Expression of Wee1 Gene in the Ascidian, Halocynthia roretzi Embryo

  • It is suggested that FGF/Ras/MEK/Erk signaling plays crucial roles in specification and cell division of the mesodermal precursor cells in ascidian embryos. To investigate how the number of cell division in tissue precursor cells is determined, we have characterized Wee1 homolog, Hr-Wee1 of the ascidian Halocynthia roretzi. We found that the Hr-Wee1 mRNA is expressed both maternally and zygotically. Maternal transcript is localized to the cytoplasm in the animal cells, while zygotic expression is seen in cells of the endoderm lineage from 32-cell to 110-cell stages. Zygotic in situ signal is detected in the A-line neural plate cells of neurulae, and in epidermal cells of the head region of tailbud embryos. Embryos treated with MEK signaling inhibitor showed a similar pattern to normal embryos in expression of Hr-Wee1. Therefore, it is likely that MEK signaling does not affect the maternal and zygotic expression of Hr-Wee1.

Plant Regeneration from Zygotic Embryos Cultures of Lilium Lancifolium Thunb. Via Bulblet Formation (참나리(Lilium lancifolium Thunb.) 접합자배로부터 소자구 형성을 통한 식물체 재생)

  • Plant regeneration system from zygotic embryos (2n=24) of Lilium lancifolium Thunb. via bulblet formation was estabished. Zygotic embryos of Lilium lancifolium formed bulblets and somatic embryos simultaneously when they cultured on MS medium supplemented with low concentration of 2,4-D. The highest frequency of bulblet and somatic embryo formation from zygotic embryos of Lilium lancifolium was 66.7% and 56.7%, respectively. The frequency of bulblet and somatic embryo formation was decreased when they cultured on MS medium over than 1 mg/L of 2,4-D. To regenerate whole plants, somatic embryos formed on zygotic embryos were transferred to MS basal medium. However somatic embryos did not fully converted into plantlets. Further incubation in the light, elongated somatic embryos formed numerous bulblets at the base of somatic embryos. Upon transfer to MS basal medium, bulblets were successfully converted into plantlets after further 4 weeks of culture in the light. After acclimatization, plantets from bulblets were transferred to soil and grown to normal plants in growth chamber (approximately $30\;{\mu}mol\;m^{-2}s^{-1}$, 16/8h photo period, $25^{\circ}C$) The chromosome analysis revealed that plants regenerated from zygotic embryos showed 2n=24. These results indicate that chromosome stability of source tissue is maintained during plant regeneration via bulblet formation.

Mass Production of Calla Lily(Zantedeschia spp. Southern Light) by the Immature Zygotic Embryo Culture (유색칼라(Zantedeschia spp. Southern Light) 미숙배 배양에 의한 다량증식)

  • In order to investigate the effects of developmental stage of embryos and plant growth regulators on mass production of Zantedeschia spp. Southern Light, immature zygotic embryos of Zantedeschia spp. Southern Light were cultured on Murashige and Skoog(1962) basal media or containing 2,4-D, NAA and BA. Globular embryos did not grow on any of the 2,4-D, NAA and BA combinations. The most suitable stage of immature zygotic embryo culture on the induction callus and multiple shoot was at early cotyledonary embryo stage, and at this stage of embryos were germinated up to 87.5%. The whitish watery callus and yellowish compact nodular callus produced on all 2,4-D, NAA and BA media. The best combination for inducing embryogenic callus was 0.5 mgL NAA and 1.0 mg/L BA. Whitish watery calli have been subcultured for more than 8 months and have retained their producing ability, Plant regeneration was only obtained by direct shoot development and yellowish compact nodular calli. Abundant plantlets were regenerated from cotyledonary stage of embryo culture on MS medium supplemented with 0.5 mg/L NAA and 1.0 mg/L BA. Supplementation of the media with 10% coconut water showed as the best concentration for plant differentiation from direct developed of shoots. The number of regenerated plants from one embryo could be seperated 25-35s plantlets. All yellowish compact callus-derived plantlets were transferred to pots containing a mixture of vermiculite, perlite and sand(1:1;1 v/v) and 100% of divided plantlets were phenotypically normal.

