Reaction Characteristics and Kinetic Analysis of Enzymatic Hydrolysis of Corn Gluten Meal Using Alkaline Protease (Alkaline Protease를 이용한 Corn Gluten Meal의 효소가수분해 반응특성 및 반응속도론적 분석)

  • Dry corn gluten meal of 70% protein content was enzymatically hydrolyzed by alkaline protease in a pH-state reactor. Such process variables as temperature, pH, and enzyme-to-substrate ratio were varied, and at each condition degree of hydrolysis was monitored and calculated. The ultimate degree of hydrolysis, which ranged between 25 and 28% based on gluten protein mass, was not significantly affected by the process variables. However, $50^{\circ}C$ and pH 9-10 appeared optimum. Kinetic analysis indicated enzyme deactivation was negligible during the hydrolysis, and the experimental data were near perfectly fitted to the model kinetic equation which was modified after neglecting enzyme deactivation term. The enzyme reaction was 1$100\times$ scaled up and basically the same hydrolysis performance was resulted. Amino acid analysis showed the hydrolyzate was relatively rich in glutamine/glutamic acid, leucine, and alanine at 19.6, 16.1, and 12.3 mole %, respectively.

  • Among the various properties of textile fabrics, the moisture-related properties are important for the textile processes or the apparel comfort characteristics. Alkaline hydrolysis results in pitting on the surface of fibers and increases the amount of hydroxyl and carboxyl end groups of the PET molecules on the fiber surface. The purpose of this study is to investigate the moisture-related properties of PET fabrics treated with quaternary ammonium compound/alkaline solution. The wetting and wicking properties of the PET fabrics were measured using the following experiments: contact angle, surface free energy, work of adhesion, vertical wicking height, moisture regain, and frictional static voltage. It was concluded that by the alkaline hydrolysis process, surface hydrophilicity and reactivity were considerably improved especially at lower levels of weight loss% and that the pitting of the fiber surface resulted in at higher levels of weight loss% was disadvantageous in moisture-related properties of PET.

  • A alkalophilic strain, Bacillus licheniformis MH31 producing an alkaline protease was isolated from mine soil of Boryeong in Korea. Production of a high level of alkaline protease was achieved 42 h after incubation when the bacterium was grown at pH 9.0 and $35^{\circ}C$ in Horikoshi medium supplemented with 0.5%(w/v) starch and 1%(w/v) skim milk as carbon and nitrogen source, respectively. The molecular weight of partially purified enzyme was estimated to be 30 kDa by SDS-PAGE and its optimum pH was pH 10. The enzyme showed optimum temperature at $50^{\circ}C$, and was stable up to $60^{\circ}C$ after 1 h incubation. The protease was strongly inhibited by 1 mM of PMSF which was known well as strong inhibitor of serine proteases, but almost not inhibited by 5 mM of EDTA and 1,10-phenanthroline. When the protein hydrolysis products of 1% skim milk by partially purified protease was compared with available commercial proteases using HPLC analysis, most of hydrolysis products were detected below molecular weight of 10,000 and the hydrolysis ratio of purified enzyme was 24.8% lower than those(above 32%) of commercial proteases.

  • Alkaline treatment gives Sea-Island type yam to produce microfiber and silk-like touch. But this treatment have some problems in dyeing and finishing process. To solve some problem occurred in dyeing and finishing of polyester fabric, the ultrasonic treatment technique was used recently. This study was carried out to confirm the effect of the ultrasonic treatment on alkaline weight loss finishing of polyester fiber under general alkaline treatment conditions; NaOH concentration 2, 3, 4, and 5%, treatment time 5, 10, 15, and 20 minutes, treatment temperature 70, 80, 90, and 99'E, respectively. On the other hand, the three way lay out method was used to test of significant obtained data from alkaline treatment. It was found that weight loss increased with increasing the NaOH concentration, temperature, and time. Also, in case of PET/Co-PET fabrics by ultrasonic, weight loss and dissolution of microfiber were superior to PET/Co-PET fabrics without ultrasonic. Tensile strength and modulus decreased with increasing NaOH concentrations and hydrolysis time. Therefore, the effect of alkali hydrolysis by ultrasonic application was better than that of the conventional method.

