Genetically engineered strains of Pseudomonas, which could degrade the various aromatic hydrocarbons, were acclimated to the synthetic wastewater with increasing substrate concentration. All of the tested strains except KUD101 showed 70-90% of the COD removal efficiencies. Acclimated strains, Pseudomonas putida KUD106, KUD107 and KUD108 were inoculated into the ordinary activated sludge and these sludges were used in the dyeing or alkylbenzene wastewater treatment system. In the case of the wastewater containing alkylbenzene compounds, COD removal efficiency was 12-14% higher than that of the ordinary activated sludge, while it was not effective for dyeing wastewater. On the other hand, the floc formation of the activated sludge inoculated with genetically engineered strains was more rapid in both wastewaters tested than that of the activated sludge by the ordinary natural strains.

The dynamics of microbial community structure of the various domestic wastewater treatment processes were examined using a novel approach of quinone profiles. The compositions of microbial quinone of 5 sites fer plant and lab-scale activated sludge were analyzed. More than 14 kinds of quinones were observed in the activated sludges tested in this study. The microbial community structure of the plant activated sludge processes a little differed from that of the lab-scale submerged MBR systems. The dominant quinones were UQ-8, UQ-10 followed $MK-8(H_4)$, MK-7 and MK-6. The molar ratio of ubiquinones to menaquinones (UQ/MK) changed from 0.81 to 1.9, indicating that aerobic bacteria dominated the microbial community of the activated sludge examined. The microbial diversity of the activated sludges calculated from the all quinone compositions was 9.5-11.9 and the microbial equability of the activated sludges was 0.64-0.79.
This study was carried out to treat textile wastewater using anaerobic sludge and aerobic fixed-bed biofilm reactor immobilized with Bacillus sp. dominated activated sludge(Bacillus sp. fraction : 81.5%). The range of influent con-centration of SCOD and soluble color were 1032-1507 mg/1, and 1239-1854 degree, respectively. Continuous treatment experiments were performed with variation of textile wastewater ratio at a same HRT. When textile wastewater ratio was 100%(HRT : 24 hours), The removal efficiency of SCOD and soluble color were 88% and 78%, respectively. When compare aerobic reactor of this study that was immobilized with Bacillus sp. dominated activated sludge to other study that was immobilized with activated sludge, SCOD and soluble color removal efficiency of this study showed a little higher efficiency than immobilized with activated sludge. The Bacillus sp. fraction of initial condition was 81.5%), but the fraction after operation was decreased to 31.8%).
Although chemical oxygen demand (COD) is an important issue for wastewater treatment, COD reduction with microalgae has been less studied compared to nitrogen or phosphorus removal. COD removal is not efficient in conventional wastewater treatment using microalgae, because the algae release organic compounds, thereby finally increasing the COD level. This study focused on enhancing COD removal and meeting the effluent standard for discharge by optimizing sludge inoculation timing, which was an important factor in forming a desirable algae/bacteria consortium for more efficient COD removal and higher biomass productivity. Activated sludge has been added to reduce COD in many studies, but its inoculation was done at the start of cultivation. However, when the sludge was added after 3 days of cultivation, at which point the COD concentration started to increase again, the algal growth and biomass productivity were higher than those of the initial sludge inoculation and control (without sludge). Algal and bacterial cell numbers measured by qPCR were also higher with sludge inoculation at 3 days later. In a semi-continuous cultivation system, a hydraulic retention time of 5 days with sludge inoculation resulted in the highest biomass productivity and N/P removal. This study achieved a further improved COD removal than the conventional microalgal wastewater treatment, by introducing bacteria in activated sludge at optimized timing.
An experimental study was carried out to evaluate the potential of flotation process for the secondary clarifier of an activated sludge process. Flotation techniques, applied in this study, include electrofloation (EF) which generated fine bubbles smaller than $35{\mu}m$ in average and diffuser flotation (DF) which generated fine bubbles smaller than $55{\mu}m$ in average. The batch experiments were done with activated sludge displaying various characteristics. It was shown that the efficiency of solids/liquid separation was reduced as the diluted sludge volume index ($DSVI_{30}$) of activated sludge increased. The dependency, however, gradually decreased as the gas to solids (G/S) ratio increased. Thickening efficiency of EF was more than 2~10 times and DF process was more than 1.5~5 times as compared with gravity sedimentation (GS). Stable sludge blanket was maintained regardless of sludge settleability when the G/S ratio was 0.019 in the EF. On the other hand, Serious deterioration in the sludge blanket was observed in the DF depends on G/S ratio and sludge settleability. And For EF and DF, the suspended solids concentration of effluent was not nearly influenced on settleability of activated sludge and more clear than GS. A biological nutrient removal (BNR) process, combined with EF as a secondary clarifier was operated for three months. The mean MLSS (mixed liquid suspended solids) concentration in the reactor and mean solids concentration of return sludge were estimated to be 5,340 mg/L and 16,770 mg/L, respectively. The water quality of effluent was considerably stable and low value was accomplished, that was, standard suspended solids concentration $0.07{\pm}0.51mg/L$ and standard turbidity $1.44{\pm}0.56NTU$. The EF could be applicable for enhancement of efficiency of activated sludge system as well as improvement of the water quality of effluent.
