Acinetobacter calcoaceticus에 의한 유지와 탄화수소의 분해 (Degradation of Fats, Oils and Hydrocarbons by Acinetobacter calcoaceticus)

  • 유지와 탄화수소를 탄소원으로 하여 Acinetobacter calcoaceticus를 배양하여 탄소원의 분해와 균체의 생육정도를 측정하고 활성오니법을 이용한 폐수정화에 이 균주의 이용가능성을 검토하였다. 실험에 사용된 모든 유지와 탄화수소가 분해되었다. 삼각플라스크에 의한 진탕배양의 경우 분해율은 어유가 26로 식물성 유지의 40-54에 비하여 분해율이 낮게 나타났다. 포화지방산은 어유 중에서 약 55, 식물성유지 중에서는 6-12 각각 함유되어 있었다. Hexadecane의 분해율은 약 60로 식물성유지에 비하여 높게 나타났다. 탄소원의 분해에 따라 균체량의 증가가 관찰되었고, jar fermentor 배양에 의하면 모든 탄소원이 80 이상 분해되었고 균체수율도 1.00% 내외로 개선되었다.배양조건에 따라 탄소원의 분해율에 영향을 받는다고 볼 수 있으며, 이는 초기 기질농도가 기질 분해율에 영향을 준 결과에서도 증명되었다. 유지 또는 탄화수소를 3% 함유한 인공폐수를 A. calcoaceticus로 우선 분해시키고, 그 분해산물을 활성오니법에 의하여 정화를 시도한 결과 탄소원의 농도는 0.06%이하로, 부유물질농도는 53mg/ml 이하로 각각 저하되어 이 균주를 폐수정화에 이용할 수 있음을 시사하였다.

Amino acid, fatty acid, and carbohydrate metabolomic profiles with ginsenoside-induced insecticidal efficacy against Ostrinia furnacalis (Guenee)

  • Background: Previous studies have shown the insecticidal efficacy of ginsenosides. In the present study, we aimed to investigate the metabolic mechanism related to the inhibitory effect of panaxadiol saponins (PDSs) against the Asian corn borer Ostrinia furnacalis (Guenee). Methods: Third instar larvae of O. furnacalis were fed normal diets with different concentrations of PDSs for 4 days. The consumption index, relative growth rate, approximate digestibility, and conversion of ingested and digested food were recorded. A targeted gas chromatographye-mass spectrometry assay was performed to detect the profiles of amino acids, fatty acids, and carbohydrates in larvae of O. furnacalis. In addition, the activity of detoxification-related enzymes was determined. Results and Conclusions: PDSs decreased the consumption index, relative growth rate, approximate digestibility, and conversion of ingested and digested food in the 3rd instar larvae of O. furnacalis in a dose-dependent manner. PDSs decreased 15 free amino acids, 16 free fatty acids, and 5 carbohydrates and increased the levels of palmitoleic acid, palmitic acid, and 9-octadecenoic acid in the 3rd instar larvae. The activity of detoxification-related enzymes, such as acetylcholinesterase, glutathione S-transferase, cytochrome P450, carboxylesterase, trehalase, acid phosphatase, and alkaline phosphatase, was reduced in a dose-dependent manner in the 3rd instar larvae exposed to PDSs. These data confirmed the inhibitory effect of PDSs against growth, food utilization, and detoxification in the 3rd instar larvae of O. furnacalis and the potential for using PDSs as an efficient tool for insect pest management for O. furnacalis larvae.