  • Algae is an abundant and potential fermentation substrate. The enzymatic hydrolysis of algae was investigated by pre-treating an alkaline hydrogen peroxide with commercial cellulase and viscozyme. Algae used in this study was the Ulva pertusa. The evaluated response was the yield of released glucose after the enzymatic hydrolysis. Alkaline hydrogen peroxide containing mixtures of 1 wt% hydrogen peroxide and 1~1.75 wt% sodium hydroxide was also used. The results show that the highest glucose conversion was obtained for Ulva pertusa using 5 wt% hydrogen peroxide at $60^{\circ}C$ for 3 h. The required amount of enzymes after the pre-treatment with alkaline hydrogen peroxide were reduced by far compared to that of untreated Ulva pertusa. Also, the amount of glucose that is released during the enzymatic hydrolysis was increased.

  • Kinetics and mechanism for alkaline hydrolysis of C. I. Disperse Blue 79(B-79) which is 4-N, N-diacetoxyethyl-2-acylamino-5-ethoxy -2′-bromo-4′,6′-dinitroazobenzene were investigated. The color strength of B-79 in acetone/water solutions of various NaOH concentrations decreased continuously. The hydrolysis rate of B-79 increased with increasing alkali concentration and appeared following first order reaction. The observed rate constants for various concentrations of B-79 showed similar values, and B-79 was hydrolyzed by first order reaction for dye concentration. Therefore, it was confirmed that the overall reaction follow second order kinetics and proceed via S/sub n/2 reaction. From the study on kinetics and spectrometric analysis, it was proposed that the rate determining step of the hydrolysis reaction of B-79 is the nucleophilic substitution reaction - that is the reaction of the rapid attack of $OH^{-}$ on the carbon atom, which is in acceptor ring, adjacent to azo group to break the C-N bond. And it was also found that the final hydrolysis products of B-79 include both the acceptor ring in the form of sodium salt and the donor ring possessing 4-N,N-dihydroxyethyl group converted from 4-N,N-diacetoxyethyl group.

  • Second-order rate constants $(k_{OH^-})$ have been measured spectrophotometrically for alkaline hydrolysis of Y-substituted phenyl phenyl carbonates (2a-j) and compared with the $k_{OH^-}$ values reported previously for the corresponding reactions of Y-substituted phenyl benzoates (1a-j). Carbonates 2a-j are 8~16 times more reactive than benzoates 1a-j. The Hammett plots correlated with $\sigma^-$ and $\sigma^o$ constants exhibit many scattered points, while the Yukawa-Tsuno plot results in excellent linear correlation with $\rho$ = 1.21 and $\gamma$ = 0.33. Thus, the reaction has been concluded to proceed through a concerted mechanism in which expulsion of the leaving group is advanced only a little. However, one cannot exclude a possibility that the current reaction proceeds through a forced concerted mechanism with a highly unstable intermediate.

  • This study is to compare characteristics of saccharification reactions applying to enzymatic hydrolysis of pretreated fallen leaves for bio-ethanol production. It experimented pretreatment of acid, alkaline in the chemical. This experiment includes preteatment of acid and alkaline in chemical, soaking, shaking and autoclaving method, which were applied to biomass. In result, the glucose production from alkaline-NaOH method was 263 mg glucose/ g biomass comparing with them of acid-HCl method. Thus, alkaline-NaOH method is superior than the acid-HCl method for chemical preteatment of fallen leaves. Also, when various chemical treatments were compared, they were all. Based on the results of this study, we found that leaves, one of biomass, are possible in pretreatment and enzymatic hydrolysis process, and they are likely to affect bio-ethanol production in the future.

  • The molecular nature of aspirin hydrolysis was studied using cyclodextrin as a biomimetic model for esterase. Cyclodextrin was selected for this purpose because it meets the necessary requirements for the hydrolysis study, Dissociation constants and catalytic rates were obtained under alkaline conditions by the kinetic method.
