Assessment of The Accuracy of The MR Abdominal Adipose Tissue Volumetry using 3D Gradient Dual Echo 2-Point DIXON Technique using CT as Reference

  • In this study, in order to determine the validity and accuracy of MR imaging of 3D gradient dual echo 2-point DIXON technique for measuring abdominal adipose tissue volume and distribution, the measurements obtained by CT were set as a reference for comparison and their correlations were evaluated. CT and MRI scans were performed on each subject (17 healthy male volunteers who were fully informed about this study) to measure abdominal adipose tissue volume. Two skilled investigators individually observed the images acquired by CT and MRI in an independent environment, and directly separated the total volume using region-based thresholding segmentation method, and based on this, the total adipose tissue volume, subcutaneous adipose tissue volume and visceral adipose tissue volume were respectively measured. The correlation of the adipose tissue volume measurements with respect to the observer was examined using the Spearman test and the inter-observer agreement was evaluated using the intra-class correlation test. The correlation of the adipose tissue volume measurements by CT and MRI imaging methods was examined by simple regression analysis. In addition, using the Bland-Altman plot, the degree of agreement between the two imaging methods was evaluated. All of the statistical analysis results showed highly statistically significant correlation (p<0.05) respectively from the results of each adipose tissue volume measurements. In conclusion, MR abdominal adipose volumetry using the technique of 3D gradient dual echo 2-point DIXON showed a very high level of concordance even when compared with the adipose tissue measuring method using CT as reference.

  • Adipose tissue plays a central role in regulating dynamic cross-talk between tissues and organs. A detailed description of molecules that are differentially expressed upon changes in adipose tissue mass is expected to increase our understanding of the molecular mechanisms that underlie obesity and related metabolic co-morbidities. Our previous studies suggest a possible link between endophilins (SH3Grb2 proteins) and changes in body weight. To explore this further, we sought to assess the distribution of endophilin A2 (EA2) in human adipose tissue and experimental animals. Human paired adipose tissue samples (subcutaneous and visceral) were collected from subjects undergoing elective abdominal surgery and abdominal liposuction. We observed elevated EA2 gene expression in the subcutaneous compared to that in the visceral human adipose tissue. EA2 gene expression negatively correlated with adiponectin and chemerin in visceral adipose tissue, and positively correlated with $TNF-{\alpha}$ in subcutaneous adipose tissue. EA2 gene expression was significantly downregulated during differentiation of preadipocytes in vitro. In conclusion, this study provides a description of EA2 distribution and emphasizes a need to study the roles of this protein during the progression of obesity.

Effect of Dietary Conjugated Linoleic Acid (CLA) on Abdominal Fat Deposition in Yellow-feather Broiler Chickens and Its Possible Mechanism

  • A total of 60 one-day-old Yellow-feather broiler chickens were allotted into treatment and control groups. The treatment group was fed with the diet supplemented with 3% conjugated linoleic acid (CLA) for 48 d, while control group was fed with the diet supplemented with 3% rapeseed oil. Chickens were slaughtered in each group at the age of 49 d, and the blood and the abdominal adipose tissue were sampled. Serum cLeptin and serum cAdiponectin were measured by ELISA. The total RNA was extracted from adipose tissue to measure the abundance of the chicken growth hormone receptor (cGHR), insulin-like growth factor 1 (cIGF-1), insulin-like growth factor I receptor (cIGF-IR), peroxisome proliferator-activated receptor gamma ($cPPAR{\gamma}$), cAdiponectin and cAdipoIR mRNA by RT-PCR using ${\beta}$-actin as an internal standard. Results showed that the CLA decreased the abdominal fat index by 20.93% (p<0.05). The level of serum cLeptin but not serum cAdiponectin was significantly increased by CLA treatment (p<0.05). CLA down-regulated the relative abundance of cGH-R mRNA and $cPPAR{\gamma}$ mRNA in abdominal adipose tissue by 24.74% (p<0.05) and 66.52% (p<0.01) respectively. However, no differences were found between CLA treatment group and control group (p>0.05) in the relative abundance of cIGF-1, cIGF-IR, cAdiponectin, and cAdipoIR mRNA in abdominal adipose tissue. The data suggested that CLA inhibited abdominal fat deposition in broiler chicken may be determined by decreasing the GHR available for GH, and by inhibiting the differentiation of preadipocytes via down-regulation of $PPAR{\gamma}$, but independent of IGF and (or) GH-IGF pathway or adiponectin action.

