Component of oxytetracycline on exfoliation of abalone, Haliotis discus hannai (옥시테트라사이클린의 전복박리 성분)

  • Exfoliation of fouling abalone, Haliotis discus hannai from shelters by commercial oxytetracycline (OTC) was observed in culture farms. In the present study, different components of OTC for exfoliation of abalone were investigated to understand how to work. Abalone were detached from shelter in 14,000 ppm of commercial OTC (main ingredients of OTC: OTC-hydrogen chloride (HCl), 50%; glucose, 49%; blue pigment, <1%), but not below 8,000 ppm. A 95% of exfoliation rate was observed in OTC-HCl (7,000 ppm, pH 2.8) but no exfoliation in OTC-HCl (7,000 ppm, pH 5.0), glucose (7,000 ppm) or blue pigment (140 ppm). Moreover 100% exfoliation rate was observed in HCl-seawater of pH 2.8. These results indicate that HCl is the component resulting in exfoliation of the fouling abalone. Abalone was detached in HCl solution (pH 2.5-3.2) within 2 min. However, a lower pH and longer treatment resulted in delayed recovery of the detached abalone. Thus, exfoliation of fouling abalone can be achieved by a low pH treatment with cautious handling.

Potential use of Bacillus amyloliquefaciens as a probiotic bacterium in abalone culture (북방전복, Haliotis discus hannai 에 대한 Bacillus amyloliquefaciens의 probiotic 효과)

  • In comparison to the numbers of such studies of fish, few studies have been carried out on the immunity, physiology and ecology of abalone, while studies on abalone disease are also extremely rare. Moreover, mass mortality of cultured abalone due to pathogenic bacteria has not been reported in the southern coast of Korea. However, Vibrio-like bacteria have been isolated from dead abalone, which indicates that a review is required in order to determine the cause of abalone mortality. Use of an antimicrobial agent to minimize the damage caused by disease in abalone farms is common, but the therapeutic effects are insignificant. Demand for probiotics has increased, but research on the development of probiotics for use in abalone culture is very rare. Therefore, the present study isolated KC16-2 from fermented kimchi soup and investigated the characteristics of the isolate as a candidate probiotic bacterium in abalone. KC16-2 was identified as Bacillus amyloliquefaciens (B. amyloliquefaciens KC16-2) based on its biochemical properties and 16S rRNA gene sequence. B. amyloliquefaciens KC16-2 showed inhibitory effects against the growth of various vibrios in vitro, and kept the numbers constant until four days after inoculation in marine water at a temperature of $15{\sim}25^{\circ}C$, indicating the possible use of KC16-2 as a probiotic, except in the winter. The growth of KC16-2 was inhibited by bile salt, but the numbers increased over time suggesting the bacteria were still alive in the abalone's digestive tract. Abalone fed with a diet including KC16-2 for 12 weeks showed good growth, but showed no significant differences from the control group. However, the mortality of the abalone supplied the probiotic diet was reduced to half that of the control group in a challenge test with Vibrio tubiashii. Therefore, we suggest that B. amyloliquefaciens KC16-2 could be used as a probiotic bacterium for control of the mortality of abalone caused by opportunistic pathogenic vibrios.

Optimum Structure and Deployment of an Abalone Reef for the Marine Ranching Creation in Jeonnam Archipelago of Korea (전남다도해형 바다목장 조성을 위한 전복초의 적정구조 및 시설방법)

  • This study describes the optimum structure and deployment of an abalone reef for marine ranching creation in Jeonnam archipelago of Korea. The structure of an abalone reef was analyzed based upon the functions of both feeding and shelter. Deployments of abalone reefs were considered based upon the stability against waves in water depth shallower than 5 m. Also, the calculation of their stabilities was made to only rocks of abalone modules, and exposure coefficient of rocks used was 1.0 and 0.5. The results show that rocks of both 0.2 and 0.5 ton are unstable under the condition that exposure coefficient is 1.0 at a depth of 5 m, but that they are stable under the condition that exposure coefficient is 0.5. It means that, if the structures(e.g. breakwater blocks) for dissipating the energy of waves and currents are provided, small rocks of 0.2 ton can be used as an abalone reef.

