The Effect of Trichosanithes kirilowii Max. Subfractions on the Insulin Activity in Streptozotocin Induced Diabetic Rats and Their Acute Toxicity (하늘타리(Trichosanthes kirilowii Max) 재분획물이 Steptozotocin 유발 당뇨 흰쥐의 인슐린 활성에 미치는 영향 및 급성독성에 관한 연구)

  • The effects of Trichosanthes kirilowii Max. subfractions, which has long been used as a wild vegetable and folk medicine, on insulin activity and acute toxicity were investigated. Diabetes melitus was induced in male Sprague-Dawley rats by the injection of streptozotocin(STZ) into the tail vein at a dose of 45mg/kg. The hexane fraction of the plant was subfractioned by silica gel column chromatography and were administered orally for 14 days and the normal and STZ-control group were orally administered with tween 80. The body weight gain was monitored and plasma levels of glucose, cholesterol, triglyceride, free fatty acid HDL-cholesterol were determined. The plasma activities of aspartate aminotransferase(AST) and alanine aminotransferase (ALT) were analysed. The subjraction A of hexane fraction lowered plasma glucose levels significantly and increased insulin activity compared to that of STZ-control. The activities of AST and ALT were decreased by administration of subfractions A and C of hexane fraction. The intakes of the hexane fraction of Trichosanthes kirilowii Max. did not showed the acute toxicity. It is suggested from the results that the subfraction A of the hexane fraction of Trichosanthes kirilowii Max. showed a hypoglycemic effect on diabetic rats and that the subfraction may be beneficial for insulin secreation.

  • Oriental pear was used as treatment of asthma, control of blood pressure tonic medicine diabetes in oriental medicine, Pear Pectin was effective on control of blood pressure in previous report. In this study, it was investigated that Woohwang with pear extractions effects on cardiovascular system as blood pressure and renin and Atrial natriuretic peptide(ANP) in plasma. The 2K1C hypertension model was prepared by constricting the left renal artery with a sliver dip. Animals were then divided into three groups, control, Woowhang:Pear Phenolic compound(1:1), Woowhang:Pear Phenolic compound(2:1),Woowhang:Pear Phenolic compound(1:2) were supplied with them. Direct-blood pressure was measured at femoral vein, Indirect-blood pressure was measured at rodent tail. The results are as follows. The blood pressure was more significantly decreased at 1:2(woohwang:pear phenolic compound)group than other groups. On 6,9,12,15days, the blood pressure was significantly decreased in 1:2(woohwang:pear phenolic compound)group. The plasma ANP was significantly increased in 1:2(woohwang:pear phenolic compound)group. It tenders to decrease in 1:2(woohwang:pear phenolic compound)group on plasma renin. Based on the above results it is assumed that oral administration of Woohwang with Pear Phenolic compound(1:2) can help the treatment of hypertension.

  • To examine the effects of lipoic acid on metabolic response to a ${\beta}$-agonist, isoproterenol, in broilers, chicks were fed dietary lipoic acid at levels of 0 (control) or 50 mg/kg for 24 d. At 27 d of age, chickens were randomly selected. Isoproterenol dissolved in 0.9% saline was injected into the wing vein at a dosage of 2 mg per kg BW; then, blood samples were taken at 0, 10, 20, 30, 60, 90, 120, and 180 min. Amounts of plasma glucose, NEFA, triglyceride and total cholesterol were determined. Dietary lipoic acid reduced only plasma total cholesterol by 25%. Following isoproterenol injection, plasma glucose in both groups increased for 20 min; then, it returned to its basal concentration. In contrast, the maximal reductions in plasma NEFA and triglyceride in both groups (20 to 30 min) were confirmed by isoproterenol injection. In addition, while glucose returned to the basal level, plasma NEFA in the lipoic acid-treated chickens increased above the basal or control value during the 60 to 180 min post-injection. The present study suggests that the dietary administration of lipoic acid elicits fatty acid mobilization in ${\beta}$-adrenergic response to isoproterenol when the basal level of plasma glucose is maintained.

