Quality Characteristics of Kochujang Prepared with Commercial Protease (시판 Protease를 이용한 고추장의 품질 특성)

  • To study the characteristics and processing of Kochujang which is rapidly fermented by commercial enzymes, three kinds of Kochujang(KP-FA, KP-FN, and KP-BN) using commercial proteases and one Kochujang(KM) using Meju were prepared and their qualities investigated. There were only small differences in pH and acidity between each Kochujang. The moisture contents were high tendency in the three kinds of Kochujangs using the commercial proteases at 20 days of fermentation. Reducing sugars had a tendency to decrease during the fermentation in the Kochujangs using the proteases. During the first half of fermentation, the Kochujangs made with proteases showed higher amino nitrogen contents than the Kochujang(KM) made using Meju. Acidic protease activity was high in KP-FA at 20 days of fermentation and neutral protease activity was high in KP-FN and KP-BN at the beginning of fermentation. The Kochujangs made using the proteases, through 20 days of fermentation, obtained high preference in the sensory evaluation for color, texture, and overall acceptability. However, the hot taste was stronger in these Kochujangs during the fermentation.

Enzymatic Extraction of Lemon Pectin by Endo-Polygalacturonase from Aspergillus niger

  • Pectin was enzymatically extracted from industrial lemon pomace by using an endo-polygalacturonase from Aspergillus niger as a processing aid and compared to pectin extraction by hot hydrochloric acid. The yield of pectin was 17.6 and 20.2% with enzymatic and acidic treatments, respectively. The molecular weight distribution did not vary greatly between the samples extracted with enzyme or acid. Large differences in charge density were observed, however, when the samples were analyzed by anionic-exchange chromatography. Pectin extracted by the enzymatic treatment indicated higher charge density than that obtained by hydrochloric acid. The higher charge density could due to the presence of endogenous lemon pectinesterase, which was activated at low pH 4.5 in situ conditions during the process of enzymatic extraction, leading to low methoxylated pectin with a higher charge density.

Changes in Chemical Components of Red Ginseng Extract Solution and Physicochemical Properties of Precipitates Formed During Pterilization and Storage (홍삼 Ext 수용액의 살균과 저장 중 성분의 변화와 생성된 침전물의 이화학적 특성)

  • Red Ginseng extracts sol'n was sterilized at 85f for 20 mins and/or stored at 4$0^{\circ}C$ for 6 months and centrifuged for 20 mins at 8,500xg in order to investigate the changes in chemical components of supernatants and the properties of precipitates. Contents of crude saponin and ginsenoside-$Rb_1$, -$Rg_1$, -Re were partially decreased during heating and storage. Starch contents were decreased from 26.81% in red ginseng extracts to 17.50-8.81% in supernatants, whereas free sugar contents were increased from 15.50% to 20.29~21.35% by heating and storage. The contents of protein and minerals in supernatants were decreased, but acidic polysaccharides and polyphenol compounds were not changed. pH values of supernatants and precipitates were decreased. The absorbances of brown color precursor and brown pigment in precipitates, detected at 285 nm and 440 nm were remarkably increased. The Overa11 data suggest that precipitates in red ginseng extracts sol'n formed during steilization and storage are provably the brown pigments resulting from Maillard reaction of amino compounds with reducing sugar which could be released from starch and protein matrix and $Cu^+$, $Ca^{2+}$ and $Fe^{3+}$ ions are implicated with the reaction incorporated.

A Study on the Bleaching and Dyeing Properties of Cotton Fabrics Treated with Ozone($O_{3}$) (Ozone($O_{3}$)을 이용한 선직물의 표백과 양색성에 관한 연구)

  • In order to reduce the amount of waste water generated from textile dyeing process and processing water, cotton fabric was bleached with ozone in ozone/water contact system, and dyed. The results obtained from the measurement of whiteness, reflectance, tensile strength, color difference, and K/S value of treated samples were as follow : 1) In ozone bleaching, factors affect whiteness most are treatment time and ozone dosage. As treatment time is lengthened, whiteness increases. Futhermore, bleaching effect is high in acidic treatment, but low in alkaline treatment. 2) As bleaching goes on, maximum absorption wave length shifts to shoter wavelengths. 3) As treatment time is lengthened or pH of treatment is low, tensile strenth of treated fabric decrease. 4) In all cases, color difference of dyed cotton fabrics in above 2% o.w.f. dyestuff concentration don't go beyond 1∼2 AN unit, compared with standard.

  • In aqueous slurry processing of silicon nitride, the interaction of dispersant and binder used as polymeric processing additives on the silicon nitride particle surface was studied to identify the effect of these processing polymeric additives on the ceramic powder processing. The adsoprtion isotherm study of anionic organic molecule as dispersant and nonionic organic molecules as binder of silicon nitricde was studied to investigate the effect of these processing organic additives on the physicochemical properties of silicon nitride particles. As anionic molecule adsorbed onto silicon nitrice surface, the IEP of silicon nitride shifted toward acidic pH and changed the stability of silicon nitride particle. However, the adsorption of binder as nonionic organic molecule onto silicon nitride surface did not changed the IEP but caused the decrease of electrostatic potentials of silicon nitride. These distinctive adsorption behaviors of organic additives on silicon nitride particles can be closely correlated to the stability of silicon nitride particles suspended in aqueous media.

