노화와 피부노화에 대한 고찰 (Aging and Skin Aging)

  • In Oriental medicine, aging is just a natural process like change of seasons. Ancient Oriental people accepted it as a natural thing to be growing older and to die at last. The science of aging has advanced dramatically. In the last 2 decades, advances in genetics and molecular biology have led to extraordinary new understandings in how cells age, how apoptosis programs cells to die, and how neuroendocrinology plays a role in the lifespan of organisms. Today, the matter of primary concern about aging is a cellular and mitochondrial damage of human body induced by reactive oxygen species(ROS). The skin aging can be divided into two areas, intrinsic(chronologic)-aging and photo-aging. There are lots of photo damage about skin aging. The skin is increasingly exposed to ultraviolet(UV) irradiation in life. Therefore, the risk of photo-oxidative damage of the skin induced by reactive oxygen species(ROS) has increased substantially. Nowadays, many people believe that they can stop or at least delay the process of aging. There are lots of treatments that promise to slow the process of aging and the associated ailments. Many of these treatments, for example, exercise, Vit E, Vit C therapy, hormone therapy, restrict diet, are gradually being subjected to clinical trials. But in spite of all efforts, researches and investigations, there is no single method or treatment which is revealed to be truly effective for delaying progress of aging. Every methods insisted on effect for delaying aging process, has its dark side. All we can do is just keeping ourself healthy until the time of death.

노인의 신체이미지와 성공적 노화의 관계에서 성장 마인드셋과 희망의 이중 매개효과 (The Dual Mediating Effects of Growth mindset and Hope in the Relationship between Body Image and Successful Aging of the Elderly)

  • 본 연구는 노인의 신체이미지가 성공적 노화에 미치는 영향에서 성장 마인드셋과 희망의 이중 매개효과를 검증하는데 목적이 있다. 이를 위해 충청남도 D시, S시 및 D광역시의 노인 422명을 대상으로 설문조사하였고 수집한 자료는 SPSS 25.0과 PROCESS macro 3.4 프로그램을 활용하여, 빈도 분석, 신뢰도 분석, 상관 분석 및 부트스트랩을 통한 이중 매개 분석을 수행하였다. 연구의 결과는 다음과 같다. 첫째, 상관분석 결과 각 변인들 간에는 통계적으로 유의미한 정적 상관관계가 있었다. 둘째, 경로분석 결과 신체이미지는 성장 마인드셋과 희망에 정적 영향을 미쳤고, 성장 마인드셋은 희망과 성공적 노화에 정적인 영향을 주었으며, 희망 또한 성공적 노화에 유의미한 영향을 미쳤다. 셋째, 이중 매개효과 분석 결과 신체이미지와 성공적 노화의 관계에서 성장 마인드셋과 희망은 이중 매개하였다. 본 연구는 노인들의 성공적인 노화를 위한 신체이미지, 성장 마인드셋 및 희망을 활용한 새로운 모델과 기초자료를 제공하였다.

동결온도가 해동후 숙성한 우육의 품질에 미치는 영향 (Effect of Freezing Temperature on the Quality of Beef Loin Aged after Thawing)

  • This study was carried out to investigate the effects of freezing temperature on the quality of thawing aged beef loin. Drip loss was higher at 3oC freezing than at 20oC freezing, showing 17.21% drip loss after 6 days aging by 3oC freezing, 14.92% drip loss 12 days aging by 20oC freezing. Cooking loss by both water bath and pan boiling were decreased with increased in aging days. The salt soluble protein extractability of the beef loin was increased until 9 days aging by both 3oC and 20oC freezing, after that was decreased. The L value of the beef loin was high until 9 days aging by 3oC freezing, after that the L value of that was decreased. And the aging at 20oC freezing was high significantly with increased aging days. The a value of the beef loin was low significantly in 6 and 9 days aging by 3oC freezing, 20oC freezing was low significantly with increased aging days. The b value of the beginning of aging was higher with increased aging days. The percentage of denatured myoglobin of the beginning of aging was the highest, then those of 3oC and 20oC freezing showed 89.70% and 88.00%, respectively. The shear force of the beef loin was decreased with aging days, but the myofibrillar fragmentation index increased with aging days. The pH of the beef loin increased until 6 days of aging by both 3oC and 20oC freezing, after that the pH decreased.

