Hypoglycemic Effect and Hepatic Detoxification Activity of Extracts from Crataegus fructus and Morus alba L. in Alcohol-treated Rats

  • To consider potentially new sources which have hypoglycemic effect and accelerating alcohol detoxification, this study was designed to investigate the effect of Crataegus fructus and Morns alba L. in alcohol-treated rats. I compared the body weight, glucose, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alcohol dehydrogenase (ADH), and aldehyde dehydrogenase (ALDH) of rats administered both alcohol and extract of experimental plants to rats treated with alcohol alone. Administration of extracts of C. fructus and M alba, respectively, resulted in a significant reduction in the blood glucose level and the activities of ADH of liver compared to the control rats, and administration of extract of M. alba showed significantly lower on bodyweight gain in the rats than in other treated rats. In contrast, the activities of ALDH of liver were increased. The activities of AST and ALT between the only alcohol-treated rats and the alcohol and experimental plants-treated rats were no significant difference. The results suggest that C. fructus and M alba have a hypoglycemic effect, and reduce liver damage by accelerating acetaldehyde metabolism in alcoholic rats, so the combined effect of C. fructus and M alba may be considered as an alternative remedy for hangovers, alcohol-induced overweight and alcohol-induced diabetes.

Effects of the Sungjuchunggan-tang(Xingjiuqinggan-tang) on alcoholic metabolism and alcoholic liver damages (주상(酒傷)에 응용되는 성주청간탕(醒酒淸肝湯)이 알코올대사 및 간장애에 미치는 영향)

  • Objectives : The aim of this study was to investigate effects of the Sungjuchunggan-tang(Xingjiuqinggan-tang), which has been used for alcoholic diseases in Oriental medicine, on alcoholic metabolism and alcoholic liver damage. Methods : We evaluated the activities of alcohol dehydrogenase(ADH)and aldehyde dehydrogenase(ALDH), and measured the levels of AST, ALT, glucose, triglyceride, HDL-cholesterol, LDL-cholesterol and phospholipid in serum of guinea pigs. Results and Conclusions : In this experiment, Sungjuchunggan-tang, Pharbitidis Semen and Puerariae Radix significantly suppressed the activity of ADH which is a precursor enzyme of acetaldehyde. Sungjuchunggan-tang and Pharbitidis Semen significantly suppressed the activity of ALDH, which is a catabolic enzyme, and increased its level. Due to alcohol consumption, level of ALT and AST were significantly reduced. These experimental results suggest that Sungjuchunggan-tang and Pharbitidis Semen inhibit the formation of acetaldehyde in the metabolism of alcohol, and affect the recovery of weakened liver functions caused by alcohol.

  • Citrus sunki Hort. ex Tanaka is one of several Jeju-native citrus fruits. A number of biological properties for citrus fruits have been suggested, however little is known about those of C. sunki. The present study tested whether fermented product of C. sunki peel (FSP) might affect the activities of enzymes regulating ethanol metabolism. Effects on body weight gain as well as on fatty liver formation were also investigated. The activities of alcohol dehydrogenase and aldehyde dehydrogenase were stimulated remarkably by FSP. Excessive ethanol-induced cytotoxicity was also prevented by FSP in HepG2 cells. FSP decreased the weight gain and fatty liver formation induced by a high-fat diet in mice. From these results, FSP might be a potent source of neutraceuticals useful for preventing ethanol-induced health problems.

Inhibition of Hepatic Triglyceride Accumulation and Stimulation of Alcohol Metabolism by the Herbal Extract Containing Phaseoli radiati semen in Rats Fed Ethanol (급성 알콜 투여 흰쥐에서 녹두 함유 복합생약제제의 간 중성지방 축적억제 및 알콜대사 촉진 효과)

  • An ethanol administration causes hepatic triglyceride accumulation in rats. To assess whether the herbal extract containing Phaseoli radiati semen(herbal extract) inhibit s the triglyceride accumulation in the liver, we determined the hepatic triglyceiide levels in rats fed ethanol and the herbal extract. In addition, the blood ethanol concentrations and the activities of hepatic alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) were measured to determine the effects of the herbal extract on alcohol metabolism in rats. The administration of the herbal extract markedly reduced the triglyceride levels elevated by ethanol in the liver as well as in the serum. The herbal extract remarkably lowered blood ethanol concentrations in a dose-dependent manner. The ADH activities decreased by ethanol were recovered to the normal level by the herbal extract treatment. Moreover, the ALDH activities slightly decreased by ethanol increased beyond the normal level by the herbal extract treatment. We conclude that the herbal extract inhibits the hepatic triglyceride accumulation and stimulates alcohol metabolism by preventing ADH and ALDH from inhbition by the ethanol administration in the rat liver.

