially for the isolation of Gram-positive cocci. A total of 15 species of Gram-negative rods was isolated and 12 of them belonged to Bacteroides.

Quality Changes of Sterilized Soybean Paste during Its Storage (살균 된장의 저장과정 중 품질변화)

  • The sterilization was attempted to improve the quality deterioration of soybean paste during its storage. For this experiment, soybean paste was sterilized at 80oC for 30 minutes and stored during 6 months at 15oC and 30oC, respectively. The total approximate composition contents were moisture 52.5%, crude protein 11.94%, crude fat 2.0%, amino nitrogen 413.3mg%, sodium chloride 11.61% and ash 15.5%. According to the increase of storage period, pH was decreased gradually because of the increase of organic acids by the metabolism of microorganisms and the acid accumulation by acid forming bacteria, but titratable acidity was increased during storage. Amino nitrogen was rapidly increased for the first one or two month storage period and maintained as the same level for the rest of them. Each amino acid contents of soybean paste, which were glutamic acid, tryptophan, proline, arginine, and aspartic acid, had much higher level than others. In color changes sterilized soybean paste(SSP) was much lower than that of raw ones(RSP). Hunter L and b values on the surface of soybean paste were decreased during storage, and the decreasing levels were higher at 30oC than at 15oC. Hunter a value, however, was increased a little in the initial storage, and thereafter it was decreased. Lactic acid bacteria, yeasts, and molds were disappeared completely by the sterilization. However, the bacteria of aerobes and anaerobes were not disappeared by this processing.
