본 연구에서는 중금속, 특히 은(銀)을 축적하는 세균을 지하수로부터 분리, 동정하고, 분리 균주의 특성 및 중금속 축적조건을 규명하여 최대축적이 일어날 수 있는 조건을 검토하였다. 서울시 지하수 10개 정점으로부터 분리된 은(銀) 축적 세균 중 가장 축적능이 뛰어난 두 균주를 선별하였으며 이들은 Pseudomonas fluorescens와 Bacillus cereus로 동정되었다. 본 분리 균주는 은(銀)의 농도가 5 ppm, 10 ppm, 20 ppm으로 높아질수록 생장 저해를 심하게 받았다. pH가 낮을수록 세포의 성장이 저하되기는 하였지만 세포 당 은(銀) 축적율은 오히려 증가하였으며 24시간 이내에 최대축적이 이루어졌다. 이때 은(銀) 축적량은 Pseudomonas fluorescens의 경우 약 1.9 $mg/g{\cdot}cell$ 이었으며 Bacillus cereus는 약 1.65 $mg/g{\cdot}cell$ 이었다. 은(銀) 처리시, Bacillus cereus에서 126 KDa, 89 KDa, 25 KDa 등 3개의 단백질이 특이적으로 증가하였으며, Pseudomonas fluorescens에서는 101 KDa, 68 KDa, 27 KDa 크기의 단백질이 감소하였으나 34 KDa 크기의 새로운 단백질이 유도됨을 확인하였다.
광산폐수, 산업폐수등으로 부터 Cd, Pb, Zn 및 Cu등의 중금속에 강한 내성을 지니고 있을 뿐만 아니라 균체내 중금속 축적능력이 우수한 중금속 내성 미생물 균주 Pseudomonas putida(Cd), Pseudomonas aeruginosa(Pb), Pseudomonas chlororaphis(Zn) 및 Pseudomonas stutzeri(Cu)를 각각 분리하여, 여러가지 물리화학적인 방법으로 세포를 전처리하여 세포구성성분을 인위적으로 조절한 후 세포내 중금속이온의 흡수 거동 및 조단백질 함량과 중금속 축적관계를 조사한 결과는 다음과 같다. 세포를 알카리로 전처리하였을 경우 세포내 중금속 축적은 매우 감소되었으며, 메탄올과 클로포름으로 전처리하였을 경우에는 중금속 축적에 큰 영향을 미치지 않았으나, 메탄올과 클로로포름으로 전처리한 후 다시 알카리로 재차 처리하였을 경우에는 중금속 축적이 매우 감소되었다. 전처리된 세포내 중금속 축적은 용출되지 않고 남아있는 조단백질 함량이 감소됨에 따라 더 얼마나 크게 감소되었으므로 세포 구성물질중 단백질이 중금속 축적에 중요한 역할을 하는 물질인 것으로 판단되었다.
It is important to identify the contaminant sources and to evaluate the fate and transport of heavy metals to crops in agricultural lands. This study was conducted to evaluate metal sources and its transfer and accumulation to crop in agricultural soils. Pollution indices were calculated and multivariate analysis was performed to identify metal sources. To evaluate transfer and accumulation of metals to crops, the contents of phytoavailable metals were evaluated by using single extraction method and the correlation between metal content and soil properties was analyzed. Also the BCF was quantitatively evaluated for investigating the metal transition to each crop grown in the research area. As a result, Cr, Ni, and Co were expected to be mainly derived from geologic factors due to weathering of certain parent rocks. The content of nickel in soils of the research area was slightly higher than that of the concern level criteria based on total concentration, but the amount transferred and accumulated in the crops was actually low. Understanding the contamination characteristics by investigating the pollution sources of heavy metals and its transfer and accumulation to crops through various evaluation techniques could provide important information for proper management of the agricultural land.
This study was undertaken to investigate the effect on the Cd accumulation in long-term Cd poisoned rats. 40 male weaning Sprague Dawley rats weighting 80-90g were divided into 4 groups (LCuLFeCd : low Cu, Fe and Cd group, ACuLFeCd : adequate Cu, low Fe and Cd group, ACuAFeCd : adequate Cu, adequate Fe and Cd group) according to Cu and Fe levels (Cu 0.5ppm, 8.5ppm : Fe 6ppm, 40ppm) for 12 weeks. There were no significant difference in water intake, feed intake, and body weight gain according to dietary Cu and Fe consumption. But the mean food intake and body weight gain of adequate Fe groups(LCuAFeCd, ACuAFeCd) were higher than those of deficient Fe groups (ACuLFeCd, LCuFeCd)in long-term Cd poisoned rats. The mean Cd levels of serum, liver, kidney, and urine in ACuAFeCd group were lower than those of Cu and /or Fe deficient groups. But the mean fecal Cd excretion of ACuAFeCd group was higher than that of Cu and/or Fe deficient groups. And the mean Cd retention amount of ACuAFeCd group was lower than those of Cu and/or Fe deficinet groups. In conclusion, these results provide an evidence that adequate Cu and Fe intakes can decrease Cd accumulation in rats. Therefore, in the point of increasing environmental Cd contamination, adequate Cu and Fe intakes must be suggested to prevent Cd accumulations.
