Aloe vera has been used in traditional medicine for the therapy of a variety of disorders, such as wounds and burns. However, few studies have examined the antioxidant capacities of A. vera plants during different growth periods. In order to investigate the effects of growth on antioxidant activity, A. vera was prepared from 2-, 4-, 6-, 8-, and 12-month-old aloe. The extracts from 6-month-old A. vera showed the highest contents of flavonoids (9.750 mg catechin equivalent/g extract) and polyphenols (23.375 mg gallic acid equivalent/g extract) and the highest ferric reducing antioxidant power (0.047 mM ferrous sulfate equivalent/mg extract). The extract from 6-month-old A. vera exhibited the highest free radical scavenging potential, and the lowest IC50 values were found for 2,2-diphenyl-1-picrylhydrazyl (0.26 mg/ml) and alkyl radicals (0.50 mg/ml). In addition, the extract from 6-month-old A. vera showed the greatest effects on cell viability in normal liver cells. Based on these findings, the extract from 6-month-old A. vera was examined further in order to determine its protective potential against tert-butyl hydroperoxide (t-BHP)-induced oxidative stress. The extract from 6-month-old A. vera at a concentration of 0.25 mg/ml showed the highest protective activity against t-BHP-induced reactive oxygen species production. These findings suggested that harvesting regimens were critical in the regulation of effects of the bioactive potential of A. vera on antioxidant activity.
The present study was carried out to investigate whether the aloe had a radioprotective effect in mice exposed to cobalt-60 gamma radiation or not. The survival ratio of mice for 30 days, hematopoiesis of blood-forming stem cells by spleen colony assay, chromosomal aberration frequency of bone marrow cells and histopathological findings of bone marrow were investigated. The survival ratios of aloe administered groups with concentration of 250, 500, 1,000 and 1,500mg for 3 days before irradiation and control group in cobalt-60 gamma irradiated mice(700rads whole body irradiation, dose rate of 50rads/min.) were 77.4, 79.3, 80.6, 90.0 and 53.1%, respectively. The survival ratios of pre-irradiation aloe administered groups were superior to those of post-irradiation aloe groups and control group. In spleen colony assay, Aloe vera administration before irradiation enhanced the recoveries of numbers of blood-forming stem cells of bone marrow of irradiated mice. There were decreased chromosomal aberrations of bone marrow cells at the first day after irradiation in aloe administered groups compared to that of control group. Histopathological findings in the bone marrow of irradiated mice were hypocellularity due to the depletion of myelocytes, abundant of fat vacuoles and these changes were weakened in aloe administered groups compared to that of control group.
This study was carried out to investigate the effect of polyamines, salt strength. sucrose and gelling agents on the regeneration of plantlets by meristem culture of Aloe arborescens Mill. and Aloe vera L.. Shoot multiplication was more effective when 10mg/ l spermine in Aloe arborescens and 1mg/ l spermidine in Aloe vera added into MS medium than when other polyamines were treated into media. A quarter strength of MS medium was effective for rooting of shoots regenerated. Higher concentration of sucrose (45g/ l) was more effective for shoot regeneration. Addition of 4g/ l gelrite into the medium was effective for induction of multiple shoots from Aloe than that of agar or other concentrations of gelrite. When plantlets regenerated from meristem culture were transferred to pot. survival rate of plantlets was 80% on perlite and was 95% on vermiculite. respectively.
Aloe is an attractive potential food source because it has functional uses. It has been widely used as a well-being food and drink. The antioxidant activity was estimated by measuring DPPH free radical scavenging activity. We investigated the quality characteristics of cookies prepared by adding various concentrations of aloe powder (0%, 1%, 3%, 5%, all w/w) as a substitute for flour. In this study, the DPPH free radical scavenging activity, moisture content, and hardness of cookies significantly increased with increasing aloe powder, while the pH, spread factor and L and b values slightly decreased. Sensory evaluation scores in terms of texture were shown to be very similar statistically, but color, flavor, taste, and overall acceptability scores were lower than the control. As a result of this study, the quality of cookies with the addition of 1% aloe powder were the most suitable in terms of taste, flavor, texture, and sensory properties.
This study was performed to investigate the effects of Aloe vera administration(500mg/kg. B.W., s.i.d., P.O.) on hematology, blood chemical values and histopathological changes of spleen in cobalt-60 gamma Irradiated mice. 1. Total leucocyte counts of aloe administered groups were less decreased and more rapidly returned to normal level than those of control group. 2. Lymphocyte counts were rapidly decreased on the 1st day after irradiation in all experimental groups, but the counts were less decreased in aloe administered groups than those of control group. 3. Erythrocyte counts, hemoglobin values and hematocrit values of aloe administered groups were maintained within normal level, but these values showed lower values on the 14th day after irradiation in control group then returned to normal level on the 21th day after irradiation. 4. There were no differences between aloe administered groups and control group In LDH, SGOT and glucose values. 5. Histopathological changes of spleen in aloe administered groups showed less lymphocytic necrosis and depletion than those of control group.
