Expression of the crylAcl Gene Under the Control of the Native or the $\alpha$-Amylase Promoters in an Acrystalliferous Bacillus thuringiensis Strain

  • Expression of the crylAcl gene of an acrystalliferous Bacillus thuringiensis strain under the control of the native or $\alpha$-amylase gene promoter was investigated. The crylAcl gene was cloned in a B. thuringiensis - E. coli shutle vector, pHT3101, undder the control of either the native promoter (pProAc) or the $\alpha$-amylase promoter from Bacillus subtilis (pAmyAc). These two recombinant plasmids were successfully expressed in B. thuringiensis subsp. kurstaki Cry B. The first transformant (ProAc/CB), harboring pProAc, expressed an about 130 kDa protein begining 24 hr after inoculations just as in the case of the wild type of B. thuringiensis subsp. kurstaki HD-73. The second pAmyAc-transformant (AmyAc/CB) began to express the gene just 6 hr after inoculation, but Western analysis showed that the activity of the $\alpha$-amylase promoter was relatively weaker than that of the native promoter. As expected, their toxicity against Plutella xylostella larvae was dependent on the amount of Cry1Acl protein expressed.

  • A plasmid vector was constructed for the expression and secretion of Bacillus macerans cyclodextrin glucanotransferase (CGTase) in Bacillus subtilis. The vector, pUBACGT, was composed of the ribosome-binding sequence, signal sequence, and cgt gene from B. macerans under the control of amyR2, the promoter of amyE gene coding for $\alpha$-amylase from B. subtilis var. natto. Bacillus subtilis LKS88, a mutant strain lacking genes for an amylase and two proteases, was used as a host for the transformation of the plasmid vector. The transformants were selected on kanamycin-containing Luria-Bertani plates. The starch hydrolyzing activity was observed on the starch-containing plates by the iodine method and cyclodextrin-forming activity was detected in the culture medium. A SDS-PAGE analysis showed that most of the expressed CGTase in the recombinant B. subtilis was secreted into the medium at a high expression level.

  • When we inject drugstuffs to a patient for a long time, it is important to control proper injection amount and flow rate. Since inproper injection amount and a flow rate would cause bad a recovery a patient, the relience of sensors which detect injection amount is an important factor for whole injection systems' performance. In this research, we've compared the suitability of three sensors for injection pump monitoring system. The three types of sensors, piezo film sensor, photo transistor made up with three transmitting photodiodes and receving photodiode, and photo array, were selected for comparing. Using suggested data processing technique and photo amy sensors, we could minimize the effect of interference, disturbance, illumanation change, and sensitivity change caused by sensor's position. According to the experiments, the photo amy showed the higher reliance than any other the three types of sensors. The developed systems could be the foundation of beginning home production of infusion pump system and available for the base model of whole monitoring and control systems.

A Study on Signification of Components in Fashion Advertising (의류광고 구성요소의 의미화 고정에 관한 연구)

  • In the study, conceiving that signifying processes like model, setting, advertisement and description are important to promote the purchase of clothes that would satisfy consumer's desire by their expressing mode, I considered the processes of components of which fashion ads consist. As for the methods to study, I regarded the results presented from prior researches of clothing & textiles and other disciplines for the components of fashion ads and objectified their image that may be interpreted subjectively: and then, I adopted to analyse them using advertisement-semiological method to make clear the signifying processes. The results are as follow: 1. Fashion ad, one of visual symbols to transfer brand image, conveys the image with which various components are combined like model, clothes, setting and description as signs. ① the image of clothes amy be differently expressed according to social, cultural norm and individual characteristics, in the case of clothes, therefore, the signified can be regarded as the transferred image by design of the clothes① sign, and the abstract conception which may be rise to mind by the image in a ceratin culture. ② Each signifier such as countenance, line of vision, attitude and hairstyle of a model conveys different image, or the signified, respectively, and it amy operate as a sign that can express the brand image symbolically. ③ The signifiers like background, color and property symbolize the advertised merchandise of clothes and define it attribute.. 2. In the case of fashion ads, key referent systems are fashion phenomena, contemporary role image, social psychology, common morality, and social, economical and milieu.

Amygism or Imagism?: Re-Vision of Amy Lowell's Discourse of Imagism

  • This paper, postulating that Lowell's Imagism is not some "Amygism" that wobbles with "emotional slither," "mushy technique" and "general floppiness" as Pound once mocked, but another kind of poetic discourse that deserves the fullest re-consideration, goes back to the very scene where Pound left for Vorticism, condescendingly allowing Lowell and her supporters to use the name "Imagism" for three years. There, it tries to illuminate how Lowell, making the most of the opportunity given to her, picked up what Pound had left behind, grafted it on the soil of America, and finally fulfilled her literary passion to awaken the common reading public to the taste for poetry reading. For the purpose, it looks into her critical reviews in Tendencies in Modern American Poetry, and stresses her creative critical efforts to re-address Pound's principles of "Imagisme." In particular, given the limit of space, it focuses only on the second principle of her Imagism and examines the modernity of her concepts of "a cadence," "suggestion," and "the real poem beyond." Then it reads "Patterns" in the context of Japanese poetry and Noh drama and analyzes the poetic patterns that Lowell made through a creative adaptation of Japanese aesthetics for Imagist poetics. In doing so, this paper aims to provide reasonable evidences to evaluate the modernity of Lowell's Imagist ars poetica and to consider her a truly serious Imagist poet worthy of a place in the history of American poetic modernism.

