As a preliminary study about the reduction of the antigenicity of whey protein isolate(WPI) by treatment of chymotrypsin, trypsin, pancreatin, and protease from Aspergillus oryzae, the properties and antigenicities of whey protein hydrolysates(WPH) were investigated. When degrees of hydrolysis (DH) were measured by use of trinitrobenzensulfonic acid(TNBS), the DH of the WPH treated by pancreatin or protease from Aspergillus oryzae$(5.05{\sim}11.47)$ were much higher than those of the tryptic or chymotryptic WPH$(15.67{\sim}20.20)$. And the pretreatments of heat$(75^{\circ}C)$, 20 min and/or pepsin resulted in higher DH of WPH, generally. When the molecular distributions of the WPH were determined by high performance size exclusion chromatography(HPSEC), the ratios of polypeptides with molecular weight more than 10kDa ranged from 12% to 36%, and the average molecular weights and the average peptide lengths of the WPH were $4,252{\sim}9,132$ dalton and $38{\sim}83$ amino acids, respectively. And there was no bitter taste in all of the WPH. Results of SDS-PAGE showed that most of intact native proteins were eliminated by the enzymatic hydrolysis but there were a few bands of peptides larger than 14.2 kDa in some WPH. When antigenicity was assayed by competitive inhibition enzyme-linked immunosorbent assay(cELISA), monovalent antigenicity of WPH to rabbit anti-WPI antiserum were lowered to $10^{-1.7}-10^{-4.9}$ times and less by the enzymatic hydrolysis. And the pretreatments of heat and pepsin resulted in the lowest antigenicicy within a group of enzymatic hydrolysis, especially in case of the pancreatic hydrolysate(PDP) whose antigenicity was found almost to be removed.
Herbicidal effectivity on perennial paddy weeds such as Sagittaria pygmaea Miq. and Eleocharis kuroguwai Ohwi was evaluated. Herbicides used were butachlor [2-chloro-2, 6-dietyl-N(butoxymethyl)-acetanilide], benthiocarb [S-(4-chlorobenzy)-N, N-diethyl-thiocarbamate], molinate (S-ethyhexahyaro-l-Hazpine-carbothiate], SW-751, Chlormethoxynil (2.4-dichlorophenyl-3-methoxy-4-nitrophenyl-ether), CNP (2.4.6-trichlorophenyl-4-nitrophenylether),oxadiazon [2-tertbutyl-4-(2.4-dichloro-S-isopropoxyphenyl)-5-OXO-1.3.4-Oxadiazoline], dinuron [1-dimethyl-benthyl)-3-pheratrylurea], bentazon [3-isopropyl-IH-2.1.3-benzothiadiazine-(4)3H-one-2.2-dioxide], ACN (3-chloro-2-amino-l.4-naphthoquinone), MCPB [4-(2-methyl-4chlorophenoxy), butyric acid], 2.4-D (sodium 2.4-dichlorophenoxy acetic acid), MCP) sodium 2-methyl-4-chlorophenoxy acetic acid), SST-5, TH 63. Graszin D (Bentazon/2.4-D) and Graszin M (Bentazon/MCP) Herbicidal effectivity was divided into three types. Type I was the complete control both leaf and tuber, and SW-751 was belonged to this type. Type II was the partial control that exhibit complete control within certain period after herbicide application. After a certain period, however, the lateral bud have the germinability and grow normally, there after. Chloromethoxynil, CNP, ACN, and Oxadiazon were belonged to this group. Type III was no control at all. For E. kuroguwai, application of CNP, Chloromethoxnil, Oxadiazon and SW-751 gave good control in the early stage shile 2.4-D, MCP, bentazon and glaszin-D controlled well the intermediate stage application. Based on this results, E. kuroguwai can be controlled by herbicide application either in the early stage or in the intermediate stage.