The Effect of Dioscorea japonica Thunb Subfractions on Blood Glucose Levels and Energy Metabolite Composition in Streptozotocin Induced Diabetic Rats (참마 재분획물이 당뇨유발 흰쥐의 혈당 및 에너지원 조성에 미치는 영향)

  • This study evaluates the effect of Dioscorea japonica Thunb subfractions on hyperglycemia and the composition of energy metabolites in diabetic rats. Diabetes emllitus was induced in male Sprague-Dawley rats by an injection of streptozotocin(STZ) dissolved in a citrate buffer into the tail vein at a dose of 45㎎/㎏ of body weight. Diabetic rats were assigned to 6 groups; STZ-control, subfraction A, B , C, D and E groups. All groups were fed an AIN-76 diet. The second butanol fraction of Dioscorea administered orally with carboxymethyl cellucose for 10 days after the STZ injection Body weight gain, diet intake and organ weights were monitored Levels of hematocrit, blood glucose, liver and muscle glycogen were measured. Levels of cholesterol, triglycerides and free fatty acids were also assayed. Body weight losses were observed by subfraction A group. Liver and kidney weights were not affected in any of the subgractioned groups. The decrease of blood glucose in daibetic rats which were fed Dioscorea japonica Thunb was significantly greater than the dicrease of blood glucose in the STZ-control group. cholesterol plasma level was not influenced in any subfraction of Dioscorea japonica Thunb. Liver triglyceride levels were significantly lowered in subfraction A compared with the STZ-control group. This study's results suggest that oral administration of subfraction C of Dioscorea japonica Thunb frction is capabl of reducing blood glucose, plasma triglyceride and free fatty acid levels, and therefore Dioscorea japonica Thunb may contain antihyperglycemic compounds.

  • The present study is involved with the prolactin secretion from anterior pituitary gland by ginseng saponin since it was handled down by tradition that ginseng might influence the milk secretion when it was given to nursing mother. To investigate the effect of saponin on the prolactin production or release from the anterior pituitary gland, cell culture study and whole animal studies were carried out. For the cell culture study, enzymatically dispersed anterior pituitary cells of rat anterior pituitary gland in HEPES buffers containing trypsin were used. Ginseng saponin was added to the culture media and the amount of prolactin produced in the cell culture media was determined by radloimmunoassay(RIA) technique. Dose-dependent increases of prolactin with ginseng saponin were observed, whereas, no change was observed without ginseng treatment. For the whole animal study, normal and castrated rats which previously cannulated into the heart via the right juglar vein were used. The prolactin concentration in plasma were determined by using the technique of RIA. In normal rats, prolactin concentration in plasma were elevated dramatically after 1 hour of ginseng saponin administration, whereas, instantaneous increases were observed in castrated rats. For prolactin assay by RIA, NIAMDD Rat Prolactin Kit and NIAMDD Rat Prolactin RP-1 were used as standard. The results indicate that ginseng saponins increase the release of prolactin from the anterior pituitary gland and production of prolactin from the cell in rats.

Pulmonary passage of canine adipose tissue-derived mesenchymal stem cells through intravenous transplantation in mouse model

  • Importance: The intravenous administration of adipose tissue-derived mesenchymal stem cells (AdMSCs) in veterinary medicine is an attractive treatment option. On the other hand, it can result in severe complications, including pulmonary thromboembolism (PTE). Objective: The present study assessed the occurrence of PTE after the intravenous infusion of canine AdMSCs (cAdMSCs) into experimental animals. Methods: Five-week-old male BALB/c hairless mice were categorized into groups labeled A to G. In the control group (A), fluorescently stained 2×106 cAdMSCs were diluted in 200 µL of suspension and injected into the tail vein as a single bolus. The remaining groups included the following: group B with 5×106 cells, group C with 3×106 cells, group D with 1×106 cells, group E with 1×106 cells injected twice with a one-day interval, group F with 2×106 cells in 100 µL of suspension, and group G with 2×106 cells in 300 µL of suspension. Results: Group D achieved a 100% survival rate, while none of the subjects in groups B and C survived (p = 0.002). Blood tests revealed a tendency for the D-dimer levels to increase as the cell dose increased (p = 0.006). The platelet count was higher in the low cell concentration groups and lower in the high cell concentration groups (p = 0.028). A histological examination revealed PTE in most deceased subjects (96.30%). Conclusions and Relevance: PTE was verified, and various variables were identified as potential contributing factors, including the cell dose, injection frequency, and suspension volume.

