To obtain direct evidence for thed involvement of C-AB13, a carrot (Daucus carota L.) homolog of VPI/Ab13, seed-specific transcription factor, in the acquisition of desiccation tolerance carrot non-embryogenic cells (NC) in which the C-AB13 gene was expressed ectopically was prepared. Non-transgenic NC, in which expression of C-AB13 was not detected, did not exhibit desiccation tolerance even after treatment with abscisic acid (ABA). In transgenic NC that expressed C-AB13, embryo-specific ABA-inducible genes (ECP genes) were expressed upon ABA-treatment. Furthermore, the transgenic NC became desiccation-tolerant upon ABA-treatment, but not tolerate desiccation without ABA-treatment. These results provide direct evidence for the involvement of C-AB13 in the ABA-induced acquisition of desiccation tolerance.

Applied Behaviour Analysis (ABA) is the applied science where strategies are derived from the principles of behaviour and are applied to improve meaningful social behaviours [3]. This study investigates the possible inclusive value of the use ABA in schools in Saudi Arabia. Interviews were conducted with two ABA therapists and a support teacher in order to address this possibility. From the research findings, it emerged how ABA is one of the cognitive-behavioural intervention models and therefore, can be generalised to other disorders or diagnoses and the applied behavioural analysis does have an inclusive value as it structures individualized activities for the increase of both personal and social skills and these activities allow the subject to work on his or her skills, which are, however, absolutely preparatory to the inclusion of the subject in the classroom context.
Effects of abscisic acid(ABA) and temperature on the anthocyanin accumulation and phenylalanine ammonia Iyase(PAL) activity were investigated in seedlings of Arabidopsis thaliana. In time course study, exogenous application of ABA $(50-1000\;{\mu}M)$ led to a noticeable increase in anthocyanin pigments which persisted over the following 5 days. Anthocyanins increased in concert with the chlorophyll loss. The activity of PAL, a key enzyme in the phenylpropanoid pathway, increased on exposure to ABA and reached maximum on the 4th day, This result shows that anthocyanin synthesis and PAL activity have a close physiological relationships. In the effects of temperatures ($10^{\circ}C,\;17^{\circ}C,\;25^{\circ}C$and $30^{\circ}C$) on anthocyanin accumulation and PAL activity in seedlings, a moderate-low temperatures ($17^{\circ}C$) enhanced both anthocyanin content and PAL activity, whereas elevated temperatures ($30^{\circ}C$) showed low levels of anthocyanin and PAL activity, suggesting a correlation between temperature-induced anthocyanin synthesis and the accumulation of PAL mRNA. Simultaneous application of ABA with temperatures Induced higher anthocyanin synthesis and PAL activity in seedlings than ABA or temperature stress alone. Moderate-low temperature with ABA exposure elicited the maximal induction of anthocyanin synthesis and PAL activity. Therefore, ABA treatment significantly increased thermotolerance in .A. thalinan seedlings. Ethephon and ABA showed similar mode of action in physiological effects on anthocyanin accumulation and PAL activity. Our data support that anthocyanins may be protective in preventing damage caused by environmental stresses and play an important role in the acquisition of freezing tolerance.
To withstand ever-changing environmental stresses, plants are equipped with phytohormone-mediated stress resistance mechanisms. Salt stress triggers abscisic acid (ABA) signaling, which enhances stress tolerance at the expense of growth. ABA is thought to inhibit the action of growth-promoting hormones, including brassinosteroids (BRs). However, the regulatory mechanisms that coordinate ABA and BR activity remain to be discovered. We noticed that ABA-treated seedlings exhibited small, round leaves and short roots, a phenotype that is characteristic of the BR signaling mutant, brassinosteroid insensitive1-9 (bri1-9). To identify genes that are antagonistically regulated by ABA and BRs, we examined published Arabidopsis microarray data sets. Of the list of genes identified, those upregulated by ABA but downregulated by BRs were enriched with a BRRE motif in their promoter sequences. After validating the microarray data using quantitative RT-PCR, we focused on RD26, which is induced by salt stress. Histochemical analysis of transgenic Arabidopsis plants expressing RD26pro:GUS revealed that the induction of GUS expression after NaCl treatment was suppressed by co-treatment with BRs, but enhanced by co-treatment with propiconazole, a BR biosynthetic inhibitor. Similarly, treatment with bikinin, an inhibitor of BIN2 kinase, not only inhibited RD26 expression, but also reduced the survival rate of the plant following exposure to salt stress. Our results suggest that ABA and BRs act antagonistically on their target genes at or after the BIN2 step in BR signaling pathways, and suggest a mechanism by which plants fine-tune their growth, particularly when stress responses and growth compete for resources.
