고형상의 모델시스템에 있어서 리놀레산의 산화에 미치는 리폭시게나아제, 카로틴, 토코페롤 및 수분활성의 영향 (Effect of Lipoxygenase, ${\beta}-Carotene$, ${\alpha}-Tocopherol$ and Water Activity on the Oxidation of Linoleic Acid in Starch-Solid Model System)

  • 고형상의 모델시스템에서 리놀레산의 산화에 영향을 미치는 여러가지 인자들 즉 리폭시게나아제, 수분활성도, ${\beta}$-카로틴, ${\alpha}$-토코페롤등에 대해 관찰하여 다음과 같은 결과를 얻었다. 리폭시게나아제에 의한 리놀레산의 산화반응은 aw 0.72에서 aw 0.25에 보다 잘 일어났다. 그러나 이 시스템에 ${\beta}$-카로틴이나 ${\alpha}$-토코페롤을 반응시키게 되면 리놀레산의 산화를 크게 저해하였으며 ${\beta}$-카로틴보다는 ${\alpha}$-토코페롤이 지질과 산화 과정에서 항산화작용이 더 컸다. 즉, 유사한 조건에서 ${\beta}$-카로틴은 24시간의 유도기간을 보여 준 반면 ${\alpha}$-토코페롤은 72시간까지도 유도기간이 지속되었다. ${\beta}$-카로틴의 항산화작용은 수분활성도의 영향을 크게 받았으며 수분활성도가 높을수록 항산화 작용이 뛰어났고 또한 ${\beta}$-카로틴의 농도가 높을수록 CDA 생성이 적었다.

Effects of Vitamins E and C on Human BreastCancer Cell Growth in the Presence of Various Fatty Acids

  • To investigate the effects of antioxidative vitamins in combination with various fatty acids on breast cancer cell proliferation, MDA-MB231 human breast cancer cells were cultured for 3 days in the serum-free Iscove's modified Dulbecco's medium (IMDM) supplemented with 1.25mg/ml delipidized bovine serum albumin and 10㎍/ml insulin. Alpha-tocopherol, ascorbic acid or both vitamins were added to the medium at the concentrations of 10 and 50μM in the presence of 3μg/ml of oletic(Oa), linoleic(LA) α-linoleinic(LNA) and docosahexaenoic acid(DHA). Cell growth was reduced significantly by α-tocopherol in a dose-dependent manner, but not affected by ascorbic aicd. The four different fatty acids did not have significant effects on cell growth, although DHA exerted inhibitory effect on the growth after 1 day. However, the each fatty acid was well incorporated into celluar lipid as such or elongated forms. Addition of α-tocopherol remarkably increased its celluar contents and reduced cellular levels of thiobarbituric acid substances (TBARS) that were elevated notably in the presence of DHA in the culture media. But ascorbic acid addition did not change much of either cellular α-tocopherol or TBARS contents. northern blot hybridization showed that tumor supressor gene ρ53 was most highly expressed by the combination of ρ-tocopherol and DHA in 8 hours of cell culture. In conclusion , the growth inhibitory effect of vitamin E suggests that breast cancer cell proliferation is reduced by the mechanism other than cytotoxicity of lipid peroxide and it is related to expressionof tumor supprosser gene p53, that can be increased by both vitamin E and n-3 fatty acid, DHA.

Retinol, α-tocopherol, and selected minerals in breast milk of lactating women with full-term infants in South Korea

  • BACKGROUND/OBJECTIVES: This study was performed to measure fat-soluble vitamins and minerals in breast milk of Korean lactating mothers who exclusively breastfed their babies. SUBJECTS/METHODS: Breast milk samples were collected from 334 mothers. Concentrations of retinol and ${\alpha}$-tocopherol were analyzed by high performance liquid chromatography ultraviolet spectrometry while concentrations of minerals were measured by inductively coupled plasma optical emission spectrometry. RESULTS: Retinol and ${\alpha}$-tocopherol contents of breast milk were $39.58{\pm}19.64{\mu}g/dL$ and $0.23{\pm}0.13mg/dL$, respectively. Average sodium, potassium, calcium, phosphorus, and magnesium levels in breast milk were $11.11{\pm}5.16$, $38.56{\pm}9.01$, $27.87{\pm}6.10$, $13.56{\pm}3.30$, and $3.05{\pm}0.65mg/dL$, respectively. Contents of trace elements such as iron, zinc, copper, and manganese were $40.26{\pm}46.21$, $98.40{\pm}62.47$, $24.09{\pm}9.03$, and $0.90{\pm}1.63{\mu}g/dL$, respectively. Fat-soluble vitamin concentration was positively correlated with total fat in milk samples, but no significant differences were observed in levels of retinol, ${\alpha}$-tocopherol, or minerals based on whether or not lactating women were taking dietary supplements. CONCLUSIONS: Micronutrient contents of breast milk samples from Korean lactating women were comparable to those of other nations. Retinol and ${\alpha}$-tocopherol levels were correlated and also with total fat in breast milk.