The Effects of Prunus on Diabetic Nephropathy Rats Induced by Unilateral Nephrectomy and Streptozotocin (도인(挑仁)이 일측 신절제와 streptozotocin으로 유발된 당뇨병성 신증 Rat에 미치는 영향)

  • Objectives: Diabetic nephropathy is the most common cause of end stage renal disease. Transforming growth factor (TGF)-${\beta}1$, type IV collagen, advanced glycation end-products (AGEs), and angiotensin II type 1 receptor (AT1) are the main factors of diabetic nephropathy. We investigated the effects of Prunus on renal function and histopathological changes of diabetic nephropathy rat model induced by unilateral nephrectomy and streptozotocin. Methods: Diabetes was induced in male Sprague-Dawley rats ($290{\pm}10g$) by injecting streptozotocin (55 mg/kg) into the tail vein after unilateral nephrectomy. Rats were divided into 3 groups (n=6): normal, control, and Prunus. After 8 weeks of oral administration of Prunus extract on the Prunus group from 3 days after streptozotocin injection, we checked weight, 24 hrs urine, blood biochemistry and renal tissue to evaluate renal function and histopathological changes by examining parameters including albuminuria, BUN, creatinine, cholesterol, low density lipoprotein (LDL), triglyceride, TGF-${\beta}1$, type IV collagen, AGEs, and AT1. We also measured mRNA expression of TGF-${\beta}1$, type IV collagen, AGEs, and AT1 by Real Time polymerase chain reaction (RT-PCR). Results: Prunus decreased the amount of 24 hrs proteinuria, and inhibited histopathological changes of diabetic nephropathy including the expression and accumulation of TGF-${\beta}1$, type IV collagen and AGEs which could promote development of diabetic nephropathy. Prunus also inhibited mRNA expression of TGF-${\beta}1$, type IV collagen. Conclusions: These findings suggest that Prunus might protect the renal function and inhibit the development of renal injury by regulating factors including TGF-${\beta}1$, type IV collagen, AGEs, except AT1, so Prunus can be used for diabetic patients to prevent the progression of diabetic nephropathy.

Studies on Local Irritation of DA-3585, A Recombinant Human Erythropoietin, in Rabbits (DA-3585(recombinant human erythropoietin)의 국소자극성에 관한 연구)

  • As a series of safety studies on DA-3585, a recombinant human erythropoietin, its local irritancy was examined in rabbits after the following treatments; application into the conjunctival sac of the eye(single), subcutaneous injection (single and -day repeated)and intravenous injection (7-day repeated.)In addition, perivascular irritation of DA-3585 was investigated in mice. In the result of ocular irritation test, 10,000IU/ml solution of DA-3585 could be considered as a non-irritating material. The local irritation of DA-3585 by a single and 7-day repeated subcutaneous injection was negligible and not so much different from that of saline. In the vascular irritancy test, macro-and microscopic observations revealed that local irritation of DA-3585 was comparable to that of saline when injected into retroauricular vein of rabbits for 7 consecutive days. Furthermore the perivascular administration of DA-3585 upto the concentration of 10,000 IU/ml did not induce any morphological abnormalities at injection sites. The results obtained from the present study suggest that the local irritancy of DA-3585 is not different from that of saline when injected through intravenous or subcutaneous route for clinical practice.

