Sequencing, Genomic Structure, Chromosomal Mapping and Association Study of the Porcine ADAMTS1 Gene with Litter Size

  • A disintegrin-like and metalloprotease (reprolysin type) with thrombospondin type 1 motif (ADAMTS1) plays a critical role in follicular rupture and represents a major advance in the proteolytic events that control ovulation. In this study, a 9,026-bp DNA sequence containing the full coding region, all 8 introns and part of the 5'and 3' untranslated region of the porcine ADAMTS1 gene was obtained. Analysis of the ADAMTS1 gene using the porcine radiation hybrid panel indicated that pig ADAMTS1 is closely linkage with microsatellite marker S0215, located on SSC13q49. The open reading frame of its cDNA covered 2,844 bp and encoded 947 amino acids. The coding region of porcine ADAMTS1 as determined by sequence alignments shared 85% and 81% identity with human and mouse cDNAs, respectively. The deduced protein contained 947 amino acids showing 85% sequence similarity both to the human and mouse proteins, respectively. Comparative sequencing of three pig breeds revealed one single nucleotide polymorphism (SNP) within exon 7 of which a G-C substitution at position 6006 changes a codon for arginine into a codon for proline. The substitution was situated within a PvuII recognition site and developed as a PCR-RFLP marker for further use in population variation investigations and association analysis with litter size. Allele frequencies of this SNP were investigated in seven pig breeds/lines. An association analysis in a new Qingping female line suggested that different ADAMTS1 genotypes have significant differences in litter size (p<0.01).

Adenovirus vector-mediated FAM176A overexpression induces cell death in human H1299 non-small cell lung cancer cells

  • FAM176A (family with sequence similarity 176 member A) is a novel molecule related to programmed cell death. A decreased expression of FAM176A has been found in several types of human tumors in including lung cancers. In the present study, we investigated the biological activities of FAM176A on the human non-small cell lung cancer cell line H1299 cells. We constructed a recombinant adenovirus 5-FAM176A vector (Ad5-FAM176A) and evaluated the expression and anti-tumor activities in vitro. Cell viability analysis revealed that the adenovirus-mediated increase of FAM176A inhibited the growth of the tumor cells in a dose- and time-dependent manner. This inhibitory effect was mediated by both autophagy and apoptosis that involved caspase activation. In addition, cell cycle analysis suggested that Ad5-FAM176A could induce cell cycle arrest at the G2/M phase, all of which suggested that adenovirus-mediated FAM176A gene transfer might present a new therapeutic approach for lung cancer treatment.

Distribution of Heavy Metals in the Soils of Hanam City. (경기도 하남시 토양의 중금속 함량 분포 조사)

  • The objectives of this study were to find out distribution of heavy metal contents in the soils of Hanam city and to provide base-line data towards development of an eco-city Hanam. One hundred surface soil (0-20 cm) samples were collected from rice paddy field, cultivated upland, forest, riverside and other areas. The samples were air-dried, sieved to pass through 2 mm sieves, followed by analyses for As, Cd, Cu, Pb and Zn by the standard method set by the ministry of environment. All the average heavy metal contents were close to background level and were much lower than concern level of the Soil Environment Conservation Act of Korea. However, some individual heavy metal contents were higher than the action level. Since natural environment including soil environment of Hanam city is being destroyed rapidly and the number of the sampling points allocated to Hanam city based on the soil contamination monitoring network of the ministry of environment is only 6, an in-depth soil survey for contamination of Hanam city is highly recommended.

Detection Characteristics of Gamma-Irradiated Seeds by using PSL, TL, ESR and GC/MS (PSL, TL, ESR 및 GC/MS 분석을 통한 감마선 조사된 유지종실류의 검지 특성 연구)

  • In this study, we investigated the applicability of the photostimulated luminescence (PSL), thermoluminescence (TL), electron spin resonance (ESR) and gas chromatography/mass spectrometry (GC/MS) methods for 5 seeds which are not allowed to be irradiated in Korea. All 5 seeds including evening primrose seed, safflower seed, rape seed, sunflower seed and flax seed were analyzed. Samples were irradiated at 1~10 kGy using a $^{60}Co$ gamma-ray irradiator. In PSL study, the photon counts of all the unirradiated samples showed negative (lower than 700). The photon counts of irradiated (1, 5, 10 kGy) samples showed positive (higher than 5,000). In TL analysis, results showed that it is possible to apply TL method to all foods containing minerals. In ESR measurements, the ESR signal (single-line) intensity of irradiated foods was higher than non-irradiated foods. The hydrocarbons 1,7-hexadecadiene ($C_{16:2}$) and 8-heptadecene ($C_{17:1}$) from oleic acid were detected only in the irradiated samples before and after the treatment at doses ${\geq}$ 1 kGy, but they were not detected in non-irradiated samples before and after treatment. These two hydrocarbons could be used as markers to identify irradiated safflower seed, rape seed, Sunflower seed and flax seed. And then, the hydrocarbons 1,7,10-hexadecatriene ($C_{16:3}$) and 6,9-heptadecadiene ($C_{17:2}$) from linoleic acid were detected in the evening primrose seed, safflower seed and sunflower seed. According to the results, PSL, TL and GC/MS methods were successfully applied to detect the irradiated foods. It is concluded that PSL, TL and GC/MS methods are suitable for detection of irradiated samples and a combined method is recommendable for enhancing the reliability of detection results.