Effect of exogeneous plant growth regulators on morphogenetic response in vitro by embryo and leaf cultures of Camellia sinensis(L.) O. Kuntze (차나무 잎과 배 배양에 있어서 식물 생장조절물질이 형태형성에 미치는 영향)

  • Morphogenetic responses were investigated by culturing embryo and leaf explants of Korean wild type tea plant, Camellia sinensis (L.) O. Kuntze. Induction of direct somatic embryogenesis as well as adventitious and/or axillary shoots was obtained from mature zygotic embryo cultures on Murashige and Skoog (MS) basal medium having 5 to $20\mu\textrm{M}$cytokinin a lone. Morphogenetic response was decreased dramatically by the addition of auxins tested. One hundred percent of induced and isolated shoots formed roots after four weeks of culture on half-strength MS or quarter-strength Schenk and Hildebrandt (SH) media supplemented with $10\mu\textrm{M}$indole-3-butyric acid (IBA). Immature zygotic embryos were shown to be a suitable explant for embryogenic callus formation in the presence of 2, 4-dichlorophenoxyacetic acid(2, 4-D) in basal medium. Mature zygotic embryo originated leaves were used to test their ability for mophogenesis by incorporating plant growth regulators such as IBA, naphthyl-1-acetic acid (NAA), and 6-benzylaminopurine (BAP). Apparently, the morphogenetic responses of the cultured explant sources on the types and/or levels of plant growth regulators tested were observed visually.

The Condition of Regeneration and Antibiotics Concentration for Gene Transformation of Zygotic Embryo in Panax ginseng C. A. Meyer (인삼(Panax ginseng C. A. Meyer)접합자 배의 형질전환을 위한 재분화 및 항생제 농도 조건)

  • To obtain multi-shoot using zygotic embryos dissected from ginseng seed, the embryos were cultured on MS medium supplemented with CPA and BA. Effective multi-shoot induction was achieved on 0.5mg/ t CPA and 1.0mg/ t BA treatment. Among the various plant growth regulator treatment, MS basal medium with 1mg/ t 2,4-D and 0.5mg/ t kinetin was more competent and could be induced 4∼6 shoots per one embryo. Also, the best condition for pre-embryoid induction from ginseng cotyledon so as to ginseng transformation appeared to 1mg/ t 2,4-D and 0.5mg/ t kinetin treatment. The kanamycin level to select transformants varied greatly by different explant tyues. The petiole explants with leaf and embryo could survived up to 100$\mu\textrm{g}$ / ml kanamycin concentration where as petiole explants without leaf died all at the level. Conclusionally, our results suggest that optimum kanamycin concentration for ginseng transformation using somatic embryos is about 75∼100$\mu\textrm{g}$ / ml concentration.

Effects of Date and Growth Regulators on the Culture of' Immature Zygotic Embryos of North American Ginseng

  • As the zygotic embryo of North American ginseng (Panax quinquefolius L.) matured during stratification over 203 days it grew from 0.75 to 5.2 mm. Embryo excision and culturing on media containing different concentrations of two growth regulators, gibberellic acid ($GA_3$, 1 to 10 ${\mu}M$) and benzyladenine (BA, 1 to 5 ${\mu}M$), during stratification, showed that shoot and root number and the shoot, root and cotyledon length increased with increased stratification time. Gibberellic acid was the more effective growth regulator for increasing shoot and root number and shoot, root and cotyledon lengths. Immature embryos (stratified for up to 63 days) needed growth regulators for further development. Cultures on $GA_3$ at the last culture date (stratified for 203 days) when embryos were mature, produced multiple shoots but there was no effect of $GA_3$ concentration. Benzyladenine inhibited shoot and root growth regardless of embryo stratification. Growth regulators had little effect on cotyledon length of mature embryos. Embryos cultured on $GA_3$ combined with BA were green on all culture dates whereas greening in the control and BA treatments increased with culture date. The BA treatments induced 100% swelling of the embryos on the final culture date while in the control and $GA_3$ treatments there was no swelling. There was little or no curling in the control and BA treatments and a linear decrease in curling with culture date in the $GA_3$ and $GA_3$ + BA treatments.