벼 품종별 미강의 Tocopherol과 Tocotrienol 함량 평가 (Genotypic Difference in Tocopherol and Tocotrienol Contents of Rice Bran)

  • 우리나라에서 생산된 벼 18품종의 미강을 품종별로 tocopherol, tocotrienol, vitamin E 함량을 분석한 결과는 다음과 같다. 1. 미강 중 tocopherol 함량은 9.12-14.76 범위에 평균은 11.14 mg/100g이었고 tocotrienol은 22.39-37.07 평균 28.03 mg/100g 이었으며, vitamin E 함량은 34.86-46.50으로 평균 39.17 mg/100g이었다. 2. tocopherol함량이 높은 품종은 서진벼 화성벼 추청벼 중안벼 등이고, tocotrienol 함량이 높은 품종은 안다 다마금 대립벼1호 진품벼 중안벼 서진벼 고시히카리 대안벼 봉광벼 등이며, vitamin E 함량이 높은 품종은 안다벼 서진벼 다마금 중안벼 고시히카리 등이었다. 3. 미강의 tocopherol(T) 및 tocotrienol($\textrm{T}_3$) 동족체함량은 $\delta\textrm{-}$T<$\beta\textrm{-}$$\textrm{T}_3$$\beta\textrm{-}$T<$\delta\textrm{-}$$\textrm{T}_3$$\gamma\textrm{-}$T<$\alpha\textrm{-}$T<$\alpha\textrm{-}$$\textrm{T}_3$$\gamma\textrm{-}$$\textrm{T}_3$ 순으로 높았다. 특히 $\gamma\textrm{-}$$\textrm{T}_3$ 의 함량은 $\alpha\textrm{-}$, $\beta\textrm{-}$, $\gamma\textrm{-}$, $\delta\textrm{-}$T의 합 보다 높았고 이러한 현상은 각각의 모든 품종에서도 같은 경향이었다. 4. 항산화력이 높고 유방암 항암에 효과가 뛰어난 $\gamma\textrm{-}$$\textrm{T}_3$ 는 평균 17.92 mg/100g으로 총 tocotrienol 중 63.9%, $\delta\textrm{-}$$\textrm{T}_3$ 는 0.88 mg/100g으로 3.1%이었고, $\gamma\textrm{-}$$\textrm{T}_3$ 함량은 안다벼가 33.44mg/100g으로 가장 높았으며 다마금 대립벼1호 고시히카리 중 안벼 대안벼 서진벼 진품벼 등의 품종에서 높았다.

Serum Concentrations of $\alpha$-Tocopherol, Carotenoids and Retinol of Normal Koreans

  • Five hundred and seventy-eight healthy subjects (351 men and 227 women) with a mean age of 44.8 years (45.2 for men, 44.3 for women) participated in this study. The serum concentrations of $\alpha$-tocopherol, \beta$-carotene, $\alpha$-carotene, zeaxanthin + lutein, cryptoxanthin, lycopene and retinol of normal Koreans were measured, and their relation to gender, age, BMI, alcohol consumption, cigarette smoking, and menopausal status were evaluated. The concentrations of $\alpha$-tocopherol, carotenoids and retinol in serum were measured simultaneously by reverse phase HPLC (multi-wavelength, gradient and computerized automatic system). Average serum levels of $\alpha$-tocopherol, \beta$-carotene and retinol were 10.1$\pm$0.41\mu$g/dl, 33.1$\pm$1.24\mu$g/dl, and 82.0$\pm$1.63\mu$g/dl for men and 11.1$\pm$0.74\mu$g/ml, 48.1$\pm$1.60\mu$g/dl and 64.5$\pm$1.96\mu$g/dl for women, respectively, Serum concentrations of $\alpha$-tocopheol \beta$-carotene, $\alpha$-carotene, cryptoxanthin and lycopene appeared to be higher in women than in men. The serum concentrations of zeaxanthin + lutein increased with the increase in age of men and those of $\alpha$-carotene and \beta$-carotene increased with the increase in age of women. For men, current smokers showed significantly lower serum concentrations of \beta$-carotene and $\alpha$-carotene than ex-smokers (p <0.05) . Current drinkers also showed significantly lower serum \beta$-carotene and zeaxanthin + lutein concentrations than ex-drinkers. For women, current smokers showed significantly love. serum concentrations of zeaxanthin+lutein and cryptoxanthin than ex-smokes (p<0.05). Men with BMI$\geq$24 showed significantly lower serum concentrations of \beta$-carotene, $\alpha$-carotene, lycopene and cryptoxanthin than men with BMI < 20. The mean concentrations of $\alpha$-tocopherol, \beta$-carotene, $\alpha$-carotene and lycopene far postmenopausal women were higher than those for premenopausal women (p<0.05). In conclusion, there were obvious differences in serum $\alpha$-tocopherol and carotenoids levels depending on gender, age, cigarette-smoking habits, alcohol consumption, BMI and menopausal status. further studies are required to establish the normal levels of these vitamins for children and the elderly and to elucidate their roles in different disease states.