Heavy metal accumulation in living organisms through food-wed can give serious damage on physiological responses for vital activities. The initiation of heavy metal supposed to begin from the bio-accumulation of plants. To establish basic data fur heavy metal contents in plants at the area without artificial contamination, both woody and herb plants at Mt. Kwang-Duk were studied. The content of heavy metals in various organs of plants were analyzed by ICP. The range of heavy metals in plants for Al, As, Cd, Co, Cr, Cu, Fe. Mn, Pb, Se and Zn were 1.019∼257.200ppm, O∼2.929ppm, 0∼0.079ppm, 0∼0.054ppm, 0.023∼3.007ppm, 0∼1.997ppm, 2.031∼148.500ppm, 1.069∼51.320ppm, O∼126.900ppm, 0.708∼4.927ppm and 0.846∼4.949ppm, respectively. The amount of heavy metals in plants are much less than that of soil except some species. In woody plants, it was detected that the metal contents of leaves were higher than that of stems especially in case of Al and Fe with statistical significance. There were significant differences between shoots and roots of herb plants in metal content of Al, Co and Fe. Those metals have more accumulated in roots comparing with shoots. Some species of plants had shown the difference tendency of heavy metal accumulation. Generally, most species had not exceeded over twice of mean value each other, and had various difference according to the kinds of heavy metals.
Searches for the nutrition requirements of three strains of Brevibacterium ammoniagenes reported in the previous paper were carried out with an aim of achieving the striking accumulation of L-glutamic acid and the large multipication of cells. It was recognized that all three strains required both biotin and thiamine, together with amino acids such as histidine or cysteine, for their good growth and extracellular L-glutamic acid accumulation. The quantity of biotin required for remarkable growth of these microorganisms was quite different from that for the maximum production of L-glutamic acid. This result, however, did not apply in the case of thiamine. It was also confirmed that, of 18 amino acids, histidine and cysteine were the msot effective organic nitrogen sources, while the most available inorganic ammonium salt resulting in a large amount of L-glutamic acid-production and considerable cell gorwth was found to be only urea. Maximum accumulation of extracellular L-glutamic acid, more than 50%(w/w) of the initial sugar content, could be obtained from fermentation in the medium containing wheat-bran extract(Brev. ammoniagenes T-1 and Brev.ammoniagenes Y-2) or rice-bran extract(Brev. ammoniagenes YR-2), which confirmed us a possibility that these bacteria might be employed for industrial fermentation of L-glutamic acid.
The reduction in the amount of asialoglycoprotein (ASGP) receptor, which resides exclusively on the plasma membrane of functioning mammalian hepatocytes, as a consequence of hepato-cellular damage has been demonstrated in various pathologic conditions of the liver. Galac-tosylated human serum albumin (GSA) is a newly developed receptor-binding agent, specific for the ASGP receptor. Tc-99m GSA binds quantitatively to liver ASGP receptors and the rate of accumulation in the liver is dependent on hepatic function represented as the amount of receptor, as well as the amount of ligand injected, its affinity to the receptor and the hepatic blood flow. The findings of Tc-99m GSA scintigraphy were reported to reflect the hepatic function of the patients with large hepatic tumors, obstructive jaundice, acute and chronic liver disease. Tc-99m GSA scintigraphy is an easy and reliable test and has the clinical potentials to evaluate the liver function in the patients with hepatic disorders.

Mathematical models of the litter accumulation, decay and turnover in the grassland and forest ecosystems of equilibrium state of the annual litter production were established to analyse the decay rates of organic and inorganic constituents of the litter. Those models were validated by an application to a Phragmites longivalvis grassland in a delta of the River Nakdong. The decay constants of cold-water-soluble fractions, other carbohydrates, hot-water-soluble fractions, cellulose, crude fat, lignin and crude protein in the litter were 0.730, 0.583, 0.555, 0.505, 0.479, 0.331 and 0.310 respectively. The amount of mineral nutrients such as N. P. K. Ca and Mg returned annually to the soil were estimated to 7.09, 1.34, 2.36, 4.37 and 0.79g/m2 respectively.
A transformed hairy root clone of Scutellaria baicalensis was established following infection with Agrobacterium rhizogenes ATCC15834. Three root clones of S baicalensis were selected by growth habit and baicalin content. The most active strain-the SR-03 clone-was examined for its growth and baicalin content under various culture conditions. The root growth and baicalin content were maximized in a Schenk and Hildebrandt medium supplemented with 4 and 6% sucrose, respectively. The accumulation of baicalin in transformed hairy roots was enhanced through exposure to various elicitors. Elicitation was attained by the addition of methyl jasmonate, salicylic acid, and various concentrations of fungal cell wall elicitors to the medium. The accumulation of baicalin in the elicited cultures ranged from 10.5 to 18.3 mg/g dry weight of the roots, which was 1.5- to 3-fold the amount attained in controls.
Electrical properties such as space charge accumulation and water tree length and physical properties such as tensile strength, elongation and degree of crosslinked polyethylene (XLPE)/ethylene n-butyl acrylate copolymer (EnBA) blends were investigated. It was found that electrical properties such as water tree length grown at a specific condition and AC breakdown strength are improved by blending the XLPE with EnBA. The EnBA having higher nBA content showed the better electrical properties in XLPE/EnBA blends. A further improvement of these properties was achieved when a small amount of crosslinking coagent was used in the preparation of XLPE/EnBA blends.
