A novel retinol derivative, polyethoxylated retinamide(Medimin A) was synthesized, as an anti-aging agent. Collagen synthesis, skin permeation, stability, and toxicity of Medimin A were evaluated and compared with those of retinol and retinyl palmitate. In vitro collagen synthesis was evaluated by quantitative assay of $[^3H]-proline$ incorporation into collagenase sensitive protein in fibroblast cultures. For in vitro skin permeation experiments, Franz diffusion cells(effective diffusion area: 1,766 $cm^2$) and the excised skin of female hairless mouse aged 8 weeks were used, The stabilities of retinoids were evaluated at two different temperature($25^{\circ}C\;and\;40^{\circ}C$) and under UV in solubilized state and in O/W emulsion. To estimate the safety, acute oral toxicity, acute dermal toxicity, primary skin irritation, acute eye irritation and human patch test were performed. The effect of Medimin A on collagen synthesis was similar to that of retinol. The skin permeability of Medimin A was higher than those of retinol and retinyl palmitate. The Medimin A was more stable than retinol and retinyl palmitate. Medimin A was nontoxic in various toxicological tests. These results suggest that Medimin A would be a good anti-aging agent for enhancing bioavailability and stability.
Tank mixing application of pesticides has been used to reduce labor and to control wide spectrum of pests, but it may cause significant pesticide poisoning on human and animals due to carelessness. The toxic response of pesticides for the tank mixtures and mixtures was investigated to determine acute toxicity and enzymatic change using experimental animals. Acute oral toxicity and acute dermal toxicity were tested by RDA test guideline. The $LD_{50}$ was calculated by probit analysis method and cholinesterase was measured with automatic analyzer. The toxicities were generally higher than estimated toxicities in tank mixing and mixture. Serum cholinesterase activity was inhibited more than expected at the dose levels of 1/5, 1/10 and 1/20 of $LD_{50}$. Therefore, the results of this study showed that acute toxicity caused by the pesticide mixtures should be considered before the tank mixing method is applied.
The acute toxicity test of high bio-active plant essential oils was conducted with Lavender, Lemon eucalyptus and Cassia oils selected to develop environment-friendly insecticides. The results of acute oral toxicity using rats showed that $LD_{50}$ of over 2,000 mg/kg bw for Lavender, Lemon eucalyptus and Cassia oils. The calculated acute dermal $LD_{50}$ value of all testing materials was over 4,000 mg/kg bw. The Skin irritation test indicated that Lavender and Lemon eucalyptus oil have no irritation while Cassia oil has a moderate irritation. For the Eye Irritation test, the result showed no irritation for Lavender and Lemon Eucalyptus oil and irritation for Cassia oils. However, the irritation was not showed for Eye Irritationwashing test of Cassia oil. Consequently, the Lavender and Lemon eucalyptus oils were showed to be low in toxicity whereas Cassia oil indicated to cause a moderate irritation on the skin and eyes.
Loess-sulfur complex has been widely used as an environmental friendly organic materials for insecticides and fungicides in crop cultivation. However, there are high concerns about skin and eye irritation for farm workers due to the high alkaline properties of loess-sulfur complex. The acute toxicity evaluation was conducted with three samples of loess-sulfur complex in different pH (pH 9, 10, 11) in order to supply the evidentiary data for selecting the optimal product among the test materials. The results of acute oral toxicity using rats showed $LD_{50}$ of over 2,000 mg/kg b.w. for all three samples of loess-sulfur complex. The calculated acute dermal $LD_{50}$ of all tested materials was over 4,000 mg/kg b.w.. The Skin and eye irritation indicated that all tested materials have no irritation. Consequently, it was suggested that loess-sulfur complex be low in acute toxicity at all different pH values (pH 9~11).
