칼슘의 섭취수준이 연령이 다른 흰쥐의 마그네슘 이용에 미치는 영향 (Effects of Ca Intake Levels on Mg Utilization in Rats of Different Ages)

  • The purpose of this study was to investigate the effects of calcium intake levels on magnesium utilization in young and adult rats fed various levels of calcium(50, 100 or 200% of requirement) for 3 weeks. Feed intake, body weight gain, and feed efficiency ratios of the young group were significantly higher than those of the adult group. In calcium levels of tissues, serum calcium levels in young group were higher than in the adult group and kidney calcium levels in HCa(high-calcium group) were higher than in other groups. There was no significant difference in magnesium intake among age and diet groups. However, magnesium excretions through feces and urine in the adult group were higher than in the young group. Therefore, magnesium retention in the young group was higher than in the adult group. Especially in the young group, fecal magnesium excretion was increased with increasing levels of dietary calcium. Serum magnesium levels were lowest in YHCa(young 8t high-calcium) group, and there was no significant difference in liver and kidney levels among the calcium-diet groups. These results indicate that high calcium consumption promotes of fecal magnesium excretion and decreased magnesium retention rates in growing rats. Therefore, it is suggested that mineral interactions should be considered in the supplementation of dietary calcium intake for growth. (Korean J Nutrition 31(9) 1404-1410, 1998)

후천적 성인 편평족 변형의 수술적 치료 (Operative Treatment of Acquired Adult Flatfoot)

  • Acquired adult flatfoot deformity is characterized by flattening of the medial longitudinal arch and dysfunction of the posteromedial soft tissues, including the posterior tibial tendon. When the non-operative treatment fails to result in improvement of symptoms, surgery should be considered. Operative techniques include flexor digitorum longus tendon transfer, calcaneal medial slide osteotomy, lateral column lengthening, and arthrodesis of the hindfoot. The principle of correcting the deformity while avoiding overcorrection and excessive stiffness is important in achievement of good outcomes in these patients.

유약호르몬류 투여가 누에 성충화 발육에 미치는 영향 (Effect of Juvenile Hormone Analogue(JHA) on the Adult Development of the Silkworm, Bombyx mori)

  • 누에 5령 유충에 유약호르몬 유연화합물(JHA)인 "마니나"를 투여하여 JHA처리 후의 누에 성장 및 성충화발육에 미치는 영향에 관하여 조사하였다. JHA는 5령 유충의 섭식기간의 연장과 이에 따른 체중의 증가를 가져왔으며 성충화 발육 기간도 1~1.5일 연장시켰다. 혈액, 피부, 소화관, 지방체, 난소 등에 관한 형태 및 단백질 성분변화를 조사한 결과 이들 조직, 기관들 역시 JHA에 의해 성충화 발육이 1-2일 정도씩 지체되고 있음이 밝혀졌다. JHA의 성충화 발육의 지연에 대한 내분비학적 고찰과 잠종생산 증대를 위한 JHA의 이용 가능성에 대하여 검토하였다.대하여 검토하였다.

Engineered adult stem cells: a promising tool for anti-cancer therapy

  • Cancers are one of the most dreaded diseases in human history and have been targeted by numerous trials including surgery, chemotherapy, radiation therapy, and anti-cancer drugs. Adult stem cells (ASCs), which can regenerate tissues and repair damage, have emerged as leading therapeutic candidates due to their homing ability toward tumor foci. Stem cells can precisely target malicious tumors, thereby minimizing the toxicity of normal cells and unfavorable side effects. ASCs, such as mesenchymal stem cells (MSCs), neural stem cells (NSCs), and hematopoietic stem cells (HSCs), are powerful tools for delivering therapeutic agents to various primary and metastatic cancers. Engineered ASCs act as a bridge between the tumor sites and tumoricidal reagents, producing therapeutic substances such as exosomes, viruses, and anti-cancer proteins encoded by several suicide genes. This review focuses on various anti-cancer therapies implemented via ASCs and summarizes the recent treatment progress and shortcomings.

