Miamiensis avidus is a scuticociliate causing mortality in olive flounder Paralichthys olivaceus. To evaluate immune response of olive flounder against M. avidus, 2.6×106cells/fish of Formalin killed cell (FKC) was intraperitoneally (i.p.) injected, and 2.4 × 106cells/㎖ of sonicated FKC was immersion immunized to 14.9 cm (26.8g) fish. Fish were immunized 2 times with 2 weeks intervals. Antiserum from immunized fish caused agglutination and immobilization of the ciliate. In ELISA test, immunized group exhibited higher titers than control group. In addition, i.p. immunized fish showed higher phagocytic ratio, phahgocytic index and chemotaxic activity. To evaluate in vivo efficacy of FKC on fish, 1.43 × 105cells/fish was i.p.. injected, and 2.2 × 105 cells/㎖ of sonicated cell was immersed into 8.6cm (6.3g) fish. After 2 times immunization with 2 weeks intervals, fish were infected with 2.0 × 104 and 2.0 × 103 cells/ml of live ciliates by immersion. After 3 weeks, cumulative mortality was lower in the i.p. immunized group and mortality was delayed in immersion immunized group. In conclusion, specific immune response of oliver flounder against M. avidus was elevated after immunization and these immune response may prevent and/or delay for the M. avidus infection to olive flounder.
Objectives: This study was conducted in order to evaluate the microbiological contamination of the indoor air of the lunchrooms at child care centers and investigate the toxin genes and toxin production ability of food-borne pathogens. Methods: A total of 64 child care centers were sampled to test total aerobic bacteria, coliform bacteria, fungi, Staphylococcus aureus, Bacillus cereus and Salmonella spp. according to the Korea Food Code. All toxin genes of pathogens were detected using the Polymerase Chain Reaction method. The Sthaph. aureus enterotoxin was detected by a Staphylococcus aureus enterotoxin-reversed passive latex agglutination kit. The heamolysin BL (HBL) and non-heamolytic enterotoxin (NHE) produced by B. cereus were detected using a B. cereus enterotoxin-reversed passive latex agglutination kit and Bacillus diarrheal enterotoxin visual immunoassay kit, respectively. Results: The means of total aerobic bacteria and coliform bacteria were $1.91{\pm}1.84$ log CFU/plate and $0.47{\pm}0.62$ log CFU/plate, respectively. The mean of fungi also showed $0.59{\pm}0.71$ log CFU/plate. Among the pathogenic bacteria tested in this study, Staphy. aureus and B. cereus were detected in four (6.3%) and 21 (32.8%) out of 64 indoor air samples from lunchrooms in child care centers, respectively. All Staphy. aureus tested in this study possessed no toxin genes and did not produce enterotoxin. The detection rate of nheABC, hblCDA, entFM and ces toxin gene in B. cereus was 100, 57.1, 76.2 and 0%, respectively. B. cereus isolates were classified into four groups according to the presence or absence of toxin genes. The nheABC gene was the major toxin gene among B. cereus tested in this study. The HBL was detected in 11 out of 21 B. cereus isolates (52.4%) and three B. cereus isolates produced NHE (14.3%). Conclusion: The results indicated that the contamination by microorganisms in the indoor air of lunchrooms was unqualified to supply safe catering in child care centers. The ongoing control of indoor air quality is required.
To avoid the self-agglutination of Staphylococcus aureus sensitized with rabbit antibody in the absence of antigen, we determined the optimum concentration of rabbit antibody for sensitization. It was analyzed by using three different kinds of S. aureus strains at various concentraions of antibody. The optimized coagglutination test using the S. aureus sensitized with rabbit antibody was applied to the diagnosis of edwardsiellosis in field and in laboratory. The presence of E. tarda as low as $10\;{\mu}g/ml$ was detected by this method. Moreover, it showed good coagglutination results against several different forms of antigens such as FKC, EDTA or heat extracted antigen of E. tarda. E. tarda strains, isolated from the flounders suffering from edwardsiellosis in fields, showed some cross-reactions to the E. tarda 219 analyzed by both agglutination and coagglutination test with rabbit anti-E, tarda 219 antibody. The degree of cross-reactions analyzed was enough to apply the coagglutination test for the diagnosis of edwardsiellosis in field. Thus, even 1,000 fold diluted tissue homogenate of infected flounder naturally contained enough E. tarda as an antigen to show good coagglutination with S. aueus sensitized with rabbit anti-E, tarda 219 antibody. The successful application of this method to the homogenate and heat extract of tissues from naturally or artificially infected flounder or tilapia preyed that coagglutination test was a simple and rapid reliable dignostic technique suitable for using in laboratory and field without any special equipments.
