Effect of fermented sarco oyster extract on age induced sarcopenia muscle repair by modulating regulatory T cells

  • Sarcopenia is an age-related, progressive skeletal muscle disorder involving the loss of muscle mass and strength. Previous studies have shown that γ-aminobutyric acid (GABA) from fermented oysters aids in regulatory T cells (Tregs) cell expansion and function by enhancing autophagy, and concomitantly mediate muscle regeneration by modulating muscle inflammation and satellite cell function. The fermentation process of oysters not only increases the GABA content but also enhances the content of branched amino acids and free amino acids that aid the level of protein absorption and muscle strength, mass, and repair. In this study, the effect of GABA-enriched fermented sarco oyster extract (FSO) on reduced muscle mass and functions via Treg modulation and enhanced autophagy in aged mice was investigated. Results showed that FSO enhanced the expression of autophagy markers (autophagy-related gene 5 [ATG5] and GABA receptor-associated protein [GABARAP]), forkhead box protein 3 (FoxP3) expression, and levels of anti-inflammatory cytokines (interleukin [IL]-10 and transforming growth factor [TGF]-β) secreted by Tregs while reducing pro-inflammatory cytokine levels (IL-17A and interferon [IFN]-γ). Furthermore, FSO increased the expression of IL-33 and its receptor IL-1 receptor-like 1 (ST2); well-known signaling pathways that increase amphiregulin (Areg) secretion and expression of myogenesis markers (myogenic factor 5, myoblast determination protein 1, and myogenin). Muscle mass and function were also enhanced via FSO. Overall, the current study suggests that FSO increased autophagy, which enhanced Treg accumulation and function, decreased muscle inflammation, and increased satellite cell function for muscle regeneration and therefore could decrease the loss of muscle mass and function with aging.

Characteristics of Beeswax-treated Hanji Aged by Closed and Ventilated Chamber (밀폐 및 배기조건에서 열화된 밀랍지의 열화특성 분석)

  • Currently a variety of efforts to conserve and restore the injured the Annal of the Joseon Dynasty are continued and many studies about the aging pathway of the beeswax-treated volume have being progressed. In this study, two chambers were set up to assess the influence of VOCs(Volatile Organic Compounds) generated in process of beeswax deterioration. One chamber was closed and the other was continuously ventilated in order to remove the VOCs. In results, the acid compounds of beeswax-treated Hanjis aged in closed chamber was more than that aged in ventilated chamber. The folding endurance of beeswax-treated Hanjis aged in closed chamber was lower than that aged in ventilated chamber. Accumulated acid compounds of beeswax-treated Hanji in closed chamber may cause accelerated aging and strength decrease. In conclusion, accumulated acid compounds can accelerate the aging of beeswax-treated Hanji and cause the severe deterioration of inside of beeswax-treated volume. Therefore, to preserve more safely beeswax-treated volumes of the Annals of the Joseon Dynasty, it is essential that VOCs emitted from beeswax-treated volume are ventilated.

Crystal Growth, Spectral, Magnetic, Antibacterial and Antifungal Studies of Co(II) and Ni(II) Complexes of 4-Nitrobenzoic Acid

  • Single crystals of cobalt(II) and nickel(II) complexes of 4-nitrobenzoic acid have been successfully grown by gel diffusion technique. The grown crystals were characterized by elemental analysis, FT-IR and UV-Visible spectroscopy. The structure of the grown crystals was elucidated using single crystal X-ray diffraction studies. The title compounds $[Co(Pnba)_2(H_2O)_4]{\cdot}2H_2O$ 1 and $[Ni(Pnba)_2(H_2O)_4]{\cdot}2H_2O$ 2 where PnbaH=4-nitrobenzoic acid, crystallize in centrosymmetric triclinic space group P-1. Magnetic susceptibility measurements reveal that the compounds are paramagnetic in nature. The mechanical strength of the grown crystals was determined by Vicker's microhardness studies. The ligand (4-nitrobenzoic acid) and the complexes have been screened for their biological activity against various bacteria and fungi. The activity data show that the biological activity of the complexes is higher than that of the ligand.

  • A series of $BeO-SiO_2$ catalysts were prepared by coprecipitation from the mixed solution of berylium chloride and sodium silicate. The addition of BeO to $SiO_2$ caused the increased of acidity, acid strength, specific surface area, and acid catalytic activity, giving a maximum at 20 mol% of BeO. On the basis of x-ray diffraction pattern, the catalysts calcined at $400{\sim}500^{\circ}C$ were found to be amorphous, having both $Br{\ddot{o}}nsted$ and Lewis acid sites. Catalytic activities for cumene dealkylation were closely correlated with the acidity of catalysts.

  • The alkaline dissolution behavior of sea-island type ultrafine nylon fiber were dependent on the concentration of NaOH and treatment time, and the most appropriate condition for alkaline dissolution was to treat with 20g/l NaOH for 30 min at $80^{\circ}C$. The dyeing properties of sea-island type ultrafine nylon fiber and regular nylon fiber were examined with 3 different types of acid dyes in this study. The dye uptakes of ultrafine nylon fiber were higher than regular nylon fiber because of large surface area per unit mass, which increased as the dye bath pH decreased. The dyeing rates on ultrafine nylon fiber were faster and dye exhaustions were higher than regular nylon fiber, however color strength and rating of wash fastness were lower. It was also found that levelling type acid dye showed fast dyeing rate on both nylon fibers than metal-complex and milling type acid dyes.

