  • In order to study the degradation pattern of propanil in solution, the environmental factors such as temperature, pH and UV irradiation effect on propanil degradation were investigated. The degradation of propanil in solution was more rapid in high temperature than in low. The production amount of DCA was increased in high temperature, and then was decreased in a certain period of time because of conversion to TCAB, but the concentration of TCAB was maintained without more degradation. Propanil was rapidly hydrolyzed in alkaline solution as well as in strong acidic solution. Degradation product, DCA was rapidly produced and condensed to TCAB in strong acidic and alkaline condition. On exposur to ultraviolet light, 90% of original propanil was degraded within 20minutes, 0.3 ppm of DCA was produced in 10 minutes, and maintained the concentration throughout irradiation times.

A Study on the Treatment of Swine Wastewater Using Titanium Dioxide Prepared by Hydrothermal Method (수열합성법으로 제조된 이산화티탄에 의한 축산폐수 처리에 관한 연구)

  • This study was performed to evaluate the application of $TiO_2$ on the photocatalytic treatment of swine wastewater. $TiO_2$ sol was prepared by hydrothermal method with the agent ratio($(C_2H_5)_2NH_2\;mol/Ti(OC_3H_7)_4\;mol)=1$ and R ratio ($H_2O\;mol/Ti(OC_3H_7)_4\;mol)=42$. The effect of parameter on the removal efficiency of swine wastewater in a batch type immobilized photocatalyst system such as initial pH, intensity of UV, dosage of $TiO_2$, air flow rate, and concentration of $H_2O_2$ was examined. Wastewater was effectively eliminated in the presence of both UV light illumination and $TiO_2$. Photocatalytic activity was higher in acidic condition compared to neutral and alkaline conditions. In addition, photocatalytic activity increased with increasing UV light intensity, dosage of $TiO_2$, the flow rate of air and the amount of $H_2O_2$ added as an oxidant, but the excess amount of $H_2O_2$ dosage decreased the removal efficiency.

Effects of Fly Ash, Gypsum, and Shell on the Chemical Properties of Soil and Growth of Chinese Cabbage in Acidic Soils (산성토양에서 석탄회,석고,패각시용이 토양화학성과 배추의 생육에 미치는 영향)

  • In order to evaluate the utility of the bituminous coal fly ash, gypsum, oyster shell as soil amendments, acid sandy loam soil with low boron content were amended in the upper 15cm with amendments, and then chinese cabbage was cultivated in fall. Amendments treated were, in metric tons per hectare, i ) none(Check) ; ii) 80 fly ash(FA) ; iii) 4shell(SH) ; iv) 56 fly ash+24gypsum (FG) ; v) 40 fiy ash + 24 gypsum +0.8 shell(FGS). On the whole, amendments imoroved soil chemical properties and contents of N, P, K, Ca, and B in leaves. Among treatmens, FA prominently neutralized soil pH and increased available $P_2O_5$ ,B but decreased Fe contents in soils. FGS also affected the increment of exchangeable Ca, Mg, and available B. Yield response in fresh weight of chinese cabbage was in order of 85% for FGS>77% for FG>66% FA>5% for SH plants. Reducing sugar and vitamin-C contents of leaves depending on treatments showed the same tendencies as that in yields, whereas crude fiber opposite to theme. In particular, FA, FG, and FGS plants showed normal growth without boron deficiency symotoms which appeared in Check and SH plants.Taken together, FGS was an effective combination enable to maximize the utility of fly ash, gypsum, and shell as soil amemdments, especially in cabbage yield and quality.

