A Missense Variant (R239Q) in CCN3 Induces Aberrant Apoptosis in the Developing Mouse Brain

  • CCN3 (also known as NOV, Nephroblastoma overexpressed) proteins are involved in various pathologies during different developmental stages. We have previously shown that intracellular levels and normal extracellular secretion of CCN3 are important for neuronal differentiation. Furthermore, we demonstrated that a single amino acid in the CCN3 TSP-1 domain is important for extracellular secretion and that palmitoylation of CCN3 is required in this process. However, the effect of abnormal CCN3 accumulation on cells remains to be studied. Here, we found mutations in the TSP-1 domain of CCN3 that led to intracellular accumulation and abnormal aggregation of CCN3. It was observed that this mutation resulted in a phenomenon similar to neurodegeneration when overexpressed in the developing mouse cortex. This mutation also confirmed the activation of apoptotic gene expression in Neuro2a cells. In addition, we confirmed the in vivo transcriptional changes induced by this mutation using microarray analysis. We observed a significant increase in the expression of Anp32a, an apoptosis-related gene. Collectively, these results indicate that a single mutation in CCN3 can lead to abnormal cell death if it shows intracellular accumulation and abnormal aggregation.

Patient-specific pluripotent stem cell-based Parkinson's disease models showing endogenous alpha-synuclein aggregation

  • After the first research declaring the generation of human induced pluripotent stem cells (hiPSCs) in 2007, several attempts have been made to model neurodegenerative disease in vitro during the past decade. Parkinson's disease (PD) is the second most common neurodegenerative disorder, which is mainly characterized by motor dysfunction. The formation of unique and filamentous inclusion bodies called Lewy bodies (LBs) is the hallmark of both PD and dementia with LBs. The key pathology in PD is generally considered to be the alpha-synuclein (${\alpha}$-syn) accumulation, although it is still controversial whether this protein aggregation is a cause or consequence of neurodegeneration. In the present work, the recently published researches which recapitulated the ${\alpha}$-syn aggregation phenomena in sporadic and familial PD hiPSC models were reviewed. Furthermore, the advantages and potentials of using patient-derived PD hiPSC with focus on ${\alpha}$-syn aggregation have been discussed.

Introduction of Whidam's Su-Gi therapy - Focused on Cervical spine (휘담식 수기요법의 소개 - 경추부를 중심으로)

  • Objective : The purpose of this study is to introduce Whidam's Su-Gi therapy(WST.) as a remarkable traditional manual therapy. Main subject : WST. is a traditional manual therapy from Do-In(導引) An-Kyo(按蹻) that treats disease by controlling Gi(Gi-Gong therapy). It makes treatment effects by pushing Sa-Gi(邪氣) which becomes aggregation-accumulation(積聚) to right places by using pointillage, stroking, and rubbing with doctor's focused intention(意念). The cervical spine connects body and brain, so treating cervical spine could be a foundation of all treatment. Discussion : WST. is a manual therapy that treats not only muscles, nerves, vessels, joints but also Oriental medicine concept-meridian, acupoint and Gi(氣). Conclusions: 1) WST. is a traditional manual therapy using Gi-Gong, 2) WST. changes Sa-Gi(邪氣) into Jung-Gi(正氣), which means curing aggregation-accumulation(積聚), 3) Cervical WST. treats a passage that connects upper and lower parts of human body; heart and brain, and 4) Cervical WST. is a treatment to activate So-Ju-Cheon(小周天), which controls human body immunity.

Hypocholesterolemic Effect of Tangerine Pulp, Sea Tangle or Prickly Pear Cactus on Lipid Level, Intestinal Cholesterol Absorption, Platelet Aggregation and Liver Tissue in Hypercholesterolemic Rats (감귤박, 다시마, 손바닥 선이장 분말을 함유한 식이의 급여가 고콜레스테롤혈증 흰쥐의 체내 지질수준과 장내 콜레스테롤 흡수, 혈소판 응집성 및 간 조직에 미치는 영향)

  • This study done to investigate the hypocholesterolemic effect of ow dietary fiber in a high cholesterol diet in aspect of cholesterol absorption and excretion using Sprague Dawley rats. After feeding diets containing 0.5% cholesterol and 5% tangerine pulp, sea tangle or prickly pear cactus for four weeks, we measured the levels of plasma and liver cholesterol and triglyceride, fecal neutral sterols and fecal radioactivity after ingestion 14(sup)C-cholesterol. We also examined platelet aggregation and histological change in liver tissues in association of hypercholesterolemia. The liver to body weight ratio was significantly(p<0.01) lower in rats fed prickly pear cactus than in other groups. The levels of plasma total cholesterol, LDL-cholesterol and triglyceride were decreased significantly(p<0.01) in the prickly pear cactus group compared with the control, while there was no difference in the liver levels of total cholesterol and triglyceride among groups. Fecal corprostanol and cholesterol were significantly(p<0.01) higher groups of control and prickly pear cactus compared to the other two groups. Radioisotope excretion after ingestion of 14(sup)C-cholesterol was higher in the control group than in tangerine pulp group or sea tangle group, with the highest in prickly pear group. Radioisotope excretion was the highest during the 2nd and 3rd days in all groups. Hematocrit and platelet aggregation were decreased in all fiber groups compared to the control, but not statistically different. Microscopic examination showed that cholesterol diet cause a fat accumulation in the liver and prickly pear cactus decreased the fat accumulation. Result indicates that prickly pear cactus has hypocholesterolemic effect by decreasing absorption and increasing excretion of cholesterol, thereby protective effect on fatty liver. Control group fed diet containing high cholesterol and low fiber seems to have a self control system in cholesterol absorption and excretion preventing hypercholesterolemia. Analysis of Variation for Drainage Structure with Flow Direction Methods on the Basis of DEM (DEM을 기반으로 한 흐름방향 모의기법에 따른 배수구조의 변동성 해석)