Quercetin induces apoptosis and cell cycle arrest in triple-negative breast cancer cells through modulation of Foxo3a activity

  • Quercetin, a plant-derived flavonoid found in fruits, vegetables and tea, has been known to possess bioactive properties such as anti-oxidant, anti-inflammatory and anti-cancer. In this study, anti-cancer effect of quercetin and its underlying mechanisms in triple-negative breast cancer cells was investigated. MTT assay showed that quercetin reduced breast cancer cell viability in a time and dose dependent manner. For this, quercetin not only increased cell apoptosis but also inhibited cell cycle progression. Moreover, quercetin increased FasL mRNA expression and p51, p21 and GADD45 signaling activities. We also observed that quercetin induced protein level, transcriptional activity and nuclear translocation of Foxo3a. Knockdown of Foxo3a caused significant reduction in the effect of quercetin on cell apoptosis and cell cycle arrest. In addition, treatment of JNK inhibitor (SP 600125) abolished quercetin-stimulated Foxo3a activity, suggesting JNK as a possible upstream signaling in regulation of Foxo3a activity. Knockdown of Foxo3a and inhibition of JNK activity reduced the signaling activities of p53, p21 and GADD45, triggered by quercetin. Taken together, our study suggests that quercetin induces apoptosis and cell cycle arrest via modification of Foxo3a signaling in triple-negative breast cancer cells.

이정환(二精丸)이 노화과정에 미치는 영향 (Effects of Ichungwhan on the Aging Process)

  • Objectives: It is well known that aging and aging-related diseases are linked to the increased level of oxidative stress caused by reactive oxygen species(ROS) and reactive nitrogen species(RNS). Nonprotein-SH decreases during aging, while substances such as ROS, nitric oxide(NO), peroxynitrite($ONOO^-$), myeloperoxidase(MPO), and dityrosine show a significant increase. This study investigated the effect of Ichungwhan on the aging process by examining its effect on the generation of the above-mentioned substances. Methods: Four comparison groups of SD rats were used. They were 6 month-old rats, 24 month-old rats, and 24 month-old rats fed with food containing 0.1% and 0.3% of Ichungwhan extract. The amount of NO, $ONOO^-$, and ROS in the rats' kidneys were examined using a fluorescence microplate reader. The reagents used for this purpose include: dihydrorhodamine 123 (DHR 123), 2',7' -dichlorodihydrofluorescein, diacetate(DCFDA), and 4,5-diaminofluorescein(DAF-2). A spectrophotometer was used to investigate the reactivity of nonprotein-SH and myeioperoxidase(MPO), using reagents such as trichloroacetic acid(TCA) and tetramethylbenzidine(TMB). The amounts of MPO protein and dityrosine were measued by western blot. Results: The observed effects of Ichungwhan on rats were as follows: increase of nonprotein-SH; effective decrease of RNS level by suppression of the generation system of $ONOO^-$ and NO; decrease of ROS level; decrease of the MPO reactivity and the subsequent reduction of amount of MPO protein; retardation of dityrosine formation. It can be hypothesized, therefore, that Ichungwhan affects both the earlier and later phases of the molecular inflammatory process, and retards the aging process. Conclusions: Empirical evidence in this study supports a role for Ichungwhan in generation mechanisms of aging process-linked substances ROS, NO, $ONOO^-$, nonprotein-SH, MPO and dityrosine. Affects contrary to the aging process observed in rats beg further empiricism to investigate potential application of Ichungwhan as a medication for age-related diseases in humans.

지역사회 노인의 성공적 노화 예측요인과 초월감의 매개효과 (Predictors of Successful Aging in Community-dwelling Elders and Mediation Effect of Transcendence)

  • 본 연구의 목적은 지역사회 노인의 성공적 노화에 미치는 영향 요인을 파악하고 초월감의 매개효과를 확인하여 고령화 사회에서의 성공적 노화를 위한 간호중재의 기초자료를 마련하고자 함이다. 연구대상은 65세 이상 지역사회에 거주하는 노인 162명이었으며, 자료분석은 PASW Statistics 18.0 프로그램을 이용하였고 다중회귀분석을 실시하였다. 분석결과, 초월감, 자녀관계, 우울, 영성, 주관적 경제수준은 성공적 노화의 유의한 영향요인으로 설명력은 77%로 나타났다. 또한 자녀관계, 우울, 영성, 주관적 경제수준과 성공적 노화의 관계에서 초월감이 매개역할을 하는 것을 확인할 수 있었다. 본 연구 결과는 지역사회 노인의 성공적 노화 수준 향상을 위한 간호중재 전략 개발에 활용할 수 있을 것이며, 특히 노년기의 초월감에 대해 재조명할 수 있었다는 데에 의의가 있다.

