Production of Alkaline Protease by Entrapped Bacillus licheniformis Cells in Repeated Batch Process

  • In this study, Bacillus licheniformis cells were immobilized by entrapment in calcium alginate beads and were used for production of alkaline protease by repeated batch process. In order to increase the stability of the beads, the immobilization procedure was optimized by statistical full factorial method, by which three factors including alginate type, calcium chloride concentration, and agitation speed were studied. Optimization of the enzyme production medium, by the Taguchi method, was also studied. The obtained results showed that optimization of the cell immobilization procedure and medium constituents significantly enhanced the production of alkaline protease. In comparison with the free-cell culture in pre-optimized medium, about 7.3-fold higher productivity was resulted after optimization of the overall procedure. Repeated batch mode of operation, using optimized conditions, resulted in continuous production of the alkaline protease for 13 batches in 19 days.

  • 발생중의 무당개구리에서의 전신 및 중신 alkaline phosphatase 활성을 Gomori 변법을 써서 관찰하였다. 전신 분비세관의 분화와 함께 세관 刷子綠에 출현한 alkaline phosphataseghkf성은  蓋완성기에 최고도에 달하였다가 제3후지 형성기에 전신의 퇴화와 함께 효소활성은 감소 소실되었다.  蓋완성기에 출현한 중신세관에서의 alkaline phosphatase 활성은 변태기를 통하여 강한 양성반응을 보였다.

  • This paper presents an overview of ONP deinking efficiency with cellulolytic enzymes and synthetic collector in alkaline pH. Deinking is a series of unit operations designed to detach ink from cellulose fibers and separate the dispersed ink from the pulp slurry. Deinking chemicals are process aids that enable expensive mill equipment used in these unit operations to operate more efficiently - often much more efficiently. We propose the blended deinking agent with cellulolutic enzymes and synthetic collector in deinking pulp of conventional alkaline method. The deinking efficiency of old news print in alkaline pH was enhanced with enzyme treatments. The brightness of deinked pulp was increased with less residual ink particles and yield of enzymatic deinked pulp was improved compared to the deinked pulp of conventional alkaline method.

  • The bone alkaline phosphatase(BALP) in 130 sera of normal dogs were assayed according to the lectin precipitation method of Rosalki and Foo. The serum BALP activities showed a wide variation as $23.27({\pm}14.73)$ IU/L in young dogs from 6weeks to 12 months old and were lower in magnitude as $9.24({\pm}3.36)$ IU/L in elder dogs from 1 years to 6years old. The serum BALP activities in normal dogs were no significant correlation in sex.

전해 알칼리수에 의한 견의 정련 (Silk Degumming by Electrolyzed Alkaline Water)

  • 명주실의 정련과정에서 다량의 세리신이 버려지고 있으므로 전해수를 이용하면 정련제의 사용이 적어 순수한 세리신을 얻을 수 있을 것으로 생각하고 전해 알칼리수에 의한 정렬법의 가능성에 관한 시험을 수행한 결과는 다음과 같다. 1. 전해 알칼리수는 밀폐하여 보관하면 8일까지도 pH와 산화환원전위력의 변화가 없었으며 보관온도에는 큰 영향을 받지 않았다. 따라서 저온에서 밀폐보관하면 실제 사용상에는 문제가 없을 것으로 생각된다. 2. 정련방법에 따른 생가 연감율은 전해수 원액으로 정련할 때 가장 크고 전해수의 pH를 11.0으로 조정한 후 정련하면 비누정련과 비슷한 정도의 효과를 얻을 수 있었다. 또 정렬방법에 따른 정련액의 pH 변화를 보면 비누액은 정련 전후의 변화가 적었으나 전해수액은 정련후 크게 저하되었다. 3. 정련에 의하여 강렬과 신도는 조금씩 감소하는 경향이었으며 pH를 11.0으로 조종하여 정련하면 비누정련과 비슷한 결과를 얻었다. 4. 이상의 결과를 종합하면 전해 알칼리수에 의한 견정련은 pH를 11.0으로 조종하여 정련함으로서 비누정련과 비슷한 연감율과 강신도 성적을 얻었으므로 전해 알칼리수에 의한 정련은 가능하리라고 생각된다.

바이오에탄올 원료로서 활용평가를 위한 낙엽의 전처리 비교 (Comparison of pretreatment of fallen leaves for application evaluation by Bio-ethanol raw material)

  • 본 연구는 바이오에탄올을 생산하고자 대표적인 임업부산물인 낙엽을 바이오매스로 하여 전처리 과정을 거친 후 효소가수분해 공정을 적용하여 당화반응 특성을 비교 하였다. 전처리 방법은 화학적으로 산(HCl), 알칼리(NaOH, $NH_3$)처리하였고 각각 침지법, 교반법, 고압멸균법을 적용하여 실험하였다. 처리 효율은 효소가수분해 진행 후 포도당 생산량으로 확인하였다. 실험결과, 알칼리 전처리에서는 산 전처리 대비 18% 높은 글루코즈 생산량을 보여 바이오에탄올 생산을 위한 임업폐기물 화학적 전처리 방법으로는 알칼리처리가 효율이 더 높은 것으로 확인되었다. 또한 화학적 처리방법 중 각 방법을 적용하여 비교한 결과, 모든 방법이 알칼리-수산화나트륨 처리하였을 때 가장 높게 나왔다. 본 연구의 결과를 바탕으로 목질계 바이오매스 중 하나인 낙엽을 기질로 전처리 및 효소가수분해 공정의 가능성을 확인하였고 차후 바이오에탄올 생산에도 영향을 미칠 것으로 예상된다.