Effects of the P/S Ratio of Dietary Lipids and Antioxidant Vitamin Supplements on the Level of Serum Lipids and Liver. Lipid Peroxidation in Rats Treated with DMBA

  • This study an analyzes the effects of the P/S ratio of dietary lipids and antioxidant vitamin supplements on serum lipids level and fatty acid profile, the degree of lipid peroxidation, and the antioxidant enzyme activities in the liver of rats treated with 7,12-dimethylbenz($\alpha$) anthracene(DMBA). P/S ratio of dietary lipids was made into 0.5, 1 and 2 by mixing palm oil, soybean oil, sesame oil and perilla oil at 10%(w/w) fat level and n-6/n-3 ratio was fixed to 4. Antioxidant vitamin of $\alpha$-tocopherol or $\beta$-carotene was supplemented in addition to vitamin mixture which was given at 1 % of the standard diet. female Sprague-Dawley strain rats, about 60 days old, were divided into three groups(LP : low P/S ratio(0.5), MP : medium P/S ratio (1.0), HP , high P/S ratio(2.0)) and each group was sub-divided into three groups(S ; standard, T ; tocopherol supplemented, C : carotene supplemented): Two weeks after feeding experimental diets, all groups were treated with a single dose of DMBA(2mg/100g BW) by gastric intubation and fed experimental diet for 9 week. The results were as follows ; 1) Serum total cholesterol(TC) level was not significantly influenced by diet but tended to be lower in HP groups compared to LP and MP groups. Triglyceride level was the highest in LP groups and the lowest in $\alpha$-tocopherol supplemented groups. 2) Thiobarbituric acid reactive substance(TBARS) level, representing lipid peroxidation in hepatic microsome, tended to be increased as the unsaturation of dietary lipids increases. $\alpha$-Tocopherol supplement significantly decreased TBARS level. 3) The activities of superoxide dismutase(SOD) and glutathione peroxidase(GSHPx) in hepatic cytosol showed the tendency to be high with increasing P/S ratio of dietary lipids. SOD activity was not significantly influenced by antioxidant vitamin, but GSHPx activity was significantly increased in $\alpha$-tocopherol supplemented groups. In summary, high polyunsaturated fat diet was effective on reducing the serum level of total cholesterol and triglyceride, while it increased unsaturation and peroxidizability of serum fatty acid. With increasing P/S ratio of dietary lipids, lipid peroxidation was increased in the liver and antioxidant enzyme system was induced to inhibit lipid peroxidation against oxidative damage. $\alpha$-Tocopherol supplement was effective in lowering lipid peoxidation, but $\beta$-carotene supplement did not exhibit antioxidant effect. 들깨의 볶음 조건에 따른 들기름의 성분 및 관능적 특성 변화 (Changes in components and sensory attribute of the oil extracted from perilla seed roasted at different roasting conditions)

