Acidic soil significantly reduces crop productivity mainly due to aluminum (Al) toxicity. Alfalfa (Medicago sativa L.) roots were exposed to aluminum stress (Al3+) in calcium chloride (CaCl2) solution (pH4.5) and root growth, physiological and antioxidant enzyme responses were investigated. The root growth (length) was significantly inhibited after 48 h of aluminum stress imposition. Histochemical staining with hematoxylin indicated significant accumulation of aluminum in Al stress-treated root tissues. Histochemical assay were also performed to detect superoxide anion, hydrogen peroxide and lipid peroxidation, which were found to be more in root tissues treated with higher aluminum concentrations. The enzymatic activity of CAT, POD and GR in root tissues was slightly increased after Al stress treatment. The result suggests that Al stress alters root growth in alfalfa and induces reactive oxygen species (ROS) production, and demonstrates that antioxidant enzymes involved in detoxification of Al-mediated oxidative stress.
To understand the adaptability of alfalfa (Medicago sativa L.) to chilling stress, we analyzed the antioxidative mechanism during seed germination. The germination rates of six alfalfa cultivars were studied comparatively at $10^{\circ}C$. Xinmu No. 1 and Northstar were selected as chilling stress-tolerant and stress-sensitive cultivars for further characterization. After chilling treatment, Xinmu No. 1 showed higher seedling growth than Northstar. Xinmu No. 1 exhibited low levels of hydrogen peroxide and lipid peroxidation compared with Northstar. In addition, shoots in Xinmu No. 1 treated with chilling showed higher activities of the superoxide dismutase, ascorbate peroxidase (APX), and catalase than those of Northstar, whereas Xinmu No. 1 showed higher APX activity in roots that Northstar. These results indicated that high antioxidation activity in Xinmu No. 1 under chilling stress is well associated with tolerance to chilling condition during germination.
This study was carried out to improve utilization of fly ash. Each animal waste was mixed with fly ash and composted This compost used at forage crops with corn, rye and alfalfa to examine to examine the fertilized efficiency and investigated productivity of forage crops, composition of this copmost and effect of fly ash on soil characteristics and composition. Content of organic matte, P2O5, K2O, CaO, MgO, Mn and B at the soil, which is given fly ash, increased. After the test crops were harvested, pH of the soil was maintained about 7 and contents of organic matter, phosphoric aicd, K, Mg, and B was increased at the soil of used fly ash. As fly ash was mixed, each DM yield of corn and rye was increased 10∼13% and 14∼21% especially alfalfa was increased 35% at the soil which is mixed fly ash with cage layer manure. As fly ash was mixed, each Crude protein (CP) of corn and rye was increased 6∼17% and about 29%, especially, as fly and cage layer manure was mixed CP of alfalfa was increased 33%. In conclusion, as fly ash is mixed with anlmal waste and use at forage crops, It makes the soil good and improve the productivity of forage crops.
Iron (Fe) is a vital element for plants and other organisms, involving in several physiological processes including respiration, chlorophyll biosynthesis, and photosynthesis. Unfortunately, how Fe accumulation regulates in response to light quality has not been well established in plants. Therefore, the aim of the study was to explore the mechanism of Fe homeostasis by light quality. In this study, we found morpho-physiological attributes were significantly improved in response to blue (λmax: 450) compared to white (λmax: 500) and red (λmax: 660) light. The root-shoot length, plant biomass, photosynthesis efficiency (Fv/Fm) and leafgreen (SPAD) significantly declined in response to white and red light. However, these parameters were improved and iron deficiency was substantially alleviated by blue light exposure in alfalfa seedlings. This study might be useful to the forage breeders and farmers for improving alfalfa yield and nutritional benefits.
An experiment was conducted to determine the effects of diets containing coarse-texture rice straw and small particle size alfalfa pellets as a part of total mixed ration (TMR) on milk productivity and chewing activity in lactating dairy cows. Sixteen multiparous Holstein dairy cows ($670{\pm}21kg$ body weight) in mid-lactation ($194.1{\pm}13.6$ days in milk) were randomly assigned to TMR containing 50% of timothy hay (TH) or TMR containing 20% of rice straw and 30% of alfalfa pellet mixture (RSAP). Geometric mean lengths of TH and RSAP were found to be 5.8 and 3.6, respectively. Dry matter intake, milk yield and milk composition were measured. Moreover, eating and ruminating times were recorded continuously using infrared digital camcorders. Milk yield and milk composition were not detected to have significant differences between TH and RSAP. Dry matter intake (DMI) did not significantly differ for cows fed with TH or RSAP. Although particle size of TH was larger than RSAP, eating, ruminating and total chewing time (min/d or min/kg of DMI) on TH and RSAP were similar. Taken together, our results suggest that using a proper amount of coarse-texture rice straw with high value nutritive alfalfa pellets may stimulate chewing activity in dairy cows without decreasing milk yield and composition even though the quantity of rice straw was 40% of TH.