Production of hGM-CSF by transformed rice cell suspension culture

  • Recombinant human GM -CSF was expressed and secreted from transgenic rice cell suspension cultures in its biologically active form. This was accomplished by transforming rice callus tissues with an expression vector, pMYN44. containing the hGM -CSF cDNA. Regulated expression and secretion of hGM -CSF from this vector achieved using the promoter, signal peptide, and terminator from a rice alfa-amylase gene Amy3D. The Amy3D gene is expressed in response to sugar deprivation. The recombinant hGM -CSF was expressed from the transgenic rice cell culture on the sugar-free medium as a yield of about 110 mg/L in the culture filtrate, which was determined by ELISA. Biological activity of hGM-CSF was confirmed by measuring the proliferation of the hGM -CSF dependent TF -1 cells.(This work was supported by a grant from the NRL program of the Korean Ministry of Science and Technology. Expression of Bacillus licheniformis $\alpha$-amylase Gene in Lactobacillus casei Strains

  • As a first step for developing Lactobacillus strains capable of fermenting starch directly, the $\alpha$-amylase gene (amyL) from Bacillus licheniformis (Kim et al., 1988. Kor. J. Appl. Microbiol. Bioeng. 16: 369-373) was introduced into Lactobacillus casei strains and the level of $\alpha$-amylase expression in transformants was examined. 3 kb EcoRI fragments encompassing amyL were subcloned into the suitable lactococcal cloning vectors (pSA3, pMG36e, and p1L2530) and then recombinant plasmids were introduced into E. coli and L. casei strains by electroporation. Only one recombinant plasmid, $pIL2530\alpha$ was able to transform few L. casei strains tested at low efficiencies. The transformation efficiencies with the plasmid into L. casei YIT 9018 and L. casei A Tee 4646 were less than $10^2/\mu$ g pIL2530\alpha$. The level of amylase activities in L. casei was five to ten-fold lower than that in E. coli cells. $p1L2530\alpha$ was stably maintained in Lactobacillus strains in the presence of Em (5 $\mu $g/ml) but without antibiotic selection, it was unstable so more than 95$%$ of cells lost plasmids after a week of daily subculturing.

BLOOD PROTEIN POLYMORPHISMS OF NATIVE AND JUNGLE FOWLS IN INDONESIA

  • In an attempt to reveal the interrelationship between fowls of jungle and native origin, their gene constitutions were compared using gene frequencies at the 16 loci controlling blood protein variations. Of the 16 loci analysed by electrophoresis, polymorphism was detected at following seven loci: Es-1, Amy-1, Akp-akp, Akp-2, Alb, Tf and 6-PGD. The other nine loci: Amy-3, Es-D, PGM, PHI, MDH, To, LDH, Hb-1 and Hb-2, were noted to be monomorphic. Genetic distance between pairs of native fowl and jungle fowls was estimated by a numerical taxonomic method. The Indonesian native fowl was genetically close to the Indonesian red jungle fowl, and the grey jungle fowl was genetically similar to the Ceylonese jungle fowl. It was also suggested that the green jungle fowl was genetically remote from the other jungle fowls and from the Indonesian native flow. The proportion of polymorphic loci (Ppoly), the expected average heterozygosity per individual $\bar{H}$, and the effective number of alleles per locus (Ne) were calculated to evaluate the genetic variabilities in the native and jungle fowls. The Indonesian native fowl exhibited slightly higher the proportion of polymorphic loci than the jungle fowls.

  • The $\alpha$-amylase gene, amyL, from Bacillus licheniformis was expressed in Lactobacillus brevis 2.14 and Escherichia coli $DH5{\alpha}$ using two different shuttle vectors, pCW4 and pSJE. E. coli transformants (TFs) harboring either $pCW4T{\alpha}$ or $pSJET{\alpha}$ produced active $\alpha$-amylase but L. brevis TFs did not, as determined by enzyme assays and zymography. But amyL transcripts were synthesized in L. brevis TFs. In terms of plasmid stability, pSJE, a theta-type replicon, was more stable than pCW4, an RCR (rolling circle replication) plasmid, in L. brevis without antibiotic selection.

  • A beamforming method is a kind of spatial filter that focus the amy's signal capturing abilities in a desired direction. In this paper, we detect the location of a source under reflective condition using the multi-dimensional MUSIC algorithm then, we can iud the image source locations from the experimental geometry, and we reconstruct the partial fields for direct wave and reflected wave by using Adaptive nulling algorithm. Numerical simulations are performed to verify its performance under various conditions.
