Biochemical Studies on Korean Fermented Foods. VIII Studies on Vitamin $B_{12}$ Contents of the Fermented Foods in Korea (한국 발효식품에 대한 생물화학적 연구 ( 제 8 보) 발효식품중의 Vitamin $B_{12}$의 함량 조사보고)

  • In order to know contents of vitamin B$_{12}$ in the fermented foods in Korea, the contents of this vitamin are studied on the following foods; kimchies (pickled vegetables) .................. 39 chukkals (fermented and salted sea foods) ...... 8 fermented soy-bean products .................... 5 the kimchies and fermented soybean products studied in this paper, are from individual homes and chukkals are from markets. The content of vitamin B$_{12}$ is estimated by the microbiological assay method using lactobacillus leichmannii ATCC 7830. Details for assay are indicated in the original part of this paper. The pseudo-vitamin B$_{12}$ substances as thymidine which is active to the growth of lactobacillus leichmannii, are eliminated by the alkali treatment method of sample solution. According to the results as indicated in the table, the following conclusions are summerized; 1. Vitamin B$_{12}$ contents of kimchies for winter season are 1.03 to 1.52 mcg% in average. The hobakk-kimchi which contain the highest content of vitamin B$_{12}$ is not popular one among Korean. 2. Chukkals contain much higher content of vitamin B$_{12}$ varying from 0.91 to 11.10mcg%. 3. The soybean fermented foods, as kanjang, denjang, kochojang, contain lower content of vitamine B$_{12}$ varying from 0.08 to 0.52 mcg% containing higher content of pseudo-vitamin. 4. Based on daily consumption of kimchi in winter season by Chai, the consumption of this vitamin through kimchi is about 3 mcgs daily per capita. This will be a quite source of this vitamin for Korean as vitamin C. as vitamin C.

  • In order to confirm on production of vitamin $B_{12}$ during the kimchi fermentation period, the variation of its content is studied in this paper As a sample of kimchi for this fermentation study due to the seasonal condition, nabakkimchi which is aseasonal one in early spring, is prepared by author. The content of vitamin $B_{12}$ is estimated by the microbiological assay method using lactobacillus leichmannii A Tee 7830. Details for assay are indicated in the original part. And the pseudo-vitamin $B_{12}$ substances as thymidine which is active to the growth of lactobacillus leichmannii, are eliminated by the alkali treatment method of sample solution, According to the results shown in table 2 and 3 and figures 1 to 4, the following conclusions are summerized; 1. Vitamin $B_{12}$ produced during the fermentation period of kimchi. And the content of this vitamin during its optimum fermentation period, is much higher content compared with the theoretically calculated amount from its materials, This has been confirmed at the repeated experiment. The trend of variation during its period is indicated at the figures. 2. The sterilized kimchi inhibited the fermentation by sterilization at the preparation, did not increased its content showing only its theoretically calculated amount.

  • Poly(vinyl chloride)(PVC) membrane electrodes based on the lipophilic neutral carrier, dibenzo-18-crown-6(DB18C6) and benzo-15-crown-5 (B15C5) as the active sensors for Tl$^+$ ion have been prepared and tested in different content of the potassium tetrakis(4-chlorophenyl)borate (KTClPB) as lipophilic salt. Dioctyl adipate (DOA), 2-nitrophenyl phenyl ether (NPPE) and o-nitrophenyl actyl ether (NPOE) were used as plasticizing solvent mediators. Electrodes exhibited good linear responses of 40∼55 mV decade$^{-1}$ for Tl$^+$ ion within the concentration ranges 10$^{-1}$∼10$^{-5}$M TlNO$_3$. Selectivity coefficients of interfering ions (alkali metal, alkaline earth metal and some transition metal ions) for Tl$^+$-ISE were determined by separate solution method and were sufficiently small for most of them. These crown ether type ion-selective electrodes are suitable for use with aqueous solution at pH > 3.