Objective: To assess the potency of calcium propionate (CAP) used as silage additive, an experiment was carried out to evaluate the effect of CAP on the nitrogen transformation, fermentation quality and aerobic stability of alfalfa silages. Methods: Alfalfa was ensiled with four levels of CAP (5, 10, 15, and 20 g/kg of fresh weight [FW]) in laboratory silos for 30 days. After opening, the silages were analyzed for the chemical and microbiological characteristics, and subjected to an aerobic stability test. Results: The increasing proportion of CAP did not affect pH, lactic acid (LA) concentrations and yeast counts, while linearly decreased counts of enterobacteria (p = 0.029), molds (p<0.001) and clostridia (p<0.001), and concentrations of acetic acid (p<0.001), propionic acid (p<0.001), butyric acid (p<0.001), and ethanol (p = 0.007), and quadratically (p = 0.001) increased lactic acid bacteria counts. With increasing the proportion of CAP, the dry matter (DM) loss (p<0.001), free amino acid N (p<0.001), ammonia N (p = 0.004), and non-protein N (p<0.001) contents were linearly reduced, whereas DM (p = 0.048), water soluble carbohydrate (p<0.001) and peptide N (p<0.001) contents were linearly increased. The highest Flieg's point was found in CAP10 (75.9), represented the best fermentation quality. All silages treated with CAP improved aerobic stability as indicated by increased stable hours compared with control. Conclusion: The addition of CAP can suppress the undesirable microorganisms during ensiling and exposure to air, thereby improving the fermentation quality and aerobic stability as well as retarding the proteolysis of alfalfa silage. It is suggested that CAP used as an additive is recommended at a level of 10 g/kg FW.
Cadmium (Cd) toxicity is a serious limitation for agricultural production. In this study, we explored tolerance mechanism associated with Cd toxicity tolerance in alfalfa plants. We used three distinct alfalfa cultivars M. sativa cv. Vernal, M. sativa cv. Zhung Mu, and M. sativa cv. Xing Jiang Daye in this study. Cd showed declined chlorophyll score in Xing Jiang Daye compared with Zhung Mu and Vernal. No significant change observed among the cultivars for root and shoot length. Atomic absorption spectroscopy analysis demonstrated a significant accumulation of Cd, Fe, S and PC in distinct alfalfa cultivars. However, Zhung Mu and Xing Jiang Daye declined Cd accumulation in root, where Fe, S and PC incremented only in Zhung Mu. It suggests that excess Cd in Zhung Mu possibly inhibited in root by the increased accumulation of Fe, S and PC. This was further confirmed by the response of Fe (MsIRT1) and S transporters (MsSULTR1;2 and MsSULTR1;3), and MsPCS1 genes associated with Fe, S and PC availability and translocation in roots and shoots. It suggests that specially the transcript signal inducing the responses to adjust Cd especially in Zhung Mu. This finding provides the essential background for further molecular breeding program for forage crops.
To investigate the effect of Mycorrhiza (Glomus intradics) and Rhizobium inoculation on the N, P utilization and growth response of Alfalfa (Medicago saliva L.) and Perennial ryegrass (Lolium perenne L.) in mixed sward, four treatments (non-inoculation, Control; Mycorrhiza inoculation: M; Rhizobium inoculation, R and Mycorrhiza and Rhizobium inoculation, M+R) were applied. The associated analyses were canied out on the early vegetative growth stage (DAS 56, 56 days after seeding) and on the early flowering stage (DAS 126). The decreased rate of total N and P content in soil, with advancing plant growth, was relatively higher in the M and M + R treatment than control. The content of availabie phosphorus in soil at DAS 126 increased by about 34 and 38 % in M and M+R treatment compared to control (189.2 mg PzOskg DM), while non-significant changes was observed in R treatment. Total N uptake and P uptake in the control at DAS 126 were 44.71 and 3.52 mglplant in mixed sward, respectively. About 71, 98 and 197 % of increases in total N uptake and 70, 72 and 11 1 % of increases in total P uptake were estimated in M, R and M+R treatment. Comparing to control, total dry matter yield significantly increased by 27, 33 and 53 %, and crude protein yield also by 78, 83 and 204 %, respectively, in M, R and M+R treatment. The present data indicated that mycorrhiza orland rhizobium inoculation improved N, P utilization of both alfalfa and perennial ryegrass plants, and consequently increased total yield (especially by dual inoculation, M+R). Comparing to control, dry matter yield significantly increased by 8, 27 and 28 %, and crude protein yield also by 21, 42 and 39 %, respectively, in M, R and M+R treatment. The present data indicated that mycorrhiza orland rhizobium inoculation improved N, P utilization, and consequently increased the yield of alfalfa. To investigate the effects of phosphate($P_2O_5$) fertilization levels on the dry matter(DM) yield, nutrition yield, and $P_2O_5$ efficiency of forages in mixed sward of alfalfa-grasses, a field experiment arranged by randomized complete block design with five treatments(0, 50, 100, 150 and 200kg $P_2O_5$/ha) was conducted at Livestock Experiment Station in Suwon, September, 1990 to Feb~ary, 1993. During two years, average DM yields of forages increased as $P_2O_5$ fertilization level was increased, but no significant difference was found between DM 10,754kg of $P_2O_5$ 150kg/ha and DM 10,845kg of $P_2O_5$ 200kg/ha. Cmde protein and energy productivity of forages increased as $P_2O_5$ fertilization level was increased, and the increasing degree was highest between $P_2O_5$ l00kg and 150kg/ha feriilization. With increasing $P_2O_5$ fertilization, mineral contents tended to increase in P, K and WCa+ Mg equivalent ratio, but not to be regular in the other elements. Efficiency of $P_2O_5$ was highest at $P_2O_5$ 150kg/ha fertilization, which produced DM 12.8kg. net energy lactation 75.4MJ. starch equivalent 6.lkStE, and total digestible nutrients 8.6kg per Ikg $P_2O_5$.