Expression of FKBP prolyl isomerase 5 gene in tissues of muscovy duck at different growth stages and its association with muscovy duck weight

  • Objective: FKBP prolyl isomerase 5 (FKBP5) has been shown to play an important role in metabolically active tissues such as skeletal muscle. However, the expression of FKBP5 in Muscovy duck tissues and its association with body weight are still unclear. Methods: In this study, real-time quantitative polymerase chain reaction was used to detect the expression of FKBP5 in different tissues of Muscovy duck at different growth stages. Further, single nucleotide polymorphisms (SNPs) were detected in the exon region of FKBP5 and were combined analyzed with the body weight of 334 Muscovy ducks. Results: FKBP5 was highly expressed in various tissues of Muscovy duck at days 17, 19, 21, 24, and 27 of embryonic development. In addition, the expression of FKBP5 in the tissues of female adult Muscovy ducks was higher than that of male Muscovy ducks. Besides, an association analysis indicated that 3 SNPs were related to body weight trait. At the g.4819252 A>G, the body weight of AG genotype was significantly higher than that of the AA and the GG genotype. At the g.4821390 G>A, the genotype GA was extremely significantly related to body weight. At the g.4830622 T>G, the body weight of TT was significantly higher than GG and TG. Conclusion: These findings indicate the possible effects of expression levels in various tissues and the SNPs of FKBP5 on Muscovy duck body weight trait. FKBP5 could be used as molecular marker for muscle development trait using early marker-assisted selection of Muscovy ducks.

닭 조직에 따른 Nebulin Isoform 단백질의 특이적 발현 (Tissue-Specific Expression of Nebulin Isoform Proteins in Chicken)

  • 대부분 척추동물에서 골격근 내 filament들의 길이 조절은 근 수축 기작의 구조를 이해하는데 중요한 단서가 된다. Nebulin은 thin filament의 전체에 걸쳐있는 거대한 단백질로 골격근에만 특이적으로 존재하는 것으로 알려져 왔다. 본 연구에서는 닭의 근육과 비근육 조직에서 nebulin isoform단백질들을 확인하기 위하여 전기영동과 immunoblot의 방법을 이용하였다. 각 조직의 단백질들은 soluble과 insoluble fraction으로 분리 준비하였다. 실험결과, 닭의 근육과 비근육 조직들에서 조직 특이성을 나타내는 다양한 nebulin isoform 단백질들이 확인되었다. Nebulin은 성계의 골격근에서 500 kDa 정도의 크기로 나타났고, nebulett은 계배와 성계의 심장근에서 107 kDa 정도로 발현되었다. 그리고 계배의 비근육 조직인 뇌에서 380 kDa 정도의 거대 단백질이 확인되었다. 이 단백질은 뇌 조직의 soluble fraction에서 인지되었다. Nebulin isoform 단백질들이 서로 다른 조직에서 발현되는 양상을 보아 서로 다른 독자적인 기능을 가질 것으로 추정된다.

면역황금표식법을 이용한 폐흡충의 발육단계별 충체조직내 항원성 부위에 관한 연구 (Antigenic localities in the tissues of Payagonimus westermani by developmental stages using immunogoldlabeling method)

  • 폐흡충(Paragonimus westermani)이 숙주의 폐조직 내에서 성숙하는 동안 숙주에게 항체생성을 유도하는 물질의 폐흡충 충체조직내 분포하는 부위와 성숙하는 동안 숙주가 생성하는 항체의 면역반응 정도를 확인하기 위하여 폐흡충 피낭유충을 실험고양이에 감염시키고 발육단계 별 충체와 면역항체를 얻어 조직항원과 면역항체를 반응시켜 환금입자를 표지하였다. 황금입자가 표지된 조직 항원을 전자현미경으로 관찰하고 황금입자의 밀도에 따라 조직성분의 항원성을 측정한 결과 감염군 면역항체를 반응시킨 경우 숙주체내에서 4 주 동안 성숙한 유약성충 조직항원은 표피충의 표피합포체에 높은 밀도의 황금입자가 표지되어 항원성이 강한 것으로 관찰되었다. 8 주 이상 성숙한 충체부터 조직항원은 표피충에 환금 입자의 표지가 현저하게 감소하여 12 주 이상 성숙한 충체 표피층은 항원성이 미약한 것으로 관찰되었다. 12 주 이상 성숙한 충체는 난황세포의 구성물질들에 각각 항원성이 특이하게 나타나며 유연조직의 분비과립과 맹관 그리고 배설낭의 구성 물질들은 충체의 성숙정도에 상관없이 항원성을 포함하고, 숙주가 생성하는 항체는 감염 후 12 주부터 16 주사이에 항체생성이 극대 화되는 것으로 생각되었다.

