Lipid Metabolism in Rats Fed Acetaminophen with Coadministration of Adzuki Bean Extract

  • The effect of water extract of adzuki beans on acetaminophen-altered lipid metabolism was examined in rats. Control group of rats was fed a basal diet, another group of rats was fed 0.5% acetaminophen (APAP group), and a third group of rats was fed 0.5% acetaminophen plus 5% adzuki bean extract (ABE group) for 4 weeks. Serum total and HDL cholesterol levels in the APAP group were significantly lower than those in the control and ABE groups. Hepatic cholesterol $7{\alpha}-hydroxylase$ and fatty acid synthase mRNA levels in the APAP and ABE groups were significantly higher and lower than in the control group, respectively. Hepatic 3-hydroxy-3-methylglutaryl-coenzyme A reductase mRNA level in the APAP group was significantly lower than in the control group, whereas that in the ABE group was significantly higher than in the APAP group. These results indicate that adzuki bean extract may improve the acetaminophen-altered serum lipid metabolism in rats.

Improving Combination Cancer Therapy by Acetaminophen and Romidepsin in Non-small Cell Lung Cancer Cells

  • Combination chemotherapy is more effective than mono-chemotherapy and is widely used in clinical practice for enhanced cancer treatment. In this study, we investigated the potential synergistic effects of acetaminophen, a common component in many cold medicines, and romidepsin, a histone deacetylase (HDAC) inhibitor, in the A549 non-small cell lung cancer (NSCLC) cell line. The combination of acetaminophen and romidepsin also exerted significant cytotoxicity and apoptosis induced by activation of caspase-3 on tumor cells in vitro. Moreover, combination therapy significantly induced increased production of chemokines that stimulate migration of activated T-cells into tumor cells. This mechanism can lead to active T-cell mediated anti-tumor immunity in addition to the direct cytotoxic chemotherapeutic effect. Activated T-cells led to enhanced cytotoxicity in drug-treated A549 cells through interaction with tumor cells. These results suggested that the interaction between the two drugs is synergistic and significant. In conclusion, our data showed that the use of romidepsin and low concentrations acetaminophen could induce effective anti-tumor effects via enhanced tumor immune and direct cytotoxic chemotherapeutic responses. The combination of acetaminophen with romidepsin should be considered as a promising strategy for the treatment of lung cancer.

Effects of acetaminophen administration on liver histopathology, serum GOT/GPT levels and circulating microRNA-122 concentration in olive flounder (Paralichthys olivaceus)

  • In human medicine, circulating microRNAs have been successfully utilized as early biomarkers for various abnormalities and disease states. Vertebrate miR-122 is a liver-specific microRNA which is expressed almost solely in hepatocytes and plays an important role in the regulation of hepatocyte function. In this study, to evaluate the potential utility of circulating miR-122 as a biomarker for liver injury in olive flounder (Paralichthys olivaceus), fish were orally intubated with two doses of acetaminophen (500 mg/kg or 1.000 mg/kg of body weight), and the expression of miR-122 in serum was quantified using real time-PCR. Histological change in liver, and the enzymatic activity of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) were also analyzed. The results showed that miR-122 was higher in acetaminophen administered groups compared to control group. The histopathological effect of acetaminophen on olive flounder liver was not distinct. The serum level of GPT and GOT was increased within 2 folds compared to control group by acetaminophen administration. However, the serum miR-122 level was increased more than 3 or 4 folds compared to the control group by administration of 1000 mg/kg of acetaminophen. These results suggest the possible use of miR-122 as an indicator of liver injury in olive flounder, even when histopathological effects are not distinctive.

  • We report on a patient who developed acute hepatic failure despite intravenous N-acetyl cysteine therapy who was treated with the Molecular Adsorbents Recirculating System (MARS). She presented 20 hours after the ingestion of 13 g of acetaminophen. The MARS is based on albumin dialysis principle which can be applied for patients with acute poisoning from drugs that have high protein-binding capacity because of its ability to selectively remove from circulation protein-bound toxins. The clinical toxicologist should be consider this technology when treating patients with hepatic failure following acetaminophen poisoning.

