시호의 어린 잎 절편으로부터 캘러스의 유도는 0.1 mg/L 2,4-D가 첨가된 MS배지에서 가장 우수하였다. 유도된 캘러스는 균일한 크기의 세포괴만을 선발하여 MS배지에 0, 0.1 mg/L, 0.5, 1.0, 2.0 mgh의 2,4-D를 첨가하여 7일간 배양한 다음, 세포괴를 부정근 유도를 위해 2,4-D가 제거된 MS배지에 계대배양하여 부정근의 형성과정 및 근원기 형성에 미치는 효과를 관찰하였다. 그 결과 부정근 형성은 0.1 mg/L 2,4-D를 처리후 호르몬 무처리의 배지에 옮겨주었을 때 세포괴당 평균 부정근수 5.2개로 가장 우수하였고 농도가 증가할수록 감소하는 경향을 보였다. 신장된 부정근의 길이에 있어서는 각 처리구별로 14-16 mm로 농도차에 따른 차이를 보이지 않았다. 처리시간에 따른 부정근의 형성에서는 수록 세포괴당 부정근의 수가 증가하였으며 120시간에 이르러서 최대치를 보였다. 이 결과 부정근 발생에 있어 적정농도의 2,4-D는 부정근 시원세포기 및 근원기 발생기에 민감한 영향을 미치며 근원기가 캘러스 표면밖으로 돌출되는 시기에는 저해적인 것으로 생각된다. 조직학적 기원에 있어서는 특정한 조직의 기원이 아닌 캘러스 표면 세포 및 내부세포로부터 최초 여러개의 시원세포가 형성되어, 캘러스 표면쪽으로 왕성한 반원형의 분열조직을 형성한 다음에 신장 되었다.
노니($Morinda$$citrifolia$) 부정근의 주요 성분인 rubiadin은 간 보호 효과가 있어 제약산업에서 높은 가치가 있다. 노니 부정근의 주요 유효성분인 rubiadin의 효율적인 추출조건을 규명하고자 용매 종류, 물과 메탄올의 비율(물, 20%, 40%, 60%, 80%, 100%), 추출시간 및 추출방법을 달리하여 추출하였다. 노니 부정근에서의 rubiadin의 함량분석은 HPLC 이용 분석조건을 확립하였으며, C-18 컬럼을 사용하여 280nm에서 메탄올과 물로 농도구배를 주어 분석하였다. 용매별 추출효율은 메탄올(0.08%) > 에탄올(0.05%) > 아세토니트릴(0.03%) > 아세톤(0.02%) 및 메틸렌클로라이드(0.02%) 순으로 증가하였다. 메탄올에 물의 혼합 비율을 달리하여 초음파 추출기로 1시간 동안 추출한 결과 60% 메탄올(0.21%) > 80% 메탄올(0.13%) > 100% 메탄올(0.07%) 및 40% 메탄올(0.07%) 순으로 효과적이었으며, 환류냉각 추출기로 2시간 동안 추출한 결과 60% 메탄올(0.21%) > 40% 메탄올(0.17%) > 80% 메탄올(0.14%) 용매처리구 순으로 효율이 좋았다. 추출방법 및 추출시간에 따른 rubiadin의 추출 효율을 비교하기 위해 환류냉각추출 및 초음파추출, 진탕추출법으로 추출하였다. 추출방법 및 추출시간에 따른 rubiadin의 추출효율은 초음파추출 및 진탕추출에서 각각 8 및 24시간 동안 추출했을 때 효율적이었다.
