Effect of Crotalaria juncea seed extracts on the estrous cycle and ovarian activity in albino mice

  • Petroleum ether, benzene and alcohol extracts of seeds of C. juncea administered orally at the dose level of 25mg/100g body weight to adult female mice for 30 days, resulted in irregular estrous cycle with prolonged estrus and metaestrus and reduced diestrus and proestrus during the experimental period. Histological studies of the ovary indicate increases in the number of atretic follicles but decreases in the number of developing follicles, Graafian follicles and corpora lutea. The total cholesterol content of the ovary is increased, whereas ascorbic acid content is decreased. The weight of the uterus and its micrometric measurement in all experimental mice are increased significantly. The alcoholic extracts showed estrogenic activity in immature mice by early opening of the vagina, premature cornification of the vaginal epithilium and increases in uterine weight. However, alcohol extract of seeds of C. juncea was more effective in causing these changes compared to other extracts. After subjecting to preliminary phytochemical screenings alcohol extract showed positive; test for alkaloids, steroids, glycosides, flavones, phenols and tannins.

  • 染色體의 構造的인 異常에 미치는 Methyl methanesulfonata (MMS)와 thymidine 相似體 (BUdR, IUdR)의 영향을 究明하기 위해 사람의 白血球 培養細胞에 2.0mM의 BUdR 또는 IUdR을 24時間 前 처리하고 이어 농도를 달리한 MMS $(0.0\\sim 2.0 mM)$를 1時間 처리한 뒤 24時間째 染色體標本을 작성하여 아래와 같은 결과를 얻었다. 1. MMS를 단독 처리한 對照區에서는 관찰된 染色體 異常型의 대부분이 예상외로 染色分體 異常型을 나타냈다. 2. BUdR 또는 IUdR의 단독처리도 染色體異常을 유발한다. 또한 MMS와 이중 처리한 경우에는 단독 처리보다 染色體異常型이 훨씬 증가한다. 따라서 thymidine 相似體는 MMS에 의한 染色體 異常型을 變化시켜주는 感受性物質로 作用함이 판명되었다.

  • The silkworm Bombyx mori L. was studied as the potential “host” of popular in Uzbekistan biocontrol ectoparasite, entomophagous Bracon hebetor Say. Being one of representatives of economic-beneficial insects, the silkworm (larvae, pupae and imago) can be used as highly sensitive test organism for revealing of neuro toxic effects of insects venom as well as of their purified components in screening assays. In comparative aspect, except a mulberry silkworm, representatives of Uzbekistan pests cotton-boll worm Helycoverpa armigera Hbn., lesser mulberry pyralid Glyphodes pyloalis Wlk., codling moth Corpocapsa pomonella L., malaria mosquito Anopheles claviger and Colorado potato beetle Leptinotarsa decemlineata Say have been subjected to insect toxic test of bracon venom gland extract (VGE) and its fractions which were obtained by gel-chromatography on Sephadex G-100. The paralyzing effect of the VGE and its fractions was shown in a various degree on the pests.

  • 1973년 7월부터 1987년 10월까지 한국연안의 32개 지역에서 채집되어 서울 대학교 자연대 동물학과에 보관되어 있던 옆새우류의 표본중에서 Ampithoidae 에 속하는 것들올 동정힌 결과 3속 12종이 밝혀졌다. 이 중에서 5종은 신종이므로 각각 Ampithoe breviPalma, Ampithoe koreana, Ampithoe youngsanensis, Peramphithoe baegryeongensis, Peramphithoe namhaensis 라고 명명 하고 기재하였다. Ampithoe valida의 동종이명으로 처리되던 Ampithoe shimizuensis를 AmPithoe valida와 구별하여 새로운 아종인 Ampithoe valida shimizuensis로 명명하였다. 이외에 4종은 한국 미기록종 이었다.