Comparison of Physicochemical Properties of Meat and Viscera with Respect to the Age of Abalone (Haliotis discus hannai) (전복(Haliotis discus hannai)의 연령에 따른 육과 내장의 이화학적 특성 비교)

  • We compared the physicochemical properties of meat and viscera with regard to the age of abalone (1-, 2-, 3- and 4-year-old abalone). With respect to total amino acids, the level was highest in 1-year-old abalone, at $19,046.00{\pm}548.53\;mg%$, whereas 4-year-old abalone had the lowest value of $15,770.44{\pm}454.19\;mg%$. In the viscera, 3-year-old abalone had the greatest level of total amino acids, at $16,575.10{\pm}477.37\;mg%$, whereas 1-year-old abalone had the lowest, at $14,947.26{\pm}430.48\;mg%$. The level of total free amino acids in meat tended to increase with age whereas that of the viscera fell. The level of polyunsaturated acids decreased with age in meat. The concentration of chondroitin sulfate in both meat and viscera tended to increase with age. This was especially noticeable in meat. The level in 1-year-old abalone, $9.03{\pm}0.23%$ (w/w), rose to $14.17{\pm}0.33%$ in 4-year-old abalone, with statistical significance. On the other hand, the collagen level in both meat and viscera decreased with age. Again, this was particularly noticeable in meat, where the concentration in 1-year-old abalone, $199.70{\pm}5.00\;mg/g$, fell remarkably to the value of $47.86{\pm}1.10\;mg/g$ in 4-year-old abalone. We have thus provided basic data for research on abalone.

Cytotoxic and Antioxidant Activities of Abalone (Haliotis discus hannai) Extracts (전복 용매 추출물의 세포독성과 항산화 활성)

  • The objective of this study was to investigate the fatty acid composition of raw and dried abalone (Haliotis discus hannai) and to determine the effect of abalone extracts on cytotoxic activity and anti-oxidant properties. Dried abalone was extracted with acetone/methylene chloride (A+M) and methanol (MeOH), and the extracts were fractionated using n-hexane, 85% aq. methanol (MeOH), butanol (BuOH), and water. Cytotoxic activity against HT-29 cancer cell lines was determined using the 3-(4,5-dimethylthiazol)-2,5-diphenyltetrazolium bromide (MTT) assay. Antioxidant activity was measured using a fluorescence sensitive dye, 2'-7' dichlorofluorescein-diacetate (DCFH-DA). The fatty acid composition of dried abalone was higher (22:6n-3) than that of raw abalone, and it had a lower percentage of 20:4n-6 than raw abalone. Analysis of cell viability showed that the crude extract treatments and fractions were cytotoxic, suppressing the growth of HT-29 cancer cell lines (p<0.05). The A+M extract showed a higher cytotoxic effect on the growth of HT-29 cells compared to the MeOH extract. Among the fractions, the 85% aq. MeOH fraction showed the strongest cytotoxicity against the growth of HT-29 cells. The highest activity in terms of scavenging reactive oxygen species (ROS) was likewise obtained with the use of 85% aq. MeOH. Our results suggest that the 85% aq. MeOH fraction has a potent inhibitory effect on the proliferation of human cancer cells.

Infestation of the Abalone, Haliotis Discus Hannai, by the Polydora under Intensive Culture Conditions in Korea (우리나라 전복 양식장의 패각 천공성 다모류 감염현황)

  • The genus Polydora(Polychaeta, Spionidae) includes many species well known for their activity as borers. They often become harmful invaders by reducing the growth rate and meat yield of, or inducing the mortality of commercially important mollusck, abalone, Haiotis discus hannai. In 2012, the frequency of the Polydora was observed with 5~99% in live abalone and 5.3~70.3% in dead abalone shells of abalone sea-caged aquaculture system, Korea. There are many nacreous blister in the ventral margin and inner of the infestated abalone by abalone in response to the Polydora. A worm bored two holes in the shell and come in and out for ingestion the organic matters. They are more than 40 mm in length and had outstanding palps with black bars, disc form pygidium and 4 eyespots. This is the first record for the statue of Polydora infestation of sea-caged cultured abalone, Haliotis discus hannai in Korea.

