Cytotoxic Activity of Leguminous Seed Extracts against Human Tumor Cell Lines

  • The cytotoxic activity of methanol extracts of 25 leguminous seeds in vitro was evaluated by sulforhodamine B assay, using the five human solid A549 lung, SK-OV-2 ovarian, SK-MEL-2 melanoma, XF-498 CNS and HCT-15 colon tumor cell lines. The responses varied with both cell line arid leguminous seed used. Extracts of Canavalia lineata and Glycine soja revealed potent cytotoxic activity against A549 arid SK-MEL-2 cell lines. Moderate activity was observed in the extracts of Cassia obtusifolia and Glyeine max var. chungtae, and C. lineata and Vigna angulasis against SK-MEL-2 and HCT-15 cell lines, respectively. The other seed extracts were ineffective against model tumor cell lines. Because of their potent cytotoxic activities, the activity of each solvent fraction from C. lineata and G. soja was determined and the potent activity was produced from their chloroform fractions. As a naturally occurring therapeutic agent, leguminous seeds described could be useful for developing new types of anti-tumor agents.

  • We have reviewed the magnetic activity in close binaries. Solar like magnetic activity indicators such as photometric spots, chromo spheric emission, coronal X-ray and radio emission, and flare activity are commonplace in many cool stars with convective envelopes. Using the UV spectra we confirmed the strength of stellar activity increases with more rapid rotation and later spectral types which corresponds to the increasing depth of the star's convective envelope. Apart from very young stellar objects such as T Tauri stars, the stars with the highest levels of activity are close binary systems composed of cool stars, i.e., the chromospherically active binaries such as RS CVn, BY Dra, W UMa and related systems. The IUE low and high dispersion spectra of V711 Tau, VW Cep and SW Lac are used for ultraviolet photometry and for a variation study of chromospheric activity. Evidence of chromospherically activity is indicated by the intensity variation of the Mg II emission line with orbital phase.

Antioxidant and Antidiabetic Activities of Extracts from Quercus serrata Thunb and Q. acutissima Carruther (도토리(Quercus serrata Thunb)와 상수리(Quercus acutissima Carruther) 추출물의 항산화와 항당뇨 활성 연구)

  • This study was conducted to analyze the antioxidant and antidiabetic activities of acorns according to the types of Quercus serrata Thunb (QST) and Q. acutissima Carruther (QAC). The total polyphenol contents of the extracts from QST and QAC were 220.59 and 320.96 mg GAE/g, respectively. The content of total polyphenol of QAC was higher than that of QAC (p<0.001). DPPH (2,2 Diphenyl 1 picrylhydrazyl) radical scavenging activity, reducing power and superoxide dismutase (SOD)-like activity were increased in a concentration-dependent manner by both acorn extracts, and QAC showed high activity in all antioxidant experiments (p<0.05). The inhibitory activities of α-glucosidase and α-amylase were also increased in a concentration-dependent manner, and QAC showed higher inhibitory activity than QST (p<0.05). Our study indicates that QST and QAC are functional food materials with high antioxidant and antidiabetic activities. In addition, QAC has a higher physiological activity than QST.

  • The histochemical, cytochemical, and immunocytochemical investigations were conducted to find out the cellulase activity in the accessory glands of the digestive system of the oriental land snail Nesiohelix samarangae under the LM, SEM, and TEM. The cellulase activity was shown in the epithelium of th digestive gland by labelling with the immunogold (protein-A gold) particles. The epithelial cells showing the cellulase activity were Type 1 and Type 3 cells out of five types of the epithelial cells of the digestive gland. None of epithelial cells of the mucus gland and the salivary gland and the salivary gland were not labeled with the immunogold particles.

Antioxidative Activity of Rice Grain using FI-CL and ESR Methods

  • Fifty-four Korean native and 28 foreign rice varieties harvested in 1998 and 1999 were examined for antioxidative activity that is measured to a chemiluminescence and superoxide radical intensity, by the flow injection chemiluminescence (FI-CL) system and an electron spin resonance (ESR) spectrophotometer, respectively. In the chemiluminescence measurement by FI-CL, radical scavenger activity did not differ significantly among rice varieties between origin types of rice varieties, and between storage periods. GinSun and Hongchoengdo, colored rice exhibited high electron scavenging effect by ESR. Therefore, these results indicate that the pigments of rice varieties may play important antioxidative roles and that it may be possible to breed rice varieties with higher antioxidative potentials.