초자화 냉동법으로 냉동.해동한 Neonatal 생쥐 난소의 생체내 동소이식 후 난포 발달에 관한 연구 (Ovarian Development of Vitrified Neonatal Ovaries after Orthotopic Transplantation into Adult Recipients)

  • Ovarian development of the vitrified neonatal ovaries after orthotopical transplantation into the ovariectomized adult recipient mouse were observed. Ovaries were collected from the neonatal females on day of birth and grouped for fresh, vitrification for 1-minute, and 3-minute. Vitrified and thawed neonatal ovaries were orthotopically transplanted into ovarian bursa of the adult mice from which endogenous ovaries have removed just prior to the transplantation (1 minute: n=25; 3 minutes n=23). Fresh ovarian tissue transplanted (n=25) mice were included as control groups. Returning of the estrus cycles and the survival and development of the transplanted ovaries were evaluated. Intact ovaries from neonatal, and four weeks old mice were used for comparison of the ovarian development as in vivo-developed control. From 2 weeks after transplantation, 64%, 36%, and 75% of the transplanted mice showed return of the estrus cycles in fresh, 1-minute, and 3-minute groups, respectively. Four weeks after transplantation, all mice were sacrificed and ovarian tissues were recovered for histological analysis. 57.1%, 33.3%, and 64.7% mice in fresh, 1-minute, and 3-minute groups, respectively, had survived ovaries with follicles at various stages of growth from primordial to preovulatory follicles. Corpus lutea were also observed. Results of the present study suggest that 1) normal folliculogenesis has initiated in vivo after vitrification, and 2) the vitrification may be used as a preservation method for ovarian tissues for establishment of ovarian tissue bank.

Expression of ATP-sensitive Potassium Channel and Sulfonylurea Receptor in Neonate and Adult Rat Tissues

  • The ATP-sensitive potassium $(K_{ATP})$) channel is a member of inward rectifier potassium channel (Kir) that is inhibited by intracellular ATP and functions in close relation to sulfonylurea receptors (SUR). Although the molecular mechanism and physiological function of $K_{ATP}$ channels are well understood, the expression pattern during development or treatment with the channel modulators such as glybenclamide is little known. In this work, we determined mRNA levels of a $K_{ATP}$ channel (Kir6.2) and a sulfonylurea receptor (SUR2) in rat tissues by RNase protection assay. Levels of Kir6.2 and SUR2 mRNA in the rat brain and skeletal muscle were higher in adult $(90{\sim}120\;days)$ than in neonate $(2{\sim}8\;days),$ whereas those in the heart were not much different between neonate $(2{\sim}8\;days)$ and adult $(90{\sim}120\;days).$ In addition, none of $K_{ATP}$ channel modulators (opener, pinacidil and nicorandil; blocker, glybenclamide) affected the Kir6.2 mRNA levels in the heart, brain and skeletal muscle. The results indicate that the expression of Kir and SUR genes can vary age-dependently, but the expression of Kir is not dependent on the long-term treatment of channel modulators. The effect of the channel modulators on mRNA level of SUR is remained to be studied further.

야생 멧돼지에서 돼지신충(swine kidney worm) 감염 예 (A case report of swine kidney worm infection in a wild feral pig)

  • This is the first case report of Stephanurus dentatus infection of a feral pig in Korea. In late April, 2000, a weakened feral pig was caught by blow gun from a very low level mountain near the Gyeongsang National University. We autopsied the feral pig in the laboratory of veterinary anatomy at the College of Veterinary Medicine. A total of 27 adult parasites, 11 females and 16 males, and numerous eggs were observed from the cysts formed in the perirenal tissues and ureters. The average size of males was $25.1{\pm}3.2mm$ long and of the females was $34.2{\pm}2.9mm$. The worms were stout, the females being about 2mm broad, and the internal organs were partly visible through the cuticle. The shape of thin-shelled eggs found in the cysts of perirenal tissues and ureter was ellisoidal and oval, and measured $40{\sim}65{\times}90{\sim}115{\mu}m$. The adult parasites were found in cysts which varied from 0.6 to 4cm in diameter, each cyst usually containing a pair of adult worms embedded in green pus. The ureter was thickened and almost occluded, with consequent hydronephrosis.
