Effect of a Alcohol Detoxification Beverage (ADB) Contained Radix puerariae and Bambusae caules in Liquamen Phyllostachyos on the Alcohol Administered Mouse (갈근(Radix Puerariae)과 죽력 (Bambusae caules in Liquamen Phyllostachyos)을 함유한 숙취해소 음료가 알코올을 투여한 생쥐에 미치는 영향)

  • This study has been carried out to investigate the effect of the beverage contained Radix puerariae (15%) and Bambusae caules in Liquamen Phyllostachyos (1%) on the alcohol administered mouse. The control group was treated with alcohol (40% alcohol, 8.0 mL/kg of body weight), experimental group 1 (Exp.1) was administered with beverage (8.0 mL/kg of body weight) before alcohol treatment, and experimental group 2 (Exp.2) was administered with beverage after alcohol treatment. Blood alcohol concentrations were decreased by 11% (in Exp. 1) and 17% (in Exp. 2) compared with control group. The concentrations of serum aspartate aminotransferase (AST) were slightly increased in Exp. 1 and Exp.2, but alanine aminotransferase (ALT) levels were not changed compared with control group. Mn-Superoxide dismutase activity was increased by about 70% in the experimental groups in comparison to control group. The Exp. 1 and Exp. 2 contained more periodic-acid Schiff (PAS)-positive materials than the control group. Electron microscopical observations showed that, cell organelles of hepatocytes were more disoriented in the control group than in Exp. 1 and Exp. 2. These results suggest that the beverage contained Radix puerariae and Bambusae caules in Liquamen Fhyllostachyos is a safe and effective therapeutic agent for alcohol abuse.

The Effects of Chungganhaeju-tang(Qingganjiejiu-tang) on Alcoholic Liver Damages by Applying Proteomics (청간해주탕(淸肝解酒湯)이 알코올 유발 간섬유화와 단백질 발현에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the effects of Chungganhaeju-tang(Qingganjiejiu-tang) on alcoholic liver damaged by applying proteomics. Materials and Methods : Sprague-Dawley rats were used in this experiment the rats were divided into the normal group, the control group(alcohol) and the sample group(CGHJT +alcohol). The ethanol was orally administered twice a day for 6 weeks in the control and sample groups. Water instead of ethanol was orally administered twice a day for 6 weeks in the normal group. CGHJT extract was orally administered once a day for 6 weeks in the sample group. The livers of each group were processed and assessed by histology, Western Blot, $Oxyblot^{TM}$, CBB and 2-dimensional electrophoresis. Results : In the histological findings of the liver, CGHJT inhibited hepatic fibrogenesis induced by alcohol. TIMP-1 decreased in the sample group assessed by western blot and statistical significance was noted by dot blotting(p<0.05). In the $Oxyblot^{TM}$, protein oxidation induced by alcohol treatment decreased with CGHJT. In the 2-dimensional electrophoresis finding, increased proteins alcohol such as HSP 60, 60kDa heat shock protein, 3-mercaptopyruvate sulfurtransferase were normalized by CGHJT. CGHJT was considered to normalize the anti-oxidation activity elevated by alcohol. In the 2-dimensional electrophoresis finding, increased oxidized proteins such as actin, prolyl 4-hydroxylase beta polypeptide, 94kDa glucose regulated protein(GRP94), heat shock protein 90-alpha(HSC86), calreticulin precursor(CRP55), ATP synthase beta chain mitochondrial precursor, caspase-8 precursor, and dihydrolipoamide succinyltransferase(E2) decreased with CGHJT. CGHJT was considered to reduce the oxidative stress of alcohol. Conclusion : Chungganhaeju-tang(Qingganjiejiu-tang) exerts an inhibitory effect against the fibrosis and protein oxidation induced by alcohol treatment of rat liver. CGHJT was considered to normalize the elevated anti-oxidation activity by alcohol and to reduce the level of oxidative stress due to alcohol.

