Screening of Antibacterial Activity from Hot Water Extracts of Indigenous Plants (자생식물 열수추출액의 항균효능 검색)

  • Hot water extracts of 141 indigenous plants were screened for antibacterial activity against 7 Gram (+) and 12 Gram (-) bacteria by agar diffusion method. Of the 141 plants tested, 55 showed antibacterial activity against one or more species of Gram (+) bacteria and especially extracts of Oenothera stricta Ledebour (Onagraceae), Quercus variabilis Blume (Fagaceae), Alnus firma (Betulaceae), Geranium thunbergii (Geraniaceae), Lagerstroemia indica (Lythraceae) showed broad spectrum antibacterial activity on over 6 strains. Of the 141 plants tested, 45 showed antibacterial activity against one or more species of Gram (-) bacteria and especially extracts of Oenothera stricta Ledebour (Onagraceae), Punica granatum (Lythraceae), Quercus variabilis Blume (Fagaceae), Schizandra chinensis Baillon (Magnoliaceae), Alnus firma (Betulaceae), Alnus hirsuta (Betulaceae), Magnolia kobus (Magnoliaceae) showed broad spectrum antibacterial activity on over 8 strains. The most active antibacterial plants were Oenothera stricta Ledebour (Onagraceae), Punica granatum (Lythraceae), Quercus variabilis Blume (Fagaceae), Schizandra chinensis Baillon (Magnoliaceae) which are edible as permitted by Korea Food & Drug Adminstration. With in-depth research, the hot water extracts of these plants can be applied as food preservatives and alternatives of antimicrobials for livestock feeds.

  • For a long-term preservation of silkworm stocks by frozen gonad storage, fundamental topics such as freezing rate and transplanting stage of the gonad, proper cryoprotectant, and super-cooling temperature and freezing point of the freezing medium were examined and following results were obtained. Proper method to anesthetize the ovary-recipient silkworm was to dip the animal to cold water for 10 minutes, and the ovary taken from the 4th instar larvae was more suitable for freezing-preservation than that from the 5th. Concerning the cryoprotectant, glycerol and DMSO were effective to prevent cryoinjury of the ovary, but sorbitol was not. The supercooling temperature and freezing point of the medium to freeze the ovary and testes were checked, and consulting with the results desirable cooling rate was confirmed. On the desirable conditions of transplanting methods, freezing rate and cryoprotectant concentration ect., the next generation was obtained when the females implanted frozen-thawed ovaries mated with normal males, but none of the normal females mated with the males implanted frozen-thawed testes laid fertilized eggs. Now it is needed to improve the connecting ration of the ducts associated with the transplanted testis to those of the hosts.

Effects of the Bambusae Caulis in Liquamen Extracted by a Different Refining Process on the Hematological and Hepatic Function of the Mouse (정제방법이 다른 죽력이 생쥐의 혈액학적 및 간 기능에 미치는 영향)

  • This study was performed to investigate the changes of the hematological and hepatic function in the mouse after the adminstration of Bambusae Caulis in Liquamen extracted by a different refining process during 30 days. The experimental groups divided seven. Control group was administered mice with 0.9% saline(4mL/kg). The experimental groups were divided 10% bamboo extract(B1, C1 and D1 experimental groups) and 30% bamboo extract(B2, C2, D2 experimental groups)administered groups(4mL/kg). Hematological results: RBC(P<0.05) and Hct(P<0.05) were significantly decreased in the D2 group. The activity of transaminase(P<0.05) of D2 group was significantly increased, but the activities of SOD(P<0.05) and catalase(P<0.01) were significantly decreased, in the D group. Histopathological observation: Ballooned hepatocytes were occurred periportal vein in the D1 and D2 groups, and necrosis of hepatic nuclei in the D2 group were observed. The results indicated that hematological and histopathological toxicity were occurred in the administered group of bamboo extract D refined by 2 times distilling at $108{\circ}C$.

