  • New antibiotics having moieties of penicillanic acid, cephalosporanic acid and ampicillin on both ends of the central polyalkylene were synthesized by reacting 6-aminopenicillanic acid (6-APAl), 7-amino cephalosporanic acid (7-ACA) and ampicillin with hexamethylene diisocyanate and sebacoyl chloride, respecetively. Antibiacterial activities of the compounds were also investigated.

Induction of $\beta$-Lactamase on $\beta$-Lactam Antibiotics Resistant Bacterium ($\beta$-Lactam계 항생물질 내성균주의 $\beta$-Lactamase 생산의 유도)

  • Bacillus subtilis J105, high resistant bacteria against $\beta$-lactam antibiotics, become higher resistant through induction of $\beta$--lactamase in the presence of $\beta$--lactam antibiotics. When there is no antibiotics in medium, the production of resistance-inductive $\beta$--lactamase reached its plateau 15 hours later. But when there is ampicillin (500$\mu\textrm{g}$/$m\ell$) in medium, the production of enzyme reached its plateau 25 hours later since cultivating bacteria, whereas it is found that enzyme 2,900 units/$m\ell$ about 20 times as much as compared with not-presence of antibiotics was actived. In addition, as the result of MIC comparing applying ampicillin-treated and non-treated strain MIC of ampicillin-treated strain is about 2~27 times higher. It is considered that this strain induce $\beta$--lactamase production by ampicil-lin-treatment, then increasing it resistance. It is found that this resistant strain induce $\beta$--lactamase production against cephalosporin antibiotics as well as peni-cillin. As the result of examining the time of adding antibiotics for each phase of growth, it is concluded that logarith-mic phase is the most effective. As the aboves, it is suggested that this strain is a peculia strain that its resistance is induced high by various $\beta$--lactam antibiotics.

Antibiotic Susceptibility of Vibrio parahaemolyticus Isolated from Commercial Marine Products (시판 어패류에서 분리한 장염 비브리오균의 항생제 감수성 특성)

  • Ninety three strains (4.2%) of Vibrio parahaemolyticus were isolated from 2,239 samples of commercial marine products during 2006-2008 in Seoul, Korea. We examined 16 antimicrobial susceptibilities of 93 V. parahaemolyticus isolates. Antibiotic resistance of V. parahaemolyticus was most frequently observed to ampicillin (93.5%), followed by cephalothin (90.3%), streptomycin (87.1%), ticarcillin (55.9%), and amikacin (40.9%). Antibiotic susceptibility was most frequently observed to nalidixic acid, chloramphenicol and ampicillin/sulbactam (100%), followed by trimethoprim/sulfamethoxazole (98.9%), gentamicin and tetracycline (82.8%), and ceftriaxone (63.4%). In addition, the isolates also displayed intermediate resistance to kanamycin (79.6%), ciprofloxacin (64.5%), amikacin (47.3%) and cefoxitin (43.0%). All isolates were resistant to more than two drugs. The most prominent multiple drug resistance was 3 drug resistance (37.6%), followed by 4 drug (24.7%), 5 drug (17.2%), and 6 drug resistance (11.8%). The most prominent multiple drug resistance pattern was the cephalothin-streptomycin-ampicillin resistance pattern (22.6%), followed by cephalothinticarcillin-streptomycin-ampicillin (18.3%) and cephalothin-ticarcillin-streptomycin-ampicillin-amikacin (9.7%). Multiple drug resistance patterns of V. parahaemolyticus from marine products require continuous monitoring.

Ampicillin treated German cockroach extract leads to reduced inflammation in human lung cells and a mouse model of Asthma

  • Cockroaches can cause allergic sensitization in humans via contact with their feces or frass. Antibiotics can affect concentration of major allergen and total bacteria production in German cockroaches (Blattella germanica). This study examined the ability of antibiotic-treated German cockroaches to induce allergic airway inflammation and the effect of antibiotics on their lipopolysaccharide and Bla g1, 2, and 5 expression levels. Specifically, we measured the ability of German cockroach extract (with or without prior antibiotic exposure) to induce allergic inflammation in human bronchial epithelial cells and a mouse model of asthma. Bacterial 16S rRNA and lipopolysaccharide levels were lower in ampicillin-treated cockroaches than in the control group. The Bla g1, Bla g2, and Bla g5 expression in ampicillin-treated cockroaches decreased at both the protein and RNA levels. In human bronchial epithelial cell lines BEAS-2B exposed to the ampicillin-treated extract, expression levels of interleukin-6 and interleukin-8 were lower than that in the control group. The total cell count and eosinophil count in bronchoalveolar lavage fluid was also lower in mice exposed to the ampicillin-treated extract than in those exposed to normal cockroach extract. Mouse lung histopathology showed reduced immune cell infiltration and mucus production in the ampicillin group. Our results showed that ampicillin treatment reduced the symbiont bacterial population and major allergen levels in German cockroaches, leading to reduced airway inflammation in mice. These results can facilitate the preparation of protein extracts for immunotherapy or diagnostics applications.

