Helianthus tuberosus has been known to inhibit the growth of weeds and other plants sharing its habitat. This study was conducted to identify the allelochemicals of Helianthus tuberosus which were extracted with water and solvents. Aqueous extracts of leaf, stem, root, tuber and tuber peel of Helianthus tuberosus except tuber did not show significant differences in phytotoxicity to alfalfa seedlings. It was considered that Helianthus tuberosus contained fewer or less potential water-soluble substances that were toxic to alfalfa. Methanol extract of leaves of Helianthus tuberosus was sequentially partitioned in increasing polarity with n-hexane, ethylacetate and n-butanol. Each extract had a yield of 148, 12, 15.7 and 9.5g, respectively. Inhibitory effects on germination of alfalfa seeds treated with four fractions were not significantly different. But the significant reductions on hypocotyl length were observed for all the solvent extracts. Among the four fractions, the ethylacetate fraction showed the most significant inhibition effect on bioassay with alfalfa. Further separation of the active ethylacetate fraction by open column chromatography led to the 25 subfractions. In bioassay of each sub-fraction with alfalfa seeds, sub-fraction No. 13 showed the most inhibitory effect on seedling growth. $^1$H NMR and gas chromatography-mass spectrometry analysis revealed that sub-fraction No. 13 was the mixture of straight-chain saturated fatty acids.
Greenhouse and laboratory studies were conducted to investigate allelopathic potential of some weed species on alfalfa(Medicago sativa L.) germination and seedling growth. In the comparison between top(leaves+stems) and root extracts, top extract exhibited greater allelopathic effects on alfalfa germination than that of root. The various weed species extract differently responded to alfalfa test species, WL-320, in terms of allelopathic effect. Top and root aqueous extracts of lambsquarter(Chenopodium album L.), giant foxtail(Setaria faberii Herrm.), redroot pigweed (Amaranthus retroflexus L.), velvetleaf(Abutilon theophrasti Medic.), crabgrass(Digitaria sanguinalis L.), canada thistle(Cirsium arvense L.) and prostrate knotweed(Polygonium aviculare L.) significantly inhibited germination, seedling length, weight, vigor, and rate of germination of alfalfa. The regression slopes of various top extracts showed that velvetleaf(b=3.69) extracts were the most inhibitory, while large crabgrass(b=2.39) extracts had the least allelopathic effect on alfalfa germination. Germination, seedling length and weight of alfalfa were inversely proportional to the concentration of dried velvetleaf extracts. Also, more of the toxic effects were observed from the dried extracts compared to the fresh extracts. Residue of velvetleaf inhibited significantly alfalfa emergence and survival percentage compared to the control. The emergence and survival percentage of alfalfa were 44%, 57% at 1.0% residue treatment, respectively. When weed residues were mixed with silica sand with incubation time, velvetleaf residue most inhibited alfalfa growth. The degree of inhibition increased as incubation time increased. An incubation for 72h caused the greatest inhibition of alfalfa growth. These results demonstrate the different allelopathic activity of weed species extracts on alfalfa and suggest that weed may affect alfalfa growth and development through the inhibitory effects of allelochemicals present in weed tissue.
To isolate, purity and identify of bioactive compounds involved in alfalfa allelopathy and/or autotoxicity, experiment was conducted. Isolation and separation procedures used from an 80% methanol extract of fresh alfalfa leaves(1kg), silica gel thin layer chromatography(TLC), followed by Droplet Counter Current Chromatography(DCCC). Preliminary identification was examined by high preformance lipid chromatography(HPLC). Four phenolic compound, salicylic acid, scopoletin, rutin, and quercetin, were identified and identified all compounds were phytotoxic to alfalfa seed germination and seedling growth. Among these compounds, quercetin treatment($10^{-3}M$) was most inhibitory to alfalfa seed germination and seedling growth. These compounds may be, at least in part, involved autotoxicity and allelopathy.
Autotoxicity restricts reseeding of new alfalfa(Medicago sativa L.) after alfalfa until autotoxic chemical(s) breaks down or is dispersed into external environments, often requiring up to a year or more. One solution for reducing autotoxicity would be to select germplasms or cultivars with tolerance to the autotoxic chemical(5) and use genetically breeding program. Bioassay of seed germination and early seedling growth was conducted to evaluate autotoxic responses of 3 varieties of alfalfa to the water-soluble extracts(at 4 and 8g/L) from alfalfa ‘Cody’leaf by using agar and filter paper medium in a petri-dish assay. Root length at 5 days after seeding was more sensitive to the extract than was hypocotyl length or seed germination, and was a better parameter of autotoxic effects of alfalfa leaf extracts. Use of an agar medium gave better sensitivity of root length than did use of filter paper. Evaluating tolerance with percent of control was more important indicator than was mean of root length because of significant variation among varieties in root length of control treatment. Bioassay ranked varieties in the following order of tolerance on the basis of relative root length; “Cody” >“ Pioneer 5373” >“ Alfagraze”. Seedling growth from old “Cody” seed was more sensitive to the autotoxic chemical(5) than was that from newly produced seed.