The Effect of Butanol Fraction of Polygonatum odoratum with Vitamin E on Blood Glucose Levels and Lipid Peroxidations in Streptozotocin-Induced Diabetic Rats (둥굴레(Polygonatum odoratum)분획물과 비타민 E 투여가 Streptozotocin 유발 당뇨 흰쥐의 혈당과 지질과산화에 미치는 영향)

  • The hypoglycemic effects of butanol(BuOH) fraction of Polygonatum odoratum with vitamin E in streptozotocin(STZ)-induced diabetic rats were investigated. Sprague-Dawley rats weighing 200-230g were devided into five groups, and four groups induced diabetes mellitus by the STZ injection(45mg/kg b.w.) into the tail vein : Normal, diabetic-control, and three diabetic experimental groups(p. odoratum group, P. od-vit. E group and Vit. E group). All groups were fed on a AIN-76 diet, and the experimental groups were orally administered with the BuOH fraction of Polygonatum odoratum(500mg/kg b.w.) and vitamin E(10mg/kg b.w.) for 14 days. The body weight, diet intake and organ weights were monitored. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol and aspartate and alanine aminotransferase activities were analyzed. The levels of glycogen in liver and muscle, cholesterol in liver were determined. The malondialdehyde(MDA) levels in liver, kidney and lung were assayed. The body weight loss was seen in P. odoratum group, P. od-vit. E group, Vit. E group and diabetic control group, while the loss in P. odoratum group was much less than that in the diabetic control group. The plasma glucose levels were significantly lowered in P. odoratum group compared to diabetic control group. The plasma insulin levels were noticeably higher in P. odoratum and Vit. E groups. The rats in P. odoratum and P. od-vit. E group showed higher liver glycogen levels than in the diabetic control group. The MDA levels in liver, kidney and lung were also significantly reduced in P. od-vit. E and Vit. E groups compared to the diabetic control group. The results suggest that the administration of BuOH fraction of Polygonatum odoratum along with vitamin E reduced blood glucose levels and peroxidative tissue damage in STZ-induced diabetic rats, showing the possibility of preventive and therapeutic use of the wild edible plant to the diabetes mellitus. The Effect of Each Fraction of Methanol Extract of Alisma canaliculatum on Blood Glucose Levels and Lipid Metabolism in Streptozotocin Induced Diabetic Rats (택사 분획물의 투여가 Streptozotocin 유발 당뇨 흰쥐의 혈당수준과 지질대사에 미치는 영향)

  • Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-245g by injection of streptozotocin(STZ) into the tail vein at a dose of 45mg/kg and were divided into seven groups ; normal, diabetic control, and five experimental groups(fractions of hexane, chloroform(CHCl$_3$), ethylacetate(EtOAc), butanol(BuOH) and water($H_2O$)). The rats of all groups were fed on AIN-93 diet and the five experimental groups were orally administered each fraction for 14 days. The body weight and diet intake were monitored daily. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol, free fatty acid and aspartate and alanine aminotransferase activities were analyzed. Diabetic rats showed the lower weight gain compared to the normal rats. The plasma glucose levels of the CHCl$_3$, and $H_2O$ fraction groups were significantly lower than the other experimental groups. The plasma insulin level of the CHCl$_3$ fraction group was much higher than that of diabetic control group. The plasma cholesterol levels were increased in all the experimental groups. The groups of hexane, BuOH and H2() fractions showed the lower plasma triglyceride levels compared to diabetic control group. The plasma free fatty acid levels were not significantly different in all groups. HDL-cholesterol levels were definitely higher in hexane, CHCl$_3$ and EtOAc fraction groups than that of diabetic control group. In conclusion, administration of CHCl$_3$ and $H_2O$ fractions of methanol extract of Alisma canaliculatum exhibited hypoglycemic effects in STZ induced diabetic rats, showing the possibility of therapeutic use of Alisma canaliculatum to the diabetes mellitus.
