Two-enzyme coupled fluorometric assay of urinary dipeptidase (이원효소 연쇄반응의 형광분석에 의한 Urinary Dipeptidase의 활성도 측정)

  • Urinary dipeptidase(Udpase) was assayed by fluorometric analysis of NADH which was produced from an indicator enzyme, L-alanine dehydrogenase. The reaction mixture was consisted of a dipeptide(L-ala-L-ala), ${\beta}-NAD^+$, L-alanine dehydrogenase in 12.5 mM sodium carbonate buffer, pH 9.0, and urinary dipeptidase which initiated the reaction. The fluorescence intensity of NADH was measured as a function of time with the excitation wavelength at 340nm and emission at 460nm. Comparison of this fluorometric method with the conventional spectrophotometric method utilizing glycyldehydrophenylalanine(Gdp) as substrate provided the correlation coefficient of 0.996 and increased the sensitivity more than ten times.

Effect of Amino Acid Substitutions of Escherichia Coli $\gamma$-Glutamylcysteine Synthetase (Escherichia coli $\gamma$-Glutamylcysteine Synthetase의 아미노산 치환 효과)

  • Two amino acid residues ($Ala^{494}$ and $Ser^{495}$ of E. coli .gamma.-glutamylcysteine synthetase have been investigated whether they are the site of feedback inhibition by site specific mutagenesis. Single substitution of $serine^{495}$ (S495F), and double substitutions of alanine$^{494}$ and $serine^{495}$ (A494G-S495F) resulted in the inactivation of the .gamma.-glutamylcysteine synthetase activity. Substitution of $alanine^{494}$ with $glycine^{494}$ resulted in a higher level of feedback inhibition. These results suggest that $serine^{495}$ in .gamma.-glutamylcysteine synthetase is required for its catalytic acitvity and $alanine^{494}$ is presumably related to the feeback inhibition site.

Effect of renal ischemia on renal function and excretion of lysine, alanine in the rabbit (신장 허혈이 토끼 신기능과 lysine 및 alanine 배설에 미치는 영향)

  • This study was carried out to determine the effect of renal ischemia on renal function and excretion of amino acid in rabbit. The animal models of renal ischemia induced experimentally by clamping the renal artery for different lengths of time. These results were summarized as follows: 1. Ischemia for 30 or 60 min produced a polyuria which is accompanied by an increase in $Na^+$ excretion. Glomerular filtration rate (GFR) and p-aminohippurate plasma($C_{PAH}$) were not altered by 30 min of ischemia, indicating that transient ischemia results in a marked tubular dysfuction before a reduction in GFR or renal blood flow. 2. Reabsorption of glucose and amino acids such as alanine and lysine was markedly reduced after 30 min of ischemia, and the effect was more pronounced after 60 min of ischemia.

Comparison of Coverage-Dependent Adsorption Structures of Alanine and Leucine on Ge(100): Bonding Configuration and Adsorption Stability

  • The bonding configuration and adsorption stability of alanine and leucine adsorbed on Ge(100)-2${\times}$1 surface were investigated and compared using core-level photoemission spectroscopy (CLPES) and density functional theory (DFT) calculations. The bonding configuration, stability, and adsorption energies were evaluated for two different coverage levels. In both cases, the C 1s, N 1s, and O 1s core-level spectra at a low coverage (0.30 ML) indicated that the carboxyl and amine groups participated in bonding with the Ge(100) surface in an "O-H dissociated-N dative bonded structure". At high coverage levels (0.60 ML), both this structure and an "O-H dissociation bonded structure" were present. As a result, we found that alanine adsorbs more easily (lower adsorption energy) than leucine on Ge(100) surfaces due to less steric hindrance of side chain.

Volatiles from the Maillard Reaction of L-Ascorbic Acid and L-Alanine at Different pHs

  • The volatiles formed from the reactions of L-ascorbic acid with L-alanine at 5 different pH (5, 6, 7, 8, or 9) and $140{\pm}2^{\circ}C$ for 2 hr was performed using solid-phase microextraction-gas chromatography-mass spectrometry (SPME-GC-MS) analysis were identified to be 25 different kinds. The reaction between L-ascorbic acid and L-alanine led mainly to the formation of pyrazines. Many of these were alkylpyrazines, such as 3-ethyl-2,5-dimethylpyrazine, 2,5-dimethylpyrazine, 2-ethyl-5-methylpyrazine, 3,5-diethyl-2-methylpyrazine, methylpyrazine, 2-ethyl-6-methylpyrazine, and 2,3-diethyl-5-methylpyrazine, other compounds identified were furans, phenols, benzoquinones, 2,4,6-trimethylpyridine, and 2-methylbenzoxazole. The studies showed that furans, such as furfural and benzofuran were formed mainly at acidic pH. In contrast, higher pH values could promote the production of pyrazines.

  • Alnine pellets were installed in a nuclear power plant for one or two operation cycles and measured by electron spin resonance (ESR) spectrometers for dosimetry. Dose and "x/y ratio", i.e., satellite peak over main center peak ratio, were measured for the returned alanine dosimeters from the nuclear power plant and compared to the values of reference alanine dosimeters exposed only to gamma rays. The variation of the x/y ratio change depended on the population of radicals from each radiation component with different LET. The gamma dose in a mixed radiation field was estimated by an additive gamma ray irradiation experiment and the measured dose rate at specified locations in the containment building.

  • L-Alanine(L-ala) and S-methyl-L-cysteine(L-mcy) cobalt(III) complexes of a flexible $N_2O_2$-type tetradentate ligand ethylenediamine-N,N'-di- -butyric acid(eddb), s-cis-[Co(eddb)(L-ala)] and s-cis-[Co(eddb)(L-mcy), have been prepared via the substitution reactions of the s-cis-$[Co(eddb)Cl_2]$-complex with, respectively, L-alanine and S-methyl-L-cysteine. Both L-alanine and S-methyl-L-cysteine are found to coordinate to the cobalt(III) ion via the nitrogen and oxygen donor atoms to give the meridional s-cis isomer. Electronic absorption, ir and pmr spectra are used to characterize the complexes obtained in this work along with elemental analysis data.