Effects of a Health Drink Containing the Extract of the Hovenia Dulcis Fruit Stalk and Theracurmin, on Ethanol-Induced Hangover

  • The fruit stalk of Hovenia dulcis (H.dulcis) is traditionally used to relieve hangovers in Korea. Theracurmin is a highly absorbable curcumin preparation which increases the bioavailability of curcumin. Curcumin is known for its antioxidant and anti-inflammatory effects. However, the role of this combination in lowering alcohol levels in the body, thereby alleviating the severity of alcohol-induced hangover has not been investigated. Therefore, we conducted a study to investigate the eliminatory effects of a health drink containing the extract of the H. dulcis fruit stalk and theracurmin (theracurmin drink) on ethanol-induced hangover in rats. The theracurmin drink delivered orally to rats 30 mins before the administration of 40% ethanol (5 g/kg body weight), lowered the concentration of ethanol and acetaldehyde in the blood samples collected 1, 3, and 5 h after ethanol administration. Furthermore, the theracurmin drink increased the activities of alcohol dehydrogenase and aldehyde dehydrogenase enzymes. The effectiveness of the theracurmin drink was thus superior to that of other health drink products, suggesting that its consumption may alleviate or prevent an alcohol-induced hangover.

Physicochemical Properties of Bokbunja Jelly Containing Fermented Sea Tangle Saccharina japonica Powder (유산균 발효다시마(Saccharina japonica)를 첨가한 복분자 젤리의 이화학적 품질특성)

  • This study investigated the physicochemical properties and physiological activities of jelly prepared from gelatin, sugar, bokbunja extract, and different amounts (0, 0.5, 1, 2 and 3%) of fermented sea tangle Saccharina japonica powder (FSP). The jelly moisture, pH, and sugar content slightly increased with increasing the FSP content. Hardness, springiness, gumminess, chewiness, and cohesiveness also increased with increasing FSP concentration. Jelly antioxidant activity did not change significantly with increasing FSP. In contrast, alcohol dehydrogenase and aldehyde dehydrogenase activities in the jellies increased significantly with increasing FSP concentration. β-secretase inhibitory activity in jellies also increased with increasing FSP concentration. Jellies containing 0.5 or 1% FSP achieved the highest overall sensory acceptance scores. Taken together, these data indicate that addition of FSP to jelly appears to improve its quality and physiological activities.

Functional analysis of genes involved in rice disease resistance

  • Several plant and microbial genes that could confer disease resistance in transgenic rice plants are being cloned and characterized. We are currently constructing transgenic rice lines that overexpress the gene products, such as a galactinol synthase, a defensin, and a bacterial ACC deaminase. Subtractive hybridization of a rice cDNA library constructed from the Xanthomonas oryzae-infected ice leaves resulted in isolation of many inducible cDNA clones including a elongation factor EF2, a oryzain alpha, a catalase, a aldehyde dehydrogenase, a S-adenosylmethionine synthetase, a caffeic acid O-methyltransferase, a glyceraldehyde-3-phosphate dehydrogenase, a light-regulated protein, nKY transcription factors, and a nucleotide diphosphate kinase. Some genes among those may be useful genetic sources for construction of disease resistant transgenic rice. Full lengths of the rice OsFIERG and a rice oryzain genomic clones were cloned, and serial deletion fragments of the promoter regions of these genes were fused with GUS reporter gene in pCAMBIA1201, respectively. Promoter activities of these constructs will be examined upon various stresses and Pathogen infections to obtain the pathogen specific inducible-promoter. Comparison of the Antioxidant and Physiological Activities of Different Solvent Extracts Derived from Enteromorpha compressa (납작파래(Enteromorpha compressa) 추출물의 항산화성 및 생리활성)

  • In this study, we investigated the antioxidant and physiological activities of the Korean marine algae, Enteromorpha compressa. Solvent extracts of E. compressa were prepared using 70% ethanol, 80% methanol, and water, with extraction yields ranging from 9.55% to 25.67%. The total polyphenol and flavonoid contents ranged from 20.76-28.41 mg/g and 2.56-18.59 mg/g, respectively. Compared with the water extract, the ethanol and methanol extracts were found to have higher antioxidant activities. All three extracts were found to promote alcohol dehydrogenase and aldehyde dehydrogenase activities in a concentration-dependent manner, whereas the methanol and ethanol extracts were established to have the highest angiotensin-converting enzyme (ACE) inhibitory activity (IC50=1.40 ㎍/mL) and β-secretase inhibitory activity (IC50=0.17 ㎍/mL), respectively. These findings thus indicate that E. compressa could have beneficial application as a supplementary antioxidant and functional constituent in food and pharmaceutical materials.