Improvement of Hardwood Pulp Yield in Continuous Kraft Cooking and Estimation of Pulp Yields Pulp yields of isothermal cooking with polysulfide and anthraquinone

  • The pulp yield was improved by about 4.5-5% when polysulfide (PS) and anthraquinone (AQ) were added to the kraft cooking liquor (white liquor). The exchange of the black liquor with fresh white liquor further increased the yield. The highest pulp yield was obtained when the PS cooking liquor containing 70% of total active alkali (AA) and 100% of AQ was used from the beginning of the reaction and the black liquor was exchanged with fresh white liquor containing the residual 30% of AA just after temperature reached $135^{\circ}C$. There was a good correlation between kraft pulp yields of a hardwood species and the ratios of the amount of xylose to glucose (X/G ratio), liberated by an acid hydrolysis of the pulps. However, the correlation was dependent on raw material wood species. Therefore, it is required in advance to establish a correlation between the yields and X/G ratios for raw material wood species of a target pulp in order to estimate pulp yield using X/G ratio. The X/G ratios of relatively high yield pulps showed higher values than those expected from the correlation. In a mill trial, the superiority of the PS-AQ isothermal cooking (ITC) process over the kraft ITC process was confirmed by examining X/G ratio of pulps obtained. The pulp yield in the PS-AQ ITC process was estimated at about 57.0%. This yield is very high, which indicates that reaction conditions of the PS-AQ ITC process are optimal.

  • A heat pipe residual heat removal system is proposed to be incorporated into the reactor driven subcritical (RDS) facility, which has been proposed by MIT Nuclear Reactor Laboratory for testing and demonstrating the Fluoride-salt-cooled High-temperature Reactor (FHR). It aims to reduce the risk of the system operation after the shutdown of the facility. One of the main components of the system is an air-cooled heat pipe heat exchanger. The alkali-metal high-temperature heat pipe was designed to meet the operation temperature and residual heat removal requirement of the facility. The heat pipe model developed in the previous work was adopted to simulate the designed heat pipe and assess the heat transport capability. 3D numerical simulation of the subcritical facility active zone was performed by the commercial CFD software STAR CCM + to investigate the operation characteristics of this proposed system. The thermal resistance network of the heat pipe was built and incorporated into the CFD model. The nominal condition, partial loss of air flow accident and partial heat pipe failure accident were simulated and analyzed. The results show that the residual heat removal system can provide sufficient cooling of the subcritical facility with a remarkable safety margin. The heat pipe can work under the recommended operation temperature range and the heat flux is below all thermal limits. The facility peak temperature is also lower than the safety limits.

Sample pre-treatment for measurement of $^{129}$I in radwastes (방사성폐기물 중 $^{129}$I 측정을 위한 시료의 전처리)

  • Many different kinds of radwastes are discharged from the nuclear power plants, and $^{129}$I is included in these radwastes. Recovery test of $^{129}$I was evaluated for different radwastes(dry active waste, sludge, spent resin and simulated evaporator bottom). Recovery of $^{129}$I for dry active waste by acid leaching with $1.8\%$ NaClO was $74.3\%$$(RSD,\;2.2\%)$ and l291 for spent rein by alkali fusion method with KOH as a flux agent was $87.7\%$$(RSD,\;0.9\%$), respectively. iodide in simulated evaporator bottom containing a high concentration of borate was adsorbed with anion exchange resin at pH 7 phosphate buffer solution. Recovery of $^{129}$I for anion exchange resin was $92.5\%$ and not affected up to 1,200 $\mu$g/mL $H_3BO)3$(as a Boron). Recovery of $^{129}$I for the spent resin from nuclear power plant was $87.2\%$ $(RSD,\;1.2\%)$.