Expression patterns of OsAREB1 revealed that expression of OsAREB1 gene can be induced by ABA, PEG and heat. Yeast one-hybrid assay demonstrated it can bind to ABA-responsive element (ABRE), which was found in most stress-induced genes. Transgenic Arabidopsis over-expressing OsAREB1 had different responses to ABA and glucose compared to wild-type plants, which suggest OsAREB1 might have a crucial role in these two signaling pathways. Further analysis indicate that OsAREB1 have multiple functions in Arabidopsis. First, OsAREB1 transgenic plants had higher resistance to drought and heat, and OsAREB1 up-regulated the ABA/stress related gene such as RD29A and RD29B. Second, it delayed plant flowering time by down-regulating the expression of flowering-related genes, such as FT, SOC1, LFY and AP1. Due to the dates, OsAREB1 may function as a positive regulator in drought/heat stresses response, but a negative regulator in flowering time in Arabidopsis.
To examine the relationship between plant hormones and the development of pepper anthracnose, we investigated the effects of several plant hormones on the progression of disease symptoms. Of the five plant hormones examined, abscisic acid (ABA) increased the lesion length and disease incidence on detached fruits of Capsicum annuum cv. Nokkwang. The simultaneous application of ABA with inoculation of Colletotrichum acutatum JC24 resulted in increased lesion length, depending the concentration of ABA applied. Additionally, application of ABA caused the development of pepper anthracnose in fruits of Capsicum baccatum cvs. PBC80 and PBC81, which were previously resistant to the disease. Furthermore, ABA administration rendered increased pathogenicity of other isolates of C. acutatum BAC02063, PECH10, and TCBNU3 obtained from the Chinese matrimony vine, peach, and tea tree, respectively. Our data suggest that exogenous ABA may result in the suppression of defense mechanisms of pepper fruits against anthracnose, which leads to a change in the susceptibility of pepper fruits and the development of pepper anthracnose.
This experiment was carried to investigate the optimum time and times of abscisic acid (ABA) treatment for the coloration in 'Kyoho' grapes. The L-phenylalanine ammonia-lyase (PAL) enzyme activities were showed highly in both ABA treatments at veraison and 10 days after veraison. However, PAL enzyme was slightly higher in ABA treatment at 10 days after veraison than in that at veraison. Anthocyanin content showed a tendency that were increased during fruit development after veraison in all treatments, and was the highest in ABA treatment at 10 days after veraison. Fructose and glucose as soluble sugars were detected by HPLC and showed little differences in all treatments. In times of ABA treatment, PAL activity showed a tendency that decreased after increased in all treatments. PAL activity in 2 times treatment of ABA was higher than other treatments. Also, anthocyanin content was highest in 2 times treatment of ABA, as 5 folds of control and folds of 1 time treatment of ABA. Both fructose and glucose contents in all ABA treatments was slightly higher than control.

The seeds of Malus sieboldii were collected in late fall of 1988 and stored in cold stratification condition for 15 weeks. Growth regulators such as $GA_3$, BA and ABA were treated to the seed to know germination capacity and seedling growth. The results were as follows. 1) Higher germination was achieved in $GA_3$ and BA plots, But ABA treatment showed lower germination percentage and physiological dwarf and also induced to the secondary dormancy condition in partly. 2) Average length of time for germination was delayed over 2 days in ABA plot when comparing with the other plots. 3) Average length of plumule and radicle also was inhibited by ABA treatment. This means lower seedling vigor. 4) Fresh and dry weight of the seeding were higher in BA plot while ABA plot was lower and undesirable seedling was produced. From above mentioned results, it is believed that ABA treatment to the seed induced the secondary dormancy and physiological dwarf pattern. Therefore, utilization of BA can be produced the normal seedling, the authors believed.
The CYP707A family genes encoding ABA 8'-hydroxylase catabolize abscisic acid (ABA), a plant stress hormone that plays an important role in stress condition, such as drought, heat, cold and salinity. Phaseic acid (PA) is a catabolic product of ABA. Recent studies have shown that PA is important for the physiological functions in plants. It is also a neuroprotective molecule that protects against ischemic brain injury in mice. To obtain enzymes for the PA production, four CaCYP707A genes (CaCYP707A1, CaCYP707A2, CaCYP707A3 and CaCYP707A4) were isolated from hot pepper. They were heterologously expressed in Escherichia coli. Among them, CaCYP707A2 showed significantly higher expression levels in both the membrane fraction and the soluble fraction. Preferred redox partners were investigated to improve the efficiency of CaCYP707A2's catalytic reaction, and NADPH-cytochrome P450 reductase (CPR) from hot pepper (CaCPR) was preferred over other redox partners (i.e., rat CPR and ferredoxin reductase/ferredoxin). The production of 8'-hydroxy ABA and PA by ABA hydroxylation activity was confirmed in CaCYP707A2 from both membrane and soluble fractions. Therefore, CaCYP707A2 is the first identified plant CYP protein that is expressed a soluble form in cytosolic fraction having stable activity. Taken together, we propose a new CYP707A protein with industrial applications for PA production without additional modifications in E. coli heterologous expression.
A new method for micro-quantification of abscisic acid from plant tissue is described. GLC-ECD method of abscisic acid pertafluorobenzyl ester synthesized by reaction of plant extract with pentafluorobenzyl bromide showed higher sensitivity than that of abscisic acid methyl ester, and the detection limit of abscisic acid by the described method was as low as 5 pg.