Nutritional Intervention for a Patient With Sleeve Gastrectomy

  • Bariatric surgery is the most effective treatment for sustained weight reduction, and it can result in substantial improvements in the severity of type 2 diabetes, metabolic syndrome, nonalcoholic fatty liver disease, and quality of life. However, sleeve gastrectomy, a weight loss surgery that removes two-thirds of the stomach, reduces appetite and nutrient absorption, impairing digestion and the absorption of nutrients like iron, vitamin B12, and protein-bound nutrients. This case study aims to demonstrate that patients undergoing sleeve gastrectomy require long-term and periodic monitoring of biochemical data, weight changes, and caloric and protein intake by a professional nutritionist to prevent malnutrition and nutritional deficiencies. In this case study, a 48-year-old woman was diagnosed with morbid obesity, hypertension, sleep apnea syndrome, and chronic gastritis. At initial evaluation, she was 160 cm tall and weighed 89 kg, with a body mass index of 34.8 kg/m2. At 1 postoperative year, she consumed 650 kcal and 25 g of protein per day, the percentage of excess weight loss was 141.1%, and body mass index was 21 kg/m2. Compared to preoperative levels, calcium and folic acid levels did not decrease after 1 postoperative year, but hemoglobin, ferritin, and vitamin B12 levels decreased. In conclusion, when patients experience rapid weight loss after sleeve gastrectomy, follow-up should be frequent and long. Dietary education should be conducted according to digestive symptoms, and oral nutritional supplements, including vitamins and minerals.

Metabolite profiling of fermented ginseng extracts by gas chromatography mass spectrometry

  • Background: Ginseng contains many small metabolites such as amino acids, fatty acids, carbohydrates, and ginsenosides. However, little is known about the relationships between microorganisms and metabolites during the entire ginseng fermentation process. We investigated metabolic changes during ginseng fermentation according to the inoculation of food-compatible microorganisms. Methods: Gas chromatography mass spectrometry (GC-MS) datasets coupled with the multivariate statistical method for the purpose of latent-information extraction and sample classification were used for the evaluation of ginseng fermentation. Four different starter cultures (Saccharomyces bayanus, Bacillus subtilis, Lactobacillus plantarum, and Leuconostoc mesenteroide) were used for the ginseng extract fermentation. Results: The principal component analysis score plot and heat map showed a clear separation between ginseng extracts fermented with S. bayanus and other strains. The highest levels of fructose, maltose, and galactose in the ginseng extracts were found in ginseng extracts fermented with B. subtilis. The levels of succinic acid and malic acid in the ginseng extract fermented with S. bayanus as well as the levels of lactic acid, malonic acid, and hydroxypruvic acid in the ginseng extract fermented with lactic acid bacteria (L. plantarum and L. mesenteroide) were the highest. In the results of taste features analysis using an electronic tongue, the ginseng extracts fermented with lactic acid bacteria were significantly distinguished from other groups by a high index of sour taste probably due to high lactic acid contents. Conclusion: These results suggest that a metabolomics approach based on GC-MS can be a useful tool to understand ginseng fermentation and evaluate the fermentative characteristics of starter cultures.

태음조위탕 엑스과립이 비만을 포함한 대사증후군 위험인자에 미치는 영향에 대한 연구: 단일군, 전향적, 다기관 임상연구 (The Effects of Taeeumjowi-tang Extract Granule on Metabolic Syndrome Risk Factors with Obesity: A Single Group, Prospective, Multi-Center Trial)

  • Objective The purpose of this study is to study the effects of Taeeumjowi-tang extract granule on metabolic syndrome with obesity in Taeeumin. Methods We studied 206 subjects who diagnosed metabolic syndrome with obesity and classified Taeeumin by a Sasang constitution analysis tool. The subjects were administered by Taeeumjowi-tang for 12 weeks. We investigated the changes of body mass index, abdominal circumference, the number of risk factors for metabolic syndrome, lipid, fasting blood sugar, Hemoglobin A1c, free fatty acid, insulin, and blood pressure. Results Body mass index and abdominal circumference, systolic and diastolic blood pressure were significantly reduced after 4 weeks, 8 weeks, and 12 weeks of administration compared to before administration. Triglyceride significantly decreased after 8 weeks and 12 weeks of dosing compared to before taking. Low density lipoprotein cholesterol, fasting blood sugar were significantly increased after 8 weeks of administration compared to before administration. High density lipoprotein cholessterol was decreased significantly after 4 weeks of administration compared to before administration. The number of risk factors for metabolic syndrome, Hemoglobin A1c increased significantly after 12 weeks of administration compared to screening. Conclusions In this study we knew that Taeeumjowi-tang extract granule is very efficient in metabolic syndrome, especially in obesity and hypertension, triglyceride, insulin resistance.