  • Neprilysin (Nep) is known to be important to degrade $A{\beta}$ derived from amyloid precursor protein (APP) by cleavage with $\beta-and\;\gamma$-secretases. In order to determine whether a correspondence between $A{\beta}-42/{\gamma}-secretase$ activity and Nep levels exists in postnatal aging of transgenic mice expressing either neuron-specific enolase (NSE)-controlled human mutant presenilin-2 (hPS2m) or APPsw alone, the levels of Nep expression and $A{\beta}-42/{\gamma}-secretase$ activity were examined age of 5, 12, and 20 months, respectively. The levels of Nep expression in both types of transgenic brains were decreased relative to those of control mice in a aging-related manner, while the level of $A{\beta}-42/{\gamma}-secretase$ activity was reversibly increased. Thus, changes in $A{\beta}-42$ may all reflect variation in amounts of Nep enzyme.

Analysis of Unmet Medical Needs according to Mental Health (정신건강유형에 따른 미충족 의료 현황 분석)

  • Objectives : This study investigated the prevalence of unmet health care needs among Korean adults and related factors. Methods : The study participants were adults over the age of 20 mental health experience from the Korea Health Panel in 2012(n=4,730). Statistical analysis methods used in this study were the ${\chi}^2$-test, Logistic Regression Analysis and other basic statistics such frequency-and percentage using SPSS version 22.0. Results : (1)Significant variables of stress: Factors were age, economic activity, subjective health status, and activity limitation. (2)Significant variables of depression: Factors were age, income class(low) and activity limitation. (3)Significant variables of suicidal impulse: Factors were age, chronic diseases, income class, and activity limitation. Conclusions : Stress, depression, and suicidal impulse can be unmet medical factors; therefore improvement measures and mental health counseling programs in response to suicide impulses, should be developed. Thus there is a need for a health sciences approach.

Simultaneous enhancement of thermostability and catalytic activity of phospholipase $A_1$ by evolutionary molecular engineering

  • The thermal stability and catalytic activity of phospholipase A$_1$ from Serratia sp. MK1 were improved by an evolutionary molecular engineering. Two thermostable mutants were isolated after sequential rounds of error-prone PCR to introduce random mutations and filter-based screening of the resultant mutant library, and identified as having six (mutant TA3) and seven (mutant TA13) amino acid substitutions, respectively. Different types of the substitutions were found in two mutants, resulting in the increase of nonploar residues (mutant TA3) or changes between side chains within polar or charged residues (mutant TA13). The wild-type and mutant enzymes were purified, and the effect of temperature on their stability and catalytic activity was investigated. The T$\sub$m/ values of TA3 and TA13 were increased by 7 and 11$^{\circ}C$, respectively. Thus, evolutionary molecular engineering was found to be an effective and efficient approach to increasing thermostability without compromising enzyme activity.

  • Moringa oleifera Lam. or munga is one of the most important plant widely cultivated in India. It belongs to family Moringaceae. This plant is widely used as nutritional herb and contains valuable pharmacological action like anti-asthmatic, anti-diabetic, hepatoprotective, anti-inflammatory, anti-fertility, anti-cancer, anti-microbial, anti-oxidant, cardiovascular, anti-ulcer, CNS activity, anti-allergic, wound healing, analgesic, and antipyretic activity, Moringa oleifera Lam. The plant is also known as Horse - radish tree, Drumstick tree. Every part of this plant contains a valuable medicinal feature. It contain rich source of the vitamin A, vitamin C and milk protein. Different types of active phytoconstituents like alkaloids, protein, quinine, saponins, flavonoids, tannin, steroids, glycosides, fixed oil and fats are present. This plant is also found in the tropical regions. Some other constituents are niazinin A, niazinin B and niazimicin A, niaziminin B. The present review discusses the phytochemical composition, medicinal uses & pharmacological activity of this plant.

Effect of oral administration of Ginsenoside-Rb2 on rotavirus infection

  • Glycosaminoglycans(PT -Gag) were isolated from the porcine testis. From the PT -Gag. we obtained two different types of Gag fractions using Dowex macroporous Resin MSA-1 column. PT-Gag-1.5% NaCl and PT -Gag-16% NaCl. Various biological activities of the GAGs were examined in aspect of anticoagulant and immunomodulating activity. The anticoagulant activity of the GAGs was evaluated by activated partial thromboplastin time (aPTT ) assay and thrombin time (TT) assay. The GAGs of porcine testis markedly increased the clotting times of both of aPTT and TT. showing that PT-Gag-16% NaCl was more effective than PT-Gag-1.5% NaCl. The immunomodulating activity of the GAGs was examined in relation to regulation of cytokine production of mutine peritoneal macrophages. Treatment with the GAGs promonently enhanced the prodution of cytokines. IFN-${\gamma}$, from macrophages. Taken together. GAGs isolated from porcine testis possess biological functions such as anticoagulant and immunomodulating activity.