The Analysis of Relationship between Abdominal Adipose Tissue Distribution and Health Risk Factors in Adolescent Obese Man (청소년기 비만 남성의 복부지방조직 분포와 건강관련 위험지표의 관련성 분석)

  • In the current study, the relationships between abdominal adipose tissue depots and skinfold thickness, body circumference and composition, and blood lipids and pressure were investigated in adolescent obese man. Total abdominal adipose tissue (TAT) and subcutaneous adipose tissue (SAT) were correlated with skinfold thickness. Especially subscapular skinfold was highly correlated with TAT and SAT, and it was found that chest skinfold was the only predictor for estimating visceral adipose tissue (VAT). Body circumference and body composition were associated with TAT and SAT, not VAT, and correlation coefficient of waist circumference was highest. On the other hand, body fat mass was highly correlated with TAT and SAT than %body fat and fat free mass. It was also shown that blood triglyceride concentration and systolic blood pressure were the only predictor for estimating VAT and SAT respectively.

Expression of Steroidogenesis-related Genes in Rat Adipose Tissues

  • Adipose tissue is one of the major endocrine gland. More recently, local production of steroids in adipocytes differentiated from mouse 3T3-L1 cell-line was reported. We hypothesized that rat adipocytes have steroidogenic machinery and the expression patterns of the components might be differentially regulated, depending on the distribution and sex. To verify this hypothesis, we collected the adipose tissues depot-and sex-specifically at postnatal day (PND) 30, and performed quantitative RT-PCRs. In overall aspects, the abundances of the transcripts were lower in the brown adipose of both sexes. $3{\beta}-HSD$ transcript levels in female abdominal and reproductive adipose, CYP17 transcript levels in female reproductive adipose, $17{\beta}-HSD$ transcript levels in female abdominal and reproductive adipose, and CYP19 transcript levels in female abdominal adipose were significantly lower than those of male counterparts. Similar to steroidogenic factors, the abundance of the $ER-{\alpha}$ transcripts were generally lower in the brown adipose of both sexes. $ER-{\beta}$ transcripts were more abundant in male white adipose depots than their female counterparts. The levels of LHR transcripts in female reproductive adipose were significantly higher than those of male counterpart. In conclusion, our study demonstrated that the expressions of steroidogenesis-related genes were depot- and sex-specifically occurred in the immature male and female rat adipose tissues. Our study suggested that the adipose tissues are not only targets but de novo synthesizing sites of sex steroid(s), though the synthesizing activities could be much less than in gonads. Further researches in this field will be helpful for understanding the adipose physiology and for medical application such as sex-specific steroid supplement therapies for older populations.

Relationship between Psychological Factors and Obesity before and after Obesity Treatment in Korean Obese Women (한국 비만 여성에서 비만 치료 전 후의 심리적 상관관계 분석)

