Activation of Heme Oxygenase-1 by Mangiferin in Human Retinal Pigment Epithelial Cells Contributes to Blocking Oxidative Damage

  • Mangiferin is a kind of natural xanthone glycosides and is known to have various pharmacological activities. However, since the beneficial efficacy of this compound has not been reported in retinal pigment epithelial (RPE) cells, this study aimed to evaluate whether mangiferin could protect human RPE ARPE-19 cells from oxidative injury mimicked by hydrogen peroxide (H2O2). The results showed that mangiferin attenuated H2O2-induced cell viability reduction and DNA damage, while inhibiting reactive oxygen species (ROS) production and preserving diminished glutathione (GSH). Mangiferin also antagonized H2O2-induced inhibition of the expression and activity of antioxidant enzymes such as manganese superoxide dismutase and GSH peroxidase, which was associated with inhibition of mitochondrial ROS production. In addition, mangiferin protected ARPE-19 cells from H2O2-induced apoptosis by increasing the Bcl-2/Bax ratio, decreasing caspase-3 activation, and blocking poly(ADP-ribose) polymerase cleavage. Moreover, mangiferin suppressed the release of cytochrome c into the cytosol, which was achieved by interfering with mitochondrial membrane disruption. Furthermore, mangiferin increased the expression and activity of heme oxygenase-1 (HO-1) and nuclear factor-erythroid-2 related factor 2 (Nrf2). However, the inhibition of ROS production, cytoprotective and anti-apoptotic effects of mangiferin were significantly attenuated by the HO-1 inhibitor, indicating that mangiferin promoted Nrf2-mediated HO-1 activity to prevent ARPE-19 cells from oxidative injury. The results of this study suggest that mangiferin, as an Nrf2 activator, has potent ROS scavenging activity and may have the potential to protect oxidative stress-mediated ocular diseases.

  • The Class III malocclusion classified in two types of Skeletal Class III and Pseudo Class III. In the case of the maxillary deficiency, the protraction H-G(facemask) with Bonded RPE can be used. For children with A-P and vertical maxillary deficiency, the preferred treatment is to move the maxilla into a more anterior and inferior position, which also increases its size as bone is added at the posterior and superior sutures. Successful forward repositioning of the maxilla can be accomplished before age 8. To resist tooth movement as much as possible, the maxillary teeth should be splinted together as a single unit. The maxillary appliance must have hooks for attachment to the facemask that are located in the canine-primary molar area above the occlusal plane. The facemask usually worn until a positive overjet of 2-5mm is achieved interincisally. Occipital chin cup is successful in those patients who can bring their incisors close to an edge-to-edge position when in centric relation. This treatment is particularly useful in patients who begin treatment with a short lower anterior facial height, as this type of treatment can lead to an increase in lower anterior facial height. If the pull of the chin cup is directed below the condyle, the force of the appliance may lead to a downward and backward rotation of the mandible.

Time for Young Adults to Reach Resting Blood Pressure after Walking as measured by an Automatic and a Mercury Sphygmomanometer (20대 성인의 보행 후 휴식시간에 따른 진동식 자동혈압계와 수은혈압계의 안정혈압 도달 시점)

  • Purpose: The purpose of this study was to identify the time for young adults to reach resting blood pressure after walking as measured by an automatic and a mercury sphygmomanometer. Methods: Participants were 33 nursing students in their 20s. Blood pressure after walking was measured every minute on both arms simultaneously for a total of 6 times after walking for 12 minutes on a treadmill at ratings of perceived exertion (RPE) of 11. Data were analyzed using paired t-test, Bland-Altman plots and repeated measures ANOVA. Results: Systolic blood pressure after walking was the same as measurements corresponding to resting blood pressure after 3 minutes of resting if measured with the automatic sphygmomanometer and 4 minutes of resting if measured with the mercury sphygmomanometer. Conclusions: In order to measure the resting blood pressure for healthy young adults who performed low-density walking for 12 minutes on flat land, the measurement needs to be made after a resting time of at least 3 minutes in the case of an automatic sphygmomanometer and 4 minutes in the case of a mercury sphygmomanometer.