Highly bio-active plant essential extracts and oils from pepper, clove bud, rosemary and origanum which are selected to develop environment-friendly insecticides was studied for their acute toxicity. The results of acute oral toxicity using rats showed $LD_{50}$ value of over 2,000 mg/kg bw for pepper, clove bud, rosemary and origanum oils. The calculated acute dermal $LD_{50}$ value of pepper was over 4,000 mg/kg bw and anther testing materials was over 4,000 mg/kg bw. The skin irritation test showed that pepper, clove bud and rosemary oils had no irritation while origanum oil had a moderate irritation. According to the eye irritation test, it showed that there was no irritation for pepper and rosemary oils, while there were irritation for clove bud and origanum oils. Consequently, pepper and rosemary oils were shown to be low in toxicity whereas clove bud oil was indicated to cause a mild eye irritability and origanum oil, causing a moderate skin and eye irritability.
Trichloroacetonitrile is used as an intermediate in insecticides, pesticides, and dyes. In Korea alone, over 10 tons are used annually. Its oral and dermal toxicity is classified as category 3 according to the globally harmonized system of classification and labelling of chemicals, and it is designated a toxic substance by the Ministry of Environment in Korea. There are no available inhalation toxicity data on trichloroacetonitrile. Thus, the present study performed inhalation tests to provide data for hazard and risk assessments. Sprague-Dawley rats were exposed to trichloroacetonitrile at concentrations of 4, 16, or 64 ppm for 6 hour per day 5 days per week for 13 weeks in a repeated study. As a result, salivation, shortness of breath, and wheezing were observed, and their body weights decreased significantly (p < 0.05) in the 16 and 64 ppm groups. All the rats in 64 ppm group were dead or moribund within 4 weeks of the exposure. Some significant changes were observed in blood hematology and serum biochemistry (e.g., prothrombin time, ratio of albumin and globulin, blood urea nitrogen, and triglycerides), but the values were within normal physiological ranges. The major target organs of trichloroacetonitrile were the nasal cavity, trachea, and lungs. The rats exposed to 16 ppm showed moderate histopathological changes in the transitional epithelium and olfactory epithelium of the nasal cavity. Nasal-associated lymphoid tissue (NALT) and respiratory epithelium were also changed. Respiratory lesions were common in the dead rats that had been exposed to the 64 ppm concentration. The dead animals also showed loss of cilia in the trachea, pneumonitis in the lung, and epithelial hyperplasia in the bronchi and bronchioles. In conclusion, the no-observed-adverse-effect level (NOAEL) was estimated to be 4 ppm. The main target organs of trichloroacetonitrile were the nasal cavity, trachea, and lungs.
As the use of cosmetics has greatly increased in a daily life, safety issues with cosmetic ingredients have drawn an attention. Drometrizole [2-(2'-hydroxy-5'-methylphenyl)benzotriazole] is categorized as a sunscreen ingredient and is used in cosmetics and non-cosmetics as a UV light absorber. No significant toxicity has been observed in acute oral, inhalation, or dermal toxicity studies. In a 13-week oral toxicity study in beagle dogs, No observed adverse effect level (NOAEL) was determined as 31.75 mg/kg bw/day in males and 34.6 mg/kg bw/day in females, based on increased serum alanine aminotransferase activity. Although drometrizole was negative for skin sensitization in two Magnusson-Kligman maximization tests in guinea pigs, there were two case reports of consumers presenting with allergic contact dermatitis. Drometrizole showed no teratogenicity in reproductive and developmental toxicity studies in which rats and mice were treated for 6 to 15 days of the gestation period. Ames tests showed that drometrizole was not mutagenic. A long-term carcinogenicity study using mice and rats showed no significant carcinogenic effect. A nail product containing 0.03% drometrizole was nonirritating, non-sensitizing and non-photosensitizing in a test with 147 human subjects. For risk assessment, the NOAEL chosen was 31.75 mg/kg bw/day in a 13-week oral toxicity study. Systemic exposure dosages were 0.27228 mg/kg bw/day and 1.90598 mg/kg bw/day for 1% and 7% drometrizole in cosmetics, respectively. Risk characterization studies demonstrated that when cosmetic products contain 1.0% of drometrizole, the margin of safety was greater than 100. Based on the risk assessment data, the MFDS revised the regulatory concentration of drometrizole from 7% to 1% in 2015. Under current regulation, drometrizole is considered to be safe for use in cosmetics. If new toxicological data are obtained in the future, the risk assessment should be carried out to update the appropriate guidelines.