Malaria transmission potential by Anopheles sinensis in the Republic of Korea

  • To evaluate the factors that determine the transmission level of vivax malaria using vectorial capacity, entomological surveys were conducted from .lune to August, 2000. From 6 nights of human-bait collection in Paju, the human biting rate (ma) was counted as 87.5 bites/man/night. The parity of Anopheles sinensis from human baiting collections fluctuated from 41% to 71% (average 48.8%) of which the rate gradually increased as time passed on: 35.2% in Jun. ; 55.0% in July; 66.2% in Aug. From this proportion of parous, we could estimate the probability of daily survival rate of An. sinensis to be 0.79 assumed with 3 days gonotrophic cycle and the expectancy of infective life through 11 days could be defined as 0.073. Blood meal analysis was performed using ELISA to determine the blood meal source. Only 0.8% of blood meals were from human hosts. We could conclude that An. sinensis is highly zoophilic (cow 61.8%) Malaria is highly unstable (stability index < 0.5) in this area. From these data, vectorial capacity VC) was determined to be 0.081. In spite of a high human biting rate (ma), malaria transmission potential is very low due to a low human blood index. Therefore, we could conclude that malaria transmission by An. sinensis is resulted by high population density, not by high transmission potential. For this reason, we need more effort to decrease vector population and vector-human contact to eradicate malaria in Korea.

Identification of Histone Deacetylase 1 Protein Complexes in Liver Cancer Cells

  • Background: Hepatocellular carcinoma is one of the leading causes of mortalities worldwide. The search for new therapeutic targets is of utmost importance for improved treatment. Altered expression of HDAC1 in hepatocellular carcinoma (HCC) and its requirement for liver formation in zebrafish, suggest that it may regulate key events in liver carcinogenesis and organogenesis. However, molecular mechanisms of HDAC1 action in liver carcinogenesis are largely unknown. The present study was conducted to identify HDAC1 interacting proteins in HepG2 cells using modified SH-double-affinity purification coupled with liquid mass spectrophotemetery. Materials and Methods: HepG2 cells were transfected with a construct containing HDAC1 with a C-terminal strepIII-HA tag as bait. Bait proteins were confirmed to be expressed in HepG2 cells by western blotting and purified by double affinity columns and protein complexes for analysis on a Thermo LTQ Orbitrap XL using a C18 nano flow ESI liquid chromatography system. Results: There were 27 proteins which showed novel interactions with HDAC1 identified only in this study, while 14 were among the established interactors. Various subunits of T complex proteins (TCP1) and prefoldin proteins (PFDN) were identified as interacting partners that showed high affinity with HDAC1 in HepG2 cells. Conclusions: The double affinity purification method adopted in this study was very successful in terms of specificity and reproducibility. The novel HDAC1 complex identified in this study could be better therapeutic target for treatment of hepatocellular carcinoma.

培養한 鷄胚筋細胞의 DNA複製 및 回復에 미치는 紫外線의 影響 (Effects of Ultraviolet Light on DNA Replication and Repair in Cultured Myoblast Cells of Chick Embryo)

  • 발생 12일째의 계배근세포를 약 7일간 배양하면서 자외선에 의한 절제회복, 광재활성 및 DNA 복제억제율을 조사하여 다음과 같은 결과를 얻었다. 자외선에 의한 절제회복은 계배근세포의 분화정도가 진행됨에 따라 감소하였으며, 이는 특히 자외선의 선량이 높을수록 뚜렷하였다. DNA 합성율은 분화가 진행된 세포일수록 현저히 감소하는 경향이었으며 각 분화단계에 따른 DNA 복제억제 현상은 배양 초기 세포인 경우 자외선 조사후 30분에서 1시간반 사이에서 가장 심하게 나타났고, 배양후기의 세포에서는 이러한 억제현상이 뚜렷하지 않았다. 또 배양 1일째 세포에서 광재활성에 의한 피리미딘 이량체의 감소율은 자외선 조사직후에 현저하였다가 그후 시간의 경과에 따른 차이는 보이지 않았다. 그러나, 절제회복만에 의한 이량체의 감소는 조사후 시간이 지남에 따라 서서히 감소하여 광재활성에 의한 이량체의 감소량과 비슷한 수준으로 접근하였다.

