Cattle feeding in South Korea has been heavily dependent on domestically produced rice straw and imported grain. Around 42% of domestically produced rice straw is utilized for forage, and the remainder is recycled to restore soil fertility. Approximately 35% of round baleages were made with rice straw. However, higher quality hay is desired over rice straw. Due to increasing stockpiles of rice, there has been an economic burden on the government to store the surplus; therefore production of annual forage crops in rice fields has been further promoted in recent years. Hay import from the USA currently constitutes more than 80% of total imported hays. The main imported hays are alfalfa (Medicago sativa), timothy (Phleum pretense), and tall fescue (Festica arundinacea). The estimated forage required for cattle feeding was approximately 5.4 million MT in 2016. Domestically produced forage sates only 43% of that value, while low quality rice straw and imported hay covered the rest of demand by 33% and 20%, respectively. As utilization of domestically produced forage is more desirable for forage-based cattle production, long-term strategies have been necessary to promote domestic production of high quality baleage. One such strategy has been utilizing the fertile soil and abundance of fallow rice fields of western region of S. Korea to produce forage crops. Italian ryegrass (Lolium multiflorum) is the most successfully produced winter annual in the region and is approximately 56% of the total winter annual forage production. Forage sorghums (Sorghum bicolor), sorghum × sudangrass hybrids, and hybrid corn (Zea mays) produce a substantial amount of warm-season forage during summer. Produced forage has been largely stored through baleage due to heavy dew and frequent rains and has been evaluated according to S. Korea's newly implemented baleage commodity evaluation system. The system weighs 50% of its total grading points on moisture content because of its importance in deliverable DM content and desirable baleage fermentation; this has proved to be an effective method. Although further improvement is required for the future of forage production in South Korea, the current government-led forage production in rice fields has been able to alleviate some of the country's shortage for quality hay.

Limiting amino acids (AAs) for milk production in dairy cows fed on a concentrate diet of corn grain and soybean meal was evaluated in this study. Four lactating and multiparous Holstein cows (in third or fourth parities, with an average body weight of 633 ± 49.2 kg), 8 to 9 weeks into their lactation period, were used in a 4 × 4 Latin square design. The experiment comprised four dietary treatments: (1) no intravenous infusion (control); (2) control plus intravenous infusion of an AA mixture of 6 g/d methionine, 19.1 g/d lysine, 13.8 g/d isoleucine, and 15.4 g/d valine (4AA); (3) control plus intravenous infusion of the AA mixture without methionine (no-Met); and (4) control plus intravenous infusion of the AA mixture without lysine (no-Lys). All animals were fed on a controlled diet (1 kg/d alfalfa hay, 10 kg/d silage, 14 kg/d concentrate mixture, ad libitum timothy hay). The AA composition of the diet and blood were determined using an automatic AA analyzer. Milk composition (protein, fat, lactose, urea nitrogen, and somatic cell counts) was determined using a MilkoScan. The results showed that feed intake for milk production did not differ from that of intravenous infusion using a limiting AA mixture. The 4AA treatment numerically had the highest milk yield (32.4 kg/d), although there was no difference when compared with the control (31.2 kg/d), no-Met (31.3 kg/d), and no-Lys (31.7 kg/d) treatments. The concentration of AAs in blood plasma of cows in all treatments, mainly isoleucine and valine, increased significantly compared with that of control. The no-Met treatment increased (p < 0.05) the concentration of lysine in the blood relative to the control and no-Lys treatments, whereas the no-Lys treatment increased (p < 0.05) the concentration of methionine relative to the control and no-Met treatments. In conclusion, milk production increased when feeding 10 g/d methionine to the cows, together with their concentrate diet of corn grain and soybean meal.
