Enhancement of scu-PA Production from Human Kidney Cells by a Novel Bioreactor (인간 신장세포로부터 새로운 배양공법에 의한 scu-PA의 생산성 향상)

  • 4.0$\times$10-3 (IU/cells/day) of maximum specific scu-PA production, which was higher than those by a 75$\textrm{cm}^2$ T-flask and conventional perfusion systems, was maintained by tube-type bioreactor at 0.35 (1/h) of perfusion rate corresponding to 0.15 (dynes/$\textrm{cm}^2$) of shear stress. The production of scu-PA is also increased as shear stress was slowly increased, which is similar to natural human blood circulation. The tube reactor proves that there may be no limitation of oxygen supply by showing 1.0 (1/h) of oxygen transfer coefficient at steady state and this system yields much lower shear stress of 0.3(dynes/$\textrm{cm}^2$) than that of 5-8(dynes/$\textrm{cm}^2$) by conventional agitation systems.

  • In this study , fluoborte solution consisting of lead fluoborate, tin fluoborate and cupric acetate was used. By addition of small amount of Cu+= ion to the solution, the Cu content of deposition layer was almost controlled less than 5%. The amount of Cu in deposition layer was almost constant without any influence of Pb++ & Sn++ in the solution, and the amount of Pb was increased by the increase of total concentration of Pb++ +Sn++ in the solution, and the amount of Pb was increased by the increase of total concentration of Pb++ +Sn++ in the solution . Agitation of plating solution & low current density result in the increase of Cu content. Analyzing of microscopic structures and etching tests of the deposited alloy, it was believed that the alloy had a lamellar structure consisting of copper rich lamellar and lead rich layer.

  • Since Korea opened the door to the Western World, traditional costume has changed. The factors of change are as follows: 1) Cultural diffusion by the cultural contact is the foundamental factor of the costume change. 2) The government made edicts of clothing reform. The formal dresses of government official were simplified several times, the edict was issued for men to cut off their hair, and military uniform and government officials' dresses were west-ernized. 3) The diffusion of modernized education by the missionary school accerlerated clothing change. Students were ahead of the abolition of Suege-Chima (which is a sort of veil used to conceal woman's face). 4) The change in the activity of women in society made the women's dresses more convenient for action. 5) The open class system and social agitation speeded up the rate of change. During the Yi Dynasty class distinction was rigid, but the government made the laws which abolished the class distiction.

Development of a Water-Spraying Type Automatic Glochids Removal System for Cactus (Opuntia humifusa) Stem

  • Purpose: A water-spraying type automatic glochids removal system for cactus (Opuntia humifusa) stem was developed, and its performance was evaluated. The system was developed to reduce intensive human labor in removing glochids from cactus stem skin without inducing damage prior to further processing into value-added products. Methods: The developed system consists of conveyor and water-spraying systems. The conveyor system delivers cactus stems through water-spraying compartments and finally to a collecting box. In order to remove the glochids, rotating nozzles spray water over all areas (i.e., front and back faces and sides) of the cactus skin under controlled water pressure. Operating conditions such as conveyor speed, water pressure, angle of water-spraying nozzles, distance between conveyor belt and rotating nozzles, and angle of cactus flipping slide were adopted from our previous study and applied on the system design and manufacturing. The performance of glochids removal was evaluated by counting the number of glochids on the cactus stem before and after processing on the system. Results: The developed system performed efficiently and effectively under the pre-studied operating conditions except for the angle of cactus flipping slide. The new system had a glochids removal ratio of 94.1% without damaging the cactus skin. Considering the original number (approximately 30-60) of glochids, the remaining number was low (1-4), and most of them were found at the side edge of the cactus stem. This system can remove glochids from 360 cactus stems in 1 hr regardless of cactus size. Conclusions: The performance of the new system in glochids removal without damaging cactus skin is superior to any other existing device (i.e., brush type, rubber-friction type, and agitation type). The system is expected to be applied in cactus (O. humifusa) processing facilities.