The response of plants growing in a landfill in the Philippines towards cadmium and chromium and its implications for future remediation of metal-contaminated soils

  • During several visits to the Cebu City landfill in the Philippines, plants were observed growing within the area, including on top of the garbage piles. Studying the response of these plants is important in assessing which can be used in remediating metal contaminated soils. This study aimed to determine whether the plants in the Cebu City landfill excluded or accumulated cadmium (Cd) and chromium (Cr) in the plant tissues. The floristic composition of the landfill was analyzed prior to the sample collection. The samples were acid-digested before the desired elements were measured using atomic absorption spectrophotometry (AAS). The Cd and Cr concentrations in the plant root-zone soil were also measured using AAS. The results indicated that the landfill substrate was generally acidic based on the results of the pH measurement. Of the 32 plant species sampled, Cyperus odoratus showed potential for Cd uptake and internal transfer; Cenchrus echinatus, Vernonia cinerea and Terminalia catappa for Cr uptake, and Cynodon dactylon for Cr internal transfer. The plants in the landfill differed in their response towards the heavy metals. To confirm the behavior of C. odoratus towards Cd, and C. echinatus, C. dactylon, V. cinerea, and T. catappa towards Cr, controlled experiments are recommended, as the plant samples analyzed were collected from the field.

  • Large amounts of aluminum hydroxide ($Al(OH)_3$) exist in water purification sludge (WPS) because of the added aluminum coagulant in water treatment process. Notably, $Al(OH)_3$ is an amphoteric compound, can be dissolved in its basic condition using sodium hydroxide to form aluminate ions ($Al(OH)_4{^-}$). However, in a process in which pH is increasing, the humid acid can be dissolved easily from WPS and will inhibit the recovery and reuse of the dissolved aluminate ions. This study attempts to fix this problem by a novel approach to separate $Al(OH)_4{^-}$ ions using nanofiltration (NF) technology. Sludge impurity in a alkaline solution is retained by the NF membrane, such that the process recovers $Al(OH)_4{^-}$ ions, and significantly decreases the organic matter or heavy metal impurities in the permeate solution. The $Al(OH)_4{^-}$ ion is an alkaline substance. Experimental results confirm that a recovered coagulant of $Al(OH)_4{^-}$ ion can effectively remove kaolin particles from slightly acidic synthetic raw water.

Biochemical and Trace Mineral Analysis of Silajit Samples From Pakistan

  • Silajit is a blackish brown pitch-like gummy substance. It is an important Unani and Ayurvedic medicine widely used in the treatment of many diseases and as nutritional supplement It is found in certain mountainous regions of Pakistan, Afghanistan, India, Nepal, Australia and Russia. Silajit samples of Pakistani origin were obtained from different sources and analyzed for their physical characteristics and biochemical & elemental composition. X-ray diffraction studies on Silajit showed that it is an amorphous substance and has no crystalline structure. Also, pH studies of Silajit (2 g/L of water) indicated that it was slightly acidic in nature and ranged from 3.45 to 7.23. Conductivity ranged from 157.7 to 330. Amino acid analysis revealed that Silajit contained lysine and alanine in higher amounts than all other amino acids and ranged from 1456 to 2240 and 68 to $1615{\eta}mole/g$, respectively. Mean concentration of arsenic, mercury, cadmium, lead, copper and zinc was 73.15, 104.92, 0.496, 3.89, 4.04 and 17.23 ppm, respectively. Silajit samples were also analyzed for calcium, potassium and sodium.

Fungal Endophytes of Alnus incana ssp. rugosa and Alnus alnobetula ssp. crispa and Their Potential to Tolerate Heavy Metals and to Promote Plant Growth

  • Soil contamination by metals is of particular interest, given that their retention times within the profile can be indefinite. Thus, phytostabilization can be viewed as a means of limiting metal toxicity in soils. Due to their ability to grow on contaminated soils, alders have repeatedly been used as key species in phytostabilization efforts. Alder ability to grow on contaminated sites stems, in part, from its association with microbial endophytes. This work emphasizes the fungal endophytes populations associated with Alnus incana ssp. rugosa and Alnus alnobetula ssp. crispa (previously A. viridis ssp. crispa) under a phytostabilization angle. Fungal endophytes were isolated from alder trees that were growing on or near disturbed environments; their tolerances to Cu, Ni, Zn, and As, and acidic pH (4.3, 3, and 2) were subsequently assessed. Cryptosporiopsis spp. and Rhizoscyphus spp. were identified as fungal endophytes of Alnus for the first time. When used as inoculants for alder, some isolates promoted plant growth, while others apparently presented antagonistic relationships with the host plant. This study reports the first step in finding the right fungal endophytic partners for two species of alder used in phytostabilization of metal-contaminated mining sites.

Natural Dyeing of Sangju Silk with Mulberry Extract Solution (오디 추출액을 이용한 상주실크의 천연염색)

  • Natural dyeing of Sangju silk for graveclothes with mulberry extract solution was studied. The anthocyanidin dye in the extracted mulberry solution was markedly influenced by pH condition, which resulted in the changed maximum light absorption from a $\lambda$max of 525nm in acidic condition to 380nm in alkaline condition possibly due to the irreversible transformation of anthocyanidin to cyanidin form of the dye. The color fastness properties to both light and washing were good when the fabrics were dyed at $80^{\circ}C$ with the mulberry-extracted solution upto twice extraction.