Use of Chemical Treatments to Reduce Tannins and Trypsin Inhibitor Contents in Salseed (Shorea robusta) Meal

  • This study investigated the effect of chemical treatments on tannins (condensed and hydrolysable) and on the trypsin inhibitor (TI) activity in salseed meal. Triplicate samples of ground salseed meal (1 kg) were mixed with 820 ml of either distilled water (pH 5.3), 0.67 M acetic acid (pH 2.4), 0.67 M sodium bicarbonate (pH 8.2) or 2% polyvinyl-pyrrolidone (PVP) solution. The material was placed in airtight plastic containers and incubated at $37^{\circ}C$ for 0, 3, 6, 12, 24, 48 and 72 h. Samples of untreated salseed meal which had not been subjected to soaking or incubation were run through the analysis to serve as control. Addition of water, acetic acid, sodium bicarbonate and PVP solutions to salseed meal and subsequent anaerobic incubation at $37^{\circ}C$ significantly reduced chemically detectable tannins. At each incubation time, alkali solution was more effective than its counterparts. The effect of acidic solution on hydrolysable tannin was least among the treatments. All the treatments reduced TI activity of salseed meal. The reduction in TI activity by these treatments was similar and ranged between 80-84%. Treatment time effected a decrease in the contents of antinutritional substances. However, the effect of the treatment with the reagents, even for zero incubation time, was quite pronounced. It may be concluded from the present results that the treatment of salseed meal with sodium bicarbonate (0.67 M) is more effective in reducing hydrolysable and condensed tannin contents than PVP, water and acid solutions. Treatment with sodium bicarbonate solution is more economical and easier to handle than acid and PVP treatments. Incubation of the treated material for 12 h is reasonably effective, economical and safe from any mould growth.

The Study on Mucin Release by Airway Goblet Cells in Primary Culture

  • Surface epithelial cells isolated from hamster tracheas and grown on a thick collagen gel become a highly enriched population of mucus-secreting cells. Epithelial cells from tracheas of hamsters were collected using enzymatic procedures and cultured under various conditions. The medium used consisted of a 1:1 mixture of medium 199 and Dulbecco's modified Eagle's (DME) medium which was conditioned before use. Insulin, transferrin, hydrocortisone, epidermal growth factor, and extract from bovine hypothalamus were used as supplement. Due to relatively low basal rates of min secretion from in vitro cultures, cultures are generally radiolabeled using $^3$H-glucosamine as a metabolic precursor. The radiolabeled mucinsreleased are quantitated by precipitation with TCA/PTA. Using this cell culture system, we investigated mucin release of goblet cells by altering the media bathing the apical surface of hamster tracheal surface epithelial(HTSE) cells. Acidic media added sulfuric acid caused sigcificant increases in mucin relesse (155${\pm}$20% at pH 4 and 146${\pm}$16% at, pH 5). Ammonium hydroxide also increased mucin release at pH 9.0(156${\pm}$17%) and pH 10(295${\pm}$9%) respectively. This additional mucin release seems to be associated with cell membrane damage as indicated by release of cellular LDH. SP stimulates secretion of mucin in cultured HTSE cells(154${\pm}$16% at 1${\times}$10$\^$-6/M and 165${\pm}$25% at 1${\times}$10$\^$-5/M. PAF at 5${\times}$10$\^$-6/M and 5${\times}$10$\^$-5/M enhanced by HTSE cells in vitro 168${\pm}$34% and 259${\pm}$30% of mucin secretion, respectively. The increase in mucin release by PAF and SP was not secondary to cell damage or necrosis. SP and PAF may be in mediating mucous secretion induced by inflammation irritantion and infection.

Flocculation Kinetics Using Fe(III) Coagulant in Advanced Water Treatment: The Effect of Sulfate Ion (상수처리시 Fe(III) 응집제를 이용한 응집동력학에 관한 연구 : 황산이온의 영향)

  • The study of flocculation kinetics is of fundamental interest in the field of water treatment, because rational study of the factors affecting the coagulation process should be based on the rate of particle growth. The effect of sulfate on flocculation kinetics were examined using ferric nitrate as a coagulant to coagulate kaolin clay in water under several experimental conditions. Both the particle size distribution data obtained from the AIA and the on-line measurement of turbidity fluctuation by the PDA were used to measure flocculation kinetics. Results show that sulfate ion added to the kaolin suspension played an important role in the flocculation process, not only improving flocculation kinetics at more acidic pH levels but also changing surface charge of particles. The kinetics of flocculation were improved mainly by the enhanced rate and extent of Fe(III) precipitation attributed to the addition of sulfate, and thereby, better interparticle collision frequency, but little by the charge reductions resulting from the sulfate addition. The increase in sulfate concentration beyond $3\times10^{-4}M (up to 2\times10^{-3}M)$ did not induce further improvement in flocculation kinetics, although the higher concentrations of sulfate ion substantially increased the negative ZP value of particles. Alteration Zoning, Mineral Assemblage and Geochemistry of the Hydrothermal Clay Deposits Related to Cretaceous Felsic Magmatism in the Haenam Area, Southwest Korea (한국 서남부, 해남지역에서 백악기 산성마그마티즘에 관련된 열수점토광상의 누대분배, 광물조합의 지구화학적 연구)

