대추 추출물의 fibrinolytic activity에 관한 연구 (Studies on Fibrinolytic Activity of Jujube (Zizyphus mauritiana) Extract)

  • 대추 추출물에서 fibrin을 가수분해 하는 생리활성 물질이 존재함을 알고 이 추출물의 열과 산에 대한 안정성 및 그 특징을 조사 하였다. Fibrin plate method 를 사용하여 측정된 fibrinolytic activity 는 대추 추출물의 첨가량이 증가할수록 높았으며, 이 추출물은 $100^{\circ}C$ 에서 10분간 열처리시에 안정한것으로 나타났다. 또한 pH 2.0, 3.0, 4.0 에서 3시간 보관후 fibrinolytic activity 를 측정한 결과 control의 80% 이상의 활성을 나타내었다. 한편 투석후 (분자량 12,000) 에는 활성이 나타나지 않았으며, 이는 대추에서 추출한 fibrin 분해 생리활성 물질이 분자량이 낮은 저분자 물질임을 알 수 있었다.

Host Plant-Antheraea mylitta Interactions and Its Effect on Reproductive and Commercial Parameters

  • Impact of food plant on reproductive and commercial parameters in Antheraea mylitta, a polyphagous insect of economic importance was studied upon feeding the insect larvae on the same host plants for six continuous generations. A. mylitta larvae were fed upon Terminalia tomentosa, Terminalia arjuna and Zizyphus jujuba and restricted them to the same host plant for six generations to document the quantitative improvement in reproductive and commercial parameters. The parameters showed significant improvement in all the host plants studied over their respective controls. Fecundity among the reproductive parameters was highly improved than others (85.9% in T. tomentosa; 58% in T. arjuna and 49.7% in Z. jujuba). Likewise in commercial parameters, the shell weight in male showed the highest improvement (by 52.9%, 45.8% and 42.1% in T. tomentosa; T. arjuna and Z. jujuba respectively). On the other hand, the shell ratio percentage in female recorded the lowest improvement. The values for all characters were recorded a decline in T. arjuna and Z. jujuba fed ones over T. tomentosa, except that of shell ratio percentage in female has registered an increase in Z jujuba fed. The study thus revealed the comparative superiority of T. tomentosa over T. arjuna and Z. jujuba.

Seasonal Variations in Tannin Profile of Tree Leaves

  • Forest tree leaves (12 different species) of semi hilly arid region of Punjab State were collected at 30-day interval throughout the year to assess the seasonal variations in tannin profile. Tannins were extracted and fractionated from fat free samples and data were analyzed statistically by $12{\times}12$ factorial design. The leaves of Anogeissus latifolia had the highest (p<0.05) concentration of total phenols (17.4%), net (15.9%) and hydrolysable (16.9%) tannins, followed by leaves of Acacia nilotica. Majority of the tree leaves selected had moderate levels (2-5%) of net tannins. Leaves of Carrisa had the highest (p<0.05) concentration of condensed tannins (CT), whereas the leaves of Anogeissus had the lowest (p<0.05) concentration of condensed tannins. The protein precipitable phenols (PPP) corresponded well with the net tannin content present in different tree leaves. Seasonal variation data revealed that in summer, net tannins and PPP decline in leaves of Bauhinia and Zizyphus whereas the net tannin content of Anogeissus and that of Carrisa increased during summer. The CT and PPP content in the leaves of Pheonix, Leucaena, Zizyphus and Ougenia increased in winter till spring season. Tree leaves generally had higher concentration of HT during summer months. It was concluded that leaves of leaves of A. nilotica, A. latifolia and L. leucocephala could serve as an excellent alternate feed stuffs for ruminants. However, leaves of Phoenix, Carrisa, Bauhinia and Dodonea should be avoided.

UHPLC/TOFHRMS analysis and anti-inflammatory effect of leaf extracts from Zizyphus jujuba in LPS-stimulated RAW264.7 cells

  • Zizyphus jujube is a plant in the buckthorn family (Rhamnaceae) that has been the subject of research into antibacterial, antifungal and anti-inflammatory properties of its fruit and seed. However, few studies have investigated its leaves. In this study, the anti-inflammatory activity of ZJL (an extract of Z. jujube leaf) was evaluated to verify its potential as an anti-inflammatory agent and SARS-CoV-2 medicine, using nitric oxide (NO) assay, RT-PCR, SDSPAGE, Western blotting, and UHPLC/TOFHRMS analysis. We found that ZJL suppresed pro-inflammatory mediators such as NO, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and tumor necrosis factor α (TNF-α) in lipopolysaccharide (LPS)-induced RAW264.7 cells. ZJL acted by inhibiting NF-KB and MAPK signaling pathway activity. We also confirmed that ZJL contains a phenol compound and flavonoids with anti-inflammatory activity such as trehalose, maleate, epigallocatechin, hyperoside, catechin, 3-O-coumaroylquinic acid, rhoifolin, gossypin, kaempferol 3-neohesperidoside, rutin, myricitrin, guaiaverin, quercitrin, quercetin, ursolic acid, and pheophorbide a. These findings suggest that ZJL may have great potential for the development of anti-inflammatory drugs and vaccines via inhibition of NF-κB and MAPK signaling in LPS-induced RAW264.7 cells.