Strain Improvement and Bioprocess Optimization for Enhanced Production of Haluronic Acid(HA) in Bioreactor Cultures of Streptococcus zooepidemicus (히알루론산 생산성 향상을 위한 Streptococcus zooepidemicus 균주 개량 및 발효조 배양공정 최적화)

  • Strain improvement and bioprocess development were undertaken to enhance hyaluronic acid(HA) production by Streptococcus zooepidemicus cells. Using a high-yielding mutant strain, statistical medium optimization was carried out in shake flask cultures, resulting in 52% increase in HA production (5.38 g/l) at the optimal medium composition relative to the parallel control cultures. For sufficient supply of dissolved oxygen (DO), which turned out to be crucial for enhanced production of HA, agitation system and speed were intensively investigated in 5 L bioreactor cultures. Increase in oxygen mass transfer coefficient (kLa) through increment of agitation speed (rpm) and 35% expansion of diameter of the newly-designed impellers showed significantly positive effects on HA production. By installing an expanded Rushton-turbine impeller for efficient break-down of sparged air, and an extended marine impeller above the Rushton-turbine impeller for efficient mixing of the air-born viscous fermentation broth, maximum amount of HA (9.79 g/l) was obtained at 450 rpm, 1.8 times higher level than that of the corresponding flask culture. Subsequently, the possibility of bioprocess scale-up to a 50 L bioreactor was investigated. Despite almost identical maximum HA production (9.11 vs 9.25 g/l), the average HA volumetric productivity (rp) of the 50 L culture turned out only 74% compared to the corresponding 5 L culture during the exponential phase, possibly caused by shear damages imposed on the producing cells at the high stirring in the 50 L culture. The scale-up process could be successfully achieved if a scale-up criterion of constant oxygen mass transfer coefficient (kLa) is applied to the 50 L pilot-scale bioreactor system.

An Analytical Study of National and International Care Label Systems of Textile and Apparel Products

  • This paper enables stakeholders involved in textile industry to gain an overview of standards used for care labelling and help establish a common standard that could be used as a universal standard. This study provides a comprehensive and detailed analytical study of care labelling standards adopted by common countries in the textile market. It was found that the development of a universal system for care labeling could enhance the trade of textile articles and assist consumers in caring for textile articles. Universal care label systems could be characterized by two main features of inclusiveness and comprehensiveness. The range of instructions and symbols presented were found different among standards. Insignificant differences in symbols' shapes were found between standards for bleaching, ironing and professional cleaning. The washing process had the widest variety of instructions; in addition, options were provided by stated standards. Different meanings were found for similar shapes in some tumble drying symbols. The study findings show the importance of enhancing text based standards or the development of an understandable format across as many cultures as possible. The unification of symbols and meanings may be needed to provide global consumers consistent guidance. The efficiency of a detailed standard that provides and covers a wide range of instructions is an important aspect. The visibility and practicality of offering variable options/symbols in one standard is an important aspect for developing a universal care label system.

Performance Enhancement of Gas-Liquid Mixed Plasma Discharge System using High Speed Agitation (고속 교반을 이용한 기-액 혼합 플라즈마방전 시스템의 성능 향상)

  • Dielectric Barrier Discharge (DBD) plasma is a new technique for use in environmental pollutant degradation, which is characterized by the production of hydroxyl radicals as the primary degradation species. Due to the short lifetime of the chemically active species generated during the plasma reaction, the dissolution of the plasma gas has a significant effect on the reaction performance. The plasma reaction performance can be enhanced by combining the basic plasma reactor with a homogenizer system in which the bubbles are destroyed and turned into micro-bubbles. For this purpose, the improvement of the dissolution of plasma gas was evaluated by measuring the RNO (N-dimethyl-4-nitrosoaniline, an indicator of the generation of OH radicals). Experiments were conducted to evaluate the effects of the diameter, rotation speed, and height of the homogenizer, pore size, and number of the diffuser and the applied voltage on the plasma reaction. The results showed that the RNO removal efficiency of the plasma reactor combined with a homogenizer is two times higher than that of the conventional one. The optimum rotor size and rotation speed of the homogenizer were 15.1 mm, and 19,700 rpm, respectively. Except for the lowest pore size distribution of $10-16{\mu}m$, the pore size of the diffuser showed little effect on RNO removal.