Studies on the Mulberry Sapling 1. Soil Characteristics of Nursery Garden Producing Nonsprouting Sapling. (뽕나무 접목묘에 관한 연구 I. 활착불량상묘 발생 묘포지의 토양특성에 관한 연구)

  • soil survey and chemical analysis on mulberry nursery garden were carried out to study the causes of the non-sprouting phenomenon occured seriously in Buy and Ogchun in 1983 and Sangju in 1984. In addition, 115 nursery garden soils taken from 21 sapling produsers in Chungbuk province were analyzed in 1984. The results were as follows, 1. Symptom of dead saplings was the highest in boron deficiency by 59% and the lowest in rot symptom. The rest of saplings by 34.9% were not detected any symptom. 2. The nursery gardens showen non-sprouting phonomenon were located along the stream. As the result, soil depth was shallow, around 20cm deep and subsoil composed with coarse sand and gravels. 3. Nursery soils were mostly strong acid, low in Ca and Mg content, especially B, whereas available phosphorus and potassium were abundant at some gardens, and deficient at others. 4. Application of red earth or borax to nursery garden, especially to paddy, increased sprouting rate. 5. Paddy nursery garden occupied by 52.8% of 60.8ha of total garden area in Chungbuk province. Ninty point five percent of the nursery garden was lower in pH than 6.5, 87.0% lower in K than 0.5me/100g, 40.8% lower in Ca than 6.5me/100g, 94.8% lower in Mg the 2.00me/100g 99.1% lower in B than 0.3ppm.

Relationship between Weather Factors and Chemical Components of Flue-cured Tobacco (기상요인과 황색종 잎담배의 화학성분과의 관계)

  • This study was conducted to investigate the relationship between weather factors during the growing season and chemical components of flue-cured tobacco. Chemical components used in this study was from 'Farm Leaf Tobacco Test' conducted at KT&G Central Research Institute from 1986 through 2003. Data of weather factors during growing season(April to July) were collected in 10 districts measured at Korea Meteorological Adminstration(KMA). Nicotine and total sugar contents, and total sugar to nicotine(TS/Nic.) ratio were increased, whereas total nitrogen to nicotine(TN/Nic.) ratio and chloride content were decreased from 1986 through 2003. Year to year variation of rainfall was the largest, followed by that of sunshine hour. Month to month variation of rainfall also was the largest, followed by that of mean daily air temperature(MDAT). Rainfall was correlated positively with relative humidity(RH), but negatively with sunshine hour. Nicotine content was correlated positively with MDAT(in July, June$\~$July, May$\~$July and average), but negatively with rainfall(in May$\~$July) and with RH(in June, July, May $\~$June, June $\~$July, April$\~$June, May $\~$July and average). Total sugar content was correlated positively with MDAT(in May), but negatively with sunshine hour(average) and RH(in June, July, June$\~$July, April$\~$June, May$\~$July and average). The positive correlation was found between total nitrogen content and sunshine hour(in April, May, April$\~$May, May$\~$July, April$\~$June and average). The negative correlation was found between TS/Nic. ratio and sunshine hour(in May$\~$July and average). TN/Nic. ratio was correlated positively with sunshine hour(in May and April$\~$May) and with RH(in July and June$\~$July), but negatively with MDAT(in July, June$\~$July, May$\~$July and average). Ether extraction content was correlated positively with MDAT(in July, June$\~$July, May$\~$July and average) and with sunshine hour(in July and June$\~$July), but negatively with rainfall(in April, July, May­July and average). Chloride content was correlated positively with sunshine hour(in April, July, April$\~$May, June$\~$July, April$\~$June, May$\~$July and average), but negatively with rainfall(in April, April$\~$June and average).