식이 DFA IV의 흰쥐 장내 단쇄지방산 생성 및 장내환경 개선효과 (Effects of Dietary DFA IV on Lumen Short Chain Fatty Acids Production and Intestinal Environment in Rats)

  • 본 연구는 미생물 발효로 생산된 DFA IV의 장내환경 개선 효과를 살펴보고자 하였다. 동물실험을 통해 식이 DFA IV가 장의 길이 및 무게와 단쇄지방산 및 젖산 생성, 장내 pH에 미치는 영향을 조사하였다. Sprague-Dawley종 흰쥐를 식이무게의 DFA IV를 0%(대조군) 또는 1%로 함유한 식이로 3주간 사육하였다. DFA IV를 공급한 군에서 대조군에 비해 맹장의 무게와 맹장벽의 무게가 유의적으로 증가하였다. DFA IV 공급에 의해 맹장 내용물의 아세트산과 부티르산 함량이 각각 대조군의 1.6배, 3.2배 증가하였고, 이에 따라 맹장과 대장의 pH도 DFA군에서 대조군에 비해 감소하였다. DFA IV는 또한 맹장의 젖산 생성을 1.5배 증가시켜, 식이로 공급한 DFA IV가 장내에서 젖산균의 증식을 촉진하였음을 보여주었다. 본 연구 결과 DFA IV 함유 식이는 장에서 발효되어 장내 환경을 개선하는 것으로 사료된다. 결론적으로 DFA IV는 장건강에 유익한 프리바이오틱 기능성 소재로 활용될 수 있으리라 기대된다.

식이로 유도한 비만마우스에서 아피오스의 내장지방 감소 효과 (Anti-Visceral Obesity Effect of Apios americana Medikus in Diet-Induced Obese Mice)

  • 본 연구는 고지방식이(열량의 40%를 지방으로 공급)로 유도한 비만마우스에게 아피오스 분말(10%, w/w)을 12주간 급여한 후 내장지방 감소 및 지방간 개선 효과를 살펴보았다. 아피오스 분말 급여로 인한 유의적인 체중 감소는 보이지 않았으나, 부고환지방과 후복막지방 무게가 유의적으로 감소하여 총 내장지방 무게는 대조군에 비해 14.3% 감소하였다. 부고환지방조직의 지방세포 크기 역시 아피오스 분말군에서 현저히 감소되었다. 혈청의 유리지방산, 중성지질과 총 콜레스테롤 함량은 실험군 간의 유의적인 차이가 없었으나 총 콜레스테롤에 대한 HDL-콜레스테롤 비와 동맥경화 지수는 아피오스 분말군이 대조군에 비해 각각 18.9%와 47.2% 유의적으로 개선되었다. 혈청의 렙틴 함량은 아피오스 분말군이 대조군에 비해 유의적으로 낮았다. 고지방식이를 급여한 마우스에 아피오스 분말 급여는 간 조직 중의 지질축적 및 유리지방산과 중성지질 함량을 유의적으로 낮추는 것으로 나타났다. 간 조직의 지질합성 관련 유전자인 Pparg, Fasn과 Dgat2의 발현이 아피오스 분말 급여로 억제되었다. 이상의 결과에서 아피오스 분말은 고지방식이로 인한 내장지방 축적과 지방간을 개선할 수 있을 것으로 평가되었다.