The Effect of Weight-support Treadmill Training on the Balance and Activity of Daily Living of Children with Spastic Diplegia

  • Purpose: This is designed to study the effect of weight-support walking training through motor learning on motor functions of children with cerebral palsy, in particular their activity of daily living and balance. Methods: Thirteen children with spastic cerebral palsy, at gross motor function classification system (GMFCS) levels III~IV, underwent treadmill walking training. It used principles of weight support, 4 times a week for 7 weeks, 10 minutes at a time, before and after neurodevelopmental physical therapy. Everyday functions were measured using Functional Independence Measure for Children (Wee-FIM). The ability to keep their balance was measured using electronic measuring equipment from good balance system and the assessment was made before and after the experiment. Results: There were significant differences (p<0.05) between pre and post experiment levels of functional independence in everyday life, in self-care activities, mobility, locomotion and social cognition. With regard to changes in standing balance, there were significant differences before and after the experiment (p<0.05) in GMFCS level III. There was a reduction in the agitation velocity in the x- and y-axes which measures the left-to-right shaking; in GMFCS level IV, velocity moment was reduced. Conclusion: Walking training using a treadmill can help improve the everyday activity and balance in children with spastic cerebral palsy. It can also be served as a useful purpose as a method of intervention in pediatric care.

New Approach for Detecting Leakage of Internal Information; Using Emotional Recognition Technology

  • Currently, the leakage of internal information has emerged as one of the most significant security concerns in enterprise computing environments. Especially, damage due to internal information leakage by insiders is more serious than that by outsiders because insiders have considerable knowledge of the system's identification and password (ID&P/W), the security system, and the main location of sensitive data. Therefore, many security companies are developing internal data leakage prevention techniques such as data leakage protection (DLP), digital right management (DRM), and system access control, etc. However, these techniques cannot effectively block the leakage of internal information by insiders who have a legitimate access authorization. The security system does not easily detect cases which a legitimate insider changes, deletes, and leaks data stored on the server. Therefore, we focused on the insider as the detection target to address this security weakness. In other words, we switched the detection target from objects (internal information) to subjects (insiders). We concentrated on biometrics signals change when an insider conducts abnormal behavior. When insiders attempt to leak internal information, they appear to display abnormal emotional conditions due to tension, agitation, and anxiety, etc. These conditions can be detected by the changes of biometrics signals such as pulse, temperature, and skin conductivity, etc. We carried out experiments in two ways in order to verify the effectiveness of the emotional recognition technology based on biometrics signals. We analyzed the possibility of internal information leakage detection using an emotional recognition technology based on biometrics signals through experiments.

Kinetics of Horseradish Peroxidase-Catalyzed Nitration of Phenol in a Biphasic System

  • The use of peroxidase in the nitration of phenols is gaining interest as compared with traditional chemical reactions. We investigated the kinetic characteristics of phenol nitration catalyzed by horseradish peroxidase (HRP) in an aqueous-organic biphasic system using n-butanol as the organic solvent and ${NO_2}^-$ and $H_2O_2$ as substrates. The reaction rate was mainly controlled by the reaction kinetics in the aqueous phase when appropriate agitation was used to enhance mass transfer in the biphasic system. The initial velocity of the reaction increased with increasing HRP concentration. Additionally, an increase in the substrate concentrations of phenol (0-2 mM in organic phase) or $H_2O_2$ (0-0.1 mM in aqueous phase) enhanced the nitration efficiency catalyzed by HRP. In contrast, high concentrations of organic solvent decreased the kinetic parameter $V_{max}/K_m$. No inhibition of enzyme activity was observed when the concentrations of phenol and $H_2O_2$ were at or below 10 mM and 0.1 mM, respectively. On the basis of the peroxidase catalytic mechanism, a double-substrate ping-pong kinetic model was established. The kinetic parameters were ${K_m}^{H_2O_2}=1.09mM$, ${K_m}^{PhOH}=9.45mM$, and $V_{max}=0.196mM/min$. The proposed model was well fit to the data obtained from additional independent experiments under the suggested optimal synthesis conditions. The kinetic model developed in this paper lays a foundation for further comprehensive study of enzymatic nitration kinetics.