This study aims at characterizing the bacteriophages isolated from activated sludge performing enhanced biological phosphorous removal (EBPR) to understand the interactions between the phage-host system and bacterial community. Sixteen bacterial isolates (E1-E16) were isolated as host bacterial strains from EBPR activated sludge for phage isolation. Forty bacteriophages based on their plaque sizes (2 plaques on E4, 4 on E8, 11 on E10, 5 on E14, 18 on E16) were obtained from filtered supernatant of the EBPR activated sludge. Each bacteriophage did not make any plaque on bacterial strains tested in this study except on its own host bacterial strain, respectively, indicating that the bacteriophages are with narrow host specificity. However, fourteen of the forty bacteriophages obtained in this study lost their virulent ability even on their own host bacteria. All of the lytic phages showed similar one-step growth patterns and had long latent period (about 9 hours) to reproduce their phage particles in their host bacterial cells. On the other hand, their probable burst sizes (6 to 48 per host cell) were large enough to actively lyse their host bacterial cells. Therefore, it could be implied that bacteriophages are also important members of the microbial community in EBPR activated sludge, and lytic phages directly decrease the population size of their host bacterial groups in EBPR activated sludge by lysis.

In activated sludge process, sludge settling condition is affected by organic loading rate or operation condition, and if settling condition is getting worse, it is common that overall process fails due to wash-out of biomass causing low concentration in the aeration tank. Also activated sludge process has such several problems as requiring large area, consuming a lot of power and producing large volume of sludge. Increased public concern over health and the environment combined with a strong desire to reduce capital, operating and maintenance costs, have created a need for innovative technologies for building new high quality effluents which vail meet 21st century crkeria. MBR(Membrane Bioreactor) process consists of a biological reactor and ultrafiltration(UF) membrane system that replaces the conventional clarifier of an activated sludge process. The main operating advantages of this system are that the quality of the effluent is independent of the settleability of the mixed liquor and that the effluent is free of suspended solids in any operating condition. It is possible to eliminate clarifier and to reduce the volume of aeration tank because it can afford to accumulate high biomass concentration in the bioreactor(20, 000~30, 000mg/L), which would not be possible in a conventional activated sludge process. Therefore, this process reduces overall treatment plant area. In addition to those advantages, Longer SRT condition enables higher sludge digestion in MBR process so the sludge volume produced is 50 to 70% lower than that of conventional activated sludge process There are two kinds of MBR process according to the allocations of membrane. One is cross flow type MBR of which module is located outside of the bioreactor and mixed liquor is driven into the membrane module. The other is submerged type MBR process of which module is submerged in the bioreactor and mixed liquor is generally sucked from the lumen side. addition to that the cake layer is often removed by the uplifting flow of bubbling air. A submerged MBR process is superior to a crossflow MBR in regard to the power consumption because suction pressure of a submerged MBR is generally lower than that of a crossflow MBR which has recirculation pump. A submerged MBR, therefore, has the potential to be applied to small wastewater treatment plants that need low cost treatment systems.
This study was conducted to evaluate the effects of pressure and dissolved oxygen concentration on the activated slut비e and to determine the optimum depth of deep shaft process. Some results from this study were summarized as follows. 1. It is considered that low sludge product in the activated sludge system maintaining high dissolved oxygen concentration is attributed to the increase of endogeneous respiration rate caused by the increase of aerobic zone in the sludge floe. 2. The increase of dissolved oxygen concentration does not affect to the increase of organic removal efficiency greatly and therefore the limiting factor is the substrate transfer into the inner part of floe. 3. The yield coefficient, Y is decreased in proportion to the increase of oxygen concentration. In this study, Y values arre ranged from 0.70 to 0.41 according to the variation of dissolved oxygen concentration from 18.0mg/$\ell$ to 258 mg/$\ell$. 4. The optimum depth of deep shaft process should be determined within the limits of non-toxicity to the microorganism and it is about loom in this study.
Treatment of domestic sewage discharged from big cafeteria kitchen, especially rice-washing water, was examined by the use of activated sludge process and phytoplankton cultivation. Only with a activated sludge process COD value decreased from around 1000 mg/ι to 100 mg/ι, but nutrients, such as nitrogen and phosphate could not be removed sufficiently phytoplankton cultivation in combination with a activate sludge process could decrease COD value down to 50 mg/ι and nutrients values were also reduced substantially. However, the initial concentration of 20 mg/ι in the rice-washing water could not be removed completely without addition of activated sludge as a nitrogen source.
In the present study, it was observed how the phage-host system that is naturally reproduced in activated sludge is affected by the host inoculation. The system of Microlunatus phosphovorus and its phages was selected as the phage-host system native to an activated sludge system operated for 19 days under sequencing anaerobic-aerobic conditions with glutamate as the main carbon source. The phage-host system related to M. phosphovorus was monitored by plaque assay for the phages and by fluorescent in situ hybridization (FISH) for the bacterial host. In addition, the whole phage structure was also monitored by pulsed-field gel electrophoresis (PFGE). During the first 9 days, the phage-host system was more or less steady at approx. 9% (FISH/ DAPI) for M. phosphovorus and approx. 10,000 PFU/ml for its lytic phages. Microlunatus phosphovorus JCM9379 was inoculated into the activated sludge on day 10. Right after the inoculation, M. phosphovorus was approx. 24% (FISH/DAPI) whereas its lytic phages dropped down to approx. 500 PFU/ ml. After the host inoculation (within 9 days), however, the phage-host system eventually reverted to its original level in each population. On the other hand, the whole phage structure was not significantly changed by M. phosphovorus inoculation but stable throughout the process operation. Only the minor change that four phage groups gradually became abundant after the host inoculation was observed.