Acrylamide gel electrophoretic pattern of 13 wild soybean (Glycine ussuriensis) was compared with that of G. gracilis and G. max var. Gwanggyo. Average Protein content (50%) of wild soybean was greater than that of C. gracilis (46%) and Gwanggyo (45%). Grain weight of wild soybean was one third of G. gracilis and one ninth of Gwanggyo. Electrophoresis of wild soybean protein showed total 16 different bands and three of which (Rm 0.09, 0.59 and 0.84) were specific and did not appeared in 86 var. of G. max which showed four specific bands (Rm 0.35, 0.45, 0.50 and 0.77) of total 17 bands. G. glacilis had all bands of Gwanggyo and two bands (Rm 0.53 and 0.59), one of which (Rm 0.59) was specific for wild soybean indicating that G. gracilis is middle type. Of 16 protein bands the third band (32%), the first band (28%) and the 5th band (13%) were main bands. Electrophoretic pattern could be sorted qualitatively into 4 groups, semiquantitatively into 6 groups and 2 or 4 groups depending on reference pattern by correlation or pattern similarity method. All sorting methods separated a wild soybean from Sogri mountain into a group and except that there were no similarity among methods but correlation methods seems more reasonable. Protein content was no relation with electrophoretic pattern but positively correlated with percent contribution of first band at 5% level suggesting that the first band may have a important role for protein synthesis.

AMP-activated protein kinase (AMPK) maintains energy homeostasis in skeletal muscle. Nonetheless, its functional role on protein synthesis with different nutrient availability has not been elucidated. Therefore, the purpose of this study is to examine the effect of AMPK activity on protein content in C2C12 myotubes incubated with low (5 mM; LG) or high (25 mM; HG) glucose media. LG stimulated (p<0.05) AMPK and acetyl CoA carboxylase (ACC) activity compare to those in HG group. Total protein content was higher in myotubes cultured with HG than in those cultured with LG and further increased by AICAR (5-amino-1-${\beta}$-D-ribofuranosyl-imidazole-4-carboxamide). Myotubes cultured with HG showed 7.5% lower UbFL (Ubiquitin Firefly Luciferase)-to-SV40 (Simian virus40) ratio compared to those in LG. Glucose level did not change the phosphorylation level of mammalian target of rapamycin (mTOR). Interestingly, administration of AICAR significantly increased phosphorylation level of mTOR in myotubes cultured with LG but not in those with HG. Overall, this data indicate that AMPK activity and protein turnover are finely regulated in response to different glucose concentration.
To understand the molecular structure of Korean garlic viruses, cDNA cloning of virus genomic RNA was attempted. Virus particles were isolated from virus-infected garlic leaves and a cDNA library was constructed from garlic virus RNA. One of these clones, S81, selected by random sequencing has been identified as a member of potexvirus group other than potyvirus and carlavirus. The clone is 873 bp long contains most of the coat protein (CP) coding region and 3'-noncoding region including poly(A) tail. A putative polyadenylation signal sequence (AAUAAA) and the hexanucleotide motif (ACUUAA), a replicational cis-acting element conserved in the 3'-noncoding region of potexvirus RNAs are noticed. The clone S81 shows about 30-40% identity in both nucleotide and amino acid sequences with CPs of potexviruses. The genome size of the virus was analysed to be 7.46 knt by Northern blot analysis, which was longer than those of other potexviruses. The open reading frame encoding CP was expressed as a fusion protein (S81CP) in Escherichia coli and the recombinant protein was purified by immobilized metal binding affinity chromatography. Polyclonal antibody was raised against S81CP in rabbit to examine the occurrence of garlic potexvirus in Korean garlic plants by immunoblot analysis. Two virus protein bands of Mr 27,000 and 29,000 from garlic leaf extract of various cultivars reacted with the antibody. It was shown that Mr 27,000 band might not be a degradation product of Mr 29,000 band, suggesting that two types of potexvirus different in size of coat protein could exist in Korean garlic plants.