  • The lipoprotein-bound carotenoids occurred in the viscera of abalone (Haliotis discus hannai) were separated and characterized to be the mixture of fatty acid esters of lutein and free lutein. The main fatty acid components of the esters were lauric (68%) and myristic acid (17%), and indicated the characteristic pattern compared with that of triglycerides and cholesterol esters in abalone. Patterns of carotenoids from the lipoproteins were, simple and this suggested selected metabolisms of various feed-originated carotenoids found in the whole acetone extract of abalone.

A Preliminary Study of a Submersible Facility for Abalone Spats (부침식 전복치패 중간육성장치 개발을 위한 초기연구)

  • The abalone shell (Haliotis) is one of the most important resources for the coastal fisheries and it is popular as an aquacultural species. Proper cultivating grounds for mid-term nursery of abalone spats are required before releasing them. It is difficult for us to find good enough aquacultural grounds to rear abalone spats to 20"30mm of shell length. Therefore, we need to study a practical and effective new type of aquacultural device for the nursing of abalone spats by using open sea areas. We can find this kind of studies from 'Marine aya No. 1' of Japan, Though they focused on the easy operation, safe working and low operating cost, it involves so much initial cost that it is difficult for us to justify such expenditure. However, with a modified small buoy system, this submersible facility needs only a horizontal frame to fulfill its essential function and the vortical part can be removed. The working boat equipped with a pump can operate this facility to keep it submerged or floated. This paper deals with the possibility of this submersible fishery facility for the mid term nursing of abalone spats in the open sea. A small version of this system to ensure low initial cost is suggested and wave and current forces were calculated for the estimation of the weight of the mooring anchor.

Analysis of Current Distribution around a Scaled-down Abalone System to determine the cause of mass mortality of abalone, Haliotis discus hannai (Ino, 1952) (북방전복 Haliotis discus hannai (Ino, 1952) 의 폐사규명을 위한 모형 양식시설 주변의 조류 분포 분석)

  • This study is aimed at figuring out the reasons of the mass mortality of abalone and the increase in its mortality rate in the sea cage. The study suggests that lack seawater circulation in an abalone aquaculture cage is an important culprit for it. We analyzed the current distribution around a 1/20 scaled-down abalone unit cage of 4 rows and 10 columns by fluid flow visualization technique (PIV : Particle Image Velocimetry). The speed of current in the model cage definitely slowed down in the first column of a unit cage. We also observed currents going down to the bottom of a water tank from the unit cages placed in the middle. The speed of wakes behind inside the row in the middle was slower than that outside the row. Water velocity inside and outside a real abalone cage at Nowha Island adjacent to Wan Island was measured to verify results from the tank test. The speed of current in front of the cage by 2 m was 0.11 m/sec while it was only 0.0009 m/sec inside the cage. It had similar findings with those of a tank test.

Preparation and characteristics of Haliotis Discus Hannai Ino (abalone) viscera Jeotgal, a Korean fermented seafood (전복내장 젓갈의 제조 및 품질특성)

  • We prepared Jeotgal with Haliotis Discus Hannai Ino (abalone) viscera and the studied the physicochemical properties. Abalone viscus was fermented with varying amounts of salt for 60 days in order to prepare for the Jeotgal. During the fermentation, we measured the change of pH, volatile basic nitrogen (VBN), amino nitrogen (AN) and protease activity. After the fermentation, we examined the composition of free amino acids and sensory evaluation. The pH decreased with the fermentation, which was not significant (from 5.5 to 6.5). After the fermentation, the highest VBN was 96.7 mg/g, while the highest AN value was 406.3 mg/g. Unlike VBN and AN, the protease activity increased and reached the highest activity at the 30th day, and then decreased afterward. Based on the results, it was deduced that higher salinity restrained the fermentation and lowered the VBN, AN and protease activity. The total free amino acids of abalone Jeotgal, which were analyzed after the fermentation, (62.75 mg/g) was more than twice the amount in the abalone viscera before the fermentation (30.37 mg/g). We prepared abalone viscera Jeotgal and studied the characteristics for the first time. This will provide us with useful information for future related researches.