  • In the present study, three clay deposits, named the Seongsan, Ogmaesan and Haenam deposits, were investigated. The altered zones are recognized in the hydrothermally altered rocks of the clay deposits from the center of the alteration to the margin: Kaolin, Kaolin-Quartz, Quartz, Sericite and Chlorite zones in the Seongsan deposits; Quartz zone, Alunite zone, Kaolin zone, Sericite zone and Chlorite zone in the Ogmaesan deposits; Quartz zone, Pyrophyllite zone, Sericite zone and Chlorite zone in the Haenam deposits. These zones can be grouped into two types of alteration: Acidic alteration such as Pyrophyllite zone, Alunite zone, Quartz zone, Kaolin zone, Kaolin-Quartz zone and a part of Sericite zone; Propylitic alteration such as Chlorite zone and a part of Sericite zone. All clay deposits belong to high-sulfidation (acid-sulfate) system. The rocks of the acidic alterations are composed of pyrophyllite, alunite, kaolin minerals, sericite, quartz and pyrite. On the basis of bulk chemical compositions, it was found that some components such as $SiO_2$, $TiO_2$, $Fe_2O_3$, FeO, MgO, CaO, $K_2O$ and $Na_2O$ were mobilized considerably from the original rocks. The mobility of these major elements is related to, and controls, mineral assemblages in each altered zone. Polytypes of sericite are determined as $2M_1$ and 1M by X-ray diffraction method. The amount of $2M_1$ is nearly equal to that of 1M in the Seongsan deposits whereas $2M_1$ is less and higher than that of 1M in the Ogmaesan and the Haenam deposits. These facts indicate that formation temperature of sericite is relatively high in the Haenam deposits, moderate in the Seongsan deposits, and low in the Ogmaesan deposits. The ratios of Na/(K+Na) for alunite in the Ogmaesan deposits determined by electron microprobe analyzer (EPMA) are higher than those in the Seongsan deposits. Thus, the alunite of the Ogmaesan deposits must have been formed from the solutions with relatively high aqueous Na/(K+Na) ratios and low pH at a high temperature rather than that of the Seongsan deposits. From all data, it is clarified that alunite is hypogene in origin, and has been formed by oxidation of hydrogen sulfide in the steam-heated environment, and that alunite has been produced by the spectacular solfataric alteration observed at the surface of some present-day hydrothermal systems.

Geochemistry of tourmalines in the Ilgwang Cu-W breccia-pipe deposit, Southeastern Gyeongsang Basin (경상남도 일광의 각력파이프형 구리(Cu)광상에서 산출되는 전기석의 지구화학)

  • A small granodiorite-quartz monzonitic stock containing sericitic and propylitic alteration assemblages hosts a Cu-W breccia-pipe deposit in the southeastern Cyeongsang basin. The mineralized breccia-pipe contains angular to subangular brecciated fragments of granitic rocks showing clast-supported textures. An assemblage of quartz, tourmalines, sulfide minerals (mainly chalcopyrite, arsenopyrite and pyrrhotite) and calcite was precipitated as a hydrothermal cement between the brecciated fragments. A tourmaline aureole surrounds the breccia pipe. Extensive tourmalinization of the granitic rocks near and within the pipe and no tourmalinization in the sedimentary and volcanic rocks. The tourmalines are marked by Fe-rich, black charcoal-like schorl (80 mol% schorl relative) nearer the schorl-dravite solid solution. The chemical changes in the hydrothermal fluid are reflected by variations in compositional Boning from cores to rims. They generally contain cores with low values of Fe/(Fe+Mg) and high values of Na/(Na+ca) relative to rims. This is because of an increase Fe and Ca contents toward rims. The main trend of these variations is a combination of the exchange vectors Ca(Fe, Mg) $(NaAl)_{- }$ $_1$ and $Fe^{3}^{+}$ $Al_{[-10]}$ $_1$ It is thought that boiling causes the loss of $H_2$ into the vapor phase resulting in the oxidation of Fe in the aqueous phase. pH of the melt would be one of important controlling factors for the tourmaline stability. The tourmalines could be precipitated when the system evolved to the acidic hydrothermal regime as most hydrothermal brines and acidic gases exsolved from the magma. The Ilgwang tourmaline crystallization is products of hypogene orthomagmatic hydrothermal processes that were strongly pipe-controlled.