We previously reported that the truncated Cel44C-$Man26A_{P558}$${\beta}$-glycosyl hydrolase protein exhibits multifunctional activities, including cellulase, xylanase, and lichenase. DNA shuffling of the truncated Cel44C-$Man26A_{P558}$ enzyme was performed to enhance the enzymatic activity of the multifunctional ${\beta}$-glycosyl hydrolase. Two mutant enzymes, M2Cel44C-$Man26A_{P558}$ that carries one mutation (P438A) and M21Cel44C-$Man26A_{P558}$ that carries two mutations (A273T and P438A) were obtained. The enzymatic activity of the M21Cel44C-$Man26A_{P558}$ double mutant was lower than enzymatic activity of the single mutant (M2Cel44C-$Man26A_{P558}$). However, both mutants displayed the enhancements in their enzyme activities that were ${\approx}1.3$- to 2.2-fold higher than the original enzymatic activity in Cel44C-$Man26A_{P558}$. In particular, the mutant M2Cel44C-$Man26A_{P558}$ exhibited an approximate 1.5- to 2.2-fold increase in the cellulase, xylanase, and lichenase activities in comparison with the control (Cel44C-$Man26A_{P558}$). The optimum cellulase, linchenase, and xylanase activities of ${\beta}$-glycosyl hydrolase were observed at pH 7.0, pH 7.0 and pH 6.0, respectively. These results, therefore, suggest that the amino acid residue Ala438 plays important roles in the enhancement of the activity of multifunctional ${\beta}$-glycosyl hydrolase.
Applications of chitosan are related to molecular weight and degree of deacetylation(DOD) of chitosan completely. The molecular weight and DOD were greatly affected by the concentration of solution time and temperature. The degree of demineralization was not significantly different at $50^{\circ}C\;and\;70^{\circ}C$ after 30 minutes. Deproteinization decreased as process time increased. The nitrogen content was reached to 6.92% after 90 minute at $80^{\circ}C$, which is similar to theoretical nitrogen content of chitin. The DOD was 82.84% after 2 hours reaction and increased as the reaction time increased in the process. Viscosity and molecular weight are increased as recycling number of concentrated NaOH solution increased. Chemical, biological and physical properties of chitosan depend on the DOD and molecular size of the molecule. Tensile strength of the films from acetic acid solutions was between $28.9{\sim}33.6$ MPa and was generally higher than that of the films from lactic acid. Elongation of the films from lactic acid was between $97.0{\sim}109.7%$ and was generally higher than that of the films from the acetic acid. Water vapor permeability of the films prepared from lcetic acid solutions was between $1.9{\sim}2.3ng{\cdot}m/m^2{\cdot}s{\cdot}Pa$ and was generally higher than that of the films from the acetic acid.

This study investigated the effects of feeding the broilers that are exposed to extreme heat stress (EHS, $33{\pm}2^{\circ}C$) with extreme heat stress diet (EHSD) containing adequate amount of soy oil and other nutrients on their growth performance. 500 broiler chickens (Ross 308) were randomized into five dietary treatment groups according to a randomized block design on the day they were hatched. Each group was further divided into four repeat pens with each repeat pen comprising 25 chickens. The five dietary treatment groups were: T1 (Normal ambient condition + basal diet (BD), T2 (EHS +BCD), T3 (EHS + extreme heat stress diet (EHSD) prepared from BD with tallow replaced with soy oil and containing molasses 2%), T4 (EHS + EHSD prepared from BD with tallow replaced with soy oil and containing molasses 2% and methionine and lysine of 1.5 times greater quantities than in BD), and T5 (EHS + EHSD prepared from BD with tallow replaced with soy oil and containing molasses 2%, methionine and lysine of 1.5 times greater quantities than in BD, and vitamin C 200 ppm) with inverse lighting. The body weight gain of the broilers increased significantly in T4 and T5 as compared with that in T1 and T2. Weights of the lymphoid organ, bursa of Fabricius, thymus, and spleen were similar between all groups. Serum concentrations of IgG, IgG and IgM were higher in T4 and T5 than inT1 and T2, but the corticosterone concentration decreased significantly in them. In T4 and T5, Lactobacillus in the cecum increased, but Escherichia, coliform, and total aerobic bacteria decreased rather significantly, compared with those in T1 and T2. Contents of acetic acid, propionic acid and total SCFA were significantly higher in T4 and T5 than in T1 and T2.