Web-enabled Healthcare System for Hypertension: Hyperlink-based Inference Approach (고혈압관리를 위한 웹 기반의 지능정보시스템: 하이퍼링크를 이용한 추론방식으로)

  • In the conduct of this study, a web-enabled healthcare system for the management of hypertension was implemented through a hyperlink-based inference approach. The hyperlink-based inference platform was implemented using the hypertext capacity of HTML which ensured accessibility, multimedia facilities, fast response, stability, ease of use and upgrade, and platform independency of expert systems. Many HTML documents, which are hyperlinked to each other based on expert rules, were uploaded beforehand to perform the hyperlink-based inference. The HTML documents were uploaded and maintained automatically by our proprietary tool called the Web-Based Inference System (WeBIS) that supports a graphical user interface (GUI) for the input and edit of decision graphs. Nevertheless, the editing task of the decision graph using the GUI tool is a time consuming and tedious chore when the knowledge engineer must perform it manually. Accordingly, this research implemented an automatic generator of the decision graph for the management of hypertension. As a result, this research suggests a methodology for the development of Web-enabled healthcare systems using the hyperlink-based inference approach and, as an example, implements a Web-enabled healthcare system for hypertension, a platform which performed especially well in the areas of speed and stability.

  • The characteristics of parasporal inclusion body from Bacillus thuringiensis subsp. kurstaki KB099 isolate which is high bioactive to the tobacco cutworm, Spodoptera litura, were examined. Parasporal inclusion of B. thuringiensis subsp. kurstaki KB099 isolate showed only 1 band at 130 kDa compared with B. thuringiensis subsp. kurstaki HD-l isolate producing 2 protein bands at 130 kDa and 60 kDa from by SDS-PAGE analysis without any enzyme treatment. Also, we confirmed that gut extract of sensitive S. litura KB099 isolate had digested only 60 kDa ${\delta}$-endotoxin protein. When the digestive enzyme of sensitive insect responsible for parasporal inclusion from KB099 and HD-l isolate was treated to each of them, protein band 60 KDa of KB099 was maintained up to 12 hours but all bands of HD-l were disappeared within 6 hours. In KB099 isolate, 6 genes (Cry1Aa, Cry1Ab, Cry1Ac, Cry1C, Cry1D and Cry1I) were identified by PCR analysis. Also, $Cry^-$ mutant of KB099 isolate was investigated by phase- contrast microscope, SDS-PAGE and PCR.

Effects of Vitamin E and Selenium on the Antioxidative Defense System in Streptozotocin-induced Diabetic Rats (Vitamin E와 Selenium이 Streptozotocin 유발 당뇨쥐의 항산화계에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin E and selenium on the antioxidative defense mechanism in the liver of streptozotocin(STZ)-induced diabetic rats. Sprague-Dawley male rats(120$\pm$10gm) were randomly assigned to one control and five STZ-diabetic groups. Diabetic groups were classified to STZ-0E (vitamin E free diet), STZ-40E(40mg vitamin E/kg of diet), STZ-400E(400mg vitamin E/kg of diet), STZ-S(0.5ppm Se/kg of diet) and STZ-400ES(400mg vitamin E and 0.5ppm Se/kg of diet) according to the level of vitamin E and selenium supplementation. Diabetes was experimentally induced by intravenous adminstration of 55mg/kg of STZ in citrate buffer(pH 4.3) after 4-weeks feedng of six experimental diets. Animals were sacrificed at the 4th day of diabetic states. Activities of the serum glutamic oxaloacetate transaminase(GOT) and the glutaminc pyruvate transaminase(GPT) in STZ-0E, STZ-40E and STZ-S rats were higher than those of control. Liver xanthine oxidase activities were similar to serum GOT and GPT. Liver superoxide dismutase(SOD) activities were higher in STZ-0E and STZ-40E groups by 33%, 22%, respectively than that of control. Glutathione S-transferase(GST) activities of liver were similar to GSH-Px activities. The contents of vitamin E in liver tissue were significantly lower STZ-0E, STZ-40E and STZ-S groups by 50%, 36%, 45% than that of control. Reduced glutathione(GSH) contents of liver were lower STZ-0E, STZ-40E, STZ-400E, STZ-S and STZ-400ES groups by 57%, 51%, 19%, 18%, 12% than that of control. Lipid peroxide values (LPO) in liver were higher 5.6, 2.3 and 2.3 times in STZ-0E, STZ-40E and STZ-S group than that of control. The present results indicate that STZ-induced diabetic rats are more sensitive to oxidative stress, leading to the acceleration of lipid peroxidation process, which can be more accelerated by feeding the low level of dietary vitamin E. In the coincident supplementation of high dietary vitamin E and selenium antioxidative enzymes activities and physiolosical antioxidants were increased more than those of the separate supplementation of vitamin E or selenium. Therefore, dietary vitamin E and selenium reduced peroxidative damage of tissue, promoting antioxidative defense mechanism against lipid peroxidation by diabetes.

