느타리버섯 자실체의 부위 및 생육시기별 지방산 조성의 변화 (Changes of Fatty Acid Composition by Various Developmental Stage and Fruit Body Section in Pleurotus ostreatus)

  • 느타리버섯(원형1호 느타리) 자실체의 부위별로 지방산 조성을 조사한 결과 linoleic acid는 갓부위에서 가장 높았고 대부위로 갈수록 낮아지는 경향이었고 stearic acid와 palmitic acid는 대부위로 갈수록 증가하는 경향이었다. 생육시기별 지방산조성은 oleic acid의 경우 버섯원기형성 후 $11{\sim}12$일 까지는 점차 증가하다가 이후부터는 감소하는 경향을 보였고 linoleic acid와 불포화지방산대비 포화지방산의 비율은 원기형성 후 $3{\sim}4$일까지는 증가하다가 그 후부터는 감소하는 경향을 나타내었다.

  • Some fatty acids(palmitic acid, stearic acid, oleic acid, linoleic acid and linolenic acid) in foxtail millet seeds were evaluated. In this study, 32 varieties of accession collected from different place of South Korea were used in order to evaluate their fatty acid composition and concentration. These samples were analyzed by GC with HP20 column. Significant variability in individual fatty acids was observed. The palmitic acid ranged from 10.9 to 13.5%, stearic acid from 2.5 to 8.3%, oleic acid from 10.0 to 15.8%, linoleic acid from 62.5 to 67.9% and linolenic acid ranged from 3.0 to 5.2%. This variability can be exploited for selection of plant and for studying the genetic variability in millet. These selections can also be utilized for genetic improvement of the crops.

  • LP가스 용접 용기의 가스 누출사고에 대한 통계적 분석을 통하여 사고 발생에 영향을 미치는 주된 요인을 조사하였다. 그 결과 대부분의 가스 누출은 용접부 국부부식에 따른 핀홀에 의한 누출이 가장 큰 비중을 차지하고 있다. 따라서, 본 논문에서는 2부구조 용기에서의 열처리조건의 문제점과 3부구조 용기의 T부위 터짐 현상에 의한 대책안을 제시하였다. 나아가, LP 가스 용기의 용접부 및 모재부에 대하여 산성비와 해수 분위기에서의 부식속도를 전기화학적 부식실험을 통하여 측정하였다. 또한, LP가스 용기의 제조공정중 분체도장 공정에 의한 도장막의 부착성 및 결함을 주사전자현미경을 통하여 분석하고 최적의 도장 두께를 제시하였다.

밤 지질(脂質) 성분(成分)의 분석(分析) -제1보. 내과육(內果肉) 및 외과육(外果肉)의 지질조성(脂質組成)- (Analysis of the Lipid Components in Chestnut (Castanea crenata) -Part I. Composition of lipid fraction of inner and outer part of chestnut-)

  • 은기(銀寄), 녹조(鹿鳥), 상피(霜被)의 3가지 밤 품종(品種)을 시료(試料)로 하여 과육(果肉)을 내과육(內果肉)과 외과육(外果肉)으로 나눈 후에 유리지질(遊離脂質) 및 결합지질(結合脂質)을 추출(抽出) 하고 그의 지질조성(脂質組成) 및 구성지방산(構成脂肪酸)을 column chromatography, gas chromatography에 의하여 분리(分離), 정량(定量)하였다. 밤의 총지질(總脂質)의 평균함량(고형물당(固刑物當))은 내과육(內果肉)에 1.84%, 외과육(外果肉)에 1.95%였으며 이 중에서 유리지질(遊離脂質)은 내과육(內果肉)에 1.26%, 외과육(外果肉)에 1.63%이나 결합지질(結合脂質)은 내과육(內果肉)에 0.58%, 외과육(外果肉)에 0.32%로서 유리지질이 결합 지질보다 $2{\sim}5$배 많았으며 외과육(外果肉)에 특히 유리지질이 많았다. 유리지질(遊離脂質) 중의 중성지질(中性脂質)은 평균 39.5%로 결합지질(結合脂質)의 4%보다 10배 정도 많았으나 외과육(外果肉)에 있어서는 유리지질(遊離脂質) 중의 중성지질이 56.6%로서 더욱 많았다. 당지질(糖脂質)의 평균함량은 유리지질에 22.0%, 결합지질에 17.0%였으며 인지질(燐脂質)의 평균함량은 유리지질에는 6.9%였으나 결합지질에는 10.5%로써 그 함량(含量)이 많았다. 밤의 유리지질(遊離脂質)을 구성하는 주요 지방산은 linoleic acid, palmitic acid 및 linolenic acid이나 결합지질(結合脂質)에는 palmitic acid가 40.1%로 가장 많았고 linoleic acid, linolenic acid가 그 다음으로 적었다. 또한 결합지질에는 포화지방산(飽和脂肪酸)이 유리지질에서 보다 2배 정도 많았다.