Effects of Dextrin and β-cyclodextrin on Protective Effect of Hovenia dulcis Fruit Extract Against Alcohol-induced Liver Damage in vivo (Dextrin과 β-cyclodextrin이 생체 내에서 헛개나무 추출물의 알코올성 손상으로부터 간보호에 미치는 영향)

  • ${\beta}$-cyclodextrin has an ability to protect compounds from oxidative reaction by collecting them within its ring-like structure. So, In harsh condition ($40^{\circ}C$), marker compound, quercetin, was dramatically reduced in Hovenia dulcis fruit extract containing dextrin at 4 and 8 week compared to 0 week, but not that containing ${\beta}$-cyclodextrin. To evaluate the effects of dextrin and ${\beta}$-cyclodextrin on protective effect of H.dulcis fruit extract against alcohol-induced liver damage, The mice were orally injected alcohol, H. dulcis fruit extract/dextrin (HD) and H. dulcis fruit extract/${\beta}$-cyclodextrin (HCD), respectively, for 7 days. The mice orally administrated with alcohol significantly enhanced the serum concentration of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and the activity of lactate dehydrogenase (LDH) in serum compared to the control group. HD and HCD significantly decreased the levels of serum ALT and AST and serum LDH activities compared to alcohol group. And also alcohol group significantly increased the level of total cholesterol compared to the control group, but HD and HCD significantly reduced it compared to the alcohol group. However, the levels of TG in blood were not significantly changed in all groups. The activities of alcohol dehydrogenase (ADH) were significantly increased in HD and HCD group although those of aldehyde dehydrogenase showed an increasing tendency. This data suggested that HD and HCD were able to induce alcohol degradation in the liver tissues. All together, the results showed that HCD demonstrated their ability to protect liver from alcohol-induced damage on equal terms with HD.

Effects of Chongkukjang Intake on Lipid Metabolism and Liver Function in Ethanol Consumed Rats (청국장을 급여한 식이가 알코올 섭취쥐의 지질대사 및 간 기능개선에 미치는 영향)

  • This study was performed to investigate the effect of chongkukjang intake on lipid metabolism and liver function in ethanol consumed rats. Twenty one Sprague-Dawley male rats aging 4 weeks old were used as experimental animals, which were divided into three dietary groups; casein diet(CA), soybean diet(SB) and chongkukjang diet(CJ). Alcohol was consumed with water as 25%(v/v) ethanol solution. After 4 weeks of experimental period, rats were sacrificed to get blood and liver samples for analysis of lipids, lipid peroxides, antioxidative enzymes and biochemical indices of liver function. The mean body weight, food intake and liver index were not significantly different among three groups. Serum level of total lipid, triglyceride and total cholesterol of chongkukjang diet group was the lowest among three groups although the difference was not significant. HDL-cholesterol level was significantly(p<0.05) higher in chongkukjang diet group than that of casein diet group. LDL-cholesterol level of chongkukjang and soybean diet group was significantly(p<0.05) lower than that of casein diet group respectively. Liver TBARS of chongkukjang and soybean diet group was significantly(p<0.05) lower than that of casein diet group respectively. The superoxide dismutase activity of chongkukjang diet group was significantly(p<0.05) higher than that of casein diet group. Catalase activity was not significantly different among three groups. As indices of liver function, glutamic oxaloacetic transminase(GOT), glutamic pyruvic transminase(GPT), $\gamma$-glutamyl transpeptidase($\gamma$-GTP) and lactate dehydrogenase(LDH) were not significantly different among three groups. Serum alcohol concentration and activities of alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) were not significantly different among three groups. The chongkukjang diet seems to give a beneficial effect for improving lipid metabolism by increasing HDL-cholesterol level and SOD activity while reducing liver TBARS level. However, effect on liver function has to be investigated further.
