Studies on Development of Antagonistic Microorganism by Cell Fusion - Biological control of disease - ) (세포융합에 의한 신 길항미생물 육종에 관한 연구 - 목초 병해의 생물학적 방제 -)

  • This study was to investigate an effective biological control of forage diseases and provide a basic data and a model in improving variety of antagonistic bacteria, with growth promoting effect on forage, through cell fusion. The results obtained were summarized as follows; 1. The antagonistic himbacterium against soil-borne phathogenic fungi Fusarium oxysporum and Rhizoctonia solani was isolated from continuous cropping himsphere soil of forage, and its biological and physiological characteristics were investigated. This bacterium was identified as Bacillus subrilis and named BS 101. Another strain for cell fusion was Bacillus thur ingiensis ssp. kurstaki HD-I(BT 37669) with insecticidal crystal. 2. The auxotropic mutants of BS 101 and BT 37669 were derived after mutagenesis using N-methyl-N'nitro- Nitrosoguanidine(NTG) to give amino acid requirement marker. n e s e auxotropic mutants of BS 101 and BT 37669 were named BS 1013(his-) and BT 69(asp-), respectively. 3. The best protoplast requirement was obtained using DM 3 medium, containing 5% casamino acid, 1 M $MgCI_2$ and 2% bovine semm albumin, to give Fusant 3, 7 and 8. BT toxin gene was not identified with fusants by Southern blotting. However, SDS-PAGE analysis of strains showed various protein patterns among fusants. 4. From the dark culture experiment, growth of forage in inoculated soil with antagonistic bacteria was delayed than that of non-inoculated soil with antagonistic bacteria in each continuous cropping soil and in each sterilized soil. On the other hand, growth duration of forage was different between continuous cropping soil and sterilized soil. 5. Seed germination of Alfalfa, Italian ryegrass and Orchardgrass were significantly improved by inoculation of antagonistic bacteria(p< 0.05).

Effect of Species and Seed Mixture on Productivity, Botanical Composition and Forage Quality in Middle Mountainous Pasture (중부산간 초지에서 초종 및 혼파조합이 목초 수량, 식생 구성 및 사료가치에 미치는 영향)

  • This study amied to determine the effect of species and seed mixture on productivity, botanical composition and forage quality in middle mountainous region (Geosan) pasture. Total seven experimental pastures (T1 : Orchardgrass, T2 : Timothy, T3 : Tall fescue, T4 : Perennial ryegrass, T5 : Kentucky bluegrass, T6 : Timothy + Orchardgrass + Tall fescue + Alfalfa, T7 : Timothy + Orchardgrass + Kentucky bluegrass + White clover) were established in autumn, 2014 and evaluated for productivity and agronomic characteristics for 2015. Plant heigh was high in Orchardgrass and Perennial ryegrass pasture. Dry matter (DM) content was high in Kentucky bluegrass. Botanical composition in $1^{st}$ harvest was high in shrub, but the grass composition was high in $2^{nd}$ and $3^{rd}$ harvest. Fresh and DM yield were higher in Orchardgrass and Perennial ryegrass (p<0.05); whreras, Kentucky bluegrass was the lowest. In seed mixture pasture, yields of T6 plot (Tall fescue) were higher than those of T7 (Kentucky bluegrass). Average CP (crude protein) content of $2^{nd}$ and $3^{rd}$ cutting time was 16.91 and 14.79%, respectively and Kentucky blue grass was the highest in $3^{rd}$ cutting time by 17.57%. IVDMD (in vitro dry matter digestibility) showed low in $1^{st}$ cutting and Kentucky bluegrass was the lowest in every cutting time. TDN (total digestible nutrient) content was the highest in $3^{rd}$ cutting and the lowest in $1^{st}$ cutting. These results indicated that Orchardgrass, Perennial ryegrass and T6 are recommendable for productivity and Timothy, Perennial ryegrass and T7 are recommendable for forage quality.

Effects of Sample Preparation Methods for the Isolation of Foodborne Pathogens from Sprout Seeds (새싹채소 종자의 전처리 방법이 식중독 세균 검출에 미치는 영향)

  • Sample preparation methods were evaluated for effectiveness in detecting foodborne pathogens from sprout seeds. The methods included: Rinse.-Test portions were rinsed with 0.1% peptone water, and the pellet after centrifugation was inoculated into pre-enrichment media; and Sprouting.-Seed samples were sprouted before pre-enrichment and sprouted seeds were inoculated into pre-enrichment media. In rinse method, E. coli was isolated from 13 of 280 sample units. In sprouting method, E. coli was isolated from 12 of 135 sample units. E. coli O157:H7, Salmonella spp., and L. monocytogenes were not detected in any of the samples. In the trials for recovering Salmonella enterica from artificially contaminated alfalfa seeds, the soak, rinse, and sprouting methods were evaluated. The detection rates of S. enterica were statistically different according to the amount of the sample tested and selective medium type (P < 0.05).
