식물 추출물 라벤더, 레몬 유칼립투스 및 계피 오일의 급성독성평가 (Evaluation of Acute Toxicity of Plant Extracts, Lavender, Lemon Eucalyptus and Cassia Essential Oil)

  • 본 연구는 생리활성이 높은 식물오일의(라벤더, 레몬유칼립투스 및 계피 오일) 친환경 살충소재의 활용가능성을 탐색하고자 급성독성(급성경구독성, 급성경피독성, 피부자극성 및 안점막자극성)시험을 수행하였다. 랫드를 이용한 급성경구독성시험결과 라벤더, 레몬 유칼립투스 및 계피 오일의 $LD_{50}$은 2,000 mg/kg bw 이상이었고, 급성경피독성시험결과 모든 시험물질의 $LD_{50}$이 4,000 mg/kg bw으로 나타났다. 피부자극성시험결과 라벤더와 레몬 유칼립투스 오일은 자극성이 없었고, 계피 오일은 중도의 자극성을 나타냈다. 안점막자극성시험결과 라벤더, 레몬 유칼립투스 오일은 자극성이 없었고, 계피오일은 중도의 자극성을 나타냈으나 세척시험 결과 자극성은 없었다. 따라서, 라벤더 및 레몬 유칼립투스 오일은 독성이 낮았으나, 계피 오일은 중도의 피부자극성 및 안점막자극성을 갖는 것으로 평가되었다.

  • With current. difficulties in Korean agricultural sector, the biotecilnoiogy is considered as the most important factor to offset the inferiority in the agriculture. New products by using the biotechnology would be commercialized and consumed by both producers and consumers. Before its production and distribution researches on new product should be undertaken in many aspects, especially in terms of economic aspects. Main objectives of this study are to measure the economic value of the new product using the agro-biotechnology, here potato microtubers, and to examine whether the stated preferences theory, the Contingent Valuation Method(CVM), could be accepted in the economic analysis for the new biotech product. Through the demand function derived from the consequences of functional relationship, the consumer's surplus was estimated.

  • Bio-venture companies have undertaken harsh environment after 2000 bubble corruption. It is unlike to be changed in the near future. The business model of near-term cash flow, financial stability and survival is a key issue to be discussed. The company shall be rearranged to achieve fast recovery from investment and have to show clear and quantitative business model to the investors. Nevertheless, bio-venture should not drop long-term value creation by sacrificing the possibility of emerging technologies. Prior to clarifing agro-bioventure business model, GMO strategy should have broad supports from stakeholders. The interests and worries of stakeholders are discussed and SWOT is analyzed. For sustainable economy of future, new technology continuously should be invested to fit ROI model of genomics-based GMO developments. Hybrid of products ${\varepsilon}$ technologies may be one of the favorites in this stormy season.

Cloning and Characterization of Squalene Synthase (SQS) Gene from Ganoderma lucidum

  • This report provides the complete nucleotide sequences of the full-length cDNA encoding squalene synthase (SQS) and its genomic DNA sequence from a triterpene-producing fungus, Ganoderma lucidum. The cDNA of the squalene synthase (SQS) (GenBank Accession Number: DQ494674) was found to contain an open reading frame (ORF) of 1,404 bp encoding a 468-amino-acid polypeptide, whereas the SQS genomic DNA sequence (GenBank Accession Number: DQ494675) consisted of 1,984 bp and contained four exons and three introns. Only one gene copy was present in the G. lucidum genome. The deduced amino acid sequence of Ganoderma lucidum squalene synthase (GI-SQS) exhibited a high homology with other fungal squalene synthase genes and contained six conserved domains. A phylogenetic analysis revealed that G. lucidum SQS belonged to the fungi SQS group, and was more closely related to the SQS of U. maydis than to those of other fungi. A gene expression analysis showed that the expression level was relatively low in mycelia incubated for 12 days, increased after 14 to 20 days of incubation, and reached a relatively high level in the mushroom primordia. Functional complementation of GI-SQS in a SQS-deficient strain of Saccharomyces cerevisiae confirmed that the cloned cDNA encoded a squalene synthase.

  • The medicinal properties of Ganoderma lucidum have been recognized in China for many centuries. Active pharmaceutical components include triterpenes. To elucidate the molecular regulation of triterpene biosynthesis in this mushroom, a 57-base pair DNA fragment encoding the fourth conserved domain SQ-4 (SMGLFLQKTNIIRDYNEDL) of squalene synthase was synthesized and cloned into the expression vector pET-32a(+). The recombinant fusion protein induced by IPTG (isopropyl-$\beta$-D-thiogalactopyranoside) was overexpressed in the Escherichia coli. Using the purified recombinant fusion protein of 20.9 kDa, a specific polyclonal antibody was obtained from immunized rabbit. Expression of squalene synthase at different development stages of Ganoderma lucidum was analyzed.