  • Objectives : Circii Herba has been used as a natural drug for the treatment of stress digestive system disease. The aim of this study is to investigate the role of Circii Herba aqua-acupuncture solution (CHAS) in experimental oxidative liver injury. Methods : In order to investigate the effects of CHAS on acute liver injury, male ICR mice were pretreated with CHAS(0.2 ml/mouse/day) at the loci of BL18 and CV12 for 6days, starved for 24hrs, and administerated acetaminophen(500 mg/kg, i.p.). After acetaminophen administeration, mice were sacrificed, and the liver was removed, rinsed with ice-cold $1.15{\%}$ KCI buffer, and homogenized at $4^{\circ}C$. Fractions(fraction Ⅰ, Ⅱ, Ⅲ) were isolated by differential centrifugation. Lipidperoxide, total SH, and glutathione(GSH) levels were measured in the Fraction Ⅰ. In addition, activities of hepatic enzyme, such as catalase, glutathione peroxidase(GSH-Px) were measured in the Fraction Ⅱ, and glutathione S-transferase(GST) was measured in the Fraction Ⅲ. Results : In vivo treatment of CHAS(BL18 and CV12) showed effective inhibition of acetaminophen induced lipid peroxidation, and showed elevations of total SH, GSH level, catalase, GSH-Px, GST activities. Conclusions : These results suggested that CHAS might suppress the formation of oxidative metabolites, and prevent acetaminophen induced hepatotoxicity.

Electrochemical Investigation of Acetaminophen with a Carbon Nano-tube Composite Film Electrode

  • Electrochemical behaviors of acetaminophen at a muti-wall carbon nano-tube composite film modified glassy carbon electrode were investigated by cyclic voltammetry, linear sweep voltammetry and chronocoulometry. Compared with that obtained at the unmodified electrode, the peak currents were enhanced significantly, and the oxidation peak shifted towards more negative potential with the reduction peak shifted positively. The peak-to-peak separation turned narrow, and suggested that the reversibility was improved greatly. Experimental parameters, such as scan rate, pH and accumulation conditions were optimized. It was found that a maximum current response can be obtained at pH = 5.0 after accumulation at -0.50 V for 80 s. The oxidation peak current was found to be linearly related to acetaminophen concentration over the range of $5.0{\times}10^{-7}\;\sim\;1.0{\times}10^{-4}$ mol $L^{-1}$ with a detection limit of $5.0{\times}10^{-8} $mol $L^{-1}$. A convenient and sensitive electrochemical method was developed for the determination of acetaminophen in a commercial paracetamol oral solution. Its practical application demonstrated that it has good selectivity and high sensitivity.

  • We made tablets by several formulations of acetaminophen and their bioavailability was compared with acetaminophen syrup. Result of this experiment, we were observed the highest bioavailability in the C tablet as same as syrup preparation, but in tablet B and tablet A we were observed the lower bioavailability compared with the syrup preparation.

  • The phormacokinetics of acetaminophen were investigated in rabbits with folate-induced renal failure. The blood level, the area under the blood concentraction curve(AUC) and the biological half-life were increased significantly, and the urinary excretion was decreased significantly as compared with those of normal rabbits. Serum creatinine concentration and AUC, creatinine clearance and renal clearance have linear relationship respectively. Dosage regimen of acetaminophen was considered to be adjusted in renal failure.

  • The metastable modification of acetaminophen was prepared in industry scale. It was found that the dissolution rate of the metastable modification was greater than that of the original powder. The metastable modification was transformed to the stable modification by grinding, but it was not transformed by compression. Starch and lactose inhibited this transformation of the metastable modification to the stable modification by grinding.

  • When acetaminophen (25mM) was introduced into the perfused rat liver, the hepatic O2 uptake was rapidly inhibited first and then later slow-down. The rapid inhibition was found to be due to mitochondrial blockade, whereas the so-called slow inhibition" was associated with microcirulatory aberrations as evidenced by inhomogneous staining of the liver tissue by trypan blue infusion (0.1%). NaCN (0.5mM) also caused rapid and slow respiratory inhibitions, giving heterogeneous trypan blue staining.ning.