한약재 주정추출물과 그 유효성분의 식물병원균에 대한 항균활성 (Antimicrobial Activity of Ethanol Extracts from Medicinal Herbs and Its Active Compound against Plant Pathogens)

  • 본 연구에서는 30종의 한약재 주정추출물을 대상으로 탄저병(Colletotrichum acutatum) 외 8종의 식물병원균에 대한 살균효과 검정을 실시하였다. 그 결과, 대추(Zizyphus jujuba) 주정추출물 및 hexane 분획물이 역병균(P. capsici), 무름병균(E. carotovorum subsp. carotovora), 세균성점무늬병균(P. syringae pv. syringae) 및 풋마름병균(R. solanacearum)에 대하여 광범위하게 살균효과를 나타내었다. 대추(Z. jujuba) hexane 분획물의 주성분을 GC/MS로 분석한 결과, eugenol(40.45%), dodecanoic acid(18.40%), β-caryophyllene(10.05%) and isoeugenol(9.85%) 임을 확인하였다. 이 중 eugenol과 isoeugenol에서 저지환 크기가 15 mm 이상인 높은 살균활성을 나타내었다. 또한 역병균(P. capsici)에 대한 균사생육억제활성과 무름병균(E. carotovorum subsp. carotovora), 세균성점무늬병균(P. syringae pv. syringae) 및 풋마름병균(R. solanacearum)에 대한 낮은 MIC 값을 확인하였다. 이로 인해 대추(Z. jujuba) 주정추출물, eugenol 및 isoeugenol은 다양한 식물병원균 방제에 이용될 수 있을 것으로 생각된다.

  • This study investigated the effect of coated feed using mixed oil on rumen fermentation characteristics. Two experiments were conducted based on materials that were mixed. First, cashew nut and soybean oils were mixed with white mineral oil. And second, different plant extracts were mixed with white mineral oil. At first experiment, inclusion levels of mixed oil on diet (0.03%, 0.1%, 0.5%) were applied as variables. A coated diet was fermented with rumen inoculum according to in vitro rumen fermentation and its parameters were investigated. In the result of first experiment, no negative effects on rumen pH were found. Significantly decreased dry matter digestibility was detected at 0.5% treatment (P<0.05). Total gas productions in control and 0.03% were significantly greater than those of others (P<0.05). Significantly reduced methane productions were found in all treatments compared to the control (P<0.05). Inclusion of mixed oil did not affect on ammonia production. Total volatile fatty acid production was also not influenced by coating with mixed oils. Rumen fermentation parameters were greatly changed according to introduced plant extracts at second experiment. The significantly lowest and greatest ammonia productions were found at treatments with Ixeris dentata and Plantago asiatica, respectively (P<0.05). The significantly greatest acetate and propionate productions were detected at treatments with Crucuma longa and Zizyphus vulgaris 1, respectively (P<0.05). All treatments, except Chrysanthemum idicum, Euyale ferox seed, Moringa leaf and fruit and Zizyphus vulgaris 1, showed significantly increased total volatile fatty acid production compared to the control (P<0.05). Only Paeonia lactiflora showed significantly lesser gas production than the control (P<0.05). In methane production, Ceramium, Zizyphus, Paeonia, Agrimonia, Torilis, Mugwort, Foeniculum, Euphorbia, Taraxacum, Artemisia, Momordica, Curcuma and Moringa reduced methane significantly compared to the control (P<0.05).

  • A number of sedative alkaloids were isolated from Sanjoin(酸棗仁), the seeds of Zizyphus vulgaris Lamark var. spinosus Bunge (Rhamnaceae) which is an important Chinese medicinal material used to treat insomnia and sometimes to treat sleepiness. Those compounds were designated as Sanjoinine-A, B, C, D, etc. depending on the order of increasing polarity. 생대추(Zizyphus Jujuba MILLER) 저장성 연구 (Storage Stability of Fresh Jujube Fruits (Zizyphus Jujuba MILLER))