THE SHEAR STRESS PROTEOME OF CHONDROCYTES (전단력이 연골세포에 미치는 영향에 관한 연구)

  • The objective of this study is screening the shear stress related proteins in chondrocytes using twodimensional electrophresis and MALDI-TOF. C-28/I2 cell line were grown. The fluid-induced shear stress(FISS) was applied using a cone viscometer at a rotational velocity of 80rpm for periods of 12 hours. Control cultures were tested under identical conditions without mechanical load application. Collected samples were used for the two-dimensional electrophoresis and MALDI-TOF. The identified proteins were calcyclin, RPE-spondin, interleukin-2, extracellular signal regulated kinase (ERK), lamin B2, porA protein, and RET-ELE1 protein. All of them showed a decreased expression. In conclusion, seven proteins were identified as a shear stress related proteins in chondrocytes. As the destruction of articular cartilage is one of main pathogenesis of TMJ internal derangement, this study will give useful information for the understanding of the molecular aspect of TMJ disease.

Increased 26S proteasome non-ATPase regulatory subunit 1 in the aqueous humor of patients with age-related macular degeneration

  • Age-related macular degeneration (AMD) is the leading cause of blindness in the world. Evidence indicates that the suppression of the ubiquitin-proteasome system (UPS) contributes to the accumulation of toxic proteins and inflammation in retinal pigment epithelium (RPE), the functional abnormalities and/or the degeneration of which are believed to be the initiators and major pathologies of AMD. To identify new protein associations with the altered UPS in AMD, we used LC-ESI-MS/MS to perform a proteomic analysis of the aqueous humor (AH) of AMD patients and matched control subjects. Six UPS-related proteins were present in the AH of the patients and control subjects. Four of the proteins, including 26S proteasome non-ATPase regulatory subunit 1 (Rpn2), were increased in patients, according to semi-quantitative proteomic profiling. An LC-MRM assay revealed a significant increase of Rpn2 in 15 AMD patients compared to the control subjects, suggesting that this protein could be a biomarker for AMD.

Evaluation of Estrogenic Effects of Phthalate Analogues Using in vitro and in vivo Screening Assays

  • Phthalate analogues are a plasticizer and solvent used in industry. Phthalates were classified in the category of "suspected" endocrine disruptors. The purpose of our study was to screen and elucidate the endocrine disrupting activity of seven phthalate analogues. E-screen assay was performed in MCF-7 human breast cancer cells with seven phthalate analogues. In this cell proliferation assay, benzyl butyl phthalate (BBP) and dibutyl phthalate (DBP) showed high estrogenic activity. Their relative proliferation efficiencies (RPE) were 109 and 106%, respectively. In vitro estrogen receptor (ER) binding assay, BBP, di-n-octyl phthalate (DOP) and dinonyl phthalate (DNP) showed weak relative binding affinity (RBA: 0.02%) compared to $17{\beta}-estradiol\;(E2)$ (RBA: 100%). In uterotrophic assay, E2 produced a significant increase, whereas four tested phthalate analogues had potential estrogenic effects in vitro did not increased in uterus weight in immature rats. From these results, we demonstrated that phthalate analogues exhibit weak estrogenic activity in vitro assays at high concentrations. Although phthalates induced an increase in MCF-7 cell proliferation by an estrogenic effect, they could not induce a uterus weight increase in vivo. From these, we may suggest that these phthalate analogues are easily metabolized to inactive forms in vivo. Further investigation in other in vitro and in vivo experimental systems might be required.

Innate Color Preference of Zebrafish and Its Use in Behavioral Analyses

  • Although innate color preference of motile organisms may provide clues to behavioral biases, it has remained a longstanding question. In this study, we investigated innate color preference of zebrafish larvae. A cross maze with different color sleeves around each arm was used for the color preference test (R; red, G; green, B; blue, Y; yellow). The findings showed that 5 dpf zebrafish larvae preferred blue over other colors (B > R > G > Y). To study innate color recognition further, tyrosinase mutants were generated using CRISPR/Cas9 system. As a model for oculocutaneous albinism (OCA) and color vision impairment, tyrosinase mutants demonstrated diminished color sensation, indicated mainly by hypopigmentation of the retinal pigment epithelium (RPE). Due to its relative simplicity and ease, color preference screening using zebrafish larvae is suitable for high-throughput screening applications. This system may potentially be applied to the analysis of drug effects on larval behavior or the detection of sensory deficits in neurological disorder models, such as autism-related disorders, using mutant larvae generated by the CRISPR/Cas9 technique.