This study was carried to develop the new material of functional food which antioxidant of natural substances of sprouts. We compared antioxdiative activity and antioxidant substances exist in sprouts. Antioxidaive activities of sprouts were measured by total polyphenolic acid contents, electron donating activity(EDA), SOD-liked activity and hydroxyl radical scavenging ability. Total polyphenolic acid content was very higher in Rape sprouts extract(Rap) than other sprout extracts. Also Rape extract was showed the most excellent antioxdiative activity in SOD liked activity at 86.94%. The EDA was ordered Rape sprouts extract(Rap)>Tatsoi sprouts extract(Tat)>Broccoli sprouts extract(Bro)>Alfalfa sprout extract(Alf). Hydroxy radical scavenging ability was the most effective in Rape sprouts extract(Rap). Therefore we could be certain that Rape sprouts extract(Rap) was the most effective in antioxidative activity from sprouts.

The evaluation of structural carbohydrate losses and its effect on silages digestibility in alfalfa (Medicago sativa L.) and orchardgrass (Dactylis glomerata L.) was studied during 5, 21 and 56 days ensiling. About 70 and 60 kg fresh matter of the two forages were ensiled in 9 silos of 120 L capacity. The digestion trials were conducted in two phases using the two grasses in two $4{\times}4$ Latin square design according to the four treatments being the grass and the three silages. There were no differences in the DM and CP contents resulting from 5 to 56 days ensiling in both forages. The water-soluble carbohydrates (WSC), hemicelllose, pectin, and energy were slightly reduced and appeared lower in 56 days silage. The ether extract and cellulose contents slightly increased as the ensiling process advanced in the two species. Hemicellulose losses of 29 and 41 g/kg DM were obtained in alfalfa and orchardgrass, respectively, 56 days after ensiling. While the cellulose losses in both species were very little, compared to that for hemicellulose, the pectin losses, 56 days after ensiling were 15 and 12 g/kg DM in alfalfa and orchardgras respectively. The total structural carbohydrates lost (ie., hemicellulose + cellulose + pectin) in g/kg DM of fresh material forage ensiled, is about four fifths the amount lost by WSC, in alfalfa and about two thirds, in orchardgrass, by 21 days ensiling after the activity of microorganism terminated, indicating that appreciable amount was used as substrate for silage fermentation. Ensiling alfalfa and orchardgrass for 0, 5, 21 and 56 days maintained a decreasing trend of 83.8, 82.5, 79.3 and 78.9% digestibility in alfalfa and 80.5, 77.0, 77.1 and 76.4% digestibility in orchardgrass. While the digestibility of cellulose and ether extract increased in silage in both species, the digestible energy values in silage were reduced from 2.6 to 2.3 and 2.9 to 2.7 Mcal/kg DM respectively in alfalfa and orchard during 5-56 days ensiling.
Objective: The effect of flavonoids from alfalfa on the microbial flora was determined using molecular techniques of 16S ribosome deoxyribonucleic acid (rDNA) analysis. Methods: Four primiparous Holstein heifers fitted with ruminal cannulas were used in a $4{\times}4$ Latin square design and fed a total mixed ration to which alfalfa flavonoids extract (AFE) was added at the rates of 0 (A, control), 20 (B), 60 (C), or 100 (D) mg per kg of heifer BW. Results: The number of operational taxonomic units in heifers given higher levels of flavonoid extract (C and D) was higher than for the two other treatments. The Shannon, Ace, and Chao indices for treatment C were significantly higher than for the other treatments (p<0.05). The number of phyla and genera increased linearly with increasing dietary supplementation of AFE (p<0.05). The principal co-ordinates analysis plot showed substantial differences in the microbial flora for the four treatments. The microbial flora in treatment A was similar to that in B, C, and D were similar by the weighted analysis. The richness of Tenericutes at the phylum level tended to increase with increasing AFE (p = 0.10). The proportion of Euryarchaeota at the phylum level increased linearly, whereas the proportion of Fusobacteria decreased linearly with increasing AFE supplementation (p = 0.04). The percentage of Mogibacterium, Pyramidobacter, and Asteroleplasma at the genus level decreased linearly with increasing AFE (p<0.05). This study was carried out to investigate the amounts of voluntary intake, digestibility and nitrogen retention in Korean native goats (KNG) fed agricultural by-products containing rice straw (RS) and apple pomace (AP) and to obtain a basic information for establishing the feeding system of KNG. The result are as follows. 1. Among the chemical composition of experimental diets, the highest values in crude protein (CP ; 18.6%) and crude ash contents (10.7%) were observed in alfalfa hay. Those of RS+AP treatment were significantly low 6.0 and 4.8%, respecitively. Acid detergent fiber (ADF), ether extract and non-structural carbohydrate (NSC) contents were shown an adverse tendency. 2. Dry matter (DM) intakes per day in KNG fed RS+commercial diet (CD), RS+AP and RS+AP+CD were significantly lower (P<0.05) 210.3, 228.3, respectively than 358.1g in alfalfa hay. 3. DM intakes per basal weight expressed as DM g/kg of BW0.75 and DM g/kg of BW(%) were highest (P<0.05) 60.5g and 3.3%, respectively in KNG fed alfalfa hay, any other treatments showed 35.6 to 42.5g and 2.0 to 2.3%, respectively and this result was similar to those of DM intake per day. 4. Digestibilities of DM, organic matter, CP, ADF, neutral detergent fiber (NDF). crude ash and ether extract in alfalfa hay and RS+AP+CD treatment were significantly higher (P<0.05) than RS+CD treatment. Those of CP, ADF and NDF of RS+AP treatment in not significantly different with these treatments. 5. NSC digestibility was higher in alfalfa hay (66.7%) than those of other treatments (44.2~52.0% ; P<0.05). 6. Nitrogen retentions expressed as N retention(g) and N retention (%) in KNG were highest (P<0.05) 0.6g and 15.6%, respectively in RS+AP+CD treatment and RS+CD treatment was lowest (P<0.05) -06g and -21.4%, respectively.