培養 鷄胚 筋原細胞의 融合에 미치는 鷄胚 抽出液 分劃의 영향 (The Effects of Fractions of Chick Embryo Extract on the Fusion of Cultured Chick Embryonic Myoblasts)

  • 鷄胚의 筋原細胞의 증식과 融合에 미치는 계배 抽出液의 영향을 조사하였다. 1. 배양액내의 계배 추출액의 농도가 높을수록 근원세포의 증식은 촉진되었으며, 반면에 근원세포 융합은 지연되었다. 2. 계배 추출액의 단백질을 Sephadex G-75로 分劃하고, 각 분획을 근원세포의 배양액에 첨가한 결과 分子量 40,000과 22,000 dalton 사이의 분획이 근원세포의 증식과 융합을 촉진시켰다. 3. 계배 추출액의 단백질을 ammonium sulfate로 분획시켜 각 분획을 근원세포의 배양액에 첨가한 결과 70% 이상 포화 용액에서 침전하는 분획이 근원세포의 증식과 융합을 현저히 증가시켰다. 이 유효 분획을 Sephadex G-75로 재차 분획하여 각 분획의 효과를 조사한 결과 근원세포의 증식과 융합을 촉진시키는 분획이 계배 추출액을 Sephadex G-75로 분획하여 얻은 유효 분획과 거의 동일한 효과를 나타내었다.

鷄胚筋原細胞의 融合에 미치는 카테콜아민의 影響 (Effects of Catecholamine on the Fusion of Chick Embryo Myoblasts in vitro)

  • 근원세포가 분화하는 과정에서 신경전달물질의 영향을 연구하기 위하여 배양한 근세포에 dopamine과 epinephrine을 처리하여 근원세포의 융합과 세포내 cAMP의 농도에 미치는 효과를 조사하였다. Dopamine $(3\\times10^{-5}M)$과 epinephrine $(3\\times10^{-5}M)$을 세포배양후 34시간에 처리했을 때 근원세포의 융합이 크게 억제되었으며, 특히 dopamine의 효과가 epinephrine보다 현저하게 나타났다. 한편, 세포내 cAMP농도는 dopamine과 epinephrine을 처리해도 거의 변화가 없었다. 근원세포의 분화에 cAMP가 관계하는지를 조사하기 위해 dbcAMP, $PGE_1$ 및 aspirin을 처리하였는데, dbcAMP $(1\\times10^{-4}M)$는 근원세포의 융합을 억제한 반면, $PGE_1 (3\\times10^{-6}M)$은 오히려 융합을 촉진하였고, PG 합성효소의 억제물질인 aspirin은 융합 억제효과를 보였다. Dopamine과 epinephrine이 근원세포의 융합과정에 작용하는 가능성있는 기작에 대해서 고찰하였다.

Survey of Dicrocoelium dendriticum infection in imported Romani and local sheep (Ovis aries), and potential epidemiological role in Saudi Arabia

  • The epidemiology of parasite infection in local and imported breeds is quite an essential topic in the meat industry and human health. This study aims to determine the prevalence of Dicrocoelium dendriticum in local sheep breeds (Naemi, Najdi, and Harri) and imported breeds from Romania (Romani breed) and the epidemiology of the infection in Saudi Arabia. Morphological description, the relationship between dicrocoeliasis and sex, age, and histological changes were also presented. A total of 6845 slaughtered sheep at Riyadh Automated slaughterhouse were investigated and followed up for 4 months between 2020-2021. It included 4,680 local breeds and 2,165 imported Romanian breeds. Fecal samples and livers and gallbladders from slaughtered animals were examined for apparent pathological lesions. The results indicated that the infection rate in slaughtered animals was 10.6% in imported Romani sheep and 0.9% in the local Naeimi breed. After identifying the parasite morphologically, negative results were obtained from examining feces, gallbladders, and livers of Najdi and Harry sheep breeds. The mean number of eggs per 20 µL/gallbladder was low (72.78 ± 17.8: 76.11 ± 5.07), medium (334.59 ± 90.6: 292.91 ± 26.63), and high (1113.2 ± 22.3: 1004 ± 143.4) in imported and Naeime sheep, respectively. Significant differences were found between gender and age (males and females were 3.67% and 6.31%; > 2 years 4.39%, 1-2 years 4.22%, and 1 year 3.53%) respectively. Histopathological lesions in the liver were more pronounced. Our survey confirmed the presence of D. dendriticum in imported Romani and local Naeimi sheep, and the potential role of imported sheep in the epidemiology of dicrocoeliasis in Saudi Arabia.