  • 들깨의 볶음 조건에 따라 착유한 들기름의 성분 변화 및 관능적 특성을 조사하였다. 이때 들깨의 볶음 온도는 150, 170, 190, $210^{\circ}C$로 볶음 시간은 10, 20, 30분으로 하였다. 볶음 조건에 따라 볶은 들깨로부터 압착법으로 들기름을 착유시 볶음 온도가 높고 볶음 시간이 길어질수록 수율은 증가하였다. 볶음 조건에 따라 볶아 착유한 들기름중의 지방산 조성은 차이가 없었으며 linolenic acid가 약 60%로 가장 주된 지방산이었고 불포화 지방산이 전체 지방산의 88% 이상이었다. 들기름중의 인 함량은 들깨의 볶음 온도가 높아지고 볶음 시간이 길어질수록 감소하는 경향을 나타냈으며 특히 $190^{\circ}C$에서 30분, $210^{\circ}C$에서 $20{\sim}30$분 볶아 착유한 경우에는 급격히 감소하였다. 한편, 들기름중의 tocopherol 함량은 들깨의 볶음 조건에 의해 영향을 받지 않는 것으로 나타났으며 v-tocopherol이 총tocopherol 함량의 약 92%로 가장 주된 tocopherol이었다. 들기름의 관능검사 결과 들깨의 볶음 조건에 의해 들기름의 향미, 맛, 색깔, 종합적 기호도등이 1% 이내의 유의차를 보였으며 $190^{\circ}C$에서 20분 볶아 착유한 들기름이 기호도에서 가장 우수한 것으로 나타났다.

천연항산화제가 분쇄돈육의 지질산화에 미치는 효과 (Effects of Natural Antioxidants on Lipid Oxidation of Ground Pork)

  • 천연항산화제의 실용화를 위한 기초자료로서 천연항산화제 상호간의 항산화력 차이를 구명하고자 분쇄돈육에 ${\alpha}-tocopherol$, GFSE, carnosine, rosemary 등 천연항산화제를 첨가하여 비가열 또는 가열처리하여 $4^{\circ}C$에서 10일간 저장하였다. 저장기간에 따른 지질의 산화 및 지방산 조성의 변화에 대한 연구결과를 요약하면 비가열 분쇄돈육을 냉장온도에서 저장할 때 육중의 지질 산화에 대해 carnosine > rosemary > ${\alpha}-tocopherol$ > GFSE의 순으로 강력한 항산화력을 나타내었다. Carnosine은 육의 pH를 상승시키는 결과를 가져왔으나, 이는 육의 가공시 유리한 성질로 작용할 수도 있어 큰 문제점은 되지 않으리라 사료된다. 또한, 가열 분쇄돈육을 냉장온도에서 저장할 때는 carnosine >rosemary > ${\alpha}-tocopherol$ > GFSE의 순으로 강력한 항산화력을 나타내었고, 열처리로 인해서 항산화력을 상실하지는 않았으나 생육에서 보다는 지방산화를 효과적으로 방지하지는 못하였다. Carnosine은 가열육에서도 pH를 상승시키는 결과를 가져왔다.

Distribution of injected fat-soluble vitamins in plasma and tissues of nursery pigs

  • Objective: The objective of this experiment was to investigate the effects of fat-soluble vitamin injection on plasma and tissue vitamin status in nursery pigs. Methods: A total of 16 pigs (initial body weight: 7.15±1.1 kg) were allotted to 2 treatments at d 7 post-weaning. Pigs were fed a corn-soybean meal-based basal diet with no supplemental vitamin A and i.m. injected with 300,000 IU of retinyl palmitate, 900 IU of d-α-tocopherol and 30,000 IU of vitamin D3 with control pigs having no vitamin injection. Blood (d 0, 3, 7, and 14 post-injection) and tissue samples (liver, brain, heart, lung, and muscle; d 7 and 14 post-injection) were collected from pigs. Retinyl palmitate, retinol, and α-tocopherol concentrations were analyzed in plasma and tissues, while plasma was assayed for 25-hydroxycholecalciferol (25-OHD3). Results: Plasma retinol and 25-OHD3 concentrations increased by the vitamin injection from d 3 to 14 post-injection (p<0.05) whereas plasma retinyl palmitate was detected only in the vitamin treatment at d 3 and 7 post-injection (115.51 and 4.97 ㎍/mL, respectively). Liver retinol, retinyl palmitate, and retinol+retinyl palmitate concentrations increased by retinyl palmitate injection at d 7 and 14 post-injection (p<0.05) whereas those were not detected in the other tissues. The d-α-tocopherol injection increased α-tocopherol concentrations in plasma at d 3 and 7 post-injection (p<0.05) and in liver, heart (p<0.10), and muscle (p<0.05) at d 7 post-injection. Conclusion: Fat-soluble vitamin injection increased plasma status of α-tocopherol, retinol, retinyl palmitate and 25-OHD3. As plasma levels decreased post-injection, vitamin A level in liver and vitamin E level in muscle, heart and liver increased. The α-tocopherol found in plasma after injection was distributed to various tissues but retinyl palmitate only to the liver.