  • Objectives : Psychological comorbidities are high in patients with obesity and are associated with a variety of medical and dietary problems. This study aims to examine the association between psychological factors and obesity. Methods : This study was performed in pre-menopausal obese($BMI{\geq}25kg/m^2$, waist circumference ${\geq}85cm$) women in Seoul, in 2008 (n=35). Every patient underwent the obesity treatment program. The program included dietary and exercise education, and abdominal mesotherapy for 6 weeks. Simple anthropometry including weight, BMI and Computed Tomography (CT) including Subcutaneous adipose tissue (SAT), Visceral adipose tissue (VAT) and Visceral adipose tissue/Subcutaneous adipose tissue ratio (VSR) were done. To assess psychological factors, the Rosenberg self-esteem scale (SES) questionnaire, Beck depression inventory (BDI) questionnaire and stress response inventory(SRI) questionnaire were administered. Results : 1. All of the obesity indicators (except VSR) decreased significantly after the obesity treatment program. 2. There was a significant relationship between self esteem (SES score) and visceral obesity (VAT and VSR) measured at the end of the program. 3. During the intervention, the more weight, BMI, and subcutaneous adipose tissue decreased, the more self-esteem (SES) increased. There was no relationship between depression (BDI) and obesity. And the change in stress response (SRI) was associated with the change of deep subcutaneous adipose tissue and total abdominal adipose tissue. Conclusions : This study proves that visceral obesity may contribute to low self-esteem, and there is a possibility that the other psychological factors could also be related with obesity in Korean obese women. Individualised antiobesity therapy may be required depending on the patient's psychological characteristics and weight loss could be helpful in order to treat psychological problem in obese patients.

Cloning and Expression of β1-Adrenergic Receptor Genes in Adipose Tissues from Korean Native Cattle (Hanwoo)

  • Bovine ${\beta}1$-adrenergic receptor (AR) cDNA was cloned using degenerative primers. Bovine ${\beta}1$-AR coded for 467 amino acids and the comparison of the deduced amino acid sequence with that of sheep showed 93.4% identity. Northern blot analysis indicated that transcript size for the bovine ${\beta}1$-AR was 3.6 kb in the adipose tissue. The expression level of three $\beta$-ARs (1, 2, and 3) in bovine abdominal, subcutaneous, and perirenal adipose tissues were examined using reverse transcription-polymerase chain reaction (RT-PCR), and the levels of ${\beta}1$- and ${\beta}3$-AR mRNA were found to be lower in the subcutaneous adipose tissue than in the abdominal and perirenal adipose tissues. These results suggest that the expression of $\beta$-ARs mRNA are differentially regulated among the adipose tissues.

Effects of Dietary Xylooligosaccharides on Lipoprotein Lipase Activity in Epididymal Adipose Tissue and Lipid Composition in Serum of Rats Fed High Fat Diets (Xylooligo당이 고지방식이 흰쥐 부고환 지방조직의 Lipoprotein Lipase 활성과 혈중 지질조성에 미치는 영향)

  • This study was conducted to examine the effects of dietary xylooligosaccharides on lipoprotein lipase (LPL) activitiy in epididymal adipose tissue and lipid composition in serum of rats fed normal or high fat diet. Male Sprague-Dawley male rats weighing 100 $\pm$ 10 g were randomly divided into four groups, two normal diets and two high fat diets containing 1% cholesterol and 10% lard. Two normal diets were classified into a basal diet (normal group) and that with 10% xylooligosaccharide diet (NX group). The high fat diet groups were classified into a HF group without xylooligosaccharides diet and HFX group supplemented with 10% xylooligosacchride diet. Experimental diets were fed ad libitum to the rats for 4 weeks and then they were sacrificed. Body weight, epididymal weight and abdominal weight in HF group were hevier than the those of normal group, but HFX group was significantly reduced compared to HF group. Relative body weight to epididymal weight and relative body weight to abdominal weight in HF group were increased to 50%, 51%, respectively, compared to normal group, but HFX group was reduced 22%, 16%, respectively, compared to HF group. The levels of serum triglyceride, total cholesterol, LDL-cholesterol and atherogenic index in HFX groups were significantly lower than those of HF group, whereas HDL-cholesterol levels were significant increased. Triglyceride contents of epididymal adipose tissue in HF group was increased to 39%, compared to normal group, but HFX group was reduced to 15.8%, compared to HF group. Cholesterol contents of epididymal adipose tissue in HF group was increased 121%, compared to normal group, but HFX group was reduced to 26%, compared to HF group. The activity of LPL in epididymal adipose tissue was increased to 259% in HF group, compared to normal group and HFX group was reduced to 66%, compared to HF group. These result of this study suggested that improved lipid metabolism observed in rats fed xylooligosaccharides may be caused by an alteration of LPL activity in epididymal adipose tissue and lipid composition in serum.