  • 본 연구에서는 강원전통장류로부터 혈전용해 활성이 우수한 효모를 분리하였다. 된장에서 분리한 AFY-1 균주는 혈전용해 활성 측정결과 양성대조구인 plasmin 보다 약 1.75배 높은 활성을 나타내었다. 분리한 효모는 18S rRNA 염기서열 및 탄소원 이용 특성 분석을 통하여 Saccharomycetales sp.로 동정되었으며, AFY-1 균주의 생육 최적온도는 $32^{\circ}C$였다. 본 연구에서 분리된 혈전용해능이 우수한 효모균주는 안전성 검증 등 추가연구를 통해 발효식품 제조용 스타터로서 활용이 가능할 것으로 기대된다.

  • The National Agricultural Biotechnology Information Center (NABIC) constructed an agricultural biology-based infrastructure and developed a biological information-based database. The major functions of the NABIC are focused on biotechnological developments for agricultural bioinformatics and providing a web-based service to construct bioinformatics workflows easily, such as protein function prediction and genome systems biology programs. The NABIC has concentrated on the functional genomics of major crops, building an integrated biotechnology database for agro-biotech information that focuses on the proteomics of major agricultural resources, such as rice, Chinese cabbage, rice Ds-tagging lines, and microorganisms.

프로바이오틱 유산균 발효에 의한 배초향 잎 추출물의 피부 항산화 및 항염증 활성 증대 (Enhancement of Skin Antioxidant and Anti-Inflammatory Potentials of Agastache rugosa Leaf Extract by Probiotic Bacterial Fermentation in Human Epidermal Keratinocytes)

  • 본 연구에서, 비발효(ARE) 및 발효(ARE-F) 배초향 잎 열수 추출물의 피부 항산화 및 항염증 효능을 비교함으로써 프로바이오틱 발효의 효과를 검토하였다. ARE-F는 Lactobacillus rhamnosus HK-9 발효에 의하여 ARE로부터 제조되었다. In vitro 항염증 효능은 지질다당류(LPS)에 의하여 자극된 HaCaT 각질세포에서 일산화질소(NO), 활성산소종(ROS) 및 유도형 일산화질소합성효소(iNOS) 분석에 의하여 평가되었다. In vitro antiradical 활성은 2,2-diphenyl-2-picrylhydrazyl radical (DPPH) 소거 측정법에 의하여 평가되었다. LPS 자극에 의하여 증가된 NO, ROS 및 iNOS 수준에 미치는 ARE-F의 감소효과가 ARE에 의한 효과보다 현저히 강하였다. 그러나, ARE와 ARE-F의 DPPH 소거 효능에는 차이가 발견되지 않았다. ARE-F가 증강된 피부 항산화 및 항염증 효능을 갖고 있는데, 이 결과는 프로바이틱세균 발효가 배초향 잎의 일부 약리학적 효능을 증가시키는 효과적인 방법일 수 있음을 암시해 주고 있다. 요약하면, 배초향 잎 추출물의 피부항산화 및 항염증 활성이 프로바이오틱 세균인 L. rhamnosus HK-9에 의한 발효에 의하여 증강한다.

Screening of novel alkaloid inhibitors for vascular endothelial growth factor in cancer cells: an integrated computational approach

  • Vascular endothelial growth factor (VEGF) is expressed at elevated levels by most cancer cells, which can stimulate vascular endothelial cell growth, survival, proliferation as well as trigger angiogenesis modulated by VEGF and VEGFR (a tyrosine kinase receptor) signaling. The angiogenic effects of the VEGF family are thought to be primarily mediated through the interaction of VEGF with VEGFR-2. Targeting this signaling molecule and its receptor is a novel approach for blocking angiogenesis. In recent years virtual high throughput screening has emerged as a widely accepted powerful technique in the identification of novel and diverse leads. The high resolution X-ray structure of VEGF has paved the way to introduce new small molecular inhibitors by structure-based virtual screening. In this study using different alkaloid molecules as potential novel inhibitors of VEGF, we proposed three alkaloid candidates for inhibiting VEGF and VEGFR mediated angiogenesis. As these three alkaloid compounds exhibited high scoring functions, which also highlights their high binding ability, it is evident that these alkaloids can be taken to further drug development pipelines for use as novel lead compounds to design new and effective drugs against cancer.