Structural Analysis of the Unusual Sugar-Containing Oligosaccharides Formed by the Selective Cleavage of Weakly Acidic Polysaccharide (약산성 다당의 선택적 분해 과정에서 얻어진 특이당 함유 Oligo당의 구조적 분석)

  • By sequential degradation using partial acid hydrolysis of a weakly acidic polysaccharide (GL-4IIb2'), two acidic oligosaccharide fragments, PA-2' and PA-1-III were isolated and their structures were characterized. PA-2' consisted of almost equal proportion of a rhamnose (Rha) and an unusual sugar, 3-deoxy-D-manno-2-octurosonic acid (Kdo). When permethylated oligosaccharide-alditol derived from PA-2' was analyzed by GC-MS, the peak gave the fragment ions at m/z 189 $(bA_1,\;6-deoxyhexose)$ and at m/z 308 $(aJ_2,\;alditol\;from\;Kdo)$. The peak also gave the characteristic ion at m/z 162 but it did not give the fragment ion at m/z 177, suggesting that Kdo is substituted at C5 but not at C4. Methylation analysis also indicated that PA-2' was composed mainly of terminal Rhap and 5-substituted Kdo. When the reduced product from PA-2' was analyzed by $^1H-NMR$, it gave a signal at 5.09 ppm due to an anomeric proton of ${\alpha}-L-Rha$. These results indicated that PA-2' mainly contained ${\alpha}-L-Rhap-(1{\rightarrow}5)-Kdo$. On the other hand, PA-1-III mainly comprised Rha and Kdo in addition to small proportions of arabinose (Ara) and 3-deoxy-D-lyxo-2-heptulosaric acid (Dha). MS analysis of permethylated oligosaccharide-alditols from PA-1-III suggested that the major peak 1P was $Rhap-(1{\rightarrow}5)-Kdo$ whereas the minor peaks 2P and 3P possessed $Araf-(1{\rightarrow}5)-Dha$ unit and these peaks were produced as epimers during reduction of carbonyl groups in Dha.

In vitro stability evaluation of coated lipase

  • Objective: The study was conducted to evaluate the stability of commercial coated lipase (CT-LIP) in vitro. Methods: The capsules were tested under different conditions with a range of temperature, pH, dry heat treatment and steaming treatment, simulated gastric fluid (SGF) and simulated intestinal fluid (SIF) in this work, respectively. Free lipase (uncoated lipase, UC-LIP) was the control group. Lipase relative activities measured in various treatments were used as a reference frame to characterize the stability. Results: The lipase activities were decreased with increasing temperatures (p<0.05), and there was a markedly decline (p<0.01) in lipase comparative activities of UC-LIP at $80^{\circ}C$ compared with CT-LIP group. Higher relative activities of lipase were observed in CT-LIP group compared with the free one under acidic ambient (pH 3 to 7) and an alkaline medium (pH 8 to 12). Residual lipase activities of CT-LIP group were increased (p<0.05) by 5.67% and 35.60% in dry heat and hydrothermal treatments, respectively. The lipase relative activity profile of CT-LIP was raised at first and dropped subsequently (p<0.05) compared with constantly reduced tendency of UC-LIP exposed to both SGF and SIF. Conclusion: The results suggest that the CT-LIP possesses relatively higher stability in comparison with the UC-LIP in vitro. The CT-LIP could retain the potential property to provide sustained release of lipase and thus improved its bioavailability in the gastrointestinal tract.