Anti-inflammatory and antifatigue effect of Korean Red Ginseng in patients with nonalcoholic fatty liver disease

  • Background: Korean Red Ginseng (KRG) is a well-known natural product with anticarcinogenic and antioxidant effects. We evaluated the antifatigue effect of KRG in patients with nonalcoholic fatty liver disease (NAFLD). Methods: Eighty patients with NAFLD were prospectively randomized to receive 3 wk of KRG or placebo in addition to counseling on healthy eating and regular exercise. Liver function test, proinflammatory cytokines, adiponectin, antioxidant activity, and fatigue score were measured and compared according to the body mass index between the KRG and placebo groups. Results: The liver function tests were significantly improved after 3 wk of treatment in both groups. The mean levels (at baseline and after treatment) of tumor necrosis factor-${\alpha}$ were $108.0pg/mL{\pm}54.8pg/mL$ and $92.7pg/mL{\pm}39.0pg/mL$ (p = 0.018) in the KRG group and $123.1pg/mL{\pm}42.1pg/mL$ and $127.5pg/mL{\pm}62.2pg/mL$ (p = 0.694) in the placebo group, respectively. There was a significant difference in change of adiponectin levels between the KRG ($7,751.2pg/mL{\pm}3,108.1pg/mL$ and $8,197.3pg/mL{\pm}2,714.5pg/mL$) and placebo groups ($7,711.6pg/mL{\pm}3,041.3pg/mL$ and $7,286.1pg/mL{\pm}5,188.7pg/mL$, p = 0.027). In patients with overweight, the fatigue score was significantly decreased in the KRG group ($35.0{\pm}13.2$ and $24.5{\pm}8.9$, p = 0.019). Conclusion: Our results show that KRG might be effective in reducing proinflammatory cytokine and fatigue in overweight patients with NAFLD, in addition to improvements in adiponectin levels.

A Potential Diagnostic Pitfall in the Differentiation of Hemorrhagic and Fatty Lesions Using Short Inversion Time Inversion Recovery: a Case Report

  • Short inversion time inversion recovery (STIR) is widely used for spinal magnetic resonance imaging (MRI) because the pulse sequence of STIR is insensitive to magnetic field inhomogeneity and can be used to scan a large field of view. In this case report, we present a case of spinal epidural hematoma with unexpected signal decrease on a STIR image. The MRI showed an epidural mass that appeared with high signal intensity on both T1- and T2-weighted images. However, a signal decrease was encountered on the STIR image. This nonspecific decrease of signal in tissue with a short T1 relaxation time that is similar to that of fat (i.e., hemorrhage) could lead to a diagnostic pitfall; one could falsely diagnose this decrease of signal as fat instead of hemorrhage. Awareness of the nonselective signal suppression achieved with STIR pulse sequences may avert an erroneous diagnosis in image interpretation.

Lipoma Causing Glossopharyngeal Neuralgia : A Case Report and Review of Literature

  • The cerebello-pontine angle lipomas causing trigeminal neuralgia or hemifacial spasm are rare. A lipoma causing glossopharyngel neuralgia is also very rare. A 46-year-old woman complained of 2-year history of severe right throat pain, with ipsilateral episodic otalgic pain. The throat pain was described as an episodic lancinating character confined to the throat. Computed tomography and magnetic resonance imaging revealed a suspicious offending posterior inferior cerebellar artery (PICA) compressing lower cranial nerves including glossopharyngeal nerve. At surgery, a soft, yellowish mass ($2{\times}3{\times}3mm$ in size) was found incorporating the lateral aspect of proximal portion of 9th and 10th cranial nerves. Only microvascular decompression of the offending PICA was performed. Additional procedure was not performed. Her severe lancinating pain remained unchanged, immediate postoperatively. The neuralgic pain disappeared over a period of several weeks. In this particular patient with a fatty neurovascular lump causing glossopharyngeal neuralgia, microvascular decompression of offending vessel alone was enough to control the neuralgic pain.