  • The main purpose of this study is to suggest and recommend the more reliable flow direction methods within the framework of DEM and power law distribution, by investigating the existing methodologies. To this end SFD (single flow direction method), MFD (multiple flow direction method) and IFD (Infinite flow direction method) are applied to analyze the determination of a flow direction for the water particles as seen in the Jeonjeokbigyo basin, and then assessed with respect to the variation of flow accumulation in that region. As the main results revealed, the study showed the different patterns of flow accumulation are found out from each applications of flow direction methods utilized in this study. This brings us to understand that as the flow dispersion on DEM increases, in this case the contributing areas to the outlet grow in sequence of SFD, IFD, MFD, but it is noted that the contribution of individual pixels into outlet decreases at that time. In what follows, especially with the MFD and IFD, the result tends to make additional hydrologic abstraction from rainfall excess, as noted due to the flow dispersion within flow paths on DEM. Based on the parameter estimation for a power law distribution, which is frequently used for identify the aggregation structure of complex system, by maximum likelihood flow accumulation can be thought of as a scale invariance factor. In this regard, the combination of flow direction methods could give rise to the more realistic water flow on DEM, as revealed through the separate flow direction methods as utilized for dispersion and aggregation effects of water flow within the available different topographies.

A Study on Spatial Aggregation Method for Path Travel Time Estimation using Hi-Pass DSRC System (하이패스 DSRC 기반의 경로통행시간 산정을 위한 공간적 집계방안 산정에 관한 연구)

  • PURPOSES : This investigational survey is to observe a proper spatial aggregation method for path travel time estimation using the hi-pass DSRC system. METHODS : The links which connect the nodes of section detectors location are used for path travel time estimation traditionally. It makes some problem such as increasing accumulation errors and processing times. In this background, the new links composition methods for spatial aggregation are considered by using some types of nodes as IC, JC, RSE combination. Path travel times estimated by new aggregation methods are compared with PBM travel times by MAE, MAPE and statistical hypothesis tests. RESULTS : The results of minimum sample size and missing rate for 5 minutes aggregation interval are satisfied except for JC link path travel time in Seoul TG~Kuemho JC. Thus, it was additionally observed for minimum sample size satisfaction. In 15, 30 minutes and 1 hour aggregation intervals, all conditions are satisfied by the minimum sample size criteria. For accuracy test and statistical hypothesis test, it has been proved that RSE, Conzone, IC, JC links have equivalent errors and statistical characteristics. CONCLUSIONS : There are some errors between the PBM and the LBM methods that come from dropping vehicles by rest areas. Consequently, this survey result means each of links compositions are available for the estimation of path travel time when PBM vehicles are missed.

The Effect of Woohwangcheongsim-won on Circulatory Disturbance in Diabetes (우황청심원이 당뇨병 Rat의 혈액순환장애에 미치는 영향)

  • Object: Death rate due to hypertension, atherosclerosis, ischemic heart disease and cerebral infarction induced by Westernized diet and increased average life span is on the rise. Decrease in blood circulation, activation of thrombus generation and intravascular lipid accumulation, cited as the principal causes of the above mentioned diseases in recent studies, result in circulatory disturbance and blood vessel obstruction leading to ischemic cell death of heart, brain and peripheral vessels. Method: We investigated the biochemical changes in microvascular permeability, aggregation of platelet and the intravascular lipid accumulation in induced-diabetic rat using Streptozotocin. We also studied the effects of Woohwangcheongsirn-won after oral administration on blood circulation, platelet function and lipid metabolism. The results are as follows: I. Woohwangcheongsim-won increased blood circulation in microvessels. 2. Woohwangcheongsim-won increased the reduced erythrocyte deformability in diabetes. 3. Woohwangcheongsim-won induced the reduction of contents of 2, 3-DPG, but failed to affect the reduced contents of ATP in erythrocyte in diabetes. 4. Woohwangcheongsim-won reduced the activity of Ca/sup 2+/-ATPase in the membrane of erythrocyte. 5. Woohwangcheongsim-won reduced the platelet aggregation evoked by platelet agglutinin factor. 6. Woohwangcheongsim-won reduced the production of platelet-derived granules. 7. Woohwangcheongsim-won reduced the production of metabolites of arachidonic acid in diabetes, and also reduced the production of increased thromboxane B2. 8. Woohwangcheongsim-won reduced the synthesis of oxidized LDL-cholesterol. In conclusion, Woohwangcheongsim-won enhanced blood circulation in microvesseles, erythrocyte deformability and inhibited the increased platelet aggregation and the synthesis of oxidized LDL-cholesterol in diabetes. Therefore Woohwangcheongsim-won is believed to positively affect blood circulation (J Korean Oriental Med 2002;23(2):164-179)