  • 식물성 오일은 다양한 생물제품 생산에 적합한 저가의 원료이다. 식물성 오일의 주성분인 oleic acid와 linoleic acid와 같은 불포화 지방산의 함량은 콩기름의 경우 각각 22%와 55%, 옥수수기름의 경우 26%와 60%, 유채기름의 경우 61%와 21%에 달한다. Hydroxy나 keto group으로 치환된 불포화 지방산들은 다른 지방산에 비해 높은 점도와 반응성 등의 특별한 화학적 특성 때문에 가소제, 계면활성제, 윤활유, 세제 등에 사용되는 유용한 산업 화합물이다. 본 연구에서는 Flavobacterium sp. strain DS5 (NRRL B-14859)를 oleic acid로부터 중간생성물로서 10-hydroxystearic acid (10-HSA)를 거쳐 10-ketostearic acid (10-KSA)로의 전환에 이용하였다. 두 개의 생물전환 생성물인 10-KSA와 10-HSA는 GC, GC-MS, $^1H-NMR$, $^{13}C-NMR$을 이용하여 정량 및 정성분석을 수행하였다. 플라스크 배양에서 10-KSA와 10-HSA의 최대 생산량은 각각 3.4 g/L와 0.5 g/L 이었다. 10-KSA 생산을 위한 최적 glucose 농도 yeast extract 농도, oleic acid 첨가시간, 첨가부피는 각각 20 g/L 이하, 5 g/L 이상, 접종 후 18 h, 0.3 ml/50 ml 이었다.

한국(韓國) 간장중(中)의 유기산(有機酸)에 대(對)하여 (Organic Acid in Korean Soy-Sauces)

  • Korean soy-sauces were orepared by the ordinary and impreved method and its analyses on the organic acid. The results obtained is as following: 1. In analysing general components of prepared soy-sauce, total acid, volatile acid and non-volatile acid were found more in improved soy-sauce than in ordinary soy-sauce. 2. Volatile organic acid were analysed by gas-chromatography method. As a result, the followings was attained: a) In the ordinary soy-sauce, formic acid, acetic acid, propionic acid, and butyric acid were detected. Butyric acid was in the highest amount and then propionic acid, acetic acid and formic acid are followed in the order. b) In the improved soy-sauce, formic acid, acetic acid, propionic acid and butyric acid were detected. Acetic acid was in the highest amount and then propionic acid, butyric acid and formic acid are followed in the nrder. 3. Non-volatile organic acid were analysed by paper partition chromatography method. As a result, the followings were attained: a) Lactic, glutaric, fumaric, malonic, malic, glycolic, oxalic, tartaric, and succinic acid and two unknown spots were detected in ordinary soysauce. Lactic acid was in the highest amount and then succinic, glycolic, oxalic, tartaric, glutaric, malic, fumaric and malonic acid are followed in the order. b) Lactic, glutaric, malonic, malic, glycolic, tartaric, succinic and tgalacturonic acid and two unknown spots were detected in the improved soy-sauce. Lactic acid was in the highest amount and then succinic, glycolic, malic, glutaric, tartaric, galacturonic and malonic acid are followed in the order. 4. ${\alpha}-keto$ acid were analysed by paper partition chromatography. As a result, the followings were attained: Pyruvic acid and ${\alpha}-keto$ glutaric acid and an unknown spot were detected in the ordinary and improved soy-sauce. Pyruvic acid was in the highest amount and then ${\alpha}-keto$ glutaric acid are ollowed in the order. 5. Stale flavor in the ordinary soy-sauces seems to be partly affected by butyric acid and propionic acid. 6. Substances influencing taste, such as lactic acid and succinic acid, were found more in improved soy-sauce than ordinary soy-sauce.