폴리에스테르섬유의 알칼리욕염색에 관한 연구 -pH변화에 따른 분산염료의 염색성을 중심으로- (A Study on the Dyeing of Polyester Fiber in Alkaline Dyebath ―Dyeing Properties of Disperse dyes According to variation of pH values―)

  • This Study was made to investigate alkaline dyeing systems for a new dyeing applicable polyester fibers. Disperse dyes for dyeing of polyester fibers were C.I. Red 60, Blue 56 and Yellow 54 as three primary colors of E type which used widely on the scene. Dyeing properties of dispers dyes on the polyester fibers are discussed according to variation of pH values for application of alkaline dyeing method compared with to ordinary acidic dyebath. Alkaline pH of the dyebath was controlled to pH 9 and 10.5 with buffer solutions using each hydrochloride and disodiumtetraborate, disodiumtetraborate and sodium hydroxide to promote the reproducibility of dyeing. Dyeing properties of dispers dyes on the polyester fibers by alkaline dyeing method compared with to ordinary acidic dyebath were discussed by estimation of color, wash-fastness, bleeding and migration of dyed polyester fabric.

Effect of Alkaline-Earth Oxide Additives on Flexural Strength of Clay-Based Membrane Supports

  • Low-cost ceramic membrane supports with pore sizes in the range of $0.52-0.62{\mu}m$ were successfully prepared by uniaxial dry compaction method using inexpensive raw materials including kaolin, bentonite, talc, sodium borate, and alkaline-earth oxides in carbonate forms (e.g., $MgCO_3$, $CaCO_3$, and $SrCO_3$). The prepared green supports were sintered at $1000^{\circ}C$ for 8 hr in air. The effect of alkaline-earth oxide additives on the flexural strength of clay-based membrane supports was investigated. The porosity of the clay-based membrane supports was found to be in the range of 33-34%. The flexural strength of the clay-based membrane supports with 1% alkaline-earth carbonates was found to be in the range of 42.8-52.7 MPa. The addition of alkaline-earth carbonates to clay-based membrane supports resulted in large increases (47-80%) in the flexural strength of the membrane supports, compared to that of membrane supports without alkaline-earth carbonates. The typical flexural strength of the clay-based membrane support with 1% $SrCO_3$ was 52.7 MPa at 33.8% porosity.

  • Alkaline .alpha.-amylase expressed in the transformant, Baciollus subtills MB809, containing alkaline amylase gene cloned from alkalophilic Bacillus sp. AL-8, was purified through for step separation processes. The purified alkaline .alpha.-amylase had molecular weight of app[roximately 59, 000 daltons on SDS-PAGE and Sephaex G-100 gel filtration. Amino acid sequence of terminal portion of the enzyme was analyzed with pure amylase eluted form the SDS-PAGE gel. N-terminal amino acid sequence of .alpha.-amylase was determined by the Edman degradation method and resulted in $NH_{2}$-ser-thr-ala-pro-ser-(ile)-lys-ala-gly-thr-(ile)-leu. For C-terminal amino acid sequencing, purified .alpha.-amylase was digested with carboxypuptidase A and B, and reverse-phase HPLC gradient elution system resulted in -thr-trp-pro-lys-COOH.

Antimicrobial efficacies of alkaline disinfectant solution and commercial disinfectants against Brucella ovis

  • Bruella spp. involves a considerable danger of public health and farm animal industry. In this study, we assessed the disinfection efficacy of alkaline disinfectant solution and three commercial farm disinfectants (quaternary ammonium compound, sodium dichloroisocyanurate, potassium monopersulphate/sodium dichloroisocyanurate) against Brucella ovis. A bactericidal efficacy test by broth dilution method was used to determine the lowest effective dilution of selected disinfectants following exposure to test bacteria for 30 minutes at $4^{\circ}C$. Disinfectants and test bacteria are diluted with distilled water (DW), hard water (HW) or organic matter suspension (OM) according to treatment condition. Three commercial disinfectant showed excellent antimicrobial activity (up to dilution of $\times200$ in OM treatment). Alkaline disinfectant solution demonstrated favorable bactericidal efficacy against B. abortus (at dilution of $\times20$ in OM treatment). Three commercial farm disinfectants possess excellent efficacy against B. ovis. Alkaline disinfectant solution has lower potency than commercial farm disinfectant but could help to limit the spread of brucellosis.
