Effects of open-field summer warming and drought on the abnormal shoot growth of Pinus densiflora seedlings (실외 여름철 온난화 및 가뭄 처리가 소나무 묘목의 이상생장 반응에 미치는 영향)

  • Pinus densiflora is a fixed-growth coniferous species that elongates its shoot once a year and finishes growing in early summer. However, it may produce additional shoots in the same year in response to external stimuli, called abnormal shoot growth. This study investigated the effects of open-field summer warming and drought on the abnormal shoot growth of P. densiflora seedlings. In March 2022, two factorial combinations were constructed, including two temperature treatments (control and 4℃ increase) and two precipitation treatments (control and drought), with five replicates for each combination. The temperature treatment was performed for 87 days from May 14 to August 8, 2022, and the precipitation treatment was performed for 33 days from May 14 to June 15, blocking 100% of the ambient rainfall. The abnormal shoot occurrence rate and leaf unfolding stages were measured in November, and the shoot and root collar diameter growth rates were calculated by comparing the seedling height and root collar diameter measured in August(after the cessation of treatment) and October(after the end of growing period) with the initial values (i.e., May 2022). The abnormal shoot occurrence rate significantly increased under the warming treatment, showing a 410.6% increase in the warming plots (38.4%) compared to the control plots (7.5%). There was no significant difference in the shoot and root collar diameter growth rate regarding warming and drought treatments. Abnormal shoots may have been affected by high temperatures by inducing early transition to the next ontogenetic stage.

Effects of Phytohormone and Activated Carbon on the Growth and Rooting of Teratoma Shoot Induced from Crown Gall Callus in Nicotiana tabacum cv. NC 2326 (연초 Crown Gall Callus유래 Teratoma Shoot의 생육과 발근에 미치는 식물호르몬과 활성탄의 영향)

  • These studies were conducted to investigate effects of phytohormone and activated carbon on the growth and rooting of teratoma shoots induced from Nicotiana tabacum cv. NC2326 transformed by Aerobacterium tumefaciens C58. GA was effective for shoot elongation and reduction of multiple shoots from teratoma shoot, however, leaves of teratoma shoot cultured on the medium with GA were pointed. ABA was also effective in promoting shoot elongation, but was not for reduction of multiple shoots. Teratoma shoot cultured on the medium with 1 n activated carbon promoted shoot elongation and inhibited the number of shoots differentiated, but was grown as abnormal shoot. Addition of 1% activated carbon and 0.5mg/l BA to culture media was effective for shoot elongation and reduction of multiple shoot and for formation of round leaves as normal leaves. Though these shoots were inoculated on the rooting medium, they could not from roots but formed multiple shoots. Boric acid, myo-inositol and sucrose were also ineffective on the rooting of teratoma shoots.

Effect of Growth Regulators on the Organ Differentiation and the Growth from the Axillary Bud of Sweetpotatoes in Vitro Culture (고구마의 액아배양에서 생장조절물질이 기관분화 및 생장에 미치는 영향)

  • This study was conducted to determine the optimum concentrations of growth regulators and their responses on the clonal propagation in axillary bud culture. Cultivars, Hongmi and Shinmi, responded differently to the levels of growth regulators, proliferation rate and shoot growth. The shoot and root of Hongmi cultivar in axillary bud culture were conspicuously induced by combination of NAA(0.1mg/l) and Kinetin(1mg/l) while Shinmi cultivar were affected by the single concentration of Kinetin(1mg/l) and BA(0.1mg/l), and also by the combination of NAA(0.1mg/l) and Kinetin(1mg/l). Better shoot growth and root initiation were obtained in the combination of NAA(0.1mg/l) and Kinetin(1mg/l) regardless of cultivars used when 5mm axillary buds were cultured. The shoots regenerated at the high levels of BA(1-5mg/l) were abnormally thicker and narrower leaves than normal plants and short in shoot height. Frequencies of abnormal plants were higher than that of the low level (0.1mg/l) of BA.