A comparison was made of the effect of different packaging materials on storability of mixed sprout vegetables (alfalfa, broccoli, cabbage, radish, and red radish) stored at $8^{\circ}C$. They were packaged by several kinds of films that were 50 fm thickness low-density polyethylene film (PE 50), 50 fm thickness polypropylene film (PP 50), $50{\mu}m$ thickness ceramic film (CE 50), $25{\mu}m$ thickness ceramic film (CE 25), $10\sim13{\mu}m$ thickness polyethylene film (wrap), and polyethylene terephthalate box with ventilation hole (box). The fresh weight of mixed sprouts packaged with different materials decreased below to 2% except box packages that decreased around 7% compared with initial weight. The atmospheres that were developed inside the different materials during storage differed significantly. The carbon dioxide and oxygen concentration of packages were kept properly with 5% in CE 25, but the PE 50 and CE 50 treatments showed higher carbon dioxide and lower oxygen concentration. So the off-flavor of mixed sprouts was more severe in the PE 50 and CE 50 treatment. The ethylene concentration of packages showed lowest in box treatment, followed by PP 50, wrap and CE 25 treatment. The overall quality of mixed sprouts was lower in PE 50 and CE 50 than CE 25 treatment after 10 days storage, supposed to be resulted from highest ethylene concentration and lowest oxygen concentration. Conclusionally, the CE 25 film that showed below 1% fresh weight loss, 5% carbon dioxide, 5% oxygen and below 4 ppm ethylene concentration in package may be a proper packaging material for mixed sprout vegetables.

Sample preparation methods were evaluated for effectiveness in detecting foodborne pathogens from sprout seeds. The methods included: Rinse.-Test portions were rinsed with 0.1% peptone water, and the pellet after centrifugation was inoculated into pre-enrichment media; and Sprouting.-Seed samples were sprouted before pre-enrichment and sprouted seeds were inoculated into pre-enrichment media. In rinse method, E. coli was isolated from 13 of 280 sample units. In sprouting method, E. coli was isolated from 12 of 135 sample units. E. coli O157:H7, Salmonella spp., and L. monocytogenes were not detected in any of the samples. In the trials for recovering Salmonella enterica from artificially contaminated alfalfa seeds, the soak, rinse, and sprouting methods were evaluated. The detection rates of S. enterica were statistically different according to the amount of the sample tested and selective medium type (P < 0.05).

This study was conducted to compare the methane ($CH_4$) production estimated by in vivo (sulfur hexafluoride tracer technique ($SF_6$)) with that of two in vitro rumen simulation (RUSITEC) and gas production (IVGPT)) techniques. Four adult dry Holstein cows, aged $7.4{\pm}3.0$ years and weighing $697{\pm}70$ kg, were used for measuring methane production from five diets by the $SF_6$ technique. The experimental diets were alfalfa hay ($D_1$), corn silage + soybean meal (SBM) (910: 90, $D_2$), Italian rye grass hay +SBM (920: 80, $D_3$), rice straw +SBM (910: 90, $D_4$) and Sudan grass hay +SBM (920: 80, $D_5$). Each diet was individually fed to all 4 cows and 5 feeding studies of 17 d each were conducted to measure the methane production. In the RUSITEC, methane production was measured from triplicate vessels for each diet .In vitro gas production was measured for each of the diets in triplicate syringes. The gas produced after 24 and 48 h was recorded and gas samples were collected in vacuum vials and the methane production was calculated after correction for standard temperature and pressure (STP). Compared to the $SF_6$ technique, estimates of methane production using the RUSITEC were lower for all diets. Methane production estimated from 24 h in vitro gas production was higher (p<0.001) on $D_1$ as compared to that measured by $SF_6$, whereas on $D_2$ to $D_5$ it was lower. Compared to $SF_6$, methane production estimated from 48 h in vitro gas production was higher on all diets. However, methane estimated from the mean of the two measurement intervals (24+48 h/2) in IVGPT was very close to that of $SF_6$ (correlation 0.98), except on $D_1$. The results of our study confirmed that IVGPT is reflective of in vivo conditions, so that it could be used to generate a database on methane production potential of various ruminant diets and to examine strategies to modify methane emissions by ruminants.