Studies on the Varietal Differences of the Electrophoretic Protein Patterns in the Pupal Haemolymph of the Silkworm, Bombyx mori (가잠용에 있어서 체액단백질의 품종간 차이에 대하여)

  • In order to know the similarity of the electrophoretic protein patterns, distributional aspects of the frequency in each protein band, differences of protein patterns between female and male, and the number of the total protein bands of each variety in the haemolymph of the silkworm pupae according to their geographical origin, the silkworm varieties, and both the sexes, variable electrophoretic protein patterns were observed by polyacrylamide gel electrophoresis, using the silkworm strains which have been reared in the sericultural experiment station. 1. The total number of electrophoretic protein bands classified were twenty-eight, and nearly fourteen bands were shown by each variety on an average. 2. According to their geographical origin, the mean values of the similarity were 20.0%, 28.3% and 14.3% in Japanese, Chinese and European respectively, and the highest similarity was observed between Chinese varieties. 3. The similarity of 129 varieties were tested and it was 22.5%, 23.5% in female and male respectively. 4. The electrophoretic protein patterns in 129 varieties were tested and variation was found in 109 patterns in female and 112 in male. 5. The frequency of the each specified protein band in female and male having variation in protein bands in each variety was found averagely 6.6%. 6. The distribution frequency of the classified 28 protein bands in both the sexes within each geographical origin was variable from 0% to 100%. 7. Among the classified 28 protein bands, HP 11 protein band was found common in all the varieties tested. 8. HP 3 and HP 4 protein bands in female showed high density in electropherograms than in male.

Effects of Artemisia selengensis Methanol Extract on Ethanol-Induced Hepatotoxicity in Rat Liver (쑥(물쑥)추출물이 에탄올에 의한 흰쥐의 간 손상에 미치는 영향)

  • This study was done to investigate the effects of Artemisia selengensis methanol extract on ethanol-induced hepatotoxicty in rat liver. Sprague-Dawley(SD) rats weighing about 150g were divided into the following 4 groups : control group(CON), Astemisia selengensis methanol extract administered group(ASE), ethanol adminstered group(ETH) and Artemisia selengenis methanol extract and ethanol administered group(ASA). Ethanol and Artemisia selengenis methanol extract were administered orally by 5m1/kg and 200mg/kg body weight per day for 6weeks, respectively. Body weight, daily food intake and percent liver weight per body weight were significantly changed by ethanol administration in comparison to control group. The activities of serum alanine aminotransferase(ALT), asparate aminotransferase(AST), and hepatic TBA-reactants increased by ethanol were decreased significantly by Artemisia selengensis methanol extract compared with ethanol group. It was also obseued that superoxide dismutase, catalase and glutathione peroxidase were not changed by Artemisia selengensis methanol extract, whereas hepatic xanthine oxidase activity was inhibitied by Artemisia selengensis methanol extract as compared to ethanol group. The glutathione contents in liver decreased by ethanol adminstration, but glutathione levels increased in ASA compared with ethanol group. These results suggest that Artemisia selengenis methanol extract have a possible protective effect on the ethanol-induced hepatotoxicity in rat liver.