Immunochemical Studies for the Characterization of Purified $(Na^+,\;K^+)-ATPase$ and Its Subunits with a Special Reference of Their Effect on Monovalent Cation Transport in Reconstituted $(Na^+,\;K^+)-ATPase$ Vesicles

  • A highly purified $(Na^+,\;K^+)-ATPase$ from the rectal gland of Squalus acanthias and from the electric organ of Electrophorus electricus has been used to raise antibodies in rabbits. The 97,000 dalton catalytic subunit and glycoprotein derived from the rectal gland of spiny shark were also used as antigens. The two $(Na^+,\;K^+)-ATPase$ holoenzymes and the two shark subunits were antigenic. In Ouchterlony double diffusion experiments, these antibodies formed precipitation bands with their antigens. Antibodies prepared against the two subunits of shark holoenzyme also formed precipitation bands with their antigens and shark holoenzyme, but not with eel holoenzyme. These observations are in good agreement with inhibitory effect of these antibodies on the catalytic activity of $(Na^+,\;K^+)-ATPase$ both from the shark and the eel, since there is very little cross-reaction between the shark anticatalytic subunit antibodies and the eel holoenzyme. The maximum antibodies titer of the anticatalytic subunit antibodies is found to be 6 weeks after the initial single exposure to this antigen. Multiple injections of the antigen increased the antibody titer. However, the time required to produce the maximum antibody titer was approximately the same. These antibodies also inhibit catalytic activity of $(Na^+,\;K^+)-ATPase$ vesicles reconstituted by a slow dialysis of cholate after solubilization of the enzyme in a presonicated mixture of cholate and phospholipid. In these reconstituted $(Na^+,\;K^+)-ATPase$ vesicles, effects of these antibodies on the fluxes of $Na^+$, $Rb^+$, and $K^+$ were investigated. Control or preimmune serum had no effect on the influx of $^{22}Na^+$ or the efflux of $^{86}Rb^+$. Immunized sera against the shark $(Na^+,\;K^+)-ATPase$ holoenzyme, its glycoprotein or catalytic subunit did inhibit the influx of $^{22}Na^+$ and the efflux of $^{86}Rb^+$. It was also demonstrated that these antibodies inhibit the coupled counter-transport of $Na^+$ and $K^+$ as studied by means of dual labeling experiments. However, this inhibitory effect of the antibodies on transport of ions in the $(Na^+,\;K^+)-ATPase$ vesicles is manifested only on the portion of energy and temperature dependent alkali metal fluxes, not on the portion of ATP and ouabain insensitive ion movement. Simultaneous determination of effects of the antibodies on ion fluxes and vesicular catalytic activity indicates that an inhibition of active ion transport in reconstituted $(Na^+,\;K^+)-ATPase$ vesicles appears to be due to the inhibitory action of the antibodies on the enzymatic activity of $(Na^+,\;K^+)-ATPase$ molecules incorporated in the vesicles. These findings that the inhibitory effects of the antibodies specific to $(Na^+,\;K^+)-ATPase$ or to its subunits on ATP and temperature sensitive monovalent cation transport in parallel with the inhibitory effect of vesicular catalytic activity by these antibodies provide direct evidence that $(Na^+,\;K^+)-ATPase$ is the molecular machinery of active cation transport in this reconstituted $(Na^+,\;K^+)-ATPase$ vesicular system.

Phytochemical-based Tannic Acid Derivatives as Draw Solutes for Forward Osmosis Process (정삼투 공정의 유도용질로서의 식물 화학물질 기반의 탄닌산 유도체)

  • Potassium tannate (TA-K), which is prepared by base treatment of the bio-renewable tannic acid (TA), was evaluated for its potential application as a draw solute for water purification by forward osmosis. The forward osmosis and recovery properties of TA-K were systematically investigated. In the application of forward osmosis through the active layer facing feed solution (AL-FS) method, the water flux of TA-K draw solution was significantly higher than that of the TA draw solution, while that of the latter was not identified. At a low concentration of 100 mM, the osmotic pressure (1,135 mOsmol/kg) of the TA-K draw solution was approximately 6.5 times that (173 mOsmol/kg) of the NaCl draw solution. Furthermore, the water flux and specific salt flux (6.14 LMH, 1.26 g/L) of the TA-K draw solution at 100 mM were approximately 2.5 and 0.5 times those of the NaCl draw solution (2.46 LMH, 2.63 g/L) at the same concentration, respectively. For reuse, TA-K was precipitated by using a metal ion and recovered through membrane filtration. This study demonstrates the applicability of a phytochemical material as a draw solute for forward osmosis.