Susceptibilities of Methicillin-Resistant Staphylococcus aureus (MRSA) Isolated from Milk of Bovine Mastitis to Antibiotics Combined with Sulbactam (젖소 유방염에서 분리된 메티실린 내성 황색포도상구균에 대한 항생제와 Sulbactam 병합의 항균효과)

  • Various kinds of antibiotic intramammary infusions are used for treatment of bovine mastitis. As antibiotic-resistant bacteria are increased, the therapeutic rate for bovine mastitis is decreased. The goal of this research is to detect significant synergic effects of combination of antibiotics with sulbactam, $\beta$-lactamase inhibitor, on methicilin-resistant Staphylococcus aureus (MRSA). We used 5 strains of MRSA isolated from bovine mastitis with clinical and subclinical signs. All of the bacteria isolated had resistance to oxacillin and showed multi-resistant patterns in the antimicrobial susceptibility tests. Minimal bactericidal concentrations of ampicillin, amoxicillin, cephalexin, ampicillin/sulbactam(2:1), amoxicillin/sulbactam (2:1), and cephalexin/sulbactam (1:1) were measured according to broth microdilution method suggested by National Committee for Clinical Laboratory Standards (NCCLS, M31-A2) to compare the synergic effects of sulbactam combination with each antibiotic alone. Ampicillin and amoxicillin showed synergic antibacterial activity to 4 and 3 respectively in 5 strains of MRSA in combination with sulbactam. This study demonstrates that ampicillin/sulbactam and amoxicillin/sulbactam can be therapeutic choices for mastitis associated with MRSA.

Appropriateness of Ampicillin/sulbactam as Prophylactic Antibiotics in Liver Transplantation (간이식 수술 전후 예방적 항생제 ampicillin/sulbactam의 적절성 평가)

  • The most common complication after liver transplantation (LT) is bacterial infection. The incidence of surgical site infections (SSIs) after LT was variable from 8.8%~37%. However, there has been no confirmed guideline in use of prophylactic antibiotics after LT. Ampicillin/sulbactam has been used as main prophylactic antibiotics after LT in Seoul National University Hospital (SNUH) according to the center protocol. The purpose of this study was to determine the incidence and risk factors for SSI after LT and to evaluate the appropriateness of prophylactic antibiotics. A total of 211 patients who underwent LT between July 2008 and June 2010 at SUNH were included. During study period, ampicillin/sulbactam was selected as prophylactic antibiotics in 140 patients (66.4%). A total of 43 patients (20.4%) developed infections and the incidence of SSI was noted in 28 patients (13.2%); 10.0% in ampicillin/sulbactam group and 19.4% in other antibiotics (p=0.049). The most common pathogen of SSI was MRSA (13 episodes, 49.4%). In multivariate analysis, choledochojejunostomy (OR: 7.0; 95% CI, 2.4-20.0) and lower serum albumin (OR: 3.7; 95% CI, 1.1-12.9) were found to be risk factors of SSIs. In conclusion, the incidence of SSIs after LT in this population was similar to those in other studies. Therefore, the prophylactic antibiotics protocol in LT at SNUH seems to be appropriate.

  • Nalidixic acid and six other drugs were studied for in vitro effectiveness against 200 strains of Escherichia coli isolated recently from healthy persons and bactericidal activity of ampicillin against one respective strain of Escherichia coli and Salmonella typhi isolated were also studied. The resutlts obtained by the plate dilution method showed the following percentage of resistance: kanamycin, 2.5%; streptomycin, 12.0%; ampicillin, 13.5%; tetracyclin, 15.5%; chloramphenicol, 17.5%; colistin sulfate, 19.5%. No strains were resistant to nalidixic acid, clearly indicating that nalidixic acid is the most effective drug tested. Ampicillin, measured by test-tube diltion method, was highly bactericidal against Salmonella typhi at the concentration of 2.5mcg/ml and against Escherichia coli at 5mcg/ml.

  • A simple, accurate and specific NMR procedure is described for the determination of amplicilin and cloxacillin mixtures in injection dosage form and capsules. The solvent was dimethysulfoxde $d_{6}$ and maleic acid was the internal standard. By integrating the peak at 2.68 ppm and 4.57 ppm, cloxacillin and ampicillin could be determined respectively. The relative proton ratio of ampicillin trihydrate and cloxacillin were 1.038 and 0.950. The coefficents of variation of amplicillin trihydrate and cloxacillin in a few commerical preparation were 1.55 % (n =9), 2.69 % (n =15).

  • Antimicrobial susceptibility of 157 Gram-negative bacilli (90 Escherichia coli. 30 Enterobacter aerogenes, 18 Klebsiella pneumoniae, 12 Proteus spp., 3 Pseudomonas aeruginosa and 4 Alcaligenes faecalis) isolated from infected bovine udders was determined by the plate dilution method. Gentamicin and oxolinic acid at a concentration of $12.5{\mu}g/ml$ were very active to all of 157 Gram-negative bacilli tested, and 98% of these strains were susceptible to nalidixic acid at a concentration of $25{\mu}g/ml$. Most of the 90 Escherichia coli strains were inhibited by chloramphenicol, ampicillin and carbenicillin and carbenicillin at a concentration of $12.5{\mu}g/ml$. None of the Klebsiella pneumoniae strains were inhibited at a concentration of $50{\mu}g/ml$ of ampicillin and carbenillin, whereas all the species of Proteus resisted a concentration of $50{\mu}g/ml$ or higher tetracycline. All the 3 strains of Pseudomonas aeruginosa were highly resistant to streptomycin, kanamycin, Ampicillin, chloramphenicol, tetracycline and nalidixic acid.

  • The clinical isolate Staphylococcus aureus SA2 was resistant to ampicillin, Chloramphenicol, clindamycin, erythromycin, gentamicin, kanamycin, methicillin, streptomycin, and tobramycin and harboured more than two kinds of plasmids. Transformation experiment demonstrated that 40.98-kb plasmid(pKH2) encoded resistance to ampicillin, clindamycin, erythromycin, kanamycin, and streptomycin. The cleavage map of a pKH2 was determined by restriction enzyme mapping techniques. Cleavage map is given for BamHI, BglI, BstEII, SalI and XhoI.