Effect of the Combination of Ethanol with Toluene Treatment for a Short Time Period on the Toluene Metabolizing Enzyme Activity (흰쥐에 Toluene과 Alcohol의 병행투여가 Toluene 대사 효소활성에 미치는 영향)

  • To elucidate the effect of acute ethanol pretreatment on some toluene metabolizing enzyme activities, rats were divided into 4groups: control, alcohol-treated, toluene-treated, rat's and toluene-treated rats pretreated with ethanol. The alcohol or toluene-treated rats showed the significantly increased activities of hepatic aniline hydroxylase(AH) and aminopyrine demethylase(AD) compared to the control group. And the toluene-treated rats pretreated with ethanol showed somewhat decreased tendency of these enzyme activities compared to only toluene-treated rats. Liver benzylalcohol or aldehyde dehydrogenase activities were higher in alcohol or toluene-treated rats than those of the control group. The toluene-treated rats showed the decreased tendency of benzylalcohol dehydrogenase activities by the pretreatment of alcohol. Furthermore, toluene treated-rats showed the markedly decreased activity of benzaldehyde dehydrogenase by the ethanol pretreat-ment. On the other hand, hepatic xanthine oxidase activity in toluene-treated animals pretreated with ethanol was significantly higher than those of the toluene alone-treated rats. These results indicate that the combination of ethanol with toluene treatment for a short period of time possibly results in decreased activity of some toluene metabolizing enzymes in rats.

Effect of Daekumeumja Herb-acupuncture on c-Fos Expression in Hippocampus of Alcohol Intoxicated Rats (대금음자 약침이 알코올 독성 흰쥐의 해마에서 c-Fos 발현에 미치는 영향)

  • Objectives : The study was conducted to investigate the effect of Deakumeumja herb-acupuncture on c-Fos expression in each area of the hippocampus of Alcohol intoxicated rats. Methods : Experimental groups were divided into five groups ; normal group, the alcohol-treated (control) group, the alcohol- 1 mg/kg Daekumeumja treated (sample A) group, the alcohol- 5 mg/kg Daekumeumja treated (sample B) group, the alcohol- 10 mg/kg Daekumeumja treated (sample C) group (n = 6 for each group). Rats of normal group were injected intraperitoneally with saline once a day for 5 consecutive days, while animals of the alcohol-treated (control) group were injected once a day with 2 g/kg of alcohol for the same duration of time. Animals of the alcohol and Daekumeumja-treated (sample A, B, C) groups were acupunctured at Chung-wan$(CV_{12})$ with 2 g/kg of alcohol and the appropriate amount of Daekumeumja extract once a day for 5 days. Each groups was evaluated by the changes of c-fos-positive neurons in each area of the hippocampus by using an image analyzer and microscope. Results: 1. In the CAI region of the hippocampus, the number of Fos-positive cells in the sample B, C groups were significantly increased compared with the control group. 2. In the CA2-3 regions of the hippocampus, the number of Fos-positive cells of the sample B, C groups were significantly increased compared with the control group. 3. In the Dentate gyrus region of the hippocampus, the number of Fos-positive cells of the sample C group was significantly increased compared with the control group. Conclusion : c-fos expression in each area of the hippocampus was reduced in alcohol-intoxicated groups. Treatment of Daekumeumja increased this reduction. In conclusion, it can be suggested that Daedumeumja possesses protective effects of the amnesia and learning disability in alcoholism.

  • Alcohol concentration via pervaporation process was performed by using commercial PDMS(polydimethylsiloxane) composite membrane and plasma treated PP(polypropylene) membranes. Effects of operating parameters of pcrvaporation process were examined. With the increase of butanol concentration in the feed, flux and selectivity increased due to the greater affinity of butanol with PDMS than that of water. As the operating temperature increased, free volume as well as the solubilities and diffusivities of alcohol and water increased to result in the greater flux and less selectivity of the membrane. As solubility parameter difference between alcohol and PDMS membrane decreased, high flux and good selectivity were obtained. When PP membrane was plasma treated with methanol, it has 6 times greater flux than PDMS membrane with equivalent separation factor. With the increase of plasma treatment time, flux and selectivity were enhanced. However, excess treatment time caused pore blocking to result in the flux decrease.

  • Neuroleptic malignant syndrome (NMS) is a rare but fatal condition, with a high mortality rate. NMS is characterized by altered mental status, fever, myoclonus, autonomic dysfunctions, and elevated creatinine phosphokinase. The clinical manifestations may be confused with alcohol-related symptoms, trauma, sepsis, postoperative agitation, or malignant hyperthermia. A 69-year-old male patient with alcohol withdrawal was admitted to the operating theatre to rule out septic shock due to mesenteric injury after multiple trauma. He was suspected NMS with abrupt increase body temperature to 41.7℃ after haloperidol administration. Active cooling and rapid fluid infusion was done during anesthesia. Delayed diagnosis and treatment of NMS lead to catastrophic result. Therefore, if the patient's past medical history is unknown or clinical symptoms develop that are suggestive of NMS, early treatment must be considered.