Thermophilic Anaerobic Acid Fermentation of Food Wastes after NaOH Addition (NaOH 첨가에 따른 음식물찌꺼기 고온 혐기성 산발효)

  • This study showed that thermophilic anaerobic acid fermentation of food wastes had an enhanced hydrolysis capability and improvement of acidification efficiency. Influence of pH on the anaerobic hydrolysis and acidogenesis was investigated to determine the proper alkalinity in the thermophilic fermentation of food wastes. The results of putting NaOH as alkali to evaluate hydrolysis and acid fermentation efficiency In acid fermentation process of food wastes showed that the food wastes pretreated with 0.05 g NaOH/g TS had the maximum 12,600 mg/L of VFAs concentration during HRT 3 days in $55^{\circ}C$ thermophilic condition and the maximum 9,700 mg/L of VFAs concentration during HRT 5 days in $35^{\circ}C$ mesophilic condition. The accomplishment of high VFAs concentration resulted from that the main component of food wastes such as cellulose, lignin and etc. is performed active chemical decomposition by alkali in thermophilic condition. The major components of VFAs produced from the thermophilic acid fermentation process of food wastes were the short chain fatty acids such as acetic acid, butyric acid, and propionic acid.

Studies on the Physico-chemical Properties and Characterization of Soil Organic Matter in Jeju Volcanic Ash Soil (제주도(濟州道) 화산회토양(火山灰土壌)의 이화학적(理化学的) 특성(特性) 및 유기물(有機物) 성상(性状)에 관(関)한 연구(硏究))

  • A series of laboratory experiment was conducted to find out the chemical composition, characterization of humic substances by physical and chemical methods and reaction of Na-pyrophosphate, $Ca(OH)_2$ and rice straw with albumin on the degradation of soil organic matter in the volcanic ask soils of the Jeju Island. Results obtained were summarized as follows: 1. The contents of organic matter, available silicon, active iron and aluminum concentration in volcanic ash the soils were remarkably higher but available phosphorous was comparatively lower than the mineral soils. In volcanic ash soil, the contents of potassium, calcium and magnessium were higher in upland soil than that of forest soil. The ratios of active $Al^{{+}{+}{+}}/Fe^{{+}{+}}$, C/P and $K/Ca^+$ Mg were apparently high in volcanic ash soils while that of $SiO_2$/O.M. was high in mineral soil. 2. The carbon/nitrogen ratio in humin, humic acid content in organic matter, and carbon contents of humin in total carbon of soil organic matter were apparently higher in the volcanic ash soils than in the mineral soils, The total nitrogen and fractions of acid or alkali soluble nitrogen were remarkably high in volcanic ash soils while mineralizable nitrogen ($NH_4$-N and $NO_3$) contents were high in mineral soils. 3. The values of K600, RF and log K were also higher in volcanic ash soils than those in mineral soils, and the absorbance in the visible range were high and color was dark in the soil of which humification was progressed Extracted humic acid from volcanic ash soil was less reactive to the oxidizing chemical reagent and was persistance to the acid or alkali hydrolysises. 4. The major oxygen-containing functional groups in humic substances of volcanic ash soils were phenolic-OH alcoholic-OH and carboxyl groups while those in mineral soil were methoxyl and carbonyl groups. 5. Absorption spectra of alkaline solution of humic acid ranged from 200 nm to maxima 500 nm. Visible spectra peaks of from humic substances in the visible region were recognized at 350, 420, 450 and 480 nm. Only one single absorbance peak was observed in the visible region at 362 nm for Heugag series and two absorbance Peak were also at 360 nm and 390 nm for Yeungrag series. 6. Evolution of carbon as $Co_2$ was increased with addition of Na-pyrophosphate in Namweon and Heugag series, and "priming effects" took place on the soil organic matter decomposition by addition of rice straw with albumin in Ido series.