This study was conducted to determine the serum antibodies against toxoplasma in the artificially infected dogs, pet and street dogs by latex agglutination (LA) and indirect fluorescent antibody (IFA) test. LA test was carried out with commercial Toxo-MT kit (Eiken chemical Co.) and IFA test was carried out with rabbit-anti-dog IgG labelled with FITC (Cappel Co.) and toxo-antigen slides prepared in laboratory. The results obtained were as follows ; 1. Antibodies to Toxoplasma gondii in the artificially infected dogs were detected firstly at the Day 8 in IFA and Day 9 in LA test after inoculation. Positive antibody reactions by these tests were declined gradually afterward but maintained up to 12 weeks. 2. In LA test serum antibody titers in 310 test sera were shown as 10 cases(32%) in 1 : 32.5(1.0%) in 1 : 64, 4(1.3%) in 1 : 128 and 2(0.7%) in 1 : 256. In IFA test serum antibody titers 310 test sera were shown as 17 cases(5.5%) in 1 : 64, 8(2.6%) in 1 : 128 and 5(1.6%) in 1 : 256. 3. In the total of 310 sera from pet and street dogs toxoplasma antibody positive rates were 21 cases (6.8%) by LA and 30 cases (9.7%) by IFA test and the positive detection rates between these two groups by LA and IFA test were not significant(p<0.05). 4. In the total of 115 sera from pet dogs toxoplasma antibody positive rates were 12 cases(10.4%) by LA and 15(13.0%) by IFA test. And in the 195 street dogs the positive rates were 9 cases(4.6%) by LA and 15(7.7%) by IFA test. Also, the positive detection rates between these two groups by LA and IFA test were not significant(p<0.05). 5. Agreement of reactivity between LA and IFA test for 310 sera was 91.3% in total of 283 cases consisting of 12 cases(3.9%) of both LA and IFA positive and 271 cases(87.4%) of LA and IFA negative. 6. LA test was almostly equivalent to the IFA test in producibility and proved to be a simple tool for the screening of toxoplasma antibody in laboratory.
A total of 377 pregnant women, 43 pelvic tumor patients and 80 of multiphysic health center persons as controls were examined by indirect latex agglutination test in order to evaluate Toxoplasma antibody titers at Kang-Nam St. Mary's Hospital in Seoul. Throughout this survey, 1 : 32 or more titers of diluted sera were regarded as positive. 1. The 377 samples of test sera in pregnant women showed negatives in 319 cases (84.6%), 1 : 2 in 44 cases (11.7%), 1:4 in 9 cases (2.4%), 1:8 in 2 cases (0.5%), 1:16 in 1 case (0.3%) and 1 : 32 in 2 cases (0.5%) respectively. 2. The 43 samples of test sera in pelvic tumor patients showed negatives in 29 cases (67.4%), 1:2 in 8 cases(18.6%), 1:4 in 1 case(2.3%), 1:16 in 2 cases (4.7%), 1:32 in 1 case(2.3%) and 1 : 128 in 2 cases (4.7%). 3. The 80 samples of test sera in multiphysic health center persons as controls negatives in 56 cases (70.0%), 1.2 in 19 cases (23.8%), 1:4 in 3 cases (3.8%), 1:8 in 1 case (1.3%) and 1 : 128 in 1 case (1.3%). 4. Among total 420 study cases, 5 cases (1.2%) showed Positives, and they were 2 cases (0.5%) of pregnant women and 3 cases (7.0%) of pelvic tumor patients. 5. One case (1.3%) out of 80 control sera showed positive result.

Edwardsiellosis has become a serious diseases problem in cultured eels for many years. This study was performed to investigate possibility of vaccination against edwardsiellosis caused by Edwardsiella tarda. We conducted a immersion and/or oral vaccination using formalin-killed E. tarda in eel Anguilla japonica. Three groups of fish ($26.8{\pm}1.2g$, $7.1{\pm}0.7g$ and $2.2{\pm}0.4g$) were used in this study. The protection (relative percentage survival, RPS) and serum antibody response (agglutination titer) were evaluated in the vaccinated fish. No correlation between agglutination titer and survival rate was observed in vaccinated fish. However, there was a satisfactory protective (RPS>50%) in vaccinated fish. Immersion (10 mg/mL, 1 hr) and immersion (10 mg/mL, 1 hr) plus oral (10 mg/g, 10 days) of $26.8{\pm}1.2g$, immersion (10 mg/mL, 1 hr) plus oral (10 mg/g, 10 days) of $7.1{\pm}0.7g$ showed RPS of 62.6%, 52.2% and 56.8%, respectively.