Generation and Selection of Promoter Trap Lines for the Investigation of Shoot Development in Arabidopsis (애기장대에 있어서 shoot 발달 연구를 위한 프로모터 trap 라인들의 제조 및 선별)

  • T-DNA-mediated transformation is a common method for generating transgenic plants with insertional mutagenesis. In order to identify important genes involved in shoot development, a system of promoter trap insertional mutagenesis was employed in Arabidopsis thaliana. For this system, an efficient promoter trap vector, pFGL561 was developed. The pFGL561 includes a basta-resistant gene, an intron with multiple splicing donor and acceptor sites, and a promoter-less GFP reporter gene. Using floral-dipping method, we made total 300 $T_1$ promoter-trap lines which were screened for GFP expression. GFP signals in the $T_1$ plants were detected with high frequency, 26.7%, and the signals were reconfirmed in $T_2$ plants. To isolate the genes that are involved in shoot development, phenotypes were analyzed in $T_2$ plants of the 19 $T_1$ lines that had GFP signals in shoot apex, and 6 $T_1$ lines were selected that had abnormal shoot development. These lines will be very useful for the investigation of shoot development.

Efficient Plant Regeneration from Mesophyll Protoplast of Arabidopsis thaliana and Morphological Characterization of Regenerants (애기장대 (Arabidopsis thaliana)의 엽육원형질체로부터 효율적인 식물체 재분화와 이들의 형태적 특성)

  • Protoplasts were isolated from the leaf mesophyll tissue of in vitro 4-weeks-old Arabidopsis thaliana and cultured in MS liquid medium supplemented with 2.0 mg/L NAA, 0.5 mg/L BAP and 9% mannitol in the dark at $25^{\circ}C$. When protoplast-derived microcolonies were dehydrated, the frequency of callus induction enhanced approximately 7-fold higher compared with non-dehydrated microcolonies in CP medium. Fifty callus lines were selected from dehydrated microcolonies. Shoots were efficiently initiated from the green spots of the selected shoot forming calli cultured on MS regeneration medium supplemented with 0.05 mg/L IAA, 7.0 mg/L 2-iP and 30 g/L sucrose under continous illumination for 4 weeks. Shoot regeneration frequencies (calli regenerating at least one shoot) were 3.5%~56%. Histological observations of shoot forming callus revealed that tracheary elements initiated from inner compact cells, and that meristemoids developed to shoot primordia and shoots. Roots were induced from these regenerating shoots on MS medium without phytohormones. These regenerants were successfully transplanted into potting soil. Morphological characterization of 50 protoplast-derived plants showed that the frequency of normal type was 78%.

Karyological Variation of Callus-derived Regenerants in Allium victorialis var. platyphyllum

  • The calli obtained from the bulbs of A. victorialis var. platyphyllum on MS based medium containing 2 mg/L 2, 4-D and 1mg/L BA. Plants from calli were regenerated on MS basal media supplemented with combinations of NAA and four kinds of cytokinin, BA, zeatin, kinetin and 2iP, and with only each cytokinin. The good response of shoot regeneration was observed in the media with combinations of NAA and BA or zeatin, and with only BA or zeatin. Shoot regenerating response in the medium with combinations of NAA and BA or zeatin, and with only BA or zeatin. Shoot regerating response in the medium with combinations of NAA and BA or zeatin was about two times higher than that in the mediuim with only BA or zeatin. From the karyotypic analysis of the regenerated plants, abnormal diploids, aneuploids and mixoploid with structural aberrations were investigated. The somaclonal variants (AV 16-04, AV 13-03) were shown the considerable differences from normal diploid regenerant (AV 18-03) in the external morphology.