숙성에 따른 한우 우둔의 풍미 전구물질 및 향기성분 변화 (The Effect of Aging on Flavor Precursors and Volatile Compounds of Top Round from Hanwoo)

  • The influence of aging on the flavor precursors and volatile compounds of top round beef was studied. The concentrations of free amino acids, nucleotides, creatine, dipeptides, and volatile compounds were measured after top round from Hanwoo was aged at $4^{\circ}C$ for 21 days. The amount of free amino acids in top round significantly increased with the increase of aging period. There was no effect of aging on the concentrations of adenosine monophosphate or inosine in top round. The inosine monophosphate content of top round significantly decreased with age, while the hypoxanthine content increased. The concentrations of creatine, carnosine, and anserine in top round were not influenced by aging. In total, 24 volatile compound were identified in aged, cooked top round. Of these, the quantities of aldehydes (propanal, pentanal, hexanal, heptanal, octanal, and nonanal), hydrocarbons (pentane and octane), 2-butanone, ethyl acetate, and pyridines (4-ethynyl-pyridine and 4-acetyl-pyridine) significantly increased after aging. We conclude that the flavor of top round can be improved by aging.

노인의 노화태도가 삶의 만족에 미치는 영향: 부모-자녀 간 결속의 다중매개효과 검증 (The Effect of the Aging Attitude on Life Satisfaction of Korean Elderly: Multi-mediation Effects of Solidarity between Parents and Children)

  • 본 연구는 노인의 노화에 대한 태도가 이들의 삶의 만족에 미치는 영향을 확인하고, 이를 매개하는 요인으로써 부모-자녀 간의 결속감의 역할을 확인하는 것을 목적으로 한다. 이를 위하여 전국의 노인 2,067명을 대상으로 설문조사를 진행하였고, SPSS macro PROCESS v.2.16을 활용하여 다중매개효과를 검증한 결과는 다음과 같다. 첫째, 노인의 노화에 대한 태도는 삶의 만족에 유의미한 영향을 미치고 있었다. 둘째, 노인의 노화태도는 부모-자녀 간의 결속감의 다섯가지 하위차원에 있어 모두 유의미한 영향을 미치고 있었다. 특히 노화에 대하여 부정적인 태도를 가질수록 부모-자녀 간의 결속이 더욱 높아지는 결과를 보이고 있었다. 셋째, 부모-자녀 간 결속의 하위차원들 중 애정적 일치적 기능적 결속이 노인의 노화태도와 삶의 만족간의 관계를 매개하는 것으로 나타났으며, 이 중 애정적 결속이 가장 큰 효과를 보이는 것으로 나타났다. 이상의 결과를 바탕으로 노인의 삶의 만족을 위한 실천적 개입방안을 제안하였다.

탄소섬유/에폭시 복합재 링 시편의 노화 특성 평가 (Aging Characteristics of Carbon Fiber/Epoxy Composite Ring Specimen)

  • 가속노화장치를 이용하여 노출시간에 따른 탄소섬유/에폭시 복합재 링 시편의 노화 특성을 평가하였다. 온도, 수분, 자외선 등의 복합적인 노출조건은 최대 3000시간까지 적용하였다. 노출시간에 따른 링 시편의 인장특성과 곡률의 영향이 고려된 굽힘특성은 하중시험장치를 이용하여 평가하였으며 노화표면은 전자현미경을 통해 관찰하였다. 연구결과에 따르면 인장강성은 환경인자의 노출시간에 큰 영향을 받지 않지만 인장강도는 노출 초기에 물리시효 현상과 경화 반응으로 다소 증가하지만 노출시간이 길어지면 열화 영향으로 감소하였다. 굽힘탄성계수와 굽힘강도는 노출초기에 다소 증가하지만 노출시간이 길어지면 감소하는 양상을 나타내었다. 또한 전자현미경을 통해 관찰된 노화표면은 환경인자의 노출시간에 따라 달라지기 때문에 환경인자의 노출시간에 따른 기계적 특성의 변화를 규명하기 위한 정보를 제공해 준다.

ACCELERATED AGING USING $FOCAS^{(R)}$-A BURNER BASED SYSTEM SIMULATING AN ENGINE

  • Accelerated aging of engine exhaust system components such as catalytic converters are traditionally performed using an engine/dynamometer test stand. $SwRI^{(R)}'s\;FOCAS^{(R)}$ system reduces or eliminates many of the engine based aging limitations. This paper will describe several studies. These include: 1) replication of engine based catalyst aging cycles with added precision and dependability; 2) catalyst aging with and without lubricating oil effects; 3) effects of lubricant phosphorus on catalyst performance; and 4) the potential to thermally age components beyond the capabilities of engine based systems. The first study includes the development of the SwRI FOCAS system to run programmed aging conditions with or without lubricating oil. A description of the subsystems is given. The second two studies used the SwRI FOCAS system to age catalysts. One study compared thermal-only aging using of the SwRI FOCAS system with equivalent aging on a traditional engine/dynamometer test stand. The other study examined the effect on catalyst performance of two lubricating oils containing different levels of phosphorus, and compared the results to field data generated using the same oils in a fleet of vehicles.