작약 뿌리 절편으로부터 유도된 캘러스 클론으로부터 부정근 발생에 대한 최적의 배양조건을 조사하기 위하여 뿌리 절편으로부터 캘러스 유도를 위해서 먼저 0.5, 1.0, 2.0, 3.0, 4.0 mg/L 농도의 오옥신(IAA, NAA, IBA, 2,4-D)과 0, 0.5, 1.0, 2.0 mg/L 농도의 싸이토카닌(kinetin, zeatin, BAP)를 조합한 MS 배지에서 배양하였다. 캘러스 클론으로부터 부정근 유도와 길이 생장은 0.1, 0.5, 1.0 mg/L 농도의 오옥신(IAA, NAA, IBA) 또는 싸이토카닌(kinetin, zeatin, BAP)을 단독으로 첨가한 배지에서 암 조건으로 그리고 0.1, 0.5, 1.0 mg/L 농도의 IBA와 zeatin을 각각 조합 첨가한 배지에서 명/암 조건으로 6주 동안 배양하였다. 캘러스 형성은 다른 조합 처리보다 1.0 mg/L 농도의 NAA와 zeatin을 조합 첨가한 배지에서 가장 효과적이었으며, 캘러스 클론으로부터 부정근 발생 수와 부정근의 길이 생장은 IBA 단독처리의 경우 각각 6.66개와 4.82 cm, zeatin 단독 처리의 경우 2.32개와 0.92 cm로 다른 호르몬에 비해 우수하였다. 특히 0.1 mg/L IBA와 0.5 mg/L zeatin을 조합 첨가한 배지에서 광 조건으로 배양할 경우 가장 많은 부정근이(14.06) 형성되었으며, 동일배지에서 암 조건으로 배양할 경우 부정근의 길이가 가장 긴 5.45 cm로 측정되어 가장 효과적인 농도와 조합임을 알 수 있었다. 이러한 작약의 기내 배양에 대한 최적의 배양 조건은 기내배양을 통해 작약 부정근의 대량 생산을 위한 배지로 사용할 수 있음을 보여 주었다.
Traditional culture technology of medicinal plants mainly depends on the field culture, which has many problems. With progress of modern culture technology, it has become possible to produce valuable secondary metabolites from medicinal plants. In this paper, we discuss about the pigment and saikosaponin production from too medicinal plants, Carthamus tinctorius and Bupleurum falcatum, through bioreactor culture system. A two-stage bioreactor culture system was established for the production of yellow and red pigments and saikosaponins by cell suspension cultures of Carthamus tinctorius and Bupleurum falcatum. In Carthamus tinctorius, balloon type airlift bioreactors and column type airlift bioreactors were employed for the tell culture and for the pigment production, respectively. The greatest pigment production was obtained on White medium supplemented with 4 mg/L kinetin, high levels of sucrose concentration and photosynthetic photon flux. In Bupleurum falcatum, adventitious roots were cultured in balloon type airlift bioreactors and the root growth was greatest on SH medium containing 5 mg/L IBA and 0.2 mg/L kinetin. HPLC analysis showed that the contents of main active saikosaponins a, c, and d in adventitious roots were almost the same as those in field cultured root.
This experiment was carried out to investigate the effects of plant growth regulators on the organogenesis from the tissue culture of Arabidopsis thaliana stem, and the origin of the callus development. When the stem segments were cultured on medium with 2mg/L IAA or NAA, adventitious roots and trichomes were differentiated after 11 days of culture. Callus vigorously formed on medium with 2/L2,4 after 7 days of culture, but adventitious roots and trichomes were not differentiated from callus after 10 days of culture. This results suggesting that picloram is very effective auxin for the callus formation and organogenesis. Callus weakly formed on 0.05mg/L kinetin, and formed on combination of auxins(2mg/L) with 0.05mg/L kinetin. But the effect of combination of auxins and kinetin the callus formation was less than 2,4-D or picloram alone. A histological examination of callus formed on picloram showed that phloram showed that phloem parenchyma cells were divided and enlarged after 2 days of culture. Cortex parenchyma cells were divided and meristematic nodules were developed from these cells after 4 days of culture. Finally, callus formed on outside of cortex and epidermis by division of meristematic nodules after 7 days of culture.
Echinacea purpurea (L.) Moench (purple cone flower) is an important medicinal plant; it can enhance immunity, relieve pain, and reduce inflammation, and also has hormonal, antiviral, and antioxidant effects. Adventitious root biomass of Echinacea purpurea was produced in commercial-scale bioreactors for use as a dietary supplement in the food industry and in traditional medicine. Biosafety and toxicological evaluations of tissue-cultured Echinacea purpurea adventitious roots (TCEPARs) were performed. Reverse mutation and chromosomal aberration tests showed no significant mutagenicity. Furthermore, repeated four-week oral dose tests performed in Sprague-Dawley rats did not show any notable changes in the general behavior of the rats, in the gross appearance of their internal organs, or in their mortality rate. There were no differences between the control group and the treatment group in parameters such as absolute body weight, hematology, blood chemistry, and absolute and relative organ weights. These findings indicate that TCEPARs are safe and nontoxic when consumed at an average dietary level and can be used as raw material for traditional medicine and the food industry.