  • When cultured on MS medium supplemented with 0.5 mg/L NAA alone or 1.0 mg/L 2,4-D and 0.5 mg/L BA, immature zygotic embryos of Stewartia koreana formed embryogenic calli and somatic embryos. In investigate effect of sucrose concentration on somatic embryo development, embryogenic calli were transferred to MS basal medium containing 1.5,3, 6 or 9% sucrose. The greatest frequency of somatic embryos was obtained on medium containing 6% sucrose. However addition of 1.5 or 9% sucrose to medium inhibited somatic embryo germination and development into normal plantlet After 5 weeks of hardening culture on medium containing 6% sucrose, somatic embryos were transferred to half strangth MS medium supplemented with 0.1% charcol, wherein these embryo developed into the normal plantlets.

Somatic embryogenesis from the axillary meristems of peanut (Arachis hypogaea L.)

  • Developmental anomalies in the plumule meristem of peanut (Arachis hypogaea L.) somatic embryos resulted in poor shoot differentiation and reduced plant recovery. Existing meristems with caulogenic potential have never been tested for embryogenesis in peanut. The present experiment was designed to test the mature zygotic embryo axis derived plumule with three meristems for somatic embryogenesis. Embryogenic masses and embryos developed from the caulogenic meristems in the axils. Exposure of 2 weeks in primary medium with $90.5{\mu}M$ 2,4-D suppressed the shoot tip differentiation temporarily which then regained the ability to form the shoot on withdrawal of 2,4-D. Exposure of 4 weeks in primary medium with $90.5{\mu}M$ 2,4-D suppressed the shoot tip differentiation irreversibly. No shoot formation was noted from the tips in any of the cultures which were in secondary medium with $13.6{\mu}M$ 2,4-D. Development of somatic embryos directly from axillary meristems was confirmed histologically. Conversion frequency of these embryos was 11%. Thus, in this report, we describe a method to obtain somatic embryos from the determined organogenic buds of the axillary meristem, by culturing the nodal explant vertically on embryo induction medium. It also displays the possibility of obtaining both embryogenic and organogenic potential in two parts of the same explant simultaneously. The possibility of extending this approach for genetic transformation in in vivo system through direct DNA delivery or Agrobacterium injection in meristems can also be explored. Using Agrobacterium rhizogenes, we have demonstrated the possibility of gene transfer in the axillary meristems of seed-derived plumule explant.

Effect of Cytokinins on the Number of Cotyledons of Sometic Embryos from Immature Zygotic Embryo Culture of Heracleum moellendorffii Hance (어수리(Heracleum moellendorffii Hance.)의 미숙배로부터 형성된 체세포배의 자엽수에 미치는 Cytokinin의 영향)

  • In order to investigate the effect of cytokinins on the cotyledonary variability of somatic embryos in Heracleum moellendorffii Hance., somatic embryos were induced from the immature zygotic embryo on MS medium containing 1.0mg/l 2, 4-D, and then transferred to 2,4-D-free and cytokinin-added medium for somatic embryogenesis after 4weeks culture. Polycotyledonary embryos were formed most abundantly(84.9%) on the 0.2mg/l BA medium as compared with the 0.2mg/l 2ip(42.6%) and kinetin(32.9%) media, and it was proportional to BA concentrations(0.01~0.1mg/l). The rate of polycotyledonary embryo formation increased proportional to the period of BA treatment and also increased more at the globular stage than at the heart, torpedo-shaped and cotyledonary stages.