This study was conducted to investigate changes in blood enzyme parameters and to evaluate the relationship between insulin-like growth factor-1 (IGF-1), antler growth and body weight during the antler growth of sika deer (Cervus nippon). Serum enzyme activity and IGF-1 concentrations were measured in blood samples collected from the jugular and femoral veins at regular intervals during the antler growth period. Blood samples were taken in the morning from fasted stags (n = 12) which were healthy and showed no clinical signs of disease. Alfalfa was available ad libitum and concentrates were given at 1% of body weight to all stags. The experimental diet was provided at 9 am with water available at all times. There were no significant differences in alkaline phosphatase, aspartate aminotransferase, and alanine aminotransferase during antler growth, but alkaline phosphatase concentrations increased with antler growth progression, and the highest alkaline phosphatase concentration was obtained 55 days after antler casting. Serum IGF-1 concentrations measured from blood samples taken from the jugular vein during antler growth, determined that levels of IGF-1 was associated with body weight and antler growth patterns. Serum IGF-1 concentrations were higher at the antler cutting date than other sampling dates. Antler length increased significantly during antler growth (p<0.001), and there was a similar trend to between right and left beams. Body weight increased with antler growth but was not significant. Consequently it appeared that serum alkaline phosphatase concentration was related to antler growth and both antler growth and body weight were associated positively with IGF-1 concentrations during antler growth.
Heat production in ruminants follows a diurnal pattern over the course of a day peaking 3 hours following afternoon feeding and then gradually declining to its lowest point prior to morning feeding. In order to clarify the cooling period most effective in reducing decreases in feed intake and milk production, experiments were carried out based on the diurnal rhythm of heat production and heat dissipation. In experiment 1, the effects of hot environment on milk production were investigated. The animals were kept first in a thermoneutral environment ($20.0^{\circ}C$, 80.0%) for 12 days, they were then transitioned to a hot environment ($32^{\circ}C$, 80.0%) for 13 days before being returned to second thermoneutral environment for a further 12 days. In experiment 2, the effectiveness of daytime cooling or nighttime cooling for improving milk production in hot environment was compared. While ten lactating Japanese Saanen goats (aged 2 years, weighing 41.0 kg) during early lactation were used in experiment 1, ten lactating goats (aged 2 years, weighing 47.5 kg) during mid-lactation were used in experiment 2. The animals were fed 300 g of concentrated feed and excessive amounts of crushed alfalfa hay cubes twice daily. Water was given ad libitum. The animals were milked twice daily. When exposed to a hot environment, milk yield and composition decreased significantly (p<0.05). Milk yield in the hot environment did not change with daytime cooling, but tended to increase with nighttime cooling. Compared to the daytime cooling, milk components percentages in the nighttime cooling were not significantly different but the milk components yields in the nighttime cooling were significantly higher (p<0.05). The results indicate that nighttime cooling is more effective than daytime cooling in the reduction of milk production declines in lactating goats exposed to a hot environment.

In goats fed on dry forage twice a day, an esophageal fistula was used to investigate the physiological factors present in the marked suppression of dry forage intake that occurs after 40 min of feeding. The animals used in this study were five large-type male esophageal- and ruminal-fistulated goats. Roughly crushed alfalfa hay cubes with any large remaining chunks removed were used as feed for this research. The study was conducted under both normal feeding conditions (NFC) and sham feeding conditions (SFC). In the NFC control, the esophageal fistulae were closed by plugs and the animals ate dry forage in the normal manner. In the SFC treatment, before starting the experiment the plugs for closing the esophageal fistula were removed and the cannulae for collecting boluses were fitted into the fistulae. Therefore, the esophageal boluses were removed via an esophageal fistula before they entered the rumen. In the NFC control, eating rates sharply decreased in the first 40 min of feeding and were subsequently maintained at low levels. However, eating rates in the SFC treatment remained high after 40 min of the feeding period had elapsed and the goats ate continuously during the 2 h feeding period. In comparison with the NFC control ($1,794{\pm}203.80\;g$/2 h), cumulative dry forage intake in the SFC treatment ($3,182{\pm}381.69\;g$/2 h) was 77.4% greater (p<0.05) upon conclusion of the 2 h feeding period. In the SFC treatment, cumulative bolus output ($6,804{\pm}469.92\;g$/2 h) was about twofold the cumulative dry forage intake due to cumulative salivary secretion volume ($3,622{\pm}104.13\;g$/2 h) upon conclusion of the 2 h feeding period. The result indicates that large amounts of secreted saliva during dry forage feeding act in conjunction with consumed feed to form the ruminal load responsible for ruminal distension. The increased plasma total protein concentrations were higher in the SFC treatment than in the NFC control. However, plasma and ruminal fluid osmolalities increased in the NFC control during and after feeding but were mostly unchanged in the SFC treatment. In comparison with the NFC control ($3,440{\pm}548.04\;g$/30 min), thirst level in the SFC treatment ($1,360{\pm}467.02\;g$/30 min) was 60.5% significantly less (p<0.05) upon conclusion of the 30 min drinking period. The results of the present study indicate that In the second hour of the 2 h feeding period, dry forage intake is regulated by factors produced when boluses enter the rumen.
