Studies on the Amylase Producing Bacteria. (partI) (Amylase 생성세균에 관한 연구 1)

  • 1. Three hundred and twenty four strains of amylase producing bacteria were isolated from various sources and a high amylase producing new strain, which was isolated from MEJU, M-181, was selected for further investigations. 2. The new strain M-181 was similar to Bacillus subtilis in the characteristics. 3. Wheat bran medium was the best one for production of amylase so for as the investigations had been done. The amylase activity of M-181 was measured D$40^{\circ}\\30^{'}$ .$deg._{30'}$ 25,000 to 26,800 on the medium of wheat bran. 4. The strain M-181 did not demand phosphate for production of amylase.

Effect of Rice Addition on Enzyme Activities of Soybean Meju Fermented by Monascus spp. (Monascus spp.를 이용한 콩 메주의 효소활성에 미치는 쌀 첨가효과)

  • This study was conducted to investigate the effect of rice powder m tile enzyme (protease, $\alpha$-amylase, $\beta$-amylase and glucoamylase) activities of soybean meju fermented by Monascus prepureus and Monascus pilosus. The activities of the enzyme in the rice meju and the soybean meju fermented by M. pilosus were higher than those by M. perpureus. Protease activity of powdered rice meju was higher than that of granular rice meju, while $\alpha$-amylase, $\beta$-amylase and glucoamylase activities were higher in granular rice meju. Protease activity in soybean meju fermented by adding of the cultured medium of Monascus strains(CMM) as a seed inocula were higher than those of the rice powder meju, while $\alpha$-amyulase, $\beta$-amylase and glucoamylase activiities were lower than those of soybean meju by CMM. The concentration of rice powder to show maximum protease activity in soybean meju was also 10% against steamed soybean. But $\alpha$-amylase activity of soybean medju by the CMM added 2% powdered rice showed lower but the activity increaed with an increase in powdered rice, whereas $\beta$-amylase and glucoamylase activiities decreased with an increase in powdered rice. Protease activity of soybean meju fermented by 10% rice meju fermented by M. pilo년 as a seed inocula was higher than that of the meju fermented by Aspergillus oryzae, whereas $\beta$-amylase and glucoamylase activities of the soybean meju showed less than 50%.

  • The amylase was0 incubated in a 9mm hole on the starch agar gel plate bored with a cork borer. When 0.1N-iodine solution sprayed on the plate, the formed uncolored zone were observed. An activity of amylase has been determined by measurement of diameter of the uncolored zone. We named this method 'cork borer method'. When amylase was incubated on the starch agar gel plate, the following points were obtained. 1) The optimum pH for the formation of the zone in case of amylases(Biotex, Spitase) which produced by Bacillus is neutrality and alkali, while that for Glucoamylase, Biodiase, and Mucorrennin which produced by Rhizopus and Mucor is from 5 to 7. 2) The diameter of the zone was increased by the incubation time and amylase activity. 3) The zone was easily formed at low level of starch concentration and was formed much bigger than at $50^{\circ}C\;than\;at\;10^{\circ}C$. From the above results, after malting the starch agar gel plate, keeping constant concentration of the starch, the measurement of amylase activity is in efficiency upon the constant reaction time and temperature.

Characteristics of Traditional Mejus of Nation-Wide Collection (전국적으로 수집한 전통식 메주의 특성 조사)

  • Typical characteristics of Mejus must be understood to get the basic data for setting up mass production system of traditional fermented soybean products. One hundred and twenty one Mejus were collected from various places and analyed. Most of shapes were rectangular and some were spherical, conical, cylindrical and doughnut types. The weight of Mejus was 0.4~4.2kg. Chemical analysis showed: moisture content, 9.73~58.22% ; pH, 4.95~8.15; acidity, 0.6~3.8% ; soluble protein content, 4.45~12.31%; soluble sugar content, 0.82~10.95%. Enzyme assay showed: $\alpha$-amylase activity, 5.0~874.2 units/g; $\beta$-amylase activity, 0.02~27.74units/g; acidic protease activity, 31.3~225.1unts/g; lipase activity 1.0~53.0units/g. Total viable cells were 3.72$\times$107~1.35$\times$1010cfu/g, and yeast and mold count 6.46$\times$104~8.91$\times$106cfu/g. respectively. $\alpha$-Amylase activity of a traditional Meju from Incheon showed the highest activity of 732.8 units/g(interior section) and 823.2units/g (exterior section). $\beta$-Amylase activity was the highest{3.57 units/g (interior sectin) and 4.25units/g (exterior section)} in Meju from Chunbuk. Acidic protease activity was the highest in sample from Seoul, whereas traditional Meju from Kyongnam showed the highest activity of 21.5units/g(interior section) and 37.5units/g(exterior sectin).

  • This study was carried out to elucidate the seasonal changes of amylase an nitrate reductase activities in Zoysia japonica Steud. The seasonal changes of the amylase activity AA) in aboveground parts appeared high activity from the beginning of May to the beginning of July and in autumn showed an activity of rapid decreasing tendency. But, the AA in winter(Nov. 15~Feb 28 was not almost detected. The seasonal changes of nitrate reductase activity(NRA) in various organs appeared a very similar figures to the seasonal changes of AA. But, the NRA in the root was not almost detected.