Effects of Feeding Betaine on Performance and Hormonal Secretion in Laying Hens

  • The effects of dietary betaine on performance, blood compositions, hepatic amino acid concentrations and hormonal secretions were examined in laying hens. Egg production was significantly higher in birds fed the 16.5 % protein diet compared to those fed 14.5 % protein diet(p<0.05), whereas dietary supplementation of betaine did not show any significant effect. The high level of protein and betaine supplementation significantly improved egg weight, egg mass and feed conversion(p<0.05), while eggshell breaking strength, eggshell thickness and Haugh unit were not influenced by betaine and dietary protein levels. Supplemental betaine did not affect serum total protein, albumin and BUN concentration. However, uric acid concentration significantly increased in 600 ppm betaine-fed groups(p<0.05). Concentrations of most hepatic amino acid were influenced by increased protein feeding and dietary betaine supplementation. Hormone studies recorded significantly higher serum and hepatocyte IGF-I concentration in 600 and 1,200 ppm betaine treatments(p<0.05) compared to those of control group. IGF-I mRNA gene expression of hepatocytes revealed statistically correlated increase in 600 and 1,200 ppm betaine-fed groups compared to the controls(p<0.05). Serum IGFBP-3 concentration was significantly elevated in 600 ppm betaine treatments. However, the secretion of IGFBP-1 in hepatocyte of laying hens fed with 600 and 1,200 ppm of betaine showed a significant decrease compared to the control group(p<0.05). Results of these study show that dietary betaine supplementation affects protein and hormone metabolism in laying hens.

Prediction of Organic Acid Chromatogram in High Performance Ion Chromatography (고성능 이온 크로마토그래피에서 유기산의 크로마토그램 예측 연구)

  • In order to predict the chromatogram for organic acid in ion chromatography, Langmuir isotherm parameters were obtained by Retention Time Method (RTM) and moment method. Ion chromatography analysis for formic acid was performed and compared with theoretically predicted profiles under isocratic condition. Band profiles were estimated with the equilibrium-dispersive model of chromatography using a PDEsolver Macsyma . The relationship between the characteristics of chromatogram and the variable operating condition in chromatography such as the flow rate, ionic strength and injection volume was studied. Satisfactory agreement was observed between the experimental and the estimated chromatograms with parameters obtained form the moment method.

Starch-Fatty Complex Modified Filler for Papermaking

  • In order to improve filler-fiber bonding in paper, starch-filler composites were prepared by a starch-fatty acid complex formation method. These composites were used as a papermaking filler to improve the physical properties of the paper, filler retention and the sizing effect. The solubility of the starch-fatty acid complex in water at different temperatures was measured. The results indicated that the starch-fatty acid complexes have very low solubility in water below $70^{\circ}C$, which can be easily coated on clay surface to modify clay-fiber bonding ability. The clay-starch composite filled handsheets showed that paper strength could increase more than $100{\sim}200%$ compared to untreated clay. It was found that ZDT of the handsheet decreased as the clay content increased when unmodified clay was used, but it increased when the starch-fatty acid composite modified filler was used. It was also found that the presence of fatty acide in the complex increased the water-repellant property of the handsheet, which can be used to aid in sizing during papermaking. Filler distribution and bonding characteristics between the composite and fiber were investigated using Scanning Electron Microscopy(SEM).

  • As pyridinoline is one of the predominant cross-lins in a mature collagen, pyridinoline formation may be an essential step during the growth process to obtain normal mechanical strength in collagen fibrils. However, the excess formation of pyridinoline in collagen will probably make the tissue stiffer, less soluble and less digestible by enzymes. We investigated the changes of pyridinoline of bone collagen and the role of ascforbic acid(AsA) on the formation of pyridinoline. The pyridinoline content of bone collagen significantly increased during incubation for 1~5 weeks at 37$^{\circ}C$ in vitro. The addition of AsA decreased pyridinoline to half the amount found in controls with 5 week incubation. When dehydroascorbic acid(DHA) and L-2, 3-diketogulonic acid (DKG), the oxidative products of AsA, were supplemented to bone collagen solution instead of AsA, the content of pyridinoline in bone collagen was about 80% or 70% that of controls, respectively. These results suggest that pyridinoline content decreases by the addition of AsA in vitro. Furthermore, it was shown that AsA in oxidized from also affected the formation of pyridinoline.

  • In the conversion of methanol, the effect of acide property of heteropoly compounds on the catalytic activity was investigated. The pretreatment of Cu-exchanged 12-tungstophosphoric acid with hydrogen enhanced both the selectivity for propane and the conversion of methanol, and the pretreatment of Al-exchanged 12-tungstophosphoric acid with water enhanced the acid strength of the catalyst. The water added into the reactant decreased the conversion of methanol, while the pretreatment temperature did not affect it but the propylene/propane ratio. Various partially-substituted Al salts of 12-tungstophosphoric acid showed different catalytic activities depending on the degree of Al-substitution.