The Optimum Methionine to Methionine Plus Cystine Ratio for Growing Pigs Determined Using Plasma Urea Nitrogen and Nitrogen Balance

  • The objective of this study was to determine the optimum ratio of methionine to methionine plus cystine for growing pigs. A nitrogen balance trial was conducted using a total of 21 barrows (Large WhiteLandrace) over two replicates. The initial body weight was $20.36{\pm}1.22kg$ (mean${\pm}$SD) in the first replicate and $23.54{\pm}1.02kg$ (mean${\pm}$SD) in the second. For each replicate, the 21 pigs were randomly assigned to one of seven dietary treatments with three observations per treatment. The diets included a methionine and cystine-deficient basal diet with all other essential nutrients meeting nutrient requirements and six diets formulated with graded levels of DL-methionine (0.00, 0.03, 0.06, 0.10, 0.13, 0.16%) and $L-Cystine{\cdot}HCl{\cdot}H_2O$ (0.19, 0.15, 0.11, 0.07, 0.04, 0.00%). This resulted in ratios of methionine to methionine plus cystine of 41.3, 29.6, 35.3, 41.2, 46.0, 51.6 and 57.5%. Each experimental period lasted 12 days consisting of a seven-day adaptation period followed by a five-day total collection of urine and feces. During the collection period, pigs were fed 900 g/day for the first replicate and 1,200 g/day for the second replicate. The feed was provided in three equal portions at 0800, 1500, and 2200 h daily. Pigs had ad libitum access to water after feeding. There was a linear (p<0.01) and quadratic (p<0.01) effect on daily gain and feed conversion as the ratio of methionine to methionine plus cystine increased. Pigs receiving the diets providing a methionine to methionine plus cystine ratio of 51.6% had the best daily gain and feed conversion. Plasma urea nitrogen was also lowest for this treatment. Nitrogen retention increased (p<0.01) as the relative proportion of methionine increased up to 51.6% and then a downward trend occurred at 57.5%. The quadratic regression model, as well as one- and two- slope regression line models, were used to determine the optimum ratio of methionine to methionine plus cystine. Eliminating the 35.3% methionine to methionine plus cystine treatment resulted in $R^2$ values in excess of 0.92. The optimal ratio of methionine to methionine plus cystine was estimated to be 54.15% for nitrogen retention and 56.72% for plasma urea nitrogen.

Comparison and Analysis of Return Current Measurement Values on High Speed Line (고속선 귀선전류 측정값 비교 및 분석)

  • A method of common earth, which is proposed by French Railway(SNCF) and is applied to high speed railway, minimizes the effect of the traction return current from tracks so that trackside electric devices can be protected and operation and maintenance staffs are kept out from injury in an electric railway environment. According to it, all the new domestic electric railway lines are replaced from existing individual earth method to the common earth method. In this paper, we analyze the correlation between common earth method and traction return current to prove whether the common earth has surely contributed to minimize the effect of the traction return current. The analysis was done based on the measurement of the traction return current at the domestic high speed railway line.

  • This study investigated the symptoms of the a reduction in output while driving on the road, or increasing of fumer out exhaust gas on inclined road while working air-conditioner in summer. Notice how the experiment in 2010, the Ministry of Environment(Chapter No. 2010-46), and how the vehicle emissions inspection was carried out. 2500cc Diesel cars used in the study were used and compare to output of engine, exhaust gas, inhalation temperature measurement Inhalation of cold air has not been supplied to all agencies when comparing the results when cold air intake temperature of the supply air-conditioning switch range control from 1 to 4, the temperature drops $98^{\circ}C$ to $78^{\circ}C$. At the momentum of switch level 4, output from 63ps to 66ps after the connection has increased 9.6 percent, the highest concentration of exhaust emissions were reduced by 42.8%. This research can contribute in part to the reduction of exhaust directly supply into the cooling air intake line, doing the output of diesel cars in the summer. In addition, construction equipment and machinery that are currently being used excluding the engine's intercooler cooling of the supply line via a separate output in the summer and help reduce exhaust emissions is expected.

  • The tilting trains can run on curve track about 30% faster than conventional train without affecting passenger comfort. As the tilting trains offer the optimum means of providing faster and more comfortable rail service with minimum of environmental disturbance and capital investment, it is widely adopted for commercial operation all over the world. Over several years, the Korea Railroad Research Institute(KRRI) and Ministry of Construction and Transportation(MOCT) have been developing 200 km/h Korean tilting train, Hanvit200. Hanvit200 adopts the pendulum type tilting mechanism and hybrid car body structure, mainly CFRP combined with steel. In this paper the vibrational characteristics of Hanvit200 was investigated through Eigen vector analysis, modal test and main line running test.

Numerical simulation of flow around two circular cylinders in various arrangements

  • The results of flow feature around two circular cylinders in various arrangements are carried out using two-dimensional simulation at Reynolds number of 200. In this work, time-averaged fluid force acting on the upstream and downstream cylinders were calculated for staggered angle ${\alpha}=0{\sim}90^{\circ}$ in the range of L/D = 1.1~5, where ${\alpha}$ is the angle between the free-stream flow and the line connecting the centers of the cylinders, L is centre-to-centre distance and D is cylinder diameter. The dependence of magnitudes and trends of fluid force coefficient on the spacing ratio L/D and ${\alpha}$ are discussed. In all arrangements of two cylinders, tandem arrangement (${\alpha}=0^{\circ}$) is the case produced a minimum drag coefficient for downstream cylinder. Moreover, the locations of separation and stagnation points or pressure coefficient on surface of the cylinder were examined.