Study on Estrogenic Activities of Phthalate Esters Using E-screen Assay and Competitive Binding Assay (E-screen Assay 및 상경적 결합반응을 이용한 Phthalate Esters의 내분비계 장애 작용 연구)

  • Phthalate esters are used extensively as a plasticizer in the manufacture of plastic products such as PVC bags and medical devices. Recently, phthalate esters have been shown to induce endocrine system mediated responses. However. only a Jew studies have been conducted for estrogenic activity of phthalate esters. In this study estrogenic activities of seven phthalate esters. butyl benzyl phthalate (BBP), di(2-ethylhexyl) phthalate (DEHP), di-n-butylphthalate (DBP), diethylphthalate (DEP), di-n-pentylphthalate (DPP), di-n-propylphthalate (DPrP) and dicyclohexylphthalate (DCHP), were examined in vitro using E-screen assay and competitive binding assay. From the E-screen assay, BBP. DEHP. DBP and DEP showed weak estrogenic activity at the concentration of 5 $\mu\textrm{M}$. The relative proliferative effect (RPE) and the relative proliferative potency (RPP) were 50~70% and 0.01%. respectively, when compared with 500 pM of 17$\beta$-estradiol (E2). In competitive binding assay with the rat uterine estrogen receptor (ER), BBP and DEP showed weak binding potency [(l/$10^4$~1/$10^5$ of E2] while DEHP and DBP scarcely bound to ER. These results suggest that some phthalate esters have weak estrogenic activities in vitro.

The Effect of Aerobic Exercise Program for Chronic Disease Patients (성인병 환자를 위한 유산소성 운동프로그램의 효과)

  • The purpose of this study is to determine the effects of exercise programs on the treatment of chronic diseases. For this purpose, nine middle-aged women suffering from such chronic diseases as obesity, hypertension and coronary heart disease(CHD) were sampled to perform a 4-month-long exercise program which required the lactate threshold(LT) level corresponding to 70% of maximal oxygen uptake(VO2max). Thereupon, their physiques, physical fitness, blood components and physiological varibles were compared pre and post of exercise program. The results were summarized as follows ; 1) After the 4-month-long exercise program, %fat was significantly decreased, while such variables as grip strength, side step, trunk flexion and leg balance were significantly increased. 2) Blood components such as TG, TC, HDL-C, LDL-C, hemoglobin and hematocrit did not show and change in after exercise program. 3) Physiological varables such as HR, SBP, DBP, HR/LT, SBP/LT, DBP/LT, RPE/LT, VE/LT, VO2/LT and VO2 max showed significant increase after 4-month exercise program. These results suggest that regular aerobic exercise in a long period might help to decrease % fat and to increase aerobic capacity in middle-aged women suffering from chronic diseases.

The Effects of an Exercise Program on Body Composition, Serum lipids, Menopausal Symptoms in Korean Menopausal Women (폐경기 여성의 운동프로그램 중재효과)

  • Purpose: The purpose of this study is to find the effects of an exercise program on body composition, serum lipids, and menopausal symptoms in Korean menopausal women. Methods: This research used none qui valent control group pretest-posttest design. The participants were 30 middle-aged women who were divided into two groups, an experimental group (n = 16) who participated in the exercise program and a control group (n = 14). The exercise program was composed of gymnastic-ball exercises with RPE $12{\sim}13$ intensity, 50 minutes' duration and three times a week for 12 weeks from April 1 to June 30, 2008. The dependent variables (body composition, serum lipids, menopause symptoms, self-efficacy, and life satisfaction) were measured before and after exercise program. Results: There were statistically significant difference in weight (F=2.92, p=.049), percent body fat (F=4.22, p=.040), total cholesterol (F=2.63, p= .017) and high density lipoprotein (F=6.74, p= .015) between the two groups. Psychological menopausal symptoms also decreased significantly (F=7.01. p=.014) after the exercise in the experimental group. Conclusions: Regular gymnastic-ball exercises are helpful for menopausal women. They have positive effects not only on body composition and serum lipids but also on the decrease of menopausal symptoms. The results of this study suggest that gymnastic-ball exercise programs are effective in health promotion of Korean middle-aged women, and thus it is necessary to develop exercise programs for women.