KNI3126 which is the eco-friendly material made by plant extracts - neem and matrine - have been evaluated for efficacies against 6 main pests and toxicity on natural enemies. Insecticidal efficacies of KNI3126 against plant hopper were above 95% at 5 days after treatment (DAT), whereas pesticidal effects against the cotton aphid were above 95% at 3 DAT and slightly decreased at 5 DAT. Insecticidal efficacy against the palm thrips resulted in lower control value as 68.1% than that of chemical insecticides. KNI3126 showed more than 95% control value against diamond back moth, suggesting that it could suppress the population of pest hard to control as eco-friendly material. Mortalities against two-spotted spider mite were about 80% at 1 DAT and over 90% at 5 DAT, respectively. For evaluation of the toxicity on natural enemies, effect against predatory natural enemy was classified as moderate selective toxicity based on the criterion of International Organization of Bio-Control (IOBC), whereas against parasitic natural enemies was found to be relatively safe. Repellent effect was weak against plant hopper, but strong against two-spotted spider mite. In the toxicity test for safety, KNI3126 was classified as the lowest level at acute oral, acute dermal and fish toxicity test and did not induce the irritancy at skin irritation test and eye irritation test.

Crude chondroitin sulfates extracted from midduck tunics (Styela clava) and munggae tunics (Halocynthia roretzi) were examined in vivo in order to be utilized as a cosmetic material which was followed by an in vitro assay. Examinations, such as acute oral toxicity, skin sensitization, acute eye irritation, and primary skin irritation, were peformed with a variety of laboratory animals. Phototoxic and photosensitization tests were not conducted since all chondroitin sulfates failed to absorb U.V. light at the range of 280 to 420 nm. In acute dermal and eye irritation, both specific clinical signs and dead cases were not demonstrated during the test period, but crude chondroitin sulfates from midduck and munggae tunics, and standard chondroitin sulfate from bovine trachea were showed 2.5, 1 and 1.25 of acute ocular irritation index (A.O.I.), respectively. In the case of skin sensitization, crude chondroitin sulfate from midduck tunics exhibited neither specific clinical signs nor dead cases in the entire course of the examination. While in acute oral toxicity, crude chondroitin sulfates from both midduck and munggae tunics found neither specific clinical signs nor dead cases during the test, and LD50 was suspected to be over 2 g/kg. Based on this study, it was proven that crude chondroitin sulfates from either midduck or munggae tunics can be used safely as a cosmetic material.
Penthiopyrad is a fungicide agent in types of pyrazole which is showing the effect of prevention in fungal disease and powdery mildew. In order to register this new pesticide, reports of acute toxicity and chronic toxicity by animal study were examined to set acceptable daily intake to evaluate hazards of consumers. Acute toxicity was low in toxic, and it did not have the effect of acute dermal toxicity, acute eye irritation, or skin sensitization. As the result of the study in chronic toxicity, the common effect of chemical appeared in the liver and thyroid which was proven as a toxic effect. Two-generation reproduction toxicity, genotoxicity, and prenatal development toxicity were not proven. As the result of carcinogenic study, increase of thyiroid follicular adenoma in the rat and the frequency of liver hepatocellar adenoma in mice were also increased. However, it was decided that the threshold value on the effect in chemicals could be controlled through study liver enzyme induction. Therefore, the ADI for penthiopyrad is 0.081 mg/kg/ bw/day, based on the NOAEL of 8.10 mg/kg bw/day of twelve-months dogs study and applying an uncertainty factor of 100.