The effect of high and low level of feed intakes on nutrient digestibility, nutrient losses through methane, energy and protein utilization by goats fed on alfalfa (Medicago sativa L.) pellets based diets was investigated in this study. Twelve castrated Japanese goats were employed in two subsequent digestion and metabolism trials. The goats were divided into three groups, offered three diets. Diet 1 consisted of 100% alfalfa pellet, Diet 2 was 70% alfalfa pellet and 30% corn, and Diet 3 was 40% alfalfa pellet and 60% corn. The two intake levels were high (1.6 times) and low (0.9 times) the maintenance requirement of total digestible nutrients (TON). Rumen ammonia nitrogen ($NH_3$-N) level of Diet 1 was lower (p<0.001) compared to Diets 2 and 3, but the values were always above the critical level (I50 mg/liter), The pH values of rumen liquor ranged from 6.02 to 7.30. Apparent digestibility of nutrient components did not show differences (p>0.05) between the two intake levels but inclusion of corn significantly altered the nutrient digestibility. Diet 3 had highest (p<0.001) dry matter (DM), organic matter (OM), ether extract (EE) and nitrogen fee extract (NFE) digestibility followed by the Diet 2 and Diet 1. The crude protein (CP) digestibility values among the three diets were in a narrow range (70.1 to 70.8%). Crude fiber (CF) digestibility for Diet 3 was slight higher (p>0.05) than that for other two diets. When alfalfa was replaced by corn, there were highly significant (p<0.001) increases in DM, OM, EE and NFE apparent digestibility and a slight increase in the CF digestibility (p>0.05). There were no differences (p>0.05) in energy losses as methane ($CH_4$) and heat production among the diets but energy loss through urine was higher for the Diet 1. The total energy loss as $CH_4$ and heat production were higher for the high intake level but the energy loss as $CH_4$ per gram DM intake were same (0.305 kcal/g) between the high and low intake level. Retained energy (RE) was higher for Diet 3 and Diet 2. Nitrogen (N) losses through feces and urine were higher (p<0.001) for Diet 1. Consequently, N retention was lower (p>0.05) for Diet 1 and higher in Diets 3 and 2. It is concluded that inclusion of corn with alfalfa increased the metabolizable energy (ME) and RE, and retained N through reducing the energy and N losses. The high level of intake reduced the rate of nutrient losses through feces and urine.

To find out the optimum mixing level of wheat bran to improve the quality of 1st cut Alfalfa silage, this trial was canied out at two-year's Alfalfa monoculture field of the National Livestock Research Institute, Suwon, Korea, from May 1955 to Oct. 1995. The results are as follows. I. According to the increase of wheat bran mixing level, DM content was high from 23.19%(nil) to 35.70%(40% mixing), pH of silage were low from 5.51(nil) to 4.45(40% mixing). but there were not significant at 20% mixing or more(P>0.05). 2. Also it was appeared to the same trend at Lactic acid content and in the Flieg's score of silage it was highest as 59 in the 20% mixing level. 3. In chemical component of silages there were not significant at 20% mixing or more in crude protein, crude fiber and ashes content, 30% mixing or more in ether extract and 10% or less in NFE content(P>0.05). 4. TDN, NEL and StE contents were low as adding level was increased. respectively, but there were not significant(P>O.OS). As mentioned above the results, desirable mixing level of wheat bran to lstcut Alfalfa to improve the quality of silage was 20% of fresh weight.