식이 섬유소 섭취상태와 변 답즙산 조성과의 관계 (The Relationship to Dietary Fiber Intake and Fecal Bile Acid Profiles)

  • Bacterial transformation of bile acids is possibly involved in colorectal carcinogenesis. n several epidemiological studies, the fecal bile acid dietary fiber are related to the indicence of colonic cancer. This study investigated the influence of age, dietary fiber intake on fecal bile acid profiles in healthy subject. The dietary fiber were assessed by mean of 24 hour dietary recall method, the subjects consist of 238 members aged 20 to 64 years old and feces are collected from the subjects. Fecal bile acids and neutral sterols were analyzed by gas chromatography. Mean dily crude fiber intake level was 7.7$\pm$1.4g(dietary fiber : 16.7$\pm$3.5g), dietary fiber intake range being 6.5-36.8g. The dietary fiber intake in elederly subject was significantly lower than in the other younger groups. Dietary fiber intakes was negatively correlated with the total bil acid concentation in feces. Probably, a decrease in dietary fiber intake results in higher fecal bile acid concentrations. The secondary bile acid concentration is related to the colon cancer, deoxycholic acid and lithocholic acid were significantly higher in elderly subjects. Concentration of fecal total bile acid, deoxycholic acid, coprostanol, coprostanone were higher in low dietary fiber intake group. These results suggest that the risk factor for colon cancer might be reduced, when dietary fibers are consummed more.

각국 인삼의 지방질성분에 관한 연구 제2보, 결합 지방질중의 지방질 및 지방산 조성 (Studies on the Lipid Components of Various Ginsengs ll. Lipid and Fatty Acid Compositions of the Bound Lipids)

  • Lipid and fatty acid compositions of the bound lipids in Panax quinquefolium (Korea, Japan and China), Panax quinquefolium (America, Canada) and Panax notoginseng (China) were studied by means of silicic acid column chromatography, thin-layer chromatography and gas-liquid chromatography. The could lipid contents in various ginsengs were 0.29 to 0.48%, in which neutral lipid fractions were 63.6 to 67.3%, glycolipid fractions 21.9 to 25.7% and phospholipid fractions 7.7 to 12.4%. The content compositions of neutral lipid fractions were lower and those of glycolipid and phospholipid fractions were higher in the bound lipids than in the free lipids from the various ginseng. The major components were fatty acids, diglycerides and free sterols in neutral lipid fractions, monogalactosyl diglyceride, sterol glucoside and esterified steryl g1ycoside in glycolipid fractions and phosphatidyl glycerol, phosphatidyl ethanolamine and phosphatidic acid in phosphoipid fractions. Seventeen fatty acids were analyzed in the four bound lipid fractions from the various ginsengs and main fatty acids were linoleic acid, palmitic acid and oleic acid. Total saturated fatty acid and palmitic acid contents were higher and total unsaturated fatty acid and linoleic acid contents lower in the total bound lipids than in the total free lipids from the various ginsengs.

Whole Cell Bioconversion of Ricinoleic Acid to 12-Ketooleic Acid by Recombinant Corynebacterium glutamicum-Based Biocatalyst

  • The biocatalytic efficiency of recombinant Corynebacterium glutamicum ATCC 13032 expressing the secondary alcohol dehydrogenase of Micrococcus luteus NCTC2665 was studied. Recombinant C. glutamicum converts ricinoleic acid to a product, identified by gas chromatography/mass spectrometry as 12-ketooleic acid (12-oxo-cis-9-octadecenoic acid). The effects of pH, reaction temperature, and non-ionic detergent on recombinant C. glutamiucm whole cell bioconversion were examined. The determined optimal conditions for production of 12-ketooleic acid are pH 8.0, 35℃, and 0.05 g/l Tween80. Under these conditions, recombinant C. glutamicum produces 3.3 mM 12-ketooleic acid, with a 72% (mol/mol) maximum conversion yield, and 1.1 g/l/h volumetric productivity in 2 h; and 3.9 mM 12-ketooleic acid, with a 74% (mol/mol) maximum conversion yield, and 0.69 g/l/h maximum volumetric productivity in 4 h of fermentation. This study constitutes the first report of significant production of 12-ketooleic acid using a recombinant Corynebacterium glutamicum-based biocatalyst.

인산형 연료전지 발전성능에 미치는 반응기체 공급량 및 공급중단의 영향 (Effects of Reactant Gas Flow Rates and Starvation on Phosphoric Acid Fuel Cell Performance)

  • Effects of reactant gas flow rates and starvation on phosphoric acid fuel cell performance were studied. As the reactant gas flow rates increased, the cell performance increased and then the cell maintained constant performance. The optimum flow rates of hydrogen, oxygen and air under galvanostatic condition of 150 mA/$\textrm{cm}^2$ are found to be 3cc/min${\cdot}\textrm{cm}^2$, 4cc/min${\cdot}\textrm{cm}^2$, and 15cc/min${\cdot}\textrm{cm}^2$, respectively. Hydrogen and oxygen starvation resulted in voltage loss of about 5mV and 0-2mV, respectively. The voltage loss was independent of starvation time. These results were discussed from the point of view of electrochemical reaction of the cell.