The effects of exogenous fibrolytic enzymes (EFE; a mixture of two preparations from Trichoderma spp., with predominant xylanase and ${\beta}$-glucanase activities, respectively) on colonization and digestion of ground barley straw and alfalfa hay by Fibrobacter succinogenes S85 and Ruminococcus flavefaciens FD1 were studied in vitro. The two levels (28 and 280 ${\mu}g$/ml) of EFE tested and both bacteria were effective at digesting NDF of hay and straw. With both substrates, more NDF hydrolysis (p<0.01) was achieved with EFE alone at 280 than at 28 ${\mu}g$/ml. A synergistic effect (p<0.01) of F. succinogenes S85 and EFE on straw digestion was observed at 28 but not 280 ${\mu}g$/ml of EFE. Strain R. flavefaciens FD1 digested more (p<0.01) hay and straw with higher EFE than with lower or no EFE, but the effect was additive rather than synergistic. Included in the incubation medium, EFE showed potential to improve fibre digestion by cellulolytic ruminal bacteria. In a second batch culture experiment using mixed rumen microbes, DM disappearance (DMD), gas production and incorporation of $^{15}N$ into particle-associated microbial N ($^{15}N$-PAMN) were higher (p<0.001) with ammoniated (5% w/w; AS) than with native (S) ground barley straw. Application of EFE to the straws increased (p<0.001) DMD and gas production at 4 and 12 h, but not at 48 h of the incubation. EFE applied onto S increased (p<0.01) $^{15}N$-PAMN at 4 h only, but EFE on AS increased (p<0.001) $^{15}N$-PAMN at all time points. Prehydrolysis increased (p<0.01) DMD from both S and AS at 4 and 12 h, but reduced (p<0.01) $^{15}N$-PAMN in the early stage (4 h) of the incubation, as compared to non-prehydrolyzed samples. Application of EFE to barley straw increased rumen bacterial colonization of the substrate, but excessive hydrolytic action of EFE prior to incubation decreased it.

The involvement of the cell-wall degrading enzymes in Rhizobium has long been an unsolved question about the infection process in the formation of root nodule. To assess the contribution of the cellulase to the nodulation of rhzobia, here we report the production of cellulase from R. meliloti TAL1372 which degrade carboxymethylcellulose (CMC) model substrate with CMC-plate method. We constructed a genomic library by cloning Sau3A-digested genomic DNA from R. meliloti TAL1372 into the BamHI site of the cosmid vector pLAFR3. Out of more than one thousand transductants of E. coli, one clone (pRC8-71) had CM-cellulase activity and contained pLAFR3 cosmid with 30 kb insert of R. meliloti DNA The product of CM-cellulase gene was analyzed by native PAGE. About 45 kD protein was considered to be a product of the gene. Tn5 mutagenesis reveals that the structural gene located in a ca. 3 kb KpnI fragment. The cellulase-minus mutants of R. meliloti TAL1372 were obtained by Tn5 mutagenesis of pRC8-71 and marker exchange techniques. Analyses of the nodulation ability of these Tn5 mutants showed that the CM-cellulase gene of R. meliloti TAL1372 may be involved in early nodulation development on alfalfa (Medicago satiua).