Background : Mycoplasma pneumonia (M. pneumonia) is a major cause of atypical pneumonia, and its incidence is predominantly at childhood and early adulthood. In contrast, the incidence of adult patients with M. pneumonia has been known to be low. Furthermore the clinical aspects of M. pneumonia are different from those of community acquired pneumonia. Thus, we evaluated the clinical aspects of M. pneumonia in the adult patients. Method : Mycoplasma antibody and cold agglutination tests were performed in patients with clinically suspected pneumonia who had abnormal infiltrations on chest P-A. The 12 patients with pneumonia, who fulfilled entry criteria of more than 1:64 of cold agglutination titer and 1:40 of mycoplasma antibody titer or four-fold increase of mycoplasma antibody titer during one week, were analyzed in terms of clinical aspects. Results : 1) Twelve patients, male 3 and female 9, were included in this study. The peak incidence was teenager. 2) M. pneumonia occured perennially, but predominantly between June to October in eight patients. 3) The main symptoms were fever, coughing, sputum. 4) The main patterns of chest P-A were bronchopneumoina in 8 cases, and involved lesion were nearly both lower lobe. Conclusion : The clinical aspects with Mycoplasma pneumonia in adult patients were different from those of community acquired pneumonia.
Effects of dietary fat and tomato on immune functions were investigated using BALB/c mice. Sixty male BALB/c mice weighing 16-17g were divided into two dietary groups. Control group was fed experimental chow (AIN-76 purified diet) supplemented with saturated fat (beef tallow) and unsaturated fat (safflower oil) at 5% level (weight basis), while treatment group was fed chow added with 0.5% (dry matter basis) of tomato (moisture content 95%) for 10 weeks. Organ weights, delayed-type hypersensitivity test, plaque-forming cell test, agglutination test, differential white cell count, and histological examination were performed at 4th, 7th, and 10th week after dietary treatment. Weight of spleen and spleen index of tomato group increased with increasing experimental period. Thymus index of tomato group was significantly higher than that of control group at 7 and 10 weeks (p<0.05). In delayed hypersensitivity test at 4th, 7th, and 10th week after dietary treatment, tomato group showed more significant increase in footpad swelling reaction 24 hr after challenge than control group (p<0.05). In plaque-forming cell test, tomato group fed 7 weeks increased more significantly than control group, while no significant difference was found between two groups at 10 weeks. Agglutination test decreased with increasing experimental period; tomato group at 10 weeks was higher than control group in antibody response to SRBC. In tomato group, Iympocyte percentage was slightly higher than that of control group, and spleen tissue showed active immune reaction caused by severe proliferation of white pulp.
The objective of this study was to see if fairly simple magnetic nano-particle treatment enhances boar semen qualities. Boar semen samples were prepared from the swine AI center and samples were divided by 4 different motility groups (1, >90%; 2. 80~90%; 3. 70~80%; 4. <70%) using computer assisted sperm analysis (CASA) evaluation. Boar semen was extended using BTS extender and same number of magnetic nano-particles as total number of spermatozoa in each sample was treated for 20 min and collected for 5 min at room temperature. Sperm qualities such as motility and viability were evaluated by the CASA before and after treatment. Sperm abnormality and degree of agglutination were also evaluated under the microscopic examination before and after treatment. There were significant changes (p<0.05) on sperm motility from all 4 different groups in the average of 7.11% after treatment. The enhancement of sperm motility changes was more clear in the groups of lower sperm motile groups (<70% and 70~80%; 19.12±1.08% and 5.67±0.71%, p<0.05). The sperm motility character in terms of curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP) and linearity (LIN, %) showed also similar pattern but motility enhancement wear more clear in below 70% motile group. Average sperm viability was increased to 4% by magnetic nano-particles (p<0.05). The percentage of sperm abnormality was also reduced significantly (p<0.05) to the range of 3.7~4.5% before after treatment. The degree of sperm agglutination was also reduced in lower motility groups by the magnetic nano-particle purification.
Total of 216 animals conserved in Seoul Grand Park were examined on the antibody titers of Toxoplasma by the indirect latex agglutination test, 20 out of 131 cases (15.3%) in mammals, 2 out of 75 cases (2.7%) in birds, and none in reptiles, according to species, 15 out of 68 species (22.1%) in mammals, 2 out of 36 species (5.6 %) in birds showed positive antibody titers when the titers of 1 : 32 or higher were regarded as positive. In mammals, it appeared as positive in 2 out of 6 cases (1 out of 3 species) in marsupials, 1 out of 15 cases (1 out of 11 species) in primates, lout of 1 case in bats, 6 out of 13 cases (5 out of 10 species) in carnivores, 1 in 12 cases (1 species out of 3) in odd-toed ungulates, 9 out of 80 cases (6 species out of 38) in even-toed ungulates, and none in rodents and in whales. In birds, 1 out of 21 cases (1 out of 7 species) in gallinaceous birds and lout of 6(5 species) in parrots appeared to have the positive antibody titers of Toxoplasma. And, none of reptiles showed positive. Frequencies of positive antibody titers were high in 1 : 64, 9 cases in mammals, followed by 1 : 32, 6 cases, 1 : 128, 3 cases, and 1 : 256, 2 cases, respectively. Two positive cases in birds appeared to be 1 : 64.