Dietary Exogenous α-Amylase Modulates the Nutrient Digestibility, Digestive Enzyme Activity, Growth-Related Gene Expression, and Diet Degradation Rate of Olive Flounder (Paralichthys olivaceus)

  • In this study, a 12-week feeding experiment was conducted to characterize the effects of exogenous α-amylase on the growth, feed utilization, digestibility, plasma α-amylase activity, feed degradation rate, and fecal particle size of olive flounder (Paralichthys olivaceus). Diet was supplemented with 0 (AA0; control), 100 (AA100), 200 (AA200), or 400 (AA400) mg/kg of α-amylase, respectively. Fish (273.1 ± 2.3 g) were stocked into 12 tanks (25 fish/1,000-L tank) and 3 tanks were randomly selected for each diet group. As a result, α-amylase was found to have no significant effects (p ≥ 0.05) on the growth, feed utilization parameters, and whole-body proximate compositions. α-Amylase-treated fish exhibited only a significant increase in the apparent digestibility coefficient of carbohydrates compared to the controls. In addition, in vitro analyses revealed that α-amylase dose-dependently increased (p < 0.05) the feed degradation rate, while photographs of the intestinal content after 2, 4, and 8 h of feeding demonstrated an improved degradation rate in the α-amylase-treated groups. Plasma α-amylase content was higher in the AA200 and AA400 groups, whereas the control group produced significantly larger-sized fecal particles (90% size class) than these two groups. In the intestine, no changes were observed in the expression levels of the immune-related TNF-α, IL-1β, IL-2, immunoglobulin-M, HSP-70, lysozyme, and amylase alpha-2A. However, growth-related genes IGF-1, IGF-2, TGF-β3, and growth hormone genes were upregulated in muscle tissues. Collectively, exogenous α-amylase has positive roles in the modulation of the digestibility coefficient, blood α-amylase concentration, growth-related gene expression, and diet degradation for improved digestion in olive flounder.

  • 1. Amylase producing bacteria have been isolated from natural sources and was identified out. The above strain is identical to Bacillus subtillis in every details of physiological and morphorogical characteristics by this investigations. 2. Sikworm chrysalis nad chlorella extracts are prove to be further effective for the stimulation of amylase production than any other sources. The former silkworm extract is more stronger in its activity. 3. It is found that effective results had been accepted by a less soybean meal addition per that of wheat bran in the mixture ratio of them, also less addition of corn is needed in the mixture ratio of between corn and whear bran. In the mixing ratio of wheat bran per silkworm or chlorella extracts, the addition of wheat bran is necessary for stimulation of amylase production. 4. Fro the stimulation of amylase production, the effective substances from natural sources and its order of activity in cultral media cultral media is proved as follows, silkworms ext.>chlorella ext.>alcohol>skim-milk and peptone>fermented milk>radish leaf ext.

Construction of a Secretory Expression Vector Producing an $\alpha$-Amylase of Yeast, Schwanniomyces occidentalis in Saccharomyces

  • Using a modified yeast secretory expression vector, $\alpha$-amylase of Schwanniomyces occidentalis was produced from Saccharomyces cerevisiae. The expression vector contains the a-amylase gene (AMY) harboring its own promoter without the regulatory region and the adenine base at the -3 position from the ATG start codon, its own signal sequence, CYC1 transcription terminator, and SV40 enhancer. The expressed $\alpha$-amylase activity from cells carrying the plasmid was approximately 26 times higher than that from the cells harboring an unmodified plasmid. When Saccharomyces diastaticus was transformed with this modified vector, a 2.5 times higher level of amylolytic activity than that from Sch. occidentalis was observed.

Production of Amylase by a Filamentous Fungus, Strain FM04, and Enzymatic Hydrolysis of Food Waste (사상균 FM04에 의한 Amylase 생산 및 음식물 쓰레기의 효소학적 가수분해)

  • A filamentous fungus, strain FM04 producing amylase was isolated from rotten yam peels and potatoes. The favorable conditions of cultivation factors such as, temperature, pH, and agitation speed of strain FM04 were 28∼30$^{\circ}C$, 5.0∼6.0, and 100 rpm, respectively. Starch was the best carbon source in the amylase production. Therefore, food wastes containing lots of starch were employed as the carbon source of the cultivation for the economical amylase production. 5.2 U/ml of amylase was obtained In the cultivation using 1 % (w/v) of food wastes. The amylase showed the highest activity at enzyme reaction conditions of 60$^{\circ}C$ and pH 4.5 and showed 90% of residual activity after the reaction at 50$^{\circ}C$ for 2 days. In the enzymatic hydrolysis reaction using 20% (w/v) of food wastes and 2.5 U/ml of amylase, 72.6 g/l of reducing sugar was obtained at the reaction condition of 50$^{\circ}C$, pH 4.5 for 2 days.

Effects of Dimethipin on α-amylase Activity of Barley Seeds (보리 종자의 α-아밀라아제 활성에 미치는 Dimethipin의 영향)

  • Effects of dimethipin on ${\alpha}$-amylase activity of barley seeds were investigated. In the treatments of $1{\mu}M\;and\;10{\mu}M$ dimethipin, the indexes of germination were reduced to 17% and 24 % respectively. After seed germination, dimethipin was added to germinated seedlings and then the seedlings were kept to measure seedling length under illumination for 7 days. In control, the length of seedling was 5.7 cm, but in the treatments of $1{\mu}M$ dimethipin and $10{\mu}M$ dimethipin, seedling lengths were 5.5 cm and 1.2 cm respectively. In the relationship between dimethipin concentrations and ${\alpha}$-amylase activities, there was a linear curve. The more dimethipin was added to the seeds, the more ${\alpha}$-amylase activities were inhibited. In the treatments of $1{\mu}M$ dimethipin and $10{\mu}M$ dimethipin, ${\alpha}$-amylase activities were reduced to 33% and 71% respectively. Dimethipin also inhibited ${\alpha}$-amylase activities increased by gibberellin and the content of soluble protein. Therefore, it could be suggested that dimethipin might inhibit directly the activities of hydrolysis enzymes including ${\alpha}$-amylase or the expression of ${\alpha}$-amylase genes as germination and seedling growth were severely disturbed.