Three experiments were conducted to evaluate the ME value, standardized ileal digestibility (SID) of amino acids (AA) of fish meal, and the effects of single cell protein (Prosin and Protide) replacing fish meal in diet on growth performance, nutrient digestibility and intestinal morphology in weaned piglets. In Exp. 1, twenty-four barrows with initial BW of $30.8{\times}2.6kg$ were allotted to one of four dietary treatments. Diet 1 contained corn as the only energy source. The other three diets replaced 20% of the corn in diet 1 with one of the three protein feeds (fish meal, Prosin and Protide), and the DE and ME contents were determined by difference. In Exp. 2, eight barrows (initial BW of $25.6{\pm}3.2kg$) were fitted with ileal T-cannulas and allotted to a replicated $4{\times}4$ Latin square design. Three cornstarch-based diets were formulated using each of the protein feeds as the sole source of AA. A nitrogen-free diet was also formulated to measure endogenous losses of AA. In Exp. 3, one hundred and eighty piglets (initial BW of $7.95{\pm}1.59kg$) weaned at $28{\times}2d$ were blocked by weight and assigned to one of five treatments for a 28-d growth performance study, each treatment was fed to six pens with six pigs (three barrows and three gilts) per pen. The five treatments consisted of the control group (CON), which was a corn-soybean meal diet containing 5% fish meal, and the other four treatments, which replaced a set amount of fish meal with either Prosin (2.5% or 5%) or Protide (2.5% or 5%). The diets were formulated to provide same nutrient levels. The results showed that on a DM basis, both of the DE and ME contents were lower in Prosin and Protide than that of fish meal (p<0.05). The SID of CP and all essential AA were greater in fish meal than in Prosin and Protide (p<0.05). The pigs fed CON diet had greater weight gain and lower feed conversion rate (FCR) than pigs fed 5% Prosin and 5% Protide diets (p<0.05). The digestibility of CP was greater in pigs fed CON, 2.5% Prosin and 2.5% Protide diets than the pigs fed 5% Prosin and 5% Protide diets (p<0.05). Villus height in jejunum and ileum, and villus height to crypt depth ratio in the jejunum were higher (p<0.05) in pigs fed CON, 2.5% Prosin and 2.5% Protide diets compared with the 5% Prosin and 5% Protide diets. Pigs fed CON diet had greater villus height to crypt depth ratio in the ileum than the pigs fed 5% Prosin and 5% Protide diets (p<0.05). In conclusion, although Prosin and Protide contained lower ME content and SID of AA than fish meal, Prosin and Protide replacing 50% of fish meal in diet with identical nutrient levels could obtain similar performance, nutrient digestibility and intestinal morphology in weaned pigs.
An experiment was conducted to compare the ME values of imported corns measured by In vivo(TME) and In vitro(MEn, ME and MEpc) methods and to investigate the true amino acid availability(TAAA) and the true nutrient availability(TNA) of imported corns. For TME assay, twenty four 57-weeks-old Hy-Line roosters were assigned to fasting group(4 roosters) and four corn groups(5 roosters each): USA; corn produced in the United States, ARG; corn produced in Argentina, CHN; regular corn produced in China, CHNP; premium corn produced in China. The MEn, ME and MEpc values were determined by equations based on chemical analysis. The TME value of USA(3,745kcal/kg) in as fed basis was significantly (P<0.01) higher than ARG(3,555kcal/kg) and CHNP(3,518kcal/kg) but was not significantly different from CHN(3,671kcal/kg). The TME value of USA(4,144kcal/kg) in DM basis was not significantly different from CHN(4,060kcal/kg) and CHNP(4,008kcal/kg) but was significantly(P<0.05) higher than ARG(4,001kcal/kg). There were significant differences in TAAA of phenylalanine, histidine and arginine among imported corns. Those of USA were highest but overall TAAA was not significantly different among imported corns. True availability of NFE of USA, ARG and CHN was significantly(P<0.05) higher than that of CHNP. However, true availability of crude protein, crude fat, crude fiber and crude ash were not significantly different among corns. The correlation coefficient between TME and MEn value was 0.91 which was significant at P<0.1 but correlation coefficient between TME and ME value and between TME and MEpc value was 0.90 and 0.83, respectively which was not significant at P<0.1. In conclusion, US corn was highest in TME values and Chinese premium corn was not significantly different from regular Chinese corn. The MEn value obtained by equation based on chemical analysis may be used as a tool to evaluate TME value of corn.

- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