  • 가공식품 원료 및 생식용 과일로서 생대추의 저장기간을 연장시키고자 $7{\pm}1^{\circ}C$에서의 CA저장과 $1,\;4,\;7{\pm}1^{\circ}C$의 온도조건에서 무포장 및 0.05 mm PE포장을 처리하여 저장하면서 품질변화 및 이화학적 성분 변화를 조사한 결과는 다음과 같다. 신선도로 본 생대추의 안전 저장 기간은 $1^{\circ}C$, PE film 포장에서 4주 였고, CA 처리구에서는 $O_2\;10%,\;CO_2\;1.5%$ 처리에서 선도가 가장 잘 유지되었다. 중량 감소는 CA처리구와 PE처리구 모두 거의 차이가 없었고 무포장 처리구에서는 저장 4주째부터 5% 내외의 중량 감소를 보여 생과로서는 상품 가치가 없었다. 색도는 저장 기간이 경과함에 따라 모든 처리구에서 L과 b값은 감소되고, a값은 증가하는 경향을 보였다. 경도는 전반적으로 저장 2주까지는 모두 증가하는 경향이었으나 CA처리구의 경우 저장 4주째 부터는 PE 처리구 및 무포장 처리구에 비해 연화가 더 느리게 진행됨을 보였다. 온도차이에 의한 경도 변화는 저온일수록 연화가 느리게 진행됨을 보였다. 초기 생대추의 당도와 적정 산도는 $26.6^{\circ}brix,\;1.18%$였으며 당도와 산도의 변화는 저장 기간 중 큰 변화가 없었다. 저장 초기 생대추의 Vitamin C 함량은 148.09 mg% 였고 저장기간에 따른 함량변화는 $1^{\circ}C$ 무포장 처리구를 제외한 모든 처리구에서 점차 감소되었으며 다른 처리구에 비해 CA 처리구에서 Vitamin C의 소실이 적어 더 안정함을 알 수 있었다.

대추나무빗자루병 매개충 "마름무늬매미충(Hishimonus sellatus Uhler)"의 기주범위 및 생태에 관한 연구 (Host range and Bionomics of the Rhombic Marked Leafhopper, Hishimonus sellatus Uhler(Homoptera: Cicadelliae) as a Vector of the Jujube Witches-Broom Mycoplasma)

  • 대추나무빗자루병 매개충인 마름무늬매미충(Hish[monus sellatus Uhler)의 생태, 혼식 및 산란 호성, 기주이동, 월동태를 조사한 결과 남부지방에서 연 5세대가 경과되었고, 제 1세대 6월 중 ~하순, 2세대 7월 중~8월 상순, 3세대 8월 중~하순, 4세대 9월 상~중순, 5세대 10월 상~중순이었고, 최대 발생기는 3화기인 8월 중~하순이었다. 계절에 따른 한 세대에 요하는 기간은 춘계 80, 하계 69, 추계 77.8일이었으며, 각 단계별 크기는 란 0.8 mm, 1령충 0.9mm, 2령충 l.4mm. 3령충 2.1mm, 4령충2.5mm, 5령충 3.2 mm 이었으며 성충 암컷 4.1mm, 수컷 3.8mm였다. 년중을 통한 기주식물의 이동을 보면 주로 뽕나무, 환삼덩굴, 대추나무에서 란으로 월동하며, 5월 중~하순경까지 뽕나무와 환삼덩굴의 신호를 계속 증가하면서 일부가 6월 하순~7월초순경에 대추나무로 이동하며 가해하는데 이들 란이 10월 중~하순 경부터 월동태로 들어간다. 산란부위는 신초, 엽병, 엽통 등이며 1마리의 암컷 산란수는 32~62개였다. 파식선호성에서는 환삼덩굴, 대추나무, 쥐똥나무, 뽕나무에서 높은 선호성(38.2S-21.64 %)을 보였고 닥나무, 개머루, 구기자나무, 구지뽕나무, 모시풀, 벼, 골담초, 일일초, 차풀 등이 4.65~2.48%였다, 산란이호성에서는 식이호성이 높은 환삼덩굴, 뽕나무, 대추나무에서 24~12개 정도였으며 성충의 생명에서는 환삼덩굴, 뽕나무, 대추나무에서 41.4~44.4일, 개멀, 차풀, 쥐똥나무, 일일초, 자운영, 샐러리에서 25일 이상이었고 기지 기주식물에서는 20일 이하이었다.

Ethanolic Extract of the Seed of Zizyphus jujuba var. spinosa Ameliorates Cognitive Impairment Induced by Cholinergic Blockade in Mice

  • In the present study, we investigated the effect of ethanolic extract of the seed of Zizyphus jujuba var. spinosa (EEZS) on cholinergic blockade-induced memory impairment in mice. Male ICR mice were treated with EEZS. The behavioral tests were conducted using the passive avoidance, the Y-maze, and the Morris water maze tasks. EEZS (100 or 200 mg/kg, p.o.) significantly ameliorated the scopolamine-induced cognitive impairment in our present behavioral tasks without changes of locomotor activity. The ameliorating effect of EEZS on scopolamine-induced memory impairment was significantly reversed by a sub-effective dose of MK-801 (0.0125 mg/kg, s.c.). In addition, single administration of EEZS in normal naive mouse enhanced latency time in the passive avoidance task. Western blot analysis was employed to confirm the mechanism of memory-ameliorating effect of EEZS. Administration of EEZS (200 mg/kg) increased the level of memory-related signaling molecules, including phosphorylation of extracellular signal-regulated kinase or cAMP response element-binding protein in the hippocampal region. Also, the time-dependent expression level of brain-derived neurotrophic factor by the administration of EEZS was markedly increased from 3 to 9 h. These results suggest that EEZS has memory-ameliorating effect on scopolamine-induced cognitive impairment, which is mediated by the enhancement of the cholinergic neurotransmitter system, in part, via NMDA receptor signaling, and that EEZS would be useful agent against cognitive dysfunction such as Alzheimer's disease.