Aloe vera gel exhibits protective effects against insulin resistance as well as lipid-lowering and anti-diabetic effects. The anti-diabetic compounds in this gel were identified as Aloe-sterols. Aloe vera gel extract (AVGE) containing Aloe-sterols has recently been produced using a new procedure. We previously reported that AVGE reduced large-sized intestinal polyps in Apc-deficient Min mice fed a high fat diet (HFD), suggesting that Aloe vera gel may protect against colorectal cancer. In the present study, we examined the effects of Aloe vera gel powder (AVGP) and AVGE on azoxymethane-induced colorectal preneoplastic aberrant crypt foci (ACF) in mice fed a HFD. Male C57BL/6J mice were given a normal diet (ND), HFD, HFD containing 0.5% carboxymethyl cellulose solution, which was used as a solvent for AVGE (HFDC), HFD containing 3% or 1% AVGP, and HFDC containing 0.0125% (H-) or 0.00375% (L-) AVGE. The number of ACF was significantly lower in mice given 3% AVGP and H-AVGE than in those given HFD or HFDC alone. Moreover, 3% AVGP, H-AVGE and L-AVGE significantly decreased the mean Ki-67 labeling index, assessed as a measure of cell proliferation in the colonic mucosa. In addition, hepatic phase II enzyme glutathione S-transferase mRNA levels were higher in the H-AVGE group than in the HFDC group. These results suggest that both AVGP and AVGE may have chemopreventive effects on colorectal carcinogenesis under the HFD condition. Furthermore, the concentration of Aloe-sterols was similar between 3% AVGP and H-AVGE, suggesting that Aloe-sterols were the main active ingredients in this experiment.
This study was carried out to investigate the in vitro and in vivo moisturizing properties and percutaneous absorption of PEG-impregnated Aloe vera gel. The PEG-i-Aloe gel was obtained from dewatering and impregnation by soaking (DIS) of Aloe vera leaf slice. The moisturizing property of the obtained sample was evaluated by moisture determination using gravimetric method in desiccator under different RH% and by water sorption-desorption test on human skin. The transdermal penetration characteristics of PEG-i-Aloe gel was investigated by Franz diffusion cell in vitro transdermal absorption method. PEG-i-Aloe gel had high moisture retention ability and could significantly lead the enhancing skin hydration status as well as reducing the skin water loss due to the film formation as a skin barrier. The skin penetration rate of PEGi- Aloe gel at steady state was 9.76 ${\mu}g/(h{\cdot}cm^2)$ and the quantity of the transdermal absorption was 144 ${\mu}g/cm^2$ in 9 hr. The penetration mechanism was well fitted with Higuchi model ($R^2$ = 0.974-0.994). The results show that PEG-i-Aloe gel has the significant moisturizing effect and strong penetration of the animal skin. It could be used as the moisturizing additive in cosmetic skin products.
Aloe(Aloe vera LINNE) has been used as a home medicine for the past several thousand in the world, and has been studied on various chronic degenerative diseases such as atherosclerosis, myocardiac infarction and hypertension. SMAP8, learning and memory impairment animal mode, were fed basic or experimental diets with 1.0% of freeze dried(FD)-Aloe powder for 8 months. This study was designed to investigate the effects of Aloe on body weight gain, grading score of senescence(GSS), triglyceride, total and LDL-cholesterol levels, and atherogenic index in serum of SAMP8, and also designed to investigate the effects of Aloe on cholesterol accumultions in mitochondria and microsome fractions of SAMP8 brain. Body weight gain was consistently lower in aloe group than in control group, but no significantly differences between them. Grading score of senescence resulted ina marked decreases pf 20% in 1.0% Aloe group compared with control group. Administrations of 1.0% aloe resulted ina marked decreases in 15% and 20% of triglyceride and cholesterol levels, respectively, and also significantly decreased in 15% of LDL-cholesterol levels and atherogenic index in serum of SAMP8 compared with control group. Cholesterol accumulations were significantly inhibited in 20% and 10% of mitochondria and microsome fractions of SAMP8 brain, respectively, by administration of 1.0% Aloe. These results suggest that administration of Aloe mau not only effectively inhibit chronic degenerative diseases in serum of SAMP8, but may also improve learning and memory impairments of SAMP8 brain.
For physiological function of aloe concentrate by ultrafiltration (UF) process, jack bean urease inhibitory activity and bentonite flocculating activity of UF aloe concentrate was investigated and compared with fresh aloe gel. Urease inhibitory activity of UF aloe concentrate ranged from 87 to 90% in 1 mL sample. Also, urease inhibitory activity of UF aloe concentrate increased about 10% by heat treatment showing the heat stability. From Lineweaver-Burk plot for UF aloe concentrate, urease inhibition pattern indicated general non-competitive inhibition. From flocculation test of UF aloe concentrate about 1% (w/v) bentonite suspension, maximum flocclulating activity of 97% was obtained at 0.5 mL addition of UF aloe concentrate/ 5 ml bentonite suspension. However, flocculating activity of 81% was obtained at 1 mL addition of UF aloe concentrate/ 5 mL bentonite suspension, which was typical flocculating behavior of polymers with re-dispersion at overdose area. FT-IR spectra of UF aloe concentrate showed the characteristic patterns of $\beta$-binding polysaccharide and less deacetylation indicating higher level of bioactive polysaccharide content.
Acemannan, a highly acetylated glucomannan, was extracted from fresh Aloe vera leaves by ethanol fractionation, resulting in a concentration increase of more than threefold. The presence of acemannan was confirmed using FTIR and 1H NMR analysis, revealing an average molecular weight of 780 kDa. The flocculating activity of the fractionated aloe gel polysaccharide was assessed through settling tests in a 1% (w/v) bentonite suspension. The results demonstrated that the aloe polysaccharide exhibited remarkable stability within a temperature range of 20~70 ℃. The maximal flocculation rate at different pH levels ranged from 93% to 97%, with an optimal dose for maximum flocculation rate between 0.25 mg/mL. Notably, the minimum dose required for flocculation was achieved at a pH of 3, attributed to the compression of electrostatic repulsion on the surface of bentonite particles. However, the flocs obtained under acidic conditions were less dense and compact, exhibiting lower sedimentation velocity compared to those formed under neutral and alkaline pH conditions. Additionally, the addition of salt showed a slight synergistic effect on flocculation, significantly enhancing the sedimentation velocity. This investigation highlights the potential of Aloe vera polysaccharide as a natural and edible flocculant, offering promising applications in various industries.