Duration of Abstinence after Discharge from Alcoholism Inpatient Treatment - Comparing Voluntarily vs. Involuntarily Admitted Patients - (알코올중독자의 입원동의 여부에 따른 퇴원후 단주기간에 관한 연구)

  • Motivation has been identified as an important factor predicting long-term outcomes of alcohol abuse treatment. Whether a patient agreed on his/her inpatient treatment at time of hospitalization could be an indicator of their motivation for treatment. This study focused on this aspect of treatment motivation and examined whether this factor predicts post-discharge abstinence. A sample of 145 individuals who were hospitalized for alcohol abuse treatment participated in the baseline data collection, of which only 66 successfully completed the 8 month follow-up telephone interview. The findings of survival analysis suggest that voluntarily admitted individuals had significantly longer duration of post-discharge abstinence compared to their counterparts. A Cox proportional hazards regression model revealed that voluntary admission and family support were significant predictors for post-discharge relapse of drinking, after controlling for the effects of marital status, living alone, and working status. Implications for social work practice are discussed.

Effect of Treating and Blanching on Qualities Preservation of Packaged Fresh-cut Yam (알코올 전 처리와 블랜칭이 포장 절편 마의 품질보존에 미치는 영향)

  • In this experiment, sliced yams (5 mm size) were used for research. Alcohol treatment and blanching treatment were used in this experiments. Color change, hardness, microorganism total count, and PPO activity of the packaged yam were measured. In result, colors of the treated yam were significantly changed in alcohol treatment with different concentration. 5% alcohol treatment is better than 10% alcohol treatment in browning inhibition of fresh-cut yam. But in blanching treatment, color change had no significant different. PPO activity of 1 min blanching packaged yam were decreased 50% and PPO activity of 5 min blanching packaged yam were decreased 85%. In the case of hardness were decreased in both experimental groups. especially 1 min, 3 min and 5 min blanching films, hardness were decreased more than 10 N. Also, both treatments are inhibited the growth of microorganisms effectively.

Study on the Anti - Hyperlipidemia and Liver Cell Protection of Korean Medicinal Herb Complex of Alcohol fed Rats

  • This study sought to investigate the ameliorating effects of a Korean medicinal herb (KMH) complex on the impacts of alcohol consumption in rat hepatocytes and in reducing the total cholesterol levels and the total lipid levels in the serum. We compared the body weight gain and ratio of the liver, the kidney to body weight, and also the serum biochemistry of the rats administered with both the alcohol and the KMH complex to the control rats treated with alcohol alone. The clinically important enzyme markers (Aspartate Aminotransferase, AST, and Alanine Aminotransferase, ALT) of rats, administered with both the alcohol and the KMH complex treatments, were compared with those in the control group. The treatment regimen (KMH complex) significantly reduced the serum AST and ALT levels, indicating the hepato-protective effects of the KMH complex. Furthermore, total cholesterol and total lipid levels were significantly reduced. These results indicate that the KMH complex may positively mediate the effects of alcohol on hepatocytes and the general liver functions.

Inhibitory Effect of Ruta chalepensis Leaf-Derived Component against Alcohol Dehydrogenase

  • Inhibitory activity of active compound isolated from Ruta chalepensis leaf was examined against alcohol dehydrogenase and, upon comparison to those of four commercially available compounds (quinoline, quinoline-3-carboxaldehyde, quinoline-3-carboxylic acid, and quinoline-4-carboxylic acid) and 1,10-phenanthroline as alcohol dehydrogenase inhibitor, was characterized as quinoline-4-caboxaldehyde by spectral analyses. Inhibitory effects ($IC_{50}$) of quinoline-4-caboxaldehyde and quinoline derivatives varied depending on chemicals and concentrations used. The $IC_{50}$ values of quinoline-4-carboxaldehyde, quinoline-3-carboxaldehyde, quinoline, quinoline-3-carboxylic acid, and quinoline-4-carboxylic acid were 0.04, 0.3, 0.8, >1, and >1 mg/mL, respectively. These results suggest inhibitory action of quinoline-4-carboxaldehyde against alcohol dehydrogenase as prospective therapeutics for treatment of alcoholic liver diseases such as alcohol hepatitis and cirrhosis resulting from chronic alcohol abuse.