Plant Regeneration and Bulblet Formation of Allium wakegi Araki

  • Allium wakegi was cultured shoot tip in the condition of light culture. The Allium wakegi added plant growth regulator was observed of plant regeneration and bulblet formation. Callus Induction and growing rate was the best of 78% when added alone 2,4-D 0.5mg/L. In the formation of shoot, its regeneration rate was 96% when added BA 0.5mg/L in the light culture condition. When BA 0.5mg/L and NAA 0.5mg/L mixed and BA 0.5 mg/L and NAA 1.0mg/L mixed, the rates were 99% and 97% respectively, and these conditions were suitable for forming shoot. In the formation of roots, when added NAA 2.0mg/L in the light culture condition, the regeneration rate was 90.6 % and the roots were abnormal. When added NAA 1.0mg/L, the rate was 82 % and the highest. In the formation of bulbs, when BA 05mg/L and NAA 1.0mg/L mixed, the root generantion and its size in the bulbs was the best compare to other treatment experiments.

Change in the Bourse Shoot and Fruit Growth due to the Gibberellins Paste in the Young Fruit of Niitaka Pear (유과기 지벨렐린 도포제 처리에 의한 '신고'배의 과대지와 과실생장 변화)

  • The object of this study was to evaluate the effect of gibberellins (GA) pasting on the bourse shoot, and not just on the fruit characteristics but also on the quality, of Niitaka pear (Pyrus pyryfolia L). The fruit stalk was treated with GA (control, 25mg/fruit stalk) 35 days after reaching full bloom. In the GA-treated tree, the occurrence of abnormal bourse shoot (52.5%) increased, and the spur bud and flower number decreased. The diameter of the GA-paste-treated fruit increased during the pear growth period with GA treatment. The diameter, length, and weight of the GA-paste-treated fruit increased at harvest time, but the hardness was lower than that of the control. The differences in soluble solid, acidity, and fruit color between the control and the GA-paste-treated fruit were not significant. Post-harvest, during the storage period, the hardness of the GA-paste-treated fruit was lower than that of the control, and its weight loss ratio was higher than that of the control on the $60^{th}$ days of storage.

  • The modification of DNA and histone plays an important role for gene expression in plant development. The objective of this research is to observe the effects of methylation on the gene expression during dedifferentiation from rice mature seeds to callus and differentiation from callus to shoots. The embryogenic callus with ability to shoot regeneration was not induced on the N6A medium supplemented with 5-azacytidine and abnormal callus with brown color was formed. When the normal rice callus was placed on the regeneration MSRA medium supplemented with 5-azacytidine, the shoot regeneration was inhibited. The results showed that 5-azacytidine, DNA demethylating agent, had negative effects on normal embryogenic callus formation and shoot regeneration. This suggested that DNA methylation of some genes was required for normal cell dedifferentiation and differentiation in tissue culture. The microarray and $GeneFishig^{TM}$ DEG screening were used to observe the gene transcript profile in callus induction and regeneration on N6A (N6 medium + 5-azaC) and MSRA (MS regeneration medium + 5-azaC). Subsets of genes were up-regulated or down-regulated in response to 5-azaC treatments. The genes related with epigenetic regulation, electron transport, nucleic acid metabolism and response to stress were up and down regulated. The different expression of some genes (germin like protein etc.) during callus induction and shoot regeneration was confirmed using RT-PCR and northern blot analysis.

Cryopreservation of in Vitro Grown Axillary Shoot-tip Meristems of Lycium chinense by Vitrification

  • In vitro-grown axillary shot-tip meristems of Lycium chinense Mill. from cold-acclimated plant were successfully cryopreserved using a vitrification technique. After loading for 15 minutes with a mixture of 2.0 M glycerol and 0.4 M sucrose ($20^{\circ}C$), small segments (1-2 mm, 3-4 mm, and 5-6 mm) were cut from axilary buds and exposed to the cryoprotectant solution containing 30% glycerol, 15% ethylen glycol,15% dimethyl sulfoxide (DMSO), and 0.4 M sucrose at $0^{\circ}C$ for 30-120 minutes prior to direct plunge into liquid nitrogen (LN). After rapid thawing ($40^{\circ}C$), the segments were washed with MS medium containing 1.2 M sucrose for 0-35 minutes, and then transferred onto recovery-growth medium. The highest survival rate (about 90%) was obtained with cold-hardening treatment, and cryopreserved explants were successfully recovered to plantlets. No abnormal morphological changes were observed with the recovered plants after cryopreservation.
